The goal of the research developed in this dissertation is to develop a more accurate segmentation method for Affymetrix microarray images. The Affymetrix microarray biotechnologies have become increasingly important in the biomedical research field. Affymetrix microarray images are widely used in disease diagnostics and disease control. They are capable of monitoring the expression levels of thousands of genes simultaneously. Hence, scientists can get a deep understanding on genomic regulation, interaction and expression by using such tools.

In this work, microfluidic devices were developed for enriching post-translational modified proteins. Post-translational modifications (PTM) of proteins play essential roles in cellular physiology and disease. The identification of protein substrates and detection of modification site helps understand PTM-mediated regulation in essential biological pathways and functions in various diseases. However, PTM proteins are typically present only at trace levels, making them difficult to identify in mass spectrometry based proteomics. Although the energy saving Warm Mix Asphalt (WMA) technology that produces asphalt mixes at lower temperatures are around for about a decade, there still exist some concerns that are hindering its wide spread implementation. The overall objective of this study is to investigate if the viscosity reductions at higher temperatures have any impacts on asphalt mix compaction and rutting performances. This study reveals that there exists a critical temperature for each asphalt binder below which viscosity will increase with addition of Sasobit®. Therefore, compaction below the critical temperature can negatively impact density.

In this dissertation, ultrasonication assisted Layer-by-Layer (LbL) technology for the preparation of multifunctional poorly water-soluble anticancer drug nanoparticles, paclitaxel and lapatinib, has been developed. Many FDA approved drugs are very low soluble in water; therefore, it is very difficult to load and control their release and targeting efficiently, which greatly confines their application. The development of this method will pave the way for the development and application of those low soluble anticancer drugs.

The power-law distribution can be used to describe various aspects of social group behavior. For mussels, sociobiological research has shown that the Lévy walk best describes their self-organizing movement strategy. A mussel's step length is drawn from a power-law distribution, and its direction is drawn from a uniform distribution. In the area of social networks, theories such as preferential attachment seek to explain why the degree distribution tends to be scale-free. The aim of this dissertation is to glean insight from these works to help solve problems in two domains: cloud computing systems and community detection.

Tissue engineered microenvironments were constructed to test the effects glial cells have on calcium information processing, and to mimic conditions in vivo for tumor invasion and residual cancer after resection of tumor. Submaximal, nM, glutamate (GLU) stimuli were applied to the engineered environments, and the resulting calcium dynamic behavior of neuronal cells was measured to help predict and interpret chaotic systems in the experimental realm. Calcium is a key signaling ion which signals through the N-methyl-D-aspartate (NMDA) glutamate receptor on the neuronal membrane. GLU binding to the NMDA receptor (NMDAR) causes a large and dynamic increase in neuronal intracellular calcium. …

Extreme wet and dry years (± 1 standard deviation, respectively), as well as the top 95 percentile (P95) of daily precipitation events, derived from tropical cyclone (TC) and nontropical cyclone (NTC) rainfall, were analyzed in coastal river basins in Southern Oaxaca, Mexico (Río Verde, Río Tehuantepec, and the Southern Coast). The study is based on daily precipitation records from 47 quality-controlled stations for the 1961 to 1990 period and TC data for the Eastern Tropical Pacific (EPAC). This paper addresses the changes in the physical characteristics (temperature and water/blood content) of breast tissue under different physiological conditions. We examined ex vivo specimens of breast tissue excised at the time of surgery to study the effects of physiological conditions on dielectric properties. We observed that the dielectric properties strongly depend on tissue physiological state. When the biological tissues undergo physiological changes, such as those due to disease or those induced by external changes such as variations in the environmental temperature, the microscopic processes deviate from their normal state and impact the overall dielectric properties. Nanomaterials with quasi-zero, one and two dimensionalities, including silicon nanoparticles, carbon nanotubes, titanium oxide particles and graphene flakes, have been incorporated into the conducting polymer polyaniline to form nanocomposite materials for making flexible supercapacitor sheets. Characterization of the capacitor sheets showed that the inclusion of the nanomaterials in polyaniline has significantly improved the energy and power capabilities of the capacitor. In particular, a specific capacitance of 477.1 F/g has been obtained. However, there have been opportunities as well. Some buying stations have closed or moved, but new buyers have come into the market and contract volumes have been stable overall. There is an international need for a variety of lanthanide elements (rare earth elements, or REEs) in modern electronic and related components. This has led to a desire to broaden production from both previously productive locations and to expand production to new sources, possibly including coal-derived fly ash. The concentration of lanthanides in coal-combustion fly ash depends on a number of factors, one of the most important being the concentration of the elements in the feed coal. We present a consequence based classification procedure for the description logics with self restriction constructor. Due to the difficulty of constructing a concept inclusion model for self restriction, we use a different proof by showing that all the completion rules can simulate all the corresponding ordered resolution inferences.

Over the past few decades, there have been issues of poor fatigue performance (the main failure mechanism) of the welded, tube-to-transverse plate connections within sign support structures. Review of the literature has indicated that a considerable amount of research has been devoted to identifying the structural response characteristics of these signs. Others have tried to identify how these connections may be repaired, retrofitted or simply better designed to sustain longer fatigue lives. However, little attention has been given to using a systematic reliability-based approach to assess the risk of fatigue-induced fracture in these structures.

In human-computer systems, such as supervisory control systems, large volumes of incoming and complex information can degrade overall system performance. Strategically integrating automation to offload tasks from the operator has been shown to increase not only human performance but also operator efficiency and safety. However, increased automation allows for increased task complexity, which can lead to high cognitive workload and degradation of situational awareness. Adaptive automation is one potential solution to resolve these issues, while maintaining the benefits of traditional automation. Liquid crystal devices are attractive for many applications such as information displays, spatial light modulators and adaptive optics, because their optical properties are electrically tunable. However, response time of liquid crystal devices is a serious concern for many applications especially for those who require large phase modulation (≥2π). The focus of this dissertation is the study of measuring light not by energy transfer as is done with a standard photodetector such as a photographic film or charged coupled device, but rather by the forces which the light exerts on matter. In this manner we are able to replace or complement standard photodetector-based light detection techniques. This thesis outlines the high intensity tabletop attosecond extreme ultraviolet laser source at the Institute for the Frontier of Attosecond Science and Technology Laboratory. First, a unique Ti:Sapphire chirped pulse amplifier laser system that delivers 14 fs pulses with 300 mJ energy at a 10 Hz repetition rate was designed and built. The broadband spectrum extending from 700 nm to 900 nm was obtained by seeding a two stage Ti:Sapphire chirped pulse power amplifier with mJ-level white light pulses from a gas filled hollow core fiber. Thulium based fiber lasers represent a promising alternative for pulse energy scaling and high peak power generation with ytterbium based systems at 1µm. Advantages of thulium arise from the operation at longer wavelengths and a large gain bandwidth (1.8-2.1µm). Nonlinear effects, such as self phase modulation, stimulated Raman scattering and stimulated Brillouin scattering generally limit peak power scaling in fiber lasers. The longer wavelength of thulium fiber lasers and large mode field areas can significantly increase the nonlinear thresholds. Pauling’s empirical bond valence-length correlation has proven valuable because it offers a quick and convenient way of checking and evaluating molecular structures and determining oxidation states from measured bond lengths. In this study, a simplified quantum-mechanical approach was used to derive Pauling’s empirical bond valence-length relationship by considering overlap of hydrogen-like orbitals. An expression for the b “empirical” fitting parameter was derived in terms of atomic-orbital exponents. A new set of orbital exponents is presented using published atomic/covalent radii and a continuous function for the effective principal quantum. This study presents a comprehensive investigation of performance and behavior of steel-fiber reinforced concrete pipes (SFRCP). The main goal of this study is to develop the material constitutive model for steel fiber reinforced concrete used in dry-cast application. To accomplish this goal a range of pipe sizes varying from 15 in. (400 mm) to 48 in. (1200 mm) in diameter and fiber content of 0.17%, 0.25%, 0.33%, 0.5%, 0.67% and 83% by volume were produced. The pipes were tested in three-edge bearing condition to obtain the load-deformation response and overall performance of the pipe. The pipes were also subjected to …
