Automated Foam Control In Single-Use Bioreactors Using The Single Use Foam Probe, Jordan Cobia, Ben Madsen, Emilee Madsen, Alex Hodge

Single-Use Technologies III: Scientific and Technological Advancements

Headspace foam in bioreactors can cause significant issues to a biological process among which are unfavorable metabolic conditions for cells, increased shear from bursting bubbles, cell death/entrainment, mass transfer interference and potential fouling of exhaust filters which can allow a point of entry for contamination or lead to pressure build-up and possible failure of the bioprocess container. This study investigates the efficacy of using a single use foam probe coupled with an automated response of an antifoam addition with an integrated DeltaV controller. Two 50L S.U.B.s run in parallel clearly illustrate differences; the first employing foam control using the foam …

Experimental Tests And Numerical Simulations For Failure Investigation On Corrugated Boxes Used On Household Appliance Packaging, Diego Fernandes Rodrigues, José Carlos Pereira

Journal of Applied Packaging Research

Packages made of corrugated paper are fundamental to the protection, transportation and handling of the appliance product market. During the storage and sales stages of a product, the package must resist compressive loads in different directions beyond moderate impacts. In this context, the objective of this work is to develop and implement a post-processor that allows the simultaneous analysis of two of the most common failure modes of packages made of corrugated paper: failure due to tensile or compressive stress limit, and failure due to local buckling, when the buckling of the faces of the corrugated paper between two peaks …

Single-Use Technologies III: Scientific and Technological Advancements

For a number of various uses (storage, mixing, freezing, transportation, formulation, and filling) biopharmaceutical solutions are stored in sterile single-use plastic bags. Material transfers can then occur between containers and contents. These migrations, of different types, depend on the physicochemical characteristics of the material (composition, pH, solubility, viscosity, molecular weight, etc.), the nature of the product (solid, semi-solid and liquid) and the conditions of the material utilization. In the case of single-use polymers, γ-irradiation sterilization of the polymer is often carried out. The interactions could be therefore influenced by the dose and the contact time between the container and the …

Judicious phosphorus (P) management is a global grand challenge and critical to achieving and maintaining water quality objectives while maintaining food production. The management of point sources has been successful in lowering P inputs to aquatic environments, but more difficult is reducing P discharges associated with diffuse sources, such as nonpoint runoff from agriculture and urban landscapes, as well as P accumulated in soils and sediments. Strategies for effective diffuse-P management are imperative. Many options are currently available, and the most cost-effective and practical choice depends on the local situation. This critical review describes how the metrics of P quantity …

Noncontact Speckle Contrast Diffuse Correlation Tomography Of Blood Flow Distributions In Tissues With Arbitrary Geometries, Siavash Mazdeyasna, Chong Huang, Mingjun Zhao, Nneamaka B. Agochukwu, Ahmed A. Bahrani, Lesley Wong, Guoqiang Yu

Biomedical Engineering Faculty Publications

A noncontact electron multiplying charge-coupled-device (EMCCD)-based speckle contrast diffuse correlation tomography (scDCT) technology has been recently developed in our laboratory, allowing for noninvasive three-dimensional measurement of tissue blood flow distributions. One major remaining constraint in the scDCT is the assumption of a semi-infinite tissue volume with a flat surface, which affects the image reconstruction accuracy for tissues with irregular geometries. An advanced photometric stereo technique (PST) was integrated into the scDCT system to obtain the surface geometry in real time for image reconstruction. Computer simulations demonstrated that a priori knowledge of tissue surface geometry is crucial for precisely reconstructing the …

The Proper Use Of Extractables Data - Aspects Beyond Extractables-Measurment, Samuel Dorey, Armin Hauk, Roberto Menzel, Thomas Loewe, Ina Pahl

Single-Use Technologies III: Scientific and Technological Advancements

Appropriate extraction techniques for SUS/SUT and methods for analysis of extractables have been intensively discussed over the last years. Today, several proposals for common methods are available and used to conduct extractables studies in the bio-pharmaceutical industry. Therefore it is expected that the number of available extractables data will significantly rise over the next years and it is worth to (re-)consider their proper use for materials and device qualification and risk assessment. While this exercise is straightforward for container closure systems (CCS), for SUS/SUT the situation is more complex. In CCS applications, a single drug product, in contact with a …

Single-Use Ph Sensor Via A Coplanar Ph Glass Electrode Design, Cd Feng, Robert Garrahy

Single-Use Technologies III: Scientific and Technological Advancements

The elimination of high temperature and high pressure sterilization processes by the advent of Single-Use-Systems (SUS) opens up a new era for the glass electrode based pH sensor. Without the requirement for pretreatment of a pH glass electrode under high temperature and pressure, the classic pH sensing technology delivers a more reliable, more rugged, and more accurate pH measurement. However, the innovative and unique SUS practice also posts new challenges, such as the long shelf-life requirement and the limitation of access to the sensor for calibration once on site,.

At Broadley-James Corporation, a new manufacturing technology has been developed to …

Physical Modeling Of Flow Nets In Groundwater And Determination Of Hydraulic Conductivity, Hannah Nicholas, Moses Karakouzian

AANAPISI Poster Presentations

The goal of this study is to physically model the paths that water particles take through soil, and estimate hydraulic conductivity for several soil configurations. Water paths, or flow lines, are shown by injecting dye into sand contained in a rectangular acrylic glass tank with a vertical barrier in the center; water is poured on one side of the tank and a pump is used to maintain constant head loss. If flow lines are formed, a flow net is to be drawn using photos of the tank and hydraulic conductivity is to be calculated.

This project consists of four phases: …
