Water quality is a crucial factor in determining the health of an environment, and in turn the health of all those living in the given environment. Early detection of potential hazards in source water used for drinking would help to avoid outbreaks of sickness and even death. Contaminants, found in contact and drinking water, having negative effects on human health may include pathogenic microorganisms, and metal ion pollutants such as lead or cadmium. Utilization of coal as a low-cost domestic energy source is becoming a subject of utmost importance due to fluctuating crude oil prices. However, coal use emits pollutants such as nitrous oxides (NOx), sulfur oxides (SOx), carbon dioxide (CO2), and particulates that have negative environmental impact and significant issues on health. Compromised liver and/or kidney function reduces the acceptable dosage of a variety of medications that can be administered to patients. These patients still have a need for drugs such as nonsteroidal anti-inflammatory drugs (NSAIDs), antivirals, and antibiotics. The project goal is to provide a drug delivery system to accommodate these reduced dosage limits with added therapeutic benefits to address symptoms of liver or kidney failure. Localized drug delivery allows for a smaller, concentrated dose rather than inundating the patient's system with the drug of interest. Acetone-butanol-ethanol (ABE) fermentation from wastewater microalgae by Clostridium saccharoperbutylacetonicum N1-4 is a novel bioprocess that utilizes waste substrate to generate valuable solvents. Butanol, the most abundant product resulting from ABE fermentation, is an environmentally safe and high performing fuel that can be utilized as a drop-in-fuel; however, high operational costs and low ABE yield present challenge in scale-up of the process. In the present research, nicotine alkaloid production by Nicotiana tabacum (tobacco) hairy roots and tropane alkaloid production by Hyoscyamus niger hairy roots were investigated. The first objective of this research was to improve the oxygen mass transfer in hairy root cultures with microbubbles. Oxygen was shown as a critical nutrient for the growth of tobacco and H. niger hairy roots. In a 1-liter fermentor, microbubble dispersion improved the oxygen mass transfer, tobacco hairy root growth, and nicotine production in the medium. Spider silk has become a popular material due to its exceptional mechanical and biocompatibility properties. The project team has developed methods of producing and improving the properties of synthetic spider silk films. Other groups have been able to produce similar films having less than impressive mechanical properties using a toxic solvent. The purpose of this study is to quantify and determine the binding of a molecular DNA probe or molecular beacon used in the detection of a known waterborne pathogen Cryptosporidium parvum. The objectives of this study are to determine the stability, sensitivity, and selectivity of the molecular beacon in detecting synthesized Cryptosporidium DNA sequences. The designed probe is based on a two-fluorophore system that is different than similar molecular beacons that utilize a fluorophore and quencher detection method. Static and dynamic coefficients of friction on structural surfaces play important roles in the power requirements and material selection for equipment used in handling and storing agricultural commodities. However, friction data on dried distillers grains with solubles (DDGS) is limited. Further, lack of a standardized method for determining friction coefficient on grain handling materials presents a challenge. The objective of this study is to optimize the growth media and genes to maximize PHB production computer simulations, and to evaluate the simulation results through experiments. In order to achieve this objective, the metabolism of E. coli that produces PHB was modeled using the existing E. coli genome-scale metabolic networks. To identify the best additional nutrient for PHB production, carbon sources, minerals, amiano acids, nitrogen sources, and sulfur sources were examined by production potential analysis. Strain design was conducted to find the unwanted reactions to overexpress PHB synthesis pathway.

Microalgae are the preferred crop for the production of biodiesel. Microalgae are microplants that have the ability to harness sunlight more efficiently than other plants and store 20-80% lipids per g of dry algae in their cells. Microalgae have the extraordinary ability to grow in brackish water or wastewater. Microalgae can be grown in municipal wastewater to uptake phosphorus and nitrogen and remediate the wastewater of these nutrients. Microalgae thus cultivated accomplishes a dual role of wastewater treatment and provides a sustainable feedstock for biofuels and other bioproducts.

Thermoreversible gels could be used to coat medical implants, to inject for subdermal treatments, or to implement in topical applications. FDA approved Pluronics® form thermoreversible gels and certain Pluronics® display inherent varying degrees of antimicrobial activity. Rdc2 is a flavin-dependent halogenase from Pochonia chlamydosporia. Through the introduction of a His6-tag to both the N- and C-termini, the isolation yield of Rdc2 from Escherichia coli using Ni-NTA affinity chromatography was increased by three-fold. In vitro reaction of Rdc2 and a flavin reductase (Fre) with seven different hydroxyquinolines revealed that 3-hydroxyquinoline (3), 5-hydroxyquinoline (5), 6-hydroxyquinoline (6), and 7-hydroxyquinoline (7) can be specifically halogenated. These products were prepared by incubating the corresponding substrates with IPTG-induced E. coli BL21(DE3)/Rdc2. The nematode Caenorhabditis elegans is a model organism for understanding sensory molecules of multicellular organisms. Ovulating hermaphrodites produce putative pheromone(s) that cause male attraction. Because pheromones are produced in such small quantities, adult conditioned-media from large-scale synchronous culture is necessary to analyze these pheromones. Current protocols for culture synchronization have volume constraints that limit large-scale synchronous cultures and current methodology for adult conditioned-media production is impractical.

Background
Humic acid (HA) has been found to affect the solubility, mineralization, and bound residue formation of polycyclic aromatic hydrocarbons (PAHs). However, most of the studies on the interaction between HA and PAH concentrated on one or two of the three phases. Few studies have provided a simple protocol to demonstrate the overall effects of HA on PAH distribution in soil systems for all three phases.

To determine whether the soil Mycobacterium isolate KMS would mineralize pyrene under rhizosphere conditions, a microcosm system was established to collect radioactive carbon dioxide released from the labeled polycyclic aromatic hydrocarbon. Microcosms were designed as sealed, flow-through systems that allowed the growth of plants. Experiments were conducted to evaluate mineralization of 14C-labeled pyrene in a sand amended with the polycyclic aromatic hydrocarbons degrading Mycobacterium isolate KMS, barley plants, or barley plants with roots colonized by isolate KMS. Background
Remediation of tert-butyl alcohol (TBA) in subsurface waters should be taken into consideration at reformulated gasoline contaminated sites since it is a biodegradation intermediate of methyl tert-butyl ether (MTBE), ethyl tert-butyl ether (ETBE), and tert-butyl formate (TBF). The effect of temperature on TBA biodegradation has not been not been published in the literature.
Methods
Biodegradation of [U 14C] TBA was determined using hyporheic zone soil microcosms.
