Abdominal gunshot wounds are amongst the deadliest injuries encountered in combat and remote emergency settings. For military medics and first responders, the challenge lies in controlling internal bleeding and preventing infection before patients can reach surgical care-often within a narrow window of time known as the “golden hour”. Currently available field treatments are either too slow, not designed for internal wounds, or fail to provide antimicrobial protection. Autoimmune diseases affect approximately 5-8% of the global population, leading to chronic symptoms, long-term treatment, and reduced quality of life. Current therapies primarily rely on lifelong administration of immunosuppressants, which are associated with severe side effects and heightened susceptibility to infections. This has spurred interest in cellular therapies that manipulate dendritic cells (DCs) effectively and can selectively treat these diseases. This work aims to improve the efficacy of dendritic cell-based therapies using cationic polymers that can electrostatically complex with nucleic acids, forming nanoparticles referred to as polyplexes, that can deliver therapeutic nucleic acid cargo. Post-operative orthopedic surgical site infections (SSIs), specifically those caused by methicillin resistant Staphylococcus aureus (MRSA), present a significant clinical challenge due to delayed detection, increased morbidity, and current rising healthcare costs. Early detection of infection remains limited due to the current methods of diagnosis, which consist of observation of the surgical site and laboratory infrastructure for conformation of infection. In the United States, due to the increasing prevalence of cardiovascular disease, over 900,000 cardiac surgeries are performed annually, with projections exceeding 1.3 million by 2029. Many post-surgical patients require continuous in-hospital cardiac monitoring, which is typically conducted using electrocardiograms (EKGs). However, traditional EKG systems such as bedside cardiac monitors and cardiac telemetry monitors rely on wired leads that can easily tangle, degrade signal quality, and require frequent readjustments or replacements due to wire breakage, and for the patient, this can cause discomfort from prolonged use. Cardiovascular diseases along with various other disease conditions are characterized by an upregulation in reactive oxygen species (ROS), which can overwhelm homeostatic processes in the body, resulting in cellular apoptosis and severe oxidative damage. The lack of clinically approved treatments to regulate or alleviate this elevated ROS presence underscores the need for novel therapeutic strategies. While antioxidants promote cell rescue through ROS scavenging, their clinical translation is impeded by insufficient bioavailable concentrations at the source of injury. Subunit vaccines present themselves as an attractive alternative to traditional approaches to vaccines, which utilizes whole organisms to trigger an immune response. While these traditional approaches have proven themselves reliable within the history of vaccinations, they generally deliver subpar robust cellular-mediated immunity and pose many safety risks. Composed of proteins and /or peptides, subunit vaccines are safer and more precise alternatives to traditional approaches. The overall goal of this honors thesis was to develop a solution synthesis of Poly(Sebacic Anhydride) with the activating agent 4-toluenesulfonyl chloride and understand the effects of varying temperature, bases, and the addition order of reagents on the reaction.
Polyanhydrides are a class of hydrophobic, degradable polymers used in controlled drug delivery for 30 years. They are synthesized through three general methods: melt polycondensation, solution polycondensation, and ring-opening polymerization. Of these three methods, solution polycondensation typically results in products with lower molecular weight. Crutches are a mobility assistive device that are often used in various situations, such as post- surgical rehabilitation and recovery after injury. The current crutch tip design that is used on today’s crutches are often unstable and do not provide quality movement on different terrains, such as snow, ice, mud, and wet surfaces. Activated carbon (AC) has long been utilized in biomedical applications due to its innate antimicrobial characteristics. However, little is understood about the mechanism of action that allows AC to prohibit the propagation of different pathogen species. Immunotherapy offers a less toxic alternative to standard cancer treatments such as chemotherapy and can have prolonged curative effects to decrease cancer recurrence. Today, many drugs and biological agents have been developed that target the immune system and elicit an antitumor/cancer response. These agents are known collectively as cancer immunotherapies. While immunotherapies have radically improved treatment outcomes for many cancer patients, there are drawbacks to using these treatments.
