Delivering effective and non-toxic doses of bioactive materials that can aid in activating tissue regeneration to wounded tissue has proven to be an enormous challenge. This study was designed to produce a potential therapeutic recombinant protein by fusing two collagen binding domains to basic fibroblast growth factors (bFGF) through a collagenase cleavage site linker, so it can release the bFGF in a wound site by the action of this enzyme. The novel fusion protein was expressed in Escherichia coli BL-21 (E. coli) using traditional flask shaker and fed-batch cultivation. Volumetric muscle loss overwhelms skeletal muscle’s ordinarily capable regenerative machinery, resulting in fibrosis and severe functional deficits which have defied clinical repair strategies. My work spans the design and preclinical evaluation of implants intended to drive the cell community of injured muscle toward a regenerative state, as well as the development of an understanding of the molecular responses of this cell community to biomaterial interventions. In the processing of biopharmaceuticals, viral clearance and viral safety are important for the development of monoclonal antibodies. Murine xenotropic leukemia virus (XMuLV) is one of the retroviruses, recommended by Food and Drug Administration (FDA) as a model virus for viral clearance via inactivation from therapeutics derived from Chinese hamster ovary cells (CHO). A robust and effective method was investigated to clear or inactivate endogenous viruses by low pH inactivation. Injectable hydrogels have been used in biomedical applications to deliver drugs and other small molecules throughout the body. Traumatic brain injuries (TBIs) are a major global health concern that have an economic impact of $60 billion in the United States in related costs annually. Developing drugs for TBI treatment is an approach that currently faces limitations involving the permeability of the blood-brain barrier (BBB). The BBB naturally limits molecules from reaching the brain as a protective mechanism against disease, acting as a barrier during drug delivery. Under normal circumstances, skeletal muscle possesses the capacity to regenerate and heal via inflammatory and myogenic pathways. In cases of severe tissue loss or certain diseases, this capacity is lost, often resulting in loss of tissue function. Extracellular matrix (ECM), the protein scaffold which houses cells in physiological tissue, has been shown to have structural and chemical properties which influence cell migration and phenotype. This results in ECM’s capacity to encourage a regenerative response when implanted into severely damaged skeletal muscle. Motivated to produce electricity with photon activated ion pumps, the main purpose of this work was to characterize the photosynthetic membrane proteins bacteriorhodopsin (proton pump) and halorhodopsin (chloride pump). The proteins were re-suspended in lipid bilayers. For this work, an experimental set-up was built which included: chambers for lipid bilayer formation and characterization, lasers for ion pump activation, and an AxoPatch electrophysiology system for small photocurrent measurement. Lipid bilayer membranes were formed using mostly folding method: folding two monolayers together. The membranes were characterized by their resistance, capacitance, and generated photocurrent. Artificial lungs are in use, but difficult issues remain in the field of membrane development related to fouling issues. Currently there are external artificial lungs circulating blood outside the body, taking out the carbon dioxide, and inserting oxygenated blood back into the body. An example of this type of machine is the ExtraCorporeal Membrane Oxygenation (ECMO) machine currently used in hospitals. However, these specific antivirals are not available in each country and can be problematic for specific patients. Chitosan is proposed as a possible broad-based antiviral, which has already demonstrated antibacterial properties, antiviral properties in plants, is used for wound healing and as a hydrogel among other medical applications. Metallic single-walled carbon nanotubes (m-SWNTs) were separated from pristine SWNTs using affinity chromatography for use in electrically conductive tissue engineering scaffolds. Approximately one third of SWNTs have metallic properties. Separations were achieved using a protocol modified from Liu & coworkers (2011) in order to improve the method for cell culture environments. Samples enriched in m-SWNTs were isolated and characterized. However, challenges still remain for the complete separation of m-SWNTs from their semiconducting counterpart (s-SWNTs) using this protocol. To reduce device loosening an improved method of tissue anchoring is required. Decellularization of skeletal muscle is a process that removes cellular components of skeletal muscle tissue while leaving behind the intact extracellular matrix (ECM). Skeletal muscle ECM is currently being studied as a biologic scaffold for repairing volumetric muscle loss (VML) because the removal of cells greatly reduces the antigenicity of the donor tissue. Decellularization usually relies on passive diffusion of detergents, surfactants and/or osmotic solutions to strip cells from the ECM. For the first time in the implantable biomaterials field, efficient hydrothermal cation exchange methods were developed suitable for mass-production of implantable material. The cation exchange conditions were performed by various methods of temperature, pressure, and time exposure. To confirm both chemical and physical alteration of the scaffold, XRD shift analysis, EDX atomic composition, and SEM imaging were conducted. The optimal results are to use aqueous solutions of 0.5 M chloride salts in sealed heated containers at 180 ̊C for eight hours.
