A Laplace Pressure Based Microfluidic Trap For Passive Droplet Trapping And Controlled Release, Melinda Simon, Robert Lin, Jeffrey Fisher, Abraham Lee Feb 2012

A Laplace Pressure Based Microfluidic Trap For Passive Droplet Trapping And Controlled Release, Melinda Simon, Robert Lin, Jeffrey Fisher, Abraham Lee

Faculty Publications

Here, we present a microfluidic droplet trap that takes advantage of the net Laplace pressure force generated when a droplet is differentially constricted. Mathematical simulations were first used to understand the working range of the component; followed by finite element modeling using the CFDsoftware package to further characterize the behavior of the system. Controlled release of the trapped droplets is also demonstrated through both a mechanical method and a chemical method that manipulates the total pressure exerted on the trapped droplet. The unique design of this trapping device also provides the capability for selection of a single dropletfrom a train, …

Optimization Of Bio-Impedance Sensor For Enhanced Detection And Characterization Of Adherent Cells, Dorielle T. Price Jan 2012

Optimization Of Bio-Impedance Sensor For Enhanced Detection And Characterization Of Adherent Cells, Dorielle T. Price

USF Tampa Graduate Theses and Dissertations

This research focuses on the detection and characterization of cells using

impedance-based techniques to understand the behavior and response of cells to internal/environmental changes. In combination with impedimetric sensing techniques, the biosensors in this work allow rapid, label-free, quantitative measurements and are very sensitive to changes in environment and cell morphology. The biosensor design and measurement setup is optimized to detect and differentiate cancer cells and healthy (normal) cells. The outcome of this work will provide a foundation for enhanced 3-dimensional tumor analysis and characterization; thus creating an avenue for earlier cancer detection and reduced healthcare costs.

Alternating Current Dielectrophoretic Manipulation Of Erythrocytes In Medical Microdevice Technology, Kaela M. Leonard Jan 2012

Alternating Current Dielectrophoretic Manipulation Of Erythrocytes In Medical Microdevice Technology, Kaela M. Leonard

Dissertations, Master's Theses and Master's Reports - Open

Medical microdevices have gained popularity in the past few decades because they allow the medical laboratory to be taken out into the field and for disease diagnostics to happen with a smaller sample volume, at a lower cost and much faster. Blood is the human body's most readily available and informative diagnostic fluid because of the wealth of information it provides about the body's general health including enzymatic, proteomic and immunological states. The purpose of this project is to optimize operating conditions and study ABO-Rh erythrocytes dielectrophoretic responses to alternating current electric signals. The end goal of this project is …

A Microfluidic Device For Impedance Spectroscopy, Ahmet Can Sabuncu Jan 2011

A Microfluidic Device For Impedance Spectroscopy, Ahmet Can Sabuncu

Mechanical & Aerospace Engineering Theses & Dissertations

Recently, microfluidics has become a versatile tool to investigate cellular biology and to build novel biomedical devices. Dielectric spectroscopy, on the other hand, allows non-invasive probing of biological cells. Information on the cell membrane, cytoplasm, and nucleus can be obtained by dielectric spectroscopy provided that appropriate tools are used in specific frequency ranges. This dissertation includes fabrication, characterization, and testing of a simple microfluidic device to measure cell dielectric properties. The dielectric measurements are performed on human T-cell leukemia (Jurkat), mouse melanoma (B16), mouse hepatoma (Hepa), and human costal chondrocyte cells. Dielectric measurements consist of measuring the complex impedance of …

Integration Of Microfluidics With Surface Plasmon Resonance, Scott B. Fratzke Aug 2010

Integration Of Microfluidics With Surface Plasmon Resonance, Scott B. Fratzke

Master's Theses

This thesis successfully integrates laminate microfluidic devices with an analytic Surface Plasmon Resonance (SPR) instrument. Integration was accomplished at low-cost using materials such as polydimethylsiloxane (PDMS), Poly(methyl methacrylate) (PMMA), Tygon tubing, and a 3-way stopcock. The main components of this thesis are the design and fabrication of the low-cost, in-house fluidics that can integrate with upstream microfluidics and the validation of the in-house fluidics using the Biosensing Instruments BI-2000 SPR instrument. The low-cost fluidics was designed and fabricated “in-house” using a novel investment casting technique that required the use of laser cutting technology to make a master cast, and candle …

Design, Fabrication, And Implementation Of A Single-Cell Capture Chamber For A Microfluidic Impedance Sensor, Joshua-Jed Doria Fadriquela Jun 2009

Design, Fabrication, And Implementation Of A Single-Cell Capture Chamber For A Microfluidic Impedance Sensor, Joshua-Jed Doria Fadriquela

Master's Theses

A microfluidic device was created for single-cell capture and analysis using polydimethylsiloxane (PDMS) channels and a glass substrate to develop a microfluidic single-cell impedance sensor for cell diagnostics. The device was fabricated using photolithography to create a master mold which in turn will use soft lithography to create the PDMS components for constant device production. The commercial software, COMSOLTM Multiphysics, was used to quantify the fluid dynamics in shallow micro-channels.

The device will be able to capture a cell and sequester it long enough to enable measurement of the impedance spectra that can characterize cell. The proposed device will be …

Mems (Micro-Electro-Mechanical-Systems) Based Microfluidic Platforms For Magnetic Cell Separation, Pulak Nath Jan 2008

Mems (Micro-Electro-Mechanical-Systems) Based Microfluidic Platforms For Magnetic Cell Separation, Pulak Nath

ETD Archive

Microfluidic platforms for magnetic cell separation were developed and investigated for isolation of magnetic particles and magnetically tagged cells from a fluidic sample. Two types of magnetic separation platforms were considered: an Isodynamic Open Gradient Magnetic Sorter (OGMS) and a multistage bio-ferrograph. Miniaturized magnets were designed using magnetostatic simulation software, microfluidic channels were fabricated using microfabrication technology and magnetic separation was investigated using video microscopy and digital image processing. The isodynamic OGMS consisted of an external magnetic circuit and a microfabricated channel (biochip) with embedded magnetic elements. The biochip is placed inside the magnetic field of the external circuit to …

Biological Cell Identification By Integrating Micro-Fluidics, Electrical Impedance Spectroscopy And Stochastic Estimation, Karl R. Schwenn

Theses and Dissertations

The integration of micro-fluidics, electrical impedance spectroscopy and stochastic estimation will lead to a device with enhanced detection capabilities. The goal of this thesis was to build a micro-fluidic electrical impedance measurement device that can be used in combination with a stochastic estimator to accurately identify living cells. A microdevice capable of making impedance measurements on individual living cells was designed and built using a series of standard microelectronic fabrication techniques. A microchannel was patterned in SU-8 photoresist between two gold microelectrodes on a two inch Pyrex 7740 wafer. The design process, the fabrication techniques for the microchannel, the fluid …

Low Concentration Microenvironments Enhance The Migration Of Neonatal Cells Of Glial Lineage, Richard A. Able, Jr., Celestin Ngnabeuye, Cade Beck, Eric C. Holland, Maribel Vazquez Jun 2002

Low Concentration Microenvironments Enhance The Migration Of Neonatal Cells Of Glial Lineage, Richard A. Able, Jr., Celestin Ngnabeuye, Cade Beck, Eric C. Holland, Maribel Vazquez

Publications and Research

Glial tumors have demonstrated abilities to sustain growth via recruitment of glial progenitor cells (GPCs), which is believed to be driven by chemotactic cues. Previous studies have illustrated that mouse GPCs of different genetic backgrounds are able to replicate the dispersion pattern seen in the human disease. How GPCs with genetic backgrounds transformed by tumor paracrine signaling respond to extracellular cues via migration is largely unexplored, and remains a limiting factor in utilizing GPCs as therapeutic targets. In this study, we utilized a microfluidic device to examine the chemotaxis of three genetically-altered mouse GPC populations towards tumor conditioned media, as …
