A serum free medium was developed for the production of recombinant antibody against Botulinum A (BoNTA) using dihydrofolate reductase deficient Chinese Hamster Ovary Cells (CHO-DG44) in suspension culture. An initial control basal medium was prepared, which was similar in composition to HAM’s F12: IMDM (1:1) supplemented with insulin, transeferrin, selenium and a lipid mixture. The vitamin concentration of the basal medium was twice that of HAM’s F12: IMDM (1:1). CHO-DG44 cells expressing S25 antibody grew from 2 x 105 cells to maximum cell density of 1.04 x 106 cells/ml after 5 days in this control medium. A central …

Structural weight and efficiency are hurdles for morphing aircraft being realizable on the full-scale level. The optimal distribution and orientation of actuators throughout an in-plane flexible morphing wing structure is investigated. The drive to minimize structural weight causes a wing to be more flexible and the location and orientation of the actuators become more critical as the structure becomes more flexible. NextGen's N-MAS morphing wing is used as a case study. The wing is modeled as a number of unit cells assembled in a scissor-like structure, each comprised of four linkages pinned together and an actuator. The flexible skin of …

This paper describes a human-in-the-loop motion-based simulator interfaced to hybrid-electric power system hardware, both of which were used to measure the duty cycle of a combat vehicle in a virtual simulation environment. The project discussed is a greatly expanded follow-on to the experiment published in [1,7]. This paper is written in the context of [1,7] and therefore highlights the enhancements. Dynamic response of inhomogeneous materials exhibits new effects, which often do not exist in homogeneous media. It is quite natural that most of studies of wave and front propagation in inhomogeneous materials are associated with numerical simulations. To develop a numerical algorithm and to perform the numerical simulations of moving fronts we need to formulate a kinetic law of progress relating the driving force and the velocity of the discontinuity. The velocity of discontinuity is determined by means of the non-equilibrium jump relations at the front. An efficient solution method for evaluating radiative transport in a foam layer is a valuable tool for predicting the properties of the layer. Two different solution methods have been investigated. First, a reverse Monte Carlo (RMC) simulation has been developed. In the RMC simulation photon bundles are traced backwards from a detector to the source where they were emitted. The RMC method takes advantage of time reflection symmetry, allowing the photons to be traced backwards in the same manner they are tracked in a standard forward Monte Carlo scheme. A computational study of the effects of a wing fence on the T-38 Talon was performed. RANS simulations were conducted using the CFD solver AVUS to examine the flow around the T-38 and the fence at a Reynolds number of 10 million. The T-38 was modeled as a half aircraft with a symmetry plane down the center line and did not include the empennage. The engine inlet and exhaust were modeled as sink and source boundary conditions using mass flow and pressure specifications. Two fence geometries placed 26" from the wing tip were tested. The United States Air Force is partnering with the United States Army as well as allied nations to develop a revolutionary advance in logistical support known as the Joint Precision Air Drop System (JPADS). The focus of this study is to develop a process to quantitatively analyze system sensitivities to various types of weather inputs and the corresponding effect on system accuracy. Weather balloons were used to provide representative "truth" to which forecast weather could be compared. Modern navigation systems can use the Global Positioning System (GPS) to accurately determine position with precision in some cases bordering on millimeters. Unfortunately, GPS technology is susceptible to jamming, interception, and unavailability indoors or underground. There are several navigation techniques that can be used to navigate during times of GPS unavailability, but there are very few that result in GPS-level precision. One method of achieving high precision navigation without GPS is to fuse data obtained from multiple sensors. The Beowulf cluster approach to parallel computation offers a potentially cheap and robust source of computational power for high complexity algorithms in robotics. The challenge is to integrate this approach with the mobility and time critical response constraints of many robotic algorithms. A hydrologic model was driven by the climate projected by 11 GCMs under two emissions scenarios (the higher emission SRES A2 and the lower emission SRES B1) to investigate whether the projected hydrologic changes by 2071–2100 have a high statistical confidence, and to determine the confidence level that the A2 and B1 emissions scenarios produce differing impacts. There are highly significant average temperature increases by 2071–2100 of 3.7°C under A2 and 2.4°C under B1; July increases are 5°C for A2 and 3°C for B1. Two high confidence hydrologic impacts are increasing winter streamflow and decreasing late spring and summer flow. …

We present a study of high-field transport in carbon nanofibers (CNFs) and breakdown phenomena due to current stress. In situ measurements with scanning transmission electron microscopy reveal that the failure mode of CNFs is strongly related to the morphology of graphite layers comprising CNFs. Comparison with carbon nanotube (CNT) breakdown is made, demonstrating that the current capacity of CNFs is described by a similar model as that of CNTs with a modification of the current capacity of each graphitic layer. The maximum current density is correlated with resistivity, leading to the conclusion that lower resistivity results in higher current capacity …

It is known that every causality-preserving transformation of Minkowski space-time is a composition of Lorentz transformations, shifts, rotations, and dilations. In principle, this result means that by only knowing the causality relation, we can determine the coordinate and metric structure on the space-time. However, strictly speaking, the theorem only says that this reconstruction is possible if we know the exact causality relation. In practice, measurements are never 100% accurate. It is therefore desirable to prove that if a transformation approximately preserves causality, then it is approximately equal to an above-described composition.

The E3 Team (Exploring Ethical decision-making in Engineering) is a group of engineering educators and educational researchers who have worked collaboratively since 2000 to understand the underlying causes of academic dishonesty in engineering undergraduate populations. The team was especially motivated by decades of others’ work showing that, when surveyed, engineering students were among those most likely to report frequently cheating. This paper summarizes some of the team’s more important findings from three major studies that surveyed a total of 1300 engineering and humanities undergraduates at eleven institutions. A new course was developed for Materials Engineering students to design, create, and install interactive, educational displays on Materials Science & Engineering for a science museum at a local K-6 charter school. The course grew out of an ASM Foundation grant “to excite young people in materials, science, and engineering careers,” and the challenge was put forth to Cal Poly students. A formal design sequence was applied to give the students the opportunity to learn about the design process, project management, and teamwork. Engineering building design focuses on optimising operational energy use and ignores the energy required to procure and construct a building. This energy, termed ‘embodied energy’, can be very significant when compared to operational energy. Therefore, it is important to minimise the embodied in buildings; this must be done at the design stage. This paper presents a comparative embodied energy analysis of two structural design solutions for a modern office building: one in concrete and one in steel. JET is an automated unit testing tool for Java classes annotated with JML specifications; JML is a formal interface specification language for Java to document the behavior of Java classes and interfaces. JET tests each method of the class under test separately. For each method, it generates a collection of test data, executes them, and decides test results (i.e., pass/fail) by using JML specifications as test oracles, thereby fully automating unit testing of Java classes. With the typical engineering program already having a high credit hour requirement, the question becomes, how to best integrate biology concepts into a packed engineering curriculum. A typical biology course is not likely to introduce the important concepts of biology to engineering students. The solution here is to develop a hybrid course that integrates chemistry and biology. The Center continues to strengthen the research climate across the institutional settings. A number of activities have been directed at this effort: internal funding of small student and faculty research projects; development of an internal funding process for doctoral student research; and presentations by researchers at Center meetings and professional conferences. The internal research program has been successful in providing relatively non-threatening experiences with proposal preparation, review, negotiation, and the conduct of small research projects. Six proposals were funded for exploratory research projects in 2005-06 and five projects were funded in 2006-07. As these projects are completed, findings are shared …

In most engineering economics courses, the emphasis is on how to compare alternatives and learn about time value of money and interest and inflation rates. The students are usually given the costs and asked to compare the alternatives. However, this is the easy part of the analysis. The hard part of real project evaluations is developing the cost and investment numbers to do the engineering economics analysis. In his unpublished lectures on general algebra, a well-known algebraist D. K. Faddeev expressed a belief that every true mathematical statement can be generalized in such a way that it becomes trivial. To the best of our knowledge, this belief has never been formalized before. However, writing effective assertions of right abstraction levels is difficult because on the one hand, detailed assertions are preferred for through testing (i.e., to detect as many errors as possible), but on the other hand abstract assertions are preferred for readability, maintainability, and reusability. As assertions become a practical tool for testing and debugging programs, this is an important and practical problem to solve for the effective use of assertions. Low cost HF RFID scanner subsystems that both deliver power and provide high bandwidth bidirectional communication channels have recently become available. These devices are anticipated to become ubiquitous in next-generation cell phones and enable a wide range of emerging e-commerce applications.
This paper considers the use of HF RFID to power and communicate with implantable medical devices. We successfully communicated with ten transponders that were implanted at three locations within a human cadaver. In our previous work, we provided a theoretical explanation for an empirical fact that it is easier to find a unique root than the multiple roots. In this short note, we strengthen that explanation by showing that finding one of many roots is also difficult.
