Pichia pastoris is an excellent host for high-level heterologous gene expression, but there is still much interest in improving the productivity of recombinant protein production. P. pastoris produces a small amount of ethanol as a by-product during the glycerol fed-batch phase and the mixed-feed induction phase (glycerol-methanol) of high cell density fermentations, regardless of the phenotype (Mutf, Mut" or Mut-). We have investigated ethanol repression of the AOXl promoter using strains, GS115 (Mutf) and MC100-3 (Mut-), expressing an AOXI-lac2 fusion. The addition of 10 mg t1 ethanol at the start of methanol induction delayed P-galactosidase production and methanol utilization …

Surface micromachining of micro-electro-mechanical systems (MEMS), like all other fabrication processes, has inherent variation that leads to uncertain material and dimensional parameters. To obtain accurate and reliable predictions of mechanism behavior, the effects of these variations need to be analyzed. This thesis expands already existing tolerance and uncertainty analysis methods to apply to micro compliant mechanisms. For simple compliant members, explicit equations can be used in uncertainty analysis. However, for a nonlinear implicit system of equations, the direct linearization method may be used to obtain sensitivities of output parameters to small changes in known variables. A fiber reinforced composite material comprising a resin matrix and primary reinforcement fibers and further comprising secondary, smaller diameter, reinforcement fibers at one or more ply interfaces, or portion thereof, provides improved interlaminar toughness, strength, and delamination resistance and without substantial increase in weight. In one embodiment, the small fibers are attached to one side of a conventional prepreg prior to lamination. The small fibers are flexible and are expected to conform to the shape and distribution of the primary reinforcing fibers at the interface.

A three-dimensional waveguiding structure for a microcavity surface-emitting laser is described in which native aluminum oxide layers provide control of intracavity waveguiding and the laser optical mode structure of the emitted beam. Microcavity lasers described herein account for the blueshift of the emission wavelength as the laser lateral dimensions are reduced to or below the emission wavelength.

In this document we describe the features of the baseline system to be used in the Distributed Speech Recognition (DSR) front end large vocabulary continuous speech recognition (LVCSR) evaluations being conducted by the Aurora Working Group of the European Telecommunications Standards Institute (ETSI). The objective of these evaluations is to determine the robustness of different front ends for use in client/server type telecommunications applications. As such, our experiments are designed to test the following focus conditions on the DARPA Wall Street Journal (WSJ0) corpus using a 5000-word closed-loop vocabulary and a bigram language model:

An isolated DNA sequence which regulates the expression of a heterologous gene composed of a mouse whey acidic protein promoter having a length of greater than about 2.4 kb extending upstream from the unique KpnI site in the mouse whey acidic protein gene is disclosed. Specifically a mouse whey acidic protein promoter of about 4.1-4.2 kb in length extending upstream from the unique KpnI site is preferred. Before the Montreal Protocol of 1987 and the subsequent phasing-out of chlorofluorocarbons (CFCs) in industrial applications, rigid polymer foams were made using these compounds as secondary blowing agents. The CFCs remain trapped in the gaseous part of the cellular foam structure, and once discarded these foams constitute a significant reservoir for the environmental release of ozone-depleting chemicals. Environmental degradation of the foam accelerates this process. Of particular interest in this work is the degradative effect of ultraviolet (UV) light on rigid polyurethane foams. A new approach for uncoupling the equations of motion typical for rotordynamical systems is presented. The method does not neglect the speed-dependent e!ects, such as gyroscopic e!ects, and can be particularly valuable in the controller design of actively controlled rotors. In the presence of hysteretic type of damping, the resulting uncoupled gyroscopic systems come with an equivalent viscous damping, equivalent in a sense of yielding the same natural frequency and decay rate. The approach is illustrated through three examples of technical interest: a Je!cott rotor with hysteretic damping, a Stodola}Green rotor, and a rotor of a small gas turbine. The …

A radio frequency transponder is provided which comprises a substrate layer, a radio frequency integrated circuit affixed to said substrate layer; and an antenna provided on said substrate layer in electrical connection with said radio frequency integrated circuit. The antenna is comprised of a conductive paste material having a resistivity of approximately 20 µΩ-cm, which is greater than the resistivity of a conventional etched copper antenna, but still sufficiently low to provide a radio frequency transponder having acceptable read range. A method of fault tolerant reconfiguration and operation of a field programmable gate array (FPGA) during normal on-line operation includes selecting a programmable logic block as a programmable logic block under test, testing the programmable logic block under test, and detecting the existence of any faults in the programmable logic block under test. An improved breakaway steel guardrail post for use in dissipation of impact energy upon impact of the post having an upper post member and a lower post member, a connecting joint, and a mechanism connected to the upper and lower post members for controlling the energy dissipation of the guardrail post about the connecting joint at a predetermined rate. A first embodiment utilizes a cable restraint lopped through openings in he guardrail posts. Other embodiments included straps and fasteners disigned to distort or fail at predetermined rates or strengths and combinations thereof.

This report covers Phase II of a three-phase pooled-fund project in Wisconsin, Iowa and Minnesota, to determine the perception/satisfaction levels of the driving public and how they correlate with the states’ physical data bases used to determine priorities for pavement improvements on rural, two-lane highways.
In Phase I, six focus groups were conducted in each state to determine the beliefs and issues about pavements that could be used to draft statewide questionnaires. This paper presents a programming language which we believe to be most appropriate for the automation of parallel data processing, especially data processing of concern to the oil industry and to the U.S. Federal Agencies involved in the analysis of Satellite Telemetry Data. In many practical situations, we are faced with a necessity to combine sophisticated mathematical knowledge about the analyzed systems with informal expert knowledge. To make this combination natural, it is desirable to reformulate the abstract mathematical knowledge in understandable intuitive terms. In this paper, we show how this can be done for an abstract metric.
One way to define a metric is to pick certain properties P1, ..., Pn, and to define a similarity between two objects x and y as the degree to which P1(x) is similar to P1(y) and P2(x) is similar to P2(y), etc.

Laboratory experiments are presented that demonstrate a novel in situ semipassive reactive barrier for the degradation of 2,4 dinitrotoluene created by coating aquifer surfaces by deposition of colloidal MnO2, which catalyzes ozone degradation and enhances contaminant oxidation. Ozone is added to the reactive barrier and is transported through the zone with the contaminants by existing hydraulic gradients. The communication presents the preliminary laboratory investigation demonstrating the viability of this method. Studies were conducted by coating Ottawa sand with colloidal MnO2. Results show that concentrations of MnO2 in the range of 0.2 mg/g can be deposited with no measurable change in …

We compare three microwave modulation methods experimentally and theoretically using a semiconductor quantum-well (QW) Fabry–Perot test laser: 1) direct microwave current modulation of the test laser (electrical modulation); 2) optical modulation by an external single-wavelength pump laser with a modulated optical injection power; and 3) electrical modulation of the test laser that is injection locked by an external single-wavelength pump laser with a constant injection power. This is the first direct comparison of the three modulation methods on the same QW laser, to the best of our knowledge. The bandwidth of optical absorption modulation is 7.7 GHz, which is 1.45 …

Nitric oxide (NO) has been shown to suppress platelet activation, induce vasodilation, inhibit smooth muscle cell proliferation, and act against infection. Activated platelets generally have opposite effects from these. They secrete platelet-derived growth factor (PDGF), which stimulates smooth muscle growth, serotonin, which is a platelet agonist, and a number of agents that promote further platelet aggregation. However, activated platelets also produce NO through the enzymatic functions of the constitutive form of nitric oxide synthase (NOS). The primary functions of this capability are not yet completely understood. Due to its low molecular weight and high diffusivity, NO is quickly transported from …

It is often important to check whether the maximum max f of a given function f on a given set B is smaller than a given number C. This "crude range testing" (CRT) problem is one of the most important problems in the practical application of interval analysis. Empirical evidence shows that the larger the difference C - max f, the easier the test. In general, the fewer global maxima, the easier the test; and finally, the further away global maxima are from each other, the easier the test. On the example of physics, we show that the traditional one-level description is not completely adequate. For a more adequate structure, a hierarchical description of uncertainty is necessary, which supplements the more traditional first-order uncertainty with second-order, third-order and more sophisticated models. In particular, the second-order approach seems to provide a bridge between probabilistic and fuzzy approaches to uncertainty.

The purpose of this descriptive study was to explore the relationship between a carrier's service/capacity share and passenger share to determine the presence of the s-curve. The author is unaware of a current, accepted analysis for understanding the s-curve with any degree of reliability. Regression analysis was used to correlate service/capacity share against passenger share for domestic, United States air carriers. Carrier ranking was then added as a predicting variable to gain further insight into the correlation between service share and passenger share.
