Heterogeneity in responses of cells to a stimulus, such as a pathogen or allergen, can potentially play an important role in deciding the fate of the responding cell population and the overall systemic response. Measuring heterogeneous responses requires tools capable of interrogating individual cells. Cell signaling studies commonly do not have single-cell resolution because of the limitations of techniques used such as Westerns, ELISAs, mass spectrometry, and DNA microarrays. Microfluidics devices are increasingly being used to overcome these limitations. Here, we report on a microfluidic platform for cell signaling analysis that combines two orthogonal single-cell measurement technologies: on-chip flow cytometry …

Low-Complexity Turbo Detection For Single-Carrier Low-Density Parity-Check-Coded Multiple-Input Multiple-Output Underwater Acoustic Communications, Longbao Wang, Jun Tao, Chengshan Xiao, T. C. Yang
Electrical and Computer Engineering Faculty Research & Creative Works
A low-complexity turbo detection scheme is proposed for single-carrier multiple-input multiple-output (MIMO) underwater acoustic (UWA) communications using low-density parity-check (LDPC) channel coding. The low complexity of the proposed detection algorithm is achieved in two aspects: first, the frequency-domain equalization technique is adopted, and it maintains a low complexity irrespective of the highly dispersive UWA channels; second, the computation of the soft equalizer output, in the form of extrinsic log-likelihood ratio, is performed with an approximating method, which further reduces the complexity. Moreover, attributed to the LDPC decoding, the turbo detection converges within only a few iterations. The proposed turbo detection …

Doctoral
The main focus of this thesis is on the design and development of novel fiber optic devices for relative humidity (RH) sensing with emphasis on high sensitivity, a wide humidity range, low temperature dependence, fast response time and good stability.
Novel RH sensors based on fiber bends are fabricated by coating the surface of the buffer stripped bent fiber with selected hygroscopic materials such as Polyethylene oxide or Agarose. It is shown that the Polyethylene oxide coated device has a high sensitivity in a narrow RH range while the Agarose coated fiber bend shows a linear RH sensitivity in a …

A gradient annealing cell has been developed for the high-throughput study of thermalannealing effects on thin-film libraries in different environments. The inexpensive gradientannealing unit permits temperature gradients as large as 28 °C/mm and can accommodate samples ranging in length from 13 mm to 51 mm. The system was validated by investigating the effects of annealing temperature on the crystallinity, resistivity, and transparency of tin-doped indium oxide deposited on a glass substrate by magnetron sputtering. The unit developed in this work will permit the rapid optimization of materials properties such as crystallinity, homogeneity, and conductivity across a variety of applications.

Maintenance of cancer stem cells (CSCs) is regulated by the tumor microenvironment. Synthetic hydrogels provide the flexibility to design three-dimensional (3D) matrices to isolate and study individual factors in the tumor microenvironment. The objective of this work was to investigate the effect of matrix modulus on tumorsphere formation by breast cancer cells and maintenance of CSCs in an inert microenvironment without the interference of other factors. In that regard, 4T1 mouse breast cancer cells were encapsulated in inert polyethylene glycol diacrylate hydrogels and the effect of matrix modulus on tumorsphere formation and expression of CSC markers was investigated. The gel …
Adaptation Of Classical Tidal Harmonic Analysis To Nonstationary Tides, With Application To River Tides, Pascal Matte, David A. Jay, Edward D. Zaron
Adaptation Of Classical Tidal Harmonic Analysis To Nonstationary Tides, With Application To River Tides, Pascal Matte, David A. Jay, Edward D. Zaron
Civil and Environmental Engineering Faculty Publications and Presentations
One of the most challenging areas in tidal analysis is the study of nonstationary signals with a tidal component, as they confront both current analysis methods and dynamical understanding. A new analysis tool has been developed, NS_TIDE, adapted to the study of nonstationary signals, in this case, river tides. It builds the nonstationary forcing directly into the tidal basis functions. It is implemented by modification of T_TIDE; however, certain concepts, particularly the meaning of a constituent and the Rayleigh criterion, are redefined to account for the smearing effects on the tidal spectral lines by nontidal energy. An error estimation procedure …

Industrial and Manufacturing Engineering
ABSTRACT
WOMEN’S HIGH HEEL DISCOMFORT ANALYSIS AND SOLUTION
Kevin Blakley and Annamarie Kepple
Women love high heeled shoes, but this love comes at the cost of comfort. Wearing high heels causes a lot of pain and, in the long term, health complications. This project tries to overcome the problem by ultimately making high heels more comfortable. In order to do this, one must first understand the reasons that high heels are uncomfortable, such as arch angle, heel height, and the changes in walking motion. Once there is a true understanding of the factors that contribute to high heel discomfort, the …
Film Cooling In Fuel Rich Environments, Jacob J. Robertson
Film Cooling In Fuel Rich Environments, Jacob J. Robertson
Theses and Dissertations
The ultra compact combustor is a high performance gas turbine design concept that portends reduced weight for future weapons platforms. A natural outcome of the design is the continual presence of fuel-rich air in the turbine component of the engine. Because modern cooling schemes for hot section turbine blades involve injecting cool, oxygen-rich air adjacent to the surface, the potential arises for reaction with the unconsumed radicals in the mainstream ow and augmented heat transfer to the blade. This outcome is contrary to the purpose of film cooling, and can lead to early life-cycle turbine failure. This study examined the …
Design And Testing Of An H2/O2 Predetonator For A Simulated Rotating Detonation Engine Channel, Stephen J. Miller
Design And Testing Of An H2/O2 Predetonator For A Simulated Rotating Detonation Engine Channel, Stephen J. Miller
Theses and Dissertations
A study is presented on the relationship between a pre-detonator and a detonation channel of an RDE. Testing was conducted on a straight narrow channel made of clear polycarbonate windows connected to an H2/O2 pre-detonator to simulate the RDE initiation scheme and allow for flow visualization. A comparison is made on decoupling distance and wave velocities for a range of pre-detonator designs, inclination angles, equivalence ratios and geometries placed within the simulated channel. Regardless of inclination angle or equivalence ratio the detonation wave decoupled within 25 mm from the pre-detonator exit into the channel. A step change in diameter 25 …

The goal of this article is dual: first, introducing a new model of accident named STAMP (systems-theoretic accident modeling and processes); then applying the model to an innovative process for the treatment of contaminated substances and the re-use of treated substances. This article is a demonstration for a need of a new tool to take into account hazards and safety within socio-technical systems.
Robust Controller Design For An Electrostatic Micromechanical Actuator, Lili Dong, Jason Edwards
Robust Controller Design For An Electrostatic Micromechanical Actuator, Lili Dong, Jason Edwards
Electrical and Computer Engineering Faculty Publications
In this paper, a robust feedback controller is developed on an electrostatic micromechanical actuator to extend the travel range of it beyond pull-in limit. The actuator system is linearized at multiple operating points, and the controller is constructed based on the linearized model. Two kinds of controller designs are developed for set-point tracking of the actuator despite the presences of sensor noise and external disturbance. One of them is a regular fourth order Active Disturbance Rejection Controller (ADRC) and is able to achieve 97% of the maximum travel range. And the other one is a novel multi-loop controller with a …
An Active Disturbance Rejection Based Approach To Vibration Suppression In Two‐Inertia Systems, Shen Zhao, Zhiqiang Gao
An Active Disturbance Rejection Based Approach To Vibration Suppression In Two‐Inertia Systems, Shen Zhao, Zhiqiang Gao
Electrical and Computer Engineering Faculty Publications
This study concerns the resonance problems found in motion control, typically described in a two‐inertia system model as compliance between the motor and the load. We reformulate the problem in the framework of active disturbance rejection control (ADRC), where the resonance is assumed to be unknown and treated as disturbance, estimated and mitigated. This allows the closed‐loop bandwidth to go well beyond the resonant frequency, which is quite difficult using existing methods. In addition, such level of performance is achieved with minimum complexity in the controller design and tuning: no parameter estimation or adaptive algorithm is needed, and the controller …

Pre-Exposure Training As A Means To Reduce Vection Induced Symptoms Of Motion Sickness, Nicholas James Stapleton
Pre-Exposure Training As A Means To Reduce Vection Induced Symptoms Of Motion Sickness, Nicholas James Stapleton
Doctoral Dissertations and Master's Theses
The focus of this study was designed to empirically examine the effect of inducing mild motion sickness as an ameliorative to a more severe motion sickness exposure. Twenty-seven participants were selected for this study based upon their susceptibility to motion sickness, that is, only people who were determined to be motion susceptible were tested. All participants were exposed to a motion sickness-inducing environment. Eighteen participants were trained to adapt to motion sickness by exposure to a milder motion sickness-inducing environment, ending either 6 hours or 24 hours prior to the more severe motion test environment.
Participants during the pre-exposure experimental …
Synthetic Array Processing Of Ocean Ambient Noise For Higher Resolution Seabed Bottom Loss Estimation, Martin Siderius, Lanfranco Muzi, Chris H. Harrison, Peter L. Nielsen
Synthetic Array Processing Of Ocean Ambient Noise For Higher Resolution Seabed Bottom Loss Estimation, Martin Siderius, Lanfranco Muzi, Chris H. Harrison, Peter L. Nielsen
Electrical and Computer Engineering Faculty Publications and Presentations
Predicting transmission loss in the ocean often strongly depends on the bottom loss. Bottom loss can be estimated using ocean noise and vertical array beam-forming [Harrison and Simons, J. Acoust. Soc. Am. 112, 1377–1389 (2002)]. With finite length arrays, the bottom loss estimate using this method can be smoothed due to beam widths. This paper describes how the noise coherence function can be synthetically expanded, which is similar to extending the length of an array. A full wave ocean noise model is used to demonstrate, in simulation, how this leads to improvements in the resolution of bottom loss estimates.

Tracking Knee Joint Functional Axes Through Tikhonov Filtering And Plűcker Coordinates, Wangdo Kim, Yoon-Hyuk Kim, António P. Veloso, Sean S. Kohles
Mechanical and Materials Engineering Faculty Publications and Presentations
Researchers have reported several compensation methods to estimate bone and joint position from a cluster of skin-mounted markers as influenced by Soft Tissue Artifacts (STA). Tikhonov Regularization Filtering (TRF) as a means to estimate Instantaneous Screw Axes (ISA) was introduced here as a means to reduce the displacement of a rigid body to its simplest geometric form. Recent studies have suggested that the ISA of the knee, i.e., Knee Functional Axes (KFA), might be closely connected to the estimation of constraint forces such as those due to medial and lateral connective tissues. The estimations of ISAs were known to be …

An In Situ Experimental Study Of Grain Growth In A Nanocrystalline Fe91ni8zr1 Alloy, Hasan Kotan, Kris A. Darling, Mostafa Saber, Ronald O. Scattergood, Carl C. Koch
Mechanical and Materials Engineering Faculty Publications and Presentations
Grain growth and microstructural evolution of thermally stabilized Fe91Ni8Zr1 were investigated by in situ and ex situ studies. Our investigations suggest that the microstructural evolution is fairly slow and the microstructure shows stabilization up to about 700 °C. Above this temperature, a certain fraction of grains grow abnormally into the nanocrystalline matrix, resulting in a bimodal microstructure and causing the complete loss of thermal stability. The reason for abnormal grain growth and the loss of thermal stability is identified as the appearance of the fcc γ-phase and consequent reduction in the total area of grain boundaries and the overall stored …

Axisymmetric Air Augmented Methanol/Gox Rocket Mixing Duct Experimental Thrust Study, Kyle Jacob Johnson
Master's Theses
A hot-flow axisymmetric Air Augmented Rocket (AAR) test apparatus was constructed to test various mixing duct configurations at static conditions. Primary flow for the AAR was provided through a liquid methanol-gaseous oxygen bipropellant rocket. Experimental thrust measurements were recorded and propellant mass flow rates and chamber conditions were calculated using an iterative solver dependant on recorded propellant line stagnation pressures. Primary rocket flow produced thrust ranging from 14 to 17.9lbf. Primary mass flow rate through testing ranged from 0.071 to 0.085lbm/s with calculated chamber pressures between 298-362psia. Calculated primary flow velocity ranged from 6,600ft/s to 8,000ft/s depending on propellant pressure …
Effects Of Ovariectomy And Anatomical Location On Osteonal Encroachment In Adult Cortical Ovine Bone, Paige Brell Ryan
Effects Of Ovariectomy And Anatomical Location On Osteonal Encroachment In Adult Cortical Ovine Bone, Paige Brell Ryan
Master's Theses
The purpose of this study is to further quantify adult ovine ovariectomized bone for new remodeling characteristics to obtain a better understanding of how remodeling is occurring and the effectiveness of this animal model for the study of postmenopausal osteoporosis. Postmenopausal osteoporosis is a major health concern and animal models to test new treatment options are needed. The ovine model is a good option because the ewes undergo Haversian remodeling, are a large sized animal, and have a similar hormone profile to humans. Ewes, however, do not undergo a natural menopause, so an ovariectomy surgery was conducted in the sheep …

Modeling Flood Reduction Scenarios For A Small Coastal Community, Evan J. Perez
Master's Theses
The Arroyo Grande Creek Watershed, an approximately 170 mi2 watershed located on the central coast of California, drains to the Pacific Ocean via the Arroyo Grande Creek that passes through several coastal cities including the community of Oceano. At the mouth of the Creek is the Arroyo Grande Lagoon, which is connected to another lagoon known as the Oceano Lagoon, by a tidal flap-gate whose hydraulics is a function of water levels in the two lagoons. Historically the Oceano Lagoon has played a part in floods that have occurred in the community of Oceano. The most recent flooding occurred …

Dc, Rf, And Thermal Characterization Of High Electric Field Induced Degradation Mechanisms In Gan-On-Si High Electron Mobility Transistors, Matthew Anthony Bloom
Dc, Rf, And Thermal Characterization Of High Electric Field Induced Degradation Mechanisms In Gan-On-Si High Electron Mobility Transistors, Matthew Anthony Bloom
Master's Theses
Gallium Nitride (GaN) high electron mobility transistors (HEMTs) are becoming increasingly popular in power amplifier systems as an alternative to bulkier vacuum tube technologies. GaN offers advantages over other III-V semiconductor heterostructures such as a large bandgap energy, a low dielectric constant, and a high critical breakdown field. The aforementioned qualities make GaN a prime candidate for high-power and radiation-hardened applications using a smaller form-factor. Several different types of semiconductor substrates have been considered for their thermal properties and cost-effectiveness, and Silicon (Si) has been of increasing interest due to a balance between both factors.

Hydrologic Modeling Of The San Joaquin Valley Watershed For Purposes Of Nitrate Analysis, Stephen Carl Clayton
Hydrologic Modeling Of The San Joaquin Valley Watershed For Purposes Of Nitrate Analysis, Stephen Carl Clayton
Master's Theses
The San Joaquin Valley is regarded as one of the most productive agricultural regions in the world. This extensive agriculture has, however, caused extensive pollution of both ground water and surface water. This thesis develops a hydrologic model of the surface and ground waters of the San Joaquin Valley. Such modeling is useful in the development and implementation of water quality regulations such as Total Maximum Daily Loads (TMDLs). A properly validated watershed simulation model can supplement data collection and can account for watershed characteristics including topography, soils, climate, land cover, anthropogenic activities, as well as simulate watershed responses including …
