Accelerating Wound Healing Rates With Cucurbita Pepo Leaf Extract Loaded Electrospun Poly(Methyl Methacrylate)/Halloysite/Chitosan/ Caco₃ Composite Nanofibers Through In Vitro And In Vivo Assessments, Samar A. Salim
Nanotechnology Research Centre
Cucurbita pepo leaf extract (CPE) was incorporated into electrospun poly(methyl methacrylate)/ halloysite/chitosan/ CaCO₃ (PMMA/Hal/CS/CaCO₃) composite nanofibers to develop a novel biomaterial for accelerating wound healing rates. The different nanofibrous scaffolds were successfully fabricated and characterized through scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD). SEM analysis revealed uniform, smooth nanofibers, while FTIR and XRD confirmed the integration of CPE into nanofiber matrix, indicating an amorphous structure and effective dispersion of the extract. In vitro agar well-diffusion and antibiofilm assays revealed that the optimized formulation exhibited potent antimicrobial activity against wound-associated pathogens. The nanofibers composite based on …

School of Podiatric Medicine - Student Research
The aim of this study is to determine the efficacy and safety profile of total navicular implants based on current literature.
Navicular insults such as high-energy trauma, osteonecrosis, and Muller-Weiss syndrome may result in avascular necrosis or a critical sized bone defect, conditions that are highly difficult to manage given current treatment modalities. Total navicular implants may offer a solution to these problems. While 3D printing of implants remains investigational, its use in highly individualized cases involving the talus has had promising results with conditions like avascular necrosis, tumors, trauma, and arthritis. The navicular is unique in its attachments to …

Volumetric Wear Analysis Of Polyethylene Orthopedic Liners Using Point Clouds, Gabriel Landi
Dartmouth College Master’s Theses
In vivo wear of ultra-high molecular weight polyethylene (UHMWPE) orthopedic bearings represents a major problem in shoulder and knee arthroplasty, with significant amounts of devices failing due to the complication itself, and even more due to associated reasons for retrieval, including aseptic loosening. However, quantification of UHMWPE wear has largely been limited to linear and in vitro volumetric analyses, obscuring the understanding of the amount of clinically relevant material loss. This study proposes new methods to measure articular, rim, and backside surfaces of retrieved shoulder and knee UHMWPE liners, centered around the coordinate measuring machine (CMM)-based collection of high density …

Engineering Plga Nanoparticle Size To Modulate Immune Responses After Spinal Cord Injury, Daniel J. Kolpek
University of Kentucky Doctoral Dissertations
Spinal cord injury is a severe and debilitating condition that often results in lifelong complications, including paralysis. Many of these chronic outcomes are driven by a sustained inflammatory response at the injury site, which limits tissue regeneration and functional recovery. As a result, therapeutic strategies that modulate this immune response have gained significant interest. Polymer based nanoparticle therapies have emerged as a promising approach due to their ability to interact with and influence immune cell behavior. Importantly, the physicochemical properties of nanoparticles can be precisely tuned to optimize these effects. Among these properties, size has been shown to influence key …

Opmed Seal: Engineering A Rapid-Response Biodegradable Device For Hemorrhage Control And Infection Prevention In Abdominal Trauma, Paula M. Cabrera, Wesley Stephen Murphy
Honors Theses
Abdominal gunshot wounds are amongst the deadliest injuries encountered in combat and remote emergency settings. For military medics and first responders, the challenge lies in controlling internal bleeding and preventing infection before patients can reach surgical care-often within a narrow window of time known as the “golden hour”. Currently available field treatments are either too slow, not designed for internal wounds, or fail to provide antimicrobial protection. To address this gap, our team developed OpMed Seal: a compact, dual-syringe delivery system that deploys a biodegradable, antibiotic-infused hydrogel directly into abdominal injuries. The system is designed for rapid hemorrhage control, localized …

Biomedical Engineering Undergraduate Honors Theses
Bone fractures and complete joint replacements are becoming more common, primarily because of accidents and age-related issues. Conventional solutions to treat these orthopedic injuries involve inert metal alloys to correct bone fractures until the healing process is complete, where a secondary revision surgery is then required to remove the implant. Issues regularly associated with these solutions include stress shielding, bone resorption, infection, inflammation, and implant loosening. Magnesium (Mg)-based nanocomposites offer promising potential as an alternative to commonly used metal alloys because of their favorable mechanical properties and biocompatibility. A particularly advantageous property of Mg is its natural biodegradation in vivo, …

End stage renal disease (ESRD) is a prominent chronic condition with dialysis as a primary treatment option. Dialysis necessitates implementation of an access point, with the gold standard being an AV fistula which possesses multiple shortcomings. Alternative access points, namely synthetic grafts, do not allow wound healing characteristics. This work aims to explore how a tissue engineered vascular graft (TEVG) may be able to address these issues by allowing a long-term access point for HD that can close puncture wounds.
The effects of growth factors on our cells were tested preliminarily by means of a scratch assay on plates of …

Honors Undergraduate Theses
Improper healing of bone fractures and limited regeneration of bone following bone tumor resection remain a prevalent complication worldwide. Long-term effects of this include joint instability, limited movement, and arthritis. Bone infections (osteomyelitis), reactive oxygen species (ROS) generation via excessive inflammation, and underlying conditions such as osteoporosis can contribute to suboptimal bone tissue regeneration. ROS accumulation stimulates osteoblast apoptosis, while promoting osteoclast differentiation. These processes inhibit mineralization and osteogenesis. Conventional therapeutics involve the utilization of drugs and bone grafts. However, these approaches pose limitations and can result in subsequent complications. Microbial infections, ROS accumulation, and excessive inflammations are factors that …

Dissertations and Theses
Protein-based self-assembling biomaterials, known as Thermo-Responsive Assembled Proteins (TRAP), present meaningful potential as drug delivery carriers. These materials enable the controlled, slow release of poorly soluble chemotherapeutic agents, such as doxorubicin (Dox), while potentially reducing systemic off-target effects. In collaboration with multiple NYU labs, this study evaluated the efficacy of two TRAP variants—TRAP and F-TRAP—as drug delivery carriers in a xenograft mouse model of glioblastoma multiforme (GBM).
The primary objective was to compare the effectiveness of TRAP-loaded Dox (TRAP-DOX) to free Dox in achieving tumor extravasation and accumulation, facilitating sustained drug release. We hypothesized that the leaky vasculature of GBM …

Investigation Using Single Point Incremental Forming (Spif) To Fabricate Patient-Specific, Titanium Orbital Floor Implants, Elizabeth M. Mamros, Lauren E. Blaha Md, Christian A. Kauffman Md
Faculty Conference Papers and Presentations
The floor of the human orbit is composed of thin bone that is prone to traumatic fracture. This leads to a loss of support for the eye, which can cause vision changes. Therefore, fractures may need surgical reconstruction using a thin, sheet-like implant. Titanium implants are available off-the-shelf in standard sizes, but fitting to each patient’s unique anatomy requires surgeons to cut, file, and bend these plates. This can be time-consuming and imprecise. To both save time and ensure a perfect fit for the patient, a custom plate can be created prior to surgery. This investigation focuses on single-point incremental …

Nanoparticles (NPs) offer promising potential as therapeutic agents for inflammation-related diseases, owing to their capabilities in drug delivery and immune modulation. In preclinical studies focusing on spinal cord injury (SCI), polymeric NPs have demonstrated the ability to reprogram innate immune cells. This reprogramming results in redirecting immune cells away from the injury site, downregulating pro-inflammatory signaling, and promoting a regenerative environment post-injury. However, to fully understand the mechanisms driving these effects and maximize therapeutic efficacy, it is crucial to assess NP interactions with innate immune cells. This review examines how the physicochemical properties of polymeric NPs influence their modulation of …

Due to high specificity and less toxicity, nanoparticles have become promising for targeted drug delivery against cancer, although they face challenges when they enter the bloodstream, making their behavior unpredictable. The deposition of proteins on nanoparticles, the protein corona, changes their biosignature, affecting their circulation, drug release potency, targeting ability, and immunogenicity. The continuous exchange of protein layers on a nanoscale is complex to analyze.
Nanoparticle tracking analysis (NTA) is a method to characterize nanoparticles in blood plasma by combining principles of light scattering microscopy and Brownian motion. In this work, time-dependent experiments with plasma were conducted to analyze the …
Locally Injectable Peptide Hydrogel For Conversion Of Temozolomide And Codelivery Of Sirna In Glioblastoma Multiforme Treatment, Megan Pitz
All Dissertations
Traditional treatment methods for glioblastoma multiforme (GBM) are largely unsuccessful, with a 5-year survival rate of 5.6%. Temozolomide (TMZ), the only chemotherapy FDA-approved for GBM treatment, is a hydrophobic prodrug that is stable in an acidic pH and begins converting to its active form at a physiological pH of 7.4. However, complete conversion of TMZ is observed only at a higher pH, and only about 30% of administered TMZ reaches the central nervous system. Therefore, there is a need for new treatment methods that maximize TMZ delivery to and efficacy at the tumor site. Self-assembling peptides are a unique class …

Graduate Theses, Dissertations, and Problem Reports (ETD)
Bioprinting, akin to 3D printing, employs cell-laden hydrogels such as GelMA (Gelatine Methacrylate) and Alginate instead of plastic or resin to create biological structures. The printing process followed by subsequent tissue maturation is known as 4D bioprinting, where the fourth dimension is time. Ensuring adequate and consistent oxygen, O2, supply within 3D bioprint volume over the entire maturation period process is crucial, as even transient hypoxia can impact cellular behavior and phenotypes. However, printing of fully developed vasculature remains an unresolved technical challenge. Several approaches to chemically (peroxides) or biologically (photosynthesis) generate oxygen during bioprint maturation have been proposed. However, …
