Nanosilicates are two-dimensional, charged nanomaterials with tunable surface chemistry that enable electrostatic interactions with proteins and nucleic acids. These properties make Laponite nanosilicate a promising platform for vaccine delivery, where coordinated antigen–adjuvant presentation and lymph node delivery are important for shaping immune responses. This thesis investigated nanosilicates as scaffolds for lysozyme-based antigen delivery and co-delivery with CpG.

Split-thickness skin grafts are widely used to treat chronic wounds. Procedure design requires surgeons to predict how much a patch of the patient's own skin expands when it is meshed with rows of slits and stretched over a larger wound area. Accurate prediction of graft expansion remains a challenge, with current models overestimating the actual expansion, leading to suboptimal outcomes. Inspired by the principles of mechanical metamaterials, we developed a model that distinguishes between the kinematic rearrangement of structural elements and their stretching, providing a more accurate prediction of skin graft expansion. Laponite® 2D Nanosilicates (NS) are disc-shaped platelets (~30 nm diameter, ~1 nm thick) with positively charged edges and negatively charged faces, capable of loading proteins, drugs, and nucleic acids via electrostatic interactions to form a house-of-cards gel that sequesters cargo. Here, we evaluated NS for sustained delivery of antigens and Toll-like receptor (TLR) agonists to extend germinal center duration and enhance antibody quality. The model antigen ovalbumin (OVA) was adsorbed onto NS at acidic pH in a ~1:1 ratio, with partial unfolding that remained stable at physiological pH. This thesis explores the micromechanical environment induced when cyclically compressing hydrogels via finite element modeling and experimentally on the impact of loading on mesenchymal stem cells (MSCs) when encapsulated withing 3D hydrogel matrices. Degenerative joint diseases, characterized by cartilage degradation, present significant challenges due to cartilage's limited self-repair capacity. Traditional electrodes used for electrophysiology recording, characterized by their hard, dry, and inanimate nature, are fundamentally mismatched with the soft, moist, and bioactive characteristics of biological tissues, leading to suboptimal skin-electrode interfaces. Hydrogel materials, mirroring the high water content and biocompatibility of biological tissues, emerge as promising candidates for epidermal electronic materials due to their adjustable physicochemical properties. This study explores the development of Mussel Foot Protein 5 (MFP5) bioadhesives to improve tendon-to-tendon and tendon-to-bone adhesive outcomes. MFP5, known for its underwater adhesive properties, was synthesized using recombinant techniques and optimized through coacervation and cross-linking. Adhesion tests demonstrated the superior performance of the optimized MFP5 adhesive on the tendon and bone samples. Improved protein concentration testing and purification methods enhanced MFP5 yield and consistency. Future research will focus on further standardization, exploring alternative purification methods, and evaluating the adhesive's performance in animal models. Antibody-antigen recognition enables antibody-conjugated nanostructures to serve as plasmonic biosensors with tunable specificity. However due to the instability of antibodies, these biosensors are susceptible to changes in the environment such as heat and aridity, leading to constraints on the transportation and handling of these sensors. Here we establish a method using a metal-organic framework crystal to preserve biosensor activity under severe environmental conditions, including exposure to high temperatures, an organic solvent and a proteolytic agent.
