Bone cements should have the ability to chemically bond to both bone and surgical metals, exhibit no chemical or thermal necrosis, and have no significant shrinkage or exotherm upon setting. Taking these properties into consideration, glass polyalkenoate cements (GPCs) have potential as bone cements. GPCs are formed by the reaction between an ion-leachable glass and an aqueous solution of polyacrylic acid (PAA) [1] and have proven antibacterial and cariostatic properties [2], which are related to their ability to release beneficial amounts of ions over time [3, 4]. The GPCs can be formulated to release ions that can have a therapeutic …

Bioactive glass particles (0.42SiO2-0.15CaO-0.23Na 2O-0.20ZnO) of varying size (<90 >μm and 425-850 μm) were synthesized and coated with silver (Ag) to produce Ag coated particles (PAg). These were compared against the uncoated analogous particles (Pcon.). Surface area analysis determined that Ag coating of the glass particles resulted in increased the surface area from 2.90 to 9.12 m2/g (90 μm) and 1.09-7.71 m2/g (425-850 μm). Scanning electron microscopy determined that the Ag coating remained at the surface and there was little diffusion through the bulk. Superparamagnetic iron oxide nanoparticles, with diameters in the range of a few tens of nanometers, display the ability to cross the blood-brain barrier and are envisioned as diagnostic and therapeutic tools in neuro-medicine. However, despite the numerous applications being explored, insufficient information is available on their potential toxic effect on neurons. While iron oxide has been shown to pose a decreased risk of toxicity, surface functionalization, often employed for targeted delivery, can significantly alter the biological response. This aspect is addressed in the present study, which investigates the response of primary cortical neurons to iron oxide nanoparticles with coatings frequently …

This paper reports on improvement of the physical and mechanical properties of Y2O3-ZrO2 ceramics through compositional optimization, the use of nanograined powders and utilization of microwave sintering. ZrO2 with 2-5 mol% Y2O3, prepared from nanopowders, was sintered in both conventional and microwave furnaces under exactly the same heating schedule. It was found that microwave sintering improves physical and mechanical properties of Y2O3-ZrO2 ceramics compared with conventional sintering. Compositions containing 2 mol% Y2O3 exhibit the greatest improvement due to retention of tetragonal ZrO …

Following spinal cord injury, axons fail to regenerate without exogenous intervention. In this study we report that aligned microfiber-based grafts foster robust regeneration of vascularized CNS tissue. Film, random, and aligned microfiber-based conduits were grafted into a 3 mm thoracic rat spinal cord gap created by complete transection. Over the course of 4 weeks, microtopography presented by aligned or random poly- l-lactic acid microfibers facilitated infiltration of host tissue, and the initial 3 mm gap was closed by endogenous cell populations. This bulk tissue response was composed of regenerating axons accompanied by morphologically aligned astrocytes. Aligned fibers promoted long distance …

Following central nervous system (CNS) injury, activated astrocytes form a glial scar that inhibits the migration of axons ultimately leading to regeneration failure. Biomaterials developed for CNS repair can provide local delivery of therapeutics and/or guidance mechanisms to encourage cell migration into damaged regions of the brain or spinal cord. Electro spun fibers are a promising type of biomaterial for CNS injury since these fibers can direct cellular and axonal migration while slowly delivering therapy to the injury site. In this study, it was hypothesized that inclusion of an anti-metabolite, 6-aminonicotinamide (6AN), within poly-l-lactic acid electro spun fibers could attenuate …

Electro spun an fibers hydrogels are two focal points of research within the biomaterial's community. The electrospinning process allows for fabrication of nanoscale, aligned topography and has been shown to direct cellular migration and outgrowth. Hydrogels exhibit benefits over traditional, rigid scaffolds in that they are injectable and can be tailored to mimic the mechanical properties of the surrounding tissue. Although both electro spun fibers and hydrogels display favorable characteristics for biomedical applications in vitro, there in vivo benefits are limited due to inherent deficiencies Therefore, to advance the field of biomaterials research, a composite material containing electro spun fibers …

To conclude, the addition of polymers to GPCs resulted in little change to the Tw and Ts, however the compressive strength was found to decrease slightly. ASAP tests revealed that the cements may undergo some structural changes as a result of these additions. In the future perhaps the addition of organic polymers suspended in the liquid phase as part of the cement may yield improved results in this type of study. © 2011 IEEE.

Severe mechanical trauma of the spinal cord induces a secondary injury response that contributes to post injury cell death and the formation of barriers inhibitory to neuronal regeneration. It is believed that engineered scaffolds produce favorable results when they mimic the properties of the t issue that they are aiming to replicate. Alginate hydrogels have been shown to be beneficial in a number of biomaterial applications and this study shows that alginate hydrogels formed in the presence of an artificial cerebrospinal fluid are able to form hydrogels with mechanical properties similar to those exhibited by neural tissue in the CNS. …

Spinal cord injury (SCI) is a debilitating problem that often results in paraplegia or quadriplegia. Fibrin hydrogels can be injected into the injured cavity and fill the irregular space. Current techniques require 60 minutes to form a hydrogel. The purpose of this study was to design a fibrin biomaterial that would solidify at a faster rate. By augmenting the existing fabrication procedure, a novel biomaterial was created that formed in 5-7 minutes but lost much of its mechanical stability. Further studies will be performed to improve the mechanical properties while maintaining the faster rate of formation observed here. © 2011 …

Silver nanoparticles were incorporated into poly-{methyl methacrylate-butyl acrylate-acrylic acid} using two different approaches. The first approach was based on dispersing the nano silver particles in acrylic latex (previously synthesized by the emulsion polymerization) and the second is the in-situ polymerization of acrylate monomers in the presence of silver nanoparticles by Mini emulsion polymerization. Mini emulsion polymerization can yield a better dispersion of nano silver in the polymeric particles because organic particles can be dispersed directly in the monomer droplets becoming encapsulated upon polymerization. Morphological investigations were performed using SEM and TEM. FTIR and thermal analyses revealed the silver nanoparticles to …

"Galactooligosaccharides (GOS) are prebiotics that are being used to influence intestinal microbiota towards health promoting, beneficial bacteria like bifidobacteria. Natural sources of GOS are not found in suitable levels to achieve the desired health effects so economically viable commercial production methods are important. Enzymatic conversion of lactose to GOS through transgalactosylation is a preferred method since the starting material is inexpensive and readily available. Enzymes used in batch production are purified from fungal or bacterial sources or remain in whole cells cultured for GOS production. Purified or unbound enzymes are single use whereas enzymes retained within whole cells can potentially …

Many commercial bone grafts cannot regenerate healthy bone in place of diseased bone. Bioactive glasses have received much attention in this regard due to the ability of their ionic dissolution products to promote cell proliferation, cell differentiation and activate gene expression. Through the incorporation of certain ions, bioactive glasses can become therapeutic for specific pathological situations. Calcium-strontium-sodium-zinc-silicate glass bone grafts have been shown to release therapeutic levels of zinc and strontium, however the in vitro compatibility of these materials is yet to be reported. In this study, the in vitro cytocompatibility of three different calcium-strontium-sodium-zinc-silicate glasses was examined as a …

Tumors frequently contain hypoxic regions that result from a shortage of oxygen due to poorly organized tumor vasculature. Cancer cells in these areas are resistant to radiation- and chemotherapy, limiting the treatment efficacy. Macrophages have inherent hypoxia-targeting ability and hold great advantages for targeted delivery of anticancer therapeutics to cancer cells in hypoxic areas. However, most anticancer drugs cannot be directly loaded into macrophages because of their toxicity. In this work, we designed a novel drug delivery vehicle by hybridizing macrophages with nanoparticles through cell surface modification. Nanoparticles immobilized on the cell surface provide numerous new sites for anticancer drug …
