Particle separation is of great interest to many biological and biomedical applications. Flow-based methods have been used to sort particles and cells. However, the main challenge with flow based particle separation systems is the need for a sheath flow for successful operation. Existence of the sheath liquid dilutes the analyte, necessitates precise flow control between sample and sheath flow, requires a complicated design to create sheath flow and separation efficiency depends on the sheath liquid composition. This dissertation has aimed to fabricate polypeptide based biomaterial and characterize physical properties. Electrospinning is used as a tool for the sample fabrication. Project focused on determining the feasibility of electrospinning of certain synthetic polypeptides and certain elastin-like peptides from aqueous feedstocks and to characterize physical properties of polymer aqueous solution, cast film and spun fibers and fiber mats. Electrowetting is an electromechanical response that can be used to change the equilibrium
shape of droplets on a surface through the application of an electric potential. By applying this potential asymmetrically to a droplet, the droplet can be moved. Typical electrowetting devices use an electrode covered by a dielectric to reduce electrochemical interactions. Successful electrowetting requires electrodes and dielectric layers that can resist damage through many cycles of voltage.

Pressure ulcers and related skin integrity threats are a significant problem in current transfer/transport systems used for spinal cord injury patients. To understand this problem twenty-three different slings with varying type, material, and features were analyzed in hopes to identify at-risk areas for skin integrity threats such as pressure ulcers. Population samples included non-disabled (otherwise referred to as "healthy") volunteers as well as SCI patients from the James A. Haley Veterans Hospital. High resolution pressure interface mapping was utilized to directly measure the interface pressures between the patient and sling interface. Sinkholes and sinkhole-related features in West-Central Florida (WCF) are commonly identified using geotechnical investigations such as standard penetration test (SPT) borings and geophysical methods such as ground penetrating radar (GPR) and electrical resistivity tomography (ERT). Geophysical investigation results can be used to locate drilling and field testing sites while geotechnical investigation can be used to ground truth geophysical results. Both methods can yield complementary information. Geotechnical investigations give important information about the type of soil, groundwater level and presence of low-density soils or voids at the test location, while geophysical investigations like GPR surveys have better spatial coverage and …

Integrated aquaculture systems (IAS) are a type of recirculating aquaculture systems (RAS) where the wastewater is treated and returned to the fish tanks. The important difference between the two is that in an IAS, wastes from the aquaculture component are recovered as fertilizer to produce an agricultural product whereas in an RAS, waste organics, nutrients and solids are treated and discharged. A pilot marine IAS at Mote Aquaculture Research Park in Sarasota, FL was studied for this project. A Wearable Motion Analysis System (WMAS) was developed to evaluate gait, particularly parameters that are indicative of mild traumatic brain injury. The WMAS consisted on six Opal IMUs attached on the sternum, waist, left and right thigh and left and right shank. Algorithms were developed to calculate the knee flexion angle, stride length and cadence parameters during slow, normal and fast gait speeds. The WMAS was validated for repeatability using a robotic arm and accuracy using the Vicon motion capture system, the gold standard for gait analysis. This thesis documents the design, manufacturing, and assembly of a flexible thermoelectric device. Such a device has immediate use in haptics, medical, and athletic applications. The governing theory behind the device is explained and a one dimensional heat transfer model is developed to estimate performance. This model and consideration for the manufacturing and assembly possibilities are the drivers behind the decisions made in design choices. Once the design was finalized, manufacturing methods for the various components were explored. Background: Jet injectors are advantageous over needle injectors by eliminating sharps hazards. The Government Accountability Office estimates 29% preventable sharp injuries with an estimated direct cost of more than $500 million out of the CDC's reported incidence of 385,000 needle stick injuries per year among US hospital healthcare workers. Yet the forces required to set and trigger devices using spring mechanisms for medication delivery have not been explored. This laboratory experiment measured forces exerted by healthcare workers (HCWs) using a particular jet injector approved by FDA in 2011.

CO hydrogenation and CO oxidation are two important processes addressing the energy and environmental issues of great interest. Both processes are carried out using metallic catalysts. The objective of this dissertation is to study the catalytic processes that govern these two reactions from a molecular perspective using quantum mechanical calculations. Density Functional Theory (DFT) has proven to be a valuable tool to study adsorption, dissociation, chain growth, reaction pathways etc., on well-defined surfaces. DFT was used to study the CO reduction reactions on promoted cobalt catalyst surfaces and CO oxidation mechanisms on cobalt surfaces.

Recirculating aquaculture systems (RAS) produce aquaculture products on land with minimal discharge of waste products and minimal water loss. High costs associated with waste treatment for RAS have triggered the growth of integrated aquaculture systems (IAS) which incorporate macrophytes (aquatic plants) into the treatment train. Clathrates are a class of new materials that have an open-framework structure that allows guest atoms or molecules to be enclosed inside of their polyhedral framework. Varying the number, weight, and size of the guest species in a particular framework allows one to alter the physical properties of the clathrate. When foundations are designed on weak clay layers, it is a common practice to modify the subgrade by installing stone columns. Currently used methods for determining the level of ground modification, represented by the percentage of soil replaced (replacement ratio), assume a rigid foundation. These analytical methods provide the designer with the potential settlement reduction based on the compressibility parameters of the subgrade and the replacement ratio. Research efforts indicated that time dependent concrete properties such as strength, heat evolution, and thermal cracking are predictable through the concept of activation energy. Eighty percent of the 780 million people worldwide that access water from an unimproved source live in rural areas. In rural areas, water systems are often managed by community based organizations and many of these systems do not provide service at the designed levels. The Sustainability Analysis Tool developed in Chapter 2 can inform decision making, characterize specific needs of rural communities in the management of their water systems, and identify weaknesses in training regimes or support mechanisms. The present work investigates, numerically, the process of melting and solidification in hollow vertical cylinders, filled with air and phase change material (PCM). The PCM used is sodium nitrate, which expands upon melting. Therefore, a void must be present within the cylinder, which is filled with air. The influence of cylinder shape on melting time is determined. The numerical model takes both conductive and convective heat transfer into account during the melting process. Estimating the quantity of water that reaches the water table following an infiltration event is vital for modeling and management of water resources. Estimating the time scale of groundwater recharge after a rainfall event is difficult because of the dependence on nonlinear soil characteristics and variability in antecedent conditions. Modeling the flow of water through the variably saturated zone is computationally intensive since it requires simulation of Richards' equation, a nonlinear partial differential equation without a closed-form analytical solution, with parametric relationships that are difficult to approximate. Bolted joints are integral parts of mechanical systems, and bolt preload loss is one of the major failure modes for bolted joint structures. Understanding the damping and frequency response to a varying preload in a single-bolted lap-joint structure can be very helpful in predicting and analyzing more complicated structures connected by these joints.

Drilled shafts are cast-in-place concrete, deep foundation elements that require high levels of quality control to ensure the borehole does not become unstable either during excavation or during concreting. Dancers spend many years practicing repetitive movements in order for their bodies to gain flexibility, strength and muscle memory. This thesis investigated the biomechanics of a dance student's knee motion during a tendu to the front in first, third, and fifth positions. A dancer will often perform 75 or more tendu closing actions during one technique class - particularly in ballet technique. During a tendu the moving foot moves along the floor but the toes never leave the floor. The tendu is used to strengthen the leg muscles, particularly the quadriceps and gluteal muscles. The National Highway Traffic Safety Administration estimates that 11.1%#37; of pedestrian fatalities and 18.5%#37; of bicyclist fatalities in the U.S. occurred in Florida in 2011, which accounts for just 6.1%#37; of the nation's population. Proper hand hygiene is the most effective and efficient method to prevent over 1.3 million deaths annually from diarrheal disease and Acute Respiratory Infections (ARIs). Hand hygiene is also indispensable in achieving the fourth Millennium Development Goal (MDG) to reduce the childhood mortality rate by 2/3rds between 1990 and 2015. Handwashing has been found in a systematic review of studies to reduce diarrhea by 47%#37; and is, thus, capable of preventing a million deaths (Curtis et. al., 2003). This study presents results from non-destructive testing to evaluate the degradation of the CFRP-masonry bond using thermal imaging. The goal of the research was to identify locations where there was evidence of bond deterioration that could subsequently be verified through destructive pull-off testing.
Four full-scale masonry walls were built outdoors at the University of South Florida in 1995 to evaluate the effectiveness of CFRP for repairing settlement damage. Two of the settlement-damaged walls were repaired using single layer, commercially available unidirectional CFRP systems that used Tonen (wall 3) and Henkel (wall 2) epoxies. The practical feasibility of using a Kelvin Probe as a novel reference electrode in the measurement of both potential and polarization pulse response of reinforcing steel in concrete is demonstrated. Potential values measured using a KP reflect greater stability and repeatability than can typically be attained with conventional reference electrodes. In this study, we explore ultra-thin insulators of organic and inorganic composition and their potential role as high-speed rectifiers. Typical applications for these structures include IR sensing, chemical detection, high speed logic circuits, and MEMS enhancements. Collective magnetic behavior in nanostructures is a phenomenon commonly observed in various magnetic systems. It arises due to competing inter/intra–particle interactions and size distribution and can manifest in phenomena like magnetic freezing, magnetic aging, and exchange bias (EB) effect. In order to probe these rather complex phenomena, conventional DC and AC magnetic measurements have been performed along with radio–frequency transverse susceptibility (TS) measurements.
