My thesis concerns analysis of data from optical and electrophysiological measurement tools for calculate potential biomarkers in three biomedical applications: (1) the gastrointestinal immaturity of infants of different ages, (2) prefrontal connectivity in healthy older adults, and (3) hemodynamic-metabolic coherence during vascular occlusion. The first application relied on electrogastrogram (EGG) measurements taken on 81 newborns in a neonatal intensive care unit. Of the 81 babies, 77 were born prematurely. By separating these babies into different gestational age groups and analyzing their EGG data, I was able to find population differences in common EGG analysis parameters. Autism Spectrum Disorder (ASD) is a condition characterized by social and behavioral impairments, affecting approximately 1 in every 44 children in the United States. Common symptoms include difficulties in communication, interpersonal interactions, and behavior. While symptoms can manifest as early as infancy, obtaining an accurate diagnosis may require multiple visits to a pediatric specialist due to the subjective nature of the assessment, which may yield varying scores from different specialists. A current trend in cardiac muscle regeneration and repair is to recreate the biophysical and biomechanical environment of the neonatal hearts, which has been shown to promote cardiac regeneration. This translational need requires thorough understanding of the biomechanical properties of the neonatal hearts. Spinal cord injury (SCI) is a common condition and is characterized by traumatic and progressive damage that results in limited neuron regeneration and thus a restricted recovery. In SCI, the vasculature is also severely damaged, which exacerbates inflammation at the injury site. Specifically, insufficient angiogenesis occurs at the site of injury due to the fibrotic pathological microenvironment1 . SCI also leads to the formation of a fluid-filled cavity and thus gliosis and fibrosis, forming a dense glial and fibrotic scar around the cavity. Microalgae biofilms have been demonstrated to recover nutrients from wastewater and serve as biomass feedstock for bioproducts. However, there is a need to develop a platform to quantitatively describe microalgae biofilm production, which can provide guidance and insights for improving biomass areal productivity and nutrient uptake efficiency. This paper proposes a unified experimental and theoretical framework to investigate algae biofilm growth on a rotating algae biofilm reactor (RABR). This study aspires to find a new screening approach to trace DNA recovery techniques to yield a higher quantity of trace DNA from larger items of evidence. It takes the path of visualizing trace DNA on items of evidence with potential DNA so analysts can swab a more localized area rather than attempting to recover trace DNA through the general swabbing technique currently used for trace DNA recovery. The relationship between reactive oxygen and nitrogen species (ROS-RNS) secretion and the concomitant biocorrosion of degradable magnesium (Mg) materials is poorly understood. We found that Mg foils implanted short term in vivo (24 h) displayed large amounts of proinflammatory F4/80+/iNOS + macrophages at the interface. We sought to investigate the interplay between biodegrading Mg materials (98.6% Mg, AZ31 & AZ61) and macrophages (RAW 264.7) stimulated with lipopolysaccharide (RAW 264.7LPS) to induce ROS-RNS secretion. Glioblastoma (GBM) brain tumors are highly aggressive gliomas due to genetic and cellular heterogeneity. Current GBM treatment consists of surgical resection of the tumor combined with radio- or chemo-therapies. While these treatments have increased the life expectancy for GBM patients up to 20 months, they have had little effect on the 5-year survival rate. The complex cellular and genetic composition of the tumor makes current treatments less effective long term. Peripheral arterial occlusive disease (PAOD) is characterized by atherosclerosis, which is the buildup of plaque, consisting largely of cholesterol, in the arterial walls. This plaque accumulation eventually blocks blood flow to the limbs, causing symptoms such as intermittent claudication and tissue death in cases of critical limb ischemia. The body compensates for the reduced perfusion by enlarging pre-existing bypass arteries, known as collaterals, in a process called arteriogenesis. The purpose of this study was to investigate and compare the corrosion mechanisms between wrought and additive-manufactured (3D-printed) copper and stainless steel. The experimental procedure consisted of measuring the open circuit potential, electrochemical impedance spectroscopy, linear sweep voltammetry, Tafel analysis, surface topology, and scanning electron microscopy for each metal within salt water, tap water, sulfuric acid, and synthetic body fluid (excluding copper in synthetic body fluid).
Overall, printed stainless steel was more corrosion-resistant than wrought stainless steel in tap water and synthetic body fluid based on OCP, LSV, and surface topology results. Additionally, printed copper was more corrosion-resistant than wrought …

Clinical studies have shown that patients undergoing renal replacement therapy are more susceptible to developing hemolysis, or the rupturing of red blood cells. Rapid hemolysis can cause symptoms such as anorexia, vomiting, and even death in severe cases. Atherosclerosis is the most prevalent pathology of cardiovascular disease with no known cure. Despite the many systemic risk factors for atherosclerosis, plaques do not form randomly in the vasculature. Instead, they form around bifurcations and the inner curvature of highly curving arterial segments that contain so-called disturbed blood flow that is low in magnitude and multidirectional over the cardiac cycle. Conversely, straight, non-bifurcated arterial segments that contain moderate-to-high and unidirectional (i.e., normal) blood flow are protected from plaque development. The severe side effects of acute dehydration and blood loss may be prevented if assessed and treated quickly. As amplifier technology has improved, small peripheral venous pressure (PVP) signals collected using a transducer on an intravenous catheter can be analyzed to monitor patient volume status. This thesis details the development of an embedded system prototype that quickly and accurately assesses volume status using PVP analysis. The volume prediction algorithm classified 10 seconds of PVP data as “dehydrated” or “resuscitated”. This paper presents an innovative and fully automatic solution of generating as-built computer-aided design (CAD) drawings for landscape architecture (LA) with three dimensional (3D) reality data scanned via drone, camera, and LiDAR. To start with the full pipeline, 2D feature images of ortho-image and elevation-map are converted from the reality data. A deep learning-based light convolutional encoder–decoder was developed, and compared with U-Net (a binary segmentation model), for image pixelwise segmentation to realize automatic site surface classification, object detection, and ground control point identification. The purpose of this study was to characterize changes in cortical activity and connectivity in stroke survivors when vibration is applied to the wrist flexor tendons during a visuomotor tracking task. Data were collected from 10 chronic stroke participants and 10 neurologically-intact controls while tracking a target through a figure-8 pattern in the horizontal plane. We investigated the structure – property – processing correlation of graphene bioscaffolds produced using three different methods. Bioscaffolds were prepared by chemical vapor deposition (CVD), sublimation of Silicon Carbide (SiC), and printed solvent assisted exfoliated graphene ink. To gain insight into the roughness and topography of graphene, AFM was performed on each bioscaffold. Raman spectroscopy mapping demonstrated differences in the I2D/IG ratio for each scaffold. Young’s modulus was determined by nanoindentation and indicated that epitaxial graphene had the highest average stiffness, followed by CVD, with printed graphene demonstrating the lowest average stiffness. Duchenne muscular dystrophy (DMD) is an X-linked muscle wasting disease caused by mutations in the Dmd gene resulting in non-functional dystrophin. Adeno-associated virus (AAV)-mediated delivery of classical CRISPR systems resulting in double-stranded breaks (DSBs) has yielded promise as a curative DMD therapeutic. However, the consequences of AAV-CRISPR require more thorough characterization. PCR-enriched short-read amplicon sequencing at cut sites is the most common method of sequencing CRISPR editing, but PCR bias results in unreliable quantification of on-target edits. We analyze orthogonal long- and short-read sequencing approaches to screen the outcomes of CRISPR technologies at the DNA and RNA-level for DMD. We …
