Stent retriever mechanical thrombectomy (SRMT) is part of the clinical standard-of-care for acute ischemic stroke (AIS) patients with large vessel occlusion (LVO). Endothelial injury (EI) is widely associated with SRMT. Studies report finding EI markers in over 50% of retrieved clots, suggesting the frequency of EI in MT patients is rather common. The mechanism of EI and its contribution to MT failure and clinical outcomes remain poorly understood because of a lack of techniques to observe EI and study it in vitro. Coronary Artery Disease (CAD) affects millions of people worldwide and remains a leading cause of morbidity and mortality. To effectively plan treatment and risk stratification, assessing CAD severity is of paramount importance. Fractional flow reserve (FFR) is a critical clinical parameter used to evaluate CAD severity, measured by the ratio of mean distal coronary pressure to mean aortic pressure during hyperemic conditions. Although non-invasive FFR estimation methods have gained popularity, computational fluid dynamics (CFD) is impractical for routine clinical use due to the time and resources required. The author of this thesis conceived and investigated an unexpected electrical signal that may not be noticed during regular electroglottography (EGG) when electrodes are conventionally placed on the anterior surface of the throat. It appears that there is a measurable electrical signal from the EGG equipment when electrodes are placed over the frontal cheek (belly of zygomatic muscle), and the tongue is elevated to contact the hard palate, or the tongue is lowered to break the contact. Therefore, this phenomenon is designated as unconventional electroglottography (UEGG, Dr. Steven Cloud, private communication, July 17, 2022).

Volumetric muscle loss (VML) overwhelms muscle’s robust capacity for regeneration. A key event in the etiology of VML injury is the bulk loss of structural cues provided by the underlying extracellular matrix (ECM). While muscle is a highly structured tissue, with cell and ECM alignment in the direction of contractile force production, the impact of scaffold alignment on recovery remains unclear. Bulk human decellularized skeletal muscle (DSM) tissues were sectioned into 10 x 1-2 mm fibers. Dissipative particle dynamics (DPD), a coarse grain simulation method, was applied to the membrane formation process of non-solvent induced phase separation (NIPS) to gain further insight on the mechanism of certain variables and how they affect the final morphology. NIPS involves two solutions, an organic polymer dissolved in an organic solvent colloquially called the dope and an aqueous coagulation bath, brought into contact with one another. The solvents then mix, causing the polymer to fall out of solution as an asymmetric membrane with a dense surface layer and a more open subsurface layer in response to the decreasing solubility. An automation program, named Jacob LCA, was created to help perform life cycle assessment (LCA). The program uses a template file to perform controlled and consistently ordered actions with the LCA program, SimaPro, and effectively removes the need for manual user input. It can be set to run using data from one or more life cycle inventory (LCI) files, which allows for rapid iteration and testing of data. The mitral valve serves a critical role in healthy cardiac function by ensuring the unidirectional flow of oxygenated blood from the left atrium into the left ventricle. It experiences the highest pressures found within the heart and its closure is the result of a complex interaction of several different structures that, furthermore, are unique to each individual. Despite the valve’s vital role however, the specific function of these constituent structures is not fully understood. Pulsed power treatment has been used to induce regulated cell death (RCD) in cells or ablate tumors in animals. A subset of pulsed power as electroporation with high voltage and pulse duration of milliseconds is used for biomedical treatment to induce pores in the plasma membrane of cells. Nanosecond Pulsed Electric Fields (nsPEFs)– an extension of electroporation, uses waveforms with pulse durations on the order of 10-900 nanoseconds. nsPEF treatment has demonstrated intracellular effects for potential biomedical applications. Surgical ablation of cardiac tissue is the definitive treatment for the most common arrhythmia, atrial fibrillation. In current medical practice, myocardial tissue is often ablated by the application of radiofrequency (RF) currents which are delivered through an electrode to create lesions that block the electrical pathways that sustain reentrant arrhythmias and restore the normal heart rhythm. Heart failure (HF) is one of the leading causes of deaths for adults worldwide, but has disproportionately significant effects on pediatric patients that are often overshadowed. Electrospinning is a versatile tool used to produce highly customizable nonwoven nanofiber mats of various fiber diameters, pore sizes, and alignment. It is possible to create electrospun mats from synthetic polymers, biobased polymers, and combinations thereof. The post-processing of the end products can occur in many ways, such as cross-linking, enzyme linking, and thermal curing, to achieve enhanced chemical and physical properties. Myosin is a motor protein that facilitates muscle contraction and movement by stepping along actin filaments using energy from ATP hydrolysis. If myosin or other motors are disrupted throughout the body, it can have many harmful effects. Hypertrophic cardiomyopathy is a disease caused by gene mutations that affect myosin heavy chains in the heart. The tissue of the heart becomes abnormally thick, which can make it more difficult to pump blood or block blood flow out of the heart. Tendon and ligament injuries are debilitating conditions across species. Poor regenerative capacities of these tissues limit restoration of original functions. The first study evaluated the effect of cellular administration on tendon/ligament injuries in horses using meta-analysis. The cellular administration was effective in restoring ultrasonographic echogenicity and increasing vascularity during early phase of healing. Additionally, it improved microstructural organization of healed tissue in terms of cellularity and fiber alignment. However, the study did not support its use for increasing rate of return to performance, expression/deposition of tendon-specific genes/proteins, or mechanical properties.

This paper proposes a framework for integrating IoT and automation in a biomedical laboratory to improve safety, optimize processes, and enhance students' learning experience. The framework incorporates a centralized control unit and distributed subsystems to control equipment and machinery and includes autonomous robotics and intelligent monitoring systems. The paper presents the results of undergraduate students' work on automating various biomedical processes, including developing an Intelligent Autonomous Monitoring (IAM) device. IAM utilizes machine learning algorithms to identify outliers in processes and safety hazards. Moreover, IAM autonomously detects and localizes biomedical tools and equipment. This paper reports on design and fabrication of a novel soft fish robot. Application of soft actuators for the fish tail will generates continuum bending motion which resembles the natural motion of the fish. However, most soft actuator mechanisms are complex and have low efficiency. Thus, to address this issue we have developed a 3D printed soft bending actuator which can be actuated with an electromotor. Diffuse Reflectance Spectroscopy (DRS) is a non-invasive optical method to characterize tissue optical properties for disease diagnosis and health monitoring. Two optical fibers are often used in a DRS system: one to deliver light to the tissue and the other to gather diffuse reflectance spectra, which provide quantitative details about the structure and composition of the tissue. Prediction of microstructure evolution during material processing is essential to control the material properties. Simulation tools for microstructure evolution prediction based on physical concepts are computationally expensive and time-consuming. Therefore, they are not practical when either there is an urgent need for microstructure morphology during the process or there is a need to generate big microstructure datasets. Essentially, microstructure evolution prediction is a spatiotemporal sequence prediction problem, where the prediction of material microstructure is difficult due to different process histories and chemistry. Syndactyly is one of the most common congenital malformations of the hand and is a result of a failed separation of adjacent digits. These digits are common among congenital limb malformations that affect 1 out of 2,000-3,000 people at birth. A reconstructive surgery would be needed to separate the digital skin, reconstruct the web space between fingers, and cover the separated digits with soft tissues. Finding the optimal shape and size for a dorsal flap in the web commissure reconstruction is important to avoid web creep. Advances in the number and type of available biomaterials have improved medical devices such as catheters, stents, pacemakers, prosthetic joints, and orthopedic devices. The introduction of a foreign material into the body comes with a risk of microbial colonization and subsequent infection. Protein modeling is a rapidly expanding field with valuable applications in the pharmaceutical industry. Accurate protein structure prediction facilitates drug design, as extensive knowledge about the atomic structure of a given protein enables scientists to target that protein in the human body. However, protein structure identification in certain types of protein images remains challenging, with medium resolution cryogenic electron microscopy (cryo-EM) protein density maps particularly difficult to analyze. Recent advancements in computational methods, namely deep learning, have improved protein modeling. Research in HCI applied to clinical interventions relies on normative assumptions about which bodies and minds are healthy, valuable, and desirable. To disrupt this normalizing drive in HCI, we define a “counterventional approach” to intervention technology design informed by critical scholarship and community perspectives. This approach is meant to unsettle normative assumptions of intervention as urgent, necessary, and curative. We begin with a historical overview of intervention in HCI and its critics. Then, through reparative readings of past HCI projects in autism intervention, we illustrate the emergent principles of a counterventional approach and how it may manifest research outcomes that …

Catechol-modified bioadhesives generate hydrogen peroxide (H2O2) during the process of curing. A robust design experiment was utilized to tune the H2O2 release profile and adhesive performance of a catechol-modified polyethylene glycol (PEG) containing silica particles (SiP). An L9 orthogonal array was used to determine the relative contributions of four factors (the PEG architecture, PEG concentration, phosphate-buffered saline (PBS) concentration, and SiP concentration) at three factor levels to the performance of the composite adhesive.
