Two primer sets were manufactured from the cytadhesin P1 gene of M. Pneumoniae in an attempt to amplify homologous sequences in M. mycoides subsp. mycoides "SC" Type in a search for a P1-like gene in the latter organism. The first primer set MP-2 amplifies a 488 bp fragment in M. pneumoniae. The second primer set MP-3 amplifies a 303 bp fragment bracketing the 13-amino acid binding domain in the same organism. Both primer sets amplified the expected fragments in all the virulent strains of M. mycoides subsp. mycoides "SC" type. The primer sets did not however amplify the expected fragments …

Listeria monocytogenes has the potential to be a serious pathogen for the compromised health segment of our population (immunocompromised, young, elderly and the pregnant). While the infectious dose is generally believed to be high 1.0 $\times$ 10$\sp6$, infectious dosages of 1.0 $\times$ 10$\sp3$ have been reported. Listeria monocytogenes poses a threat because of several factors: ubiquity in nature, isolation from fresh/salt waters, soil, ruminates and 17% of the human population. Listeria monocytogenes can colonize surfaces within 20 minutes, evade sanitizing, and is capable of growth from 37$\sp\circ$C to below refrigeration temperature. The present research was performed to characterize immunoreactive (ir) pituitary cells in the equine species. Two experiments were performed to measure the prevalence of single- and double-hormone labeled gonadotropes, lactotropes, somatotropes, and mammosomatotropes. In Experiments 1 and 2, pituitaries from three intact pony mares were processed for ultrastructural evaluation of cells stained for luteinizing hormone (LH), follicle-stimulating hormone (FSH), prolactin (PRL), and growth hormone (GH). Primary antibodies and gold-conjugated antibodies were used to double-stain ir-cells. Dairy Herd Improvement Records As Replacement Of Technician Breeding Receipt Database For Routine Estimates Of Non-Return Rates For Al Bulls In Technician-Devoid Areas., Rick A. Baron

The objective of this study was to find whether DHI data could be used to estimate sire non-return rates to replace current technician data estimates. Bull weighted least squares means for non-return rates were calculated separately for five overlapping 60-90 day service periods from each data source. Models included stud, sire, service number, and linear and quadratic form of breeding month for both data sources, service unit for technician and lactation for DHI data. Sire and lactation were not significant $(P > .10).$ Technician differences $(P < .05)$ were in service unit, stud, service number, and linear and quadratic service month in all but one service period. DHI differed $(P < .05)$ for service number, month (linear and quadratic), and stud in two service periods. Technician R-square values were 0.23 to 0.28 versus 0.94 to 0.96 for DHI. Sire estimated non-return rates were weighted using the inverted estimator standard error squared and compared. Sire, stud, data source, service period, and appropriate interactions were modeled. Weighted bull non-returns differed $(P < .01)$ in magnitude across data sources. Stud, data source by stud, and sire within stud by data source were significant $(P .10).$ Four fertility categories based on mean and standard deviation of the weighted estimates were formed within the data sources across service periods. These categories were correlated (0.5 $>$ r $>$ 0.9) and 52.9 to 87.4% congruent within data source for adjacent …

A monoclonal antibody (MAb)-based western blot analysis (WB) was developed for detection of a 26-28 kD coproantigen in Fasciola hepatica infected cattle. Using WB the coproantigen was detected in feces from experimentally infected calves with 22 flukes or more. Using the biotin-streptavidin modification of WB, antigen was detected in the feces from experimentally-infected calves with 10 flukes or more as early as 6 weeks post-infection. The MAbs did not react with the ES of Paramphistomum sp. or Moniezia sp, but showed slight cross-reactivity with ES from F. gigantica, and there was moderate cross-reactivity with Fascioloides magna ES. This report characterizes several aspects of water allocation tradeoffs between fish species listed under the Federal Endangered Species Act and agriculture in the American West. The geographic intersection between endangered/threatened (E/T) fish and agricultural production reliant on surface water for irrigation is identified. Three findings are: (1) 235 counties, representing 22 percent of the West's counties, contain irrigated production that relies on water from rivers with E/T fish, (2) areas generating the highest revenues per acre from crop production are those most dependent on surface water irrigation, and (3) these same areas are also most likely to be drawing water …

A simulation/optimization (S/0) model to aid managing multiobjective wastewater loading to streams while maintaining adequate downstream water quality is presented. The conflicting objectives are to maximize the human and dairy cattle populations from which treated wastewater can be discharged to the river system. Nonindustrial municipal (domestic) wastewater undergoes primary and secondary treatment by a sewage treatment plant (STP) before entering as a steady point source. Dairy wastewater is treated by overland flow (OLF) land treatment before entering the stream as a controlled steady diffuse source. Farmers' preferences are important in the development of farm bills. In that spirit, the Department of Agricultural Economics at the University of Nebraska-Lincoln and the Nebraska Agricultural Statistics Service recently asked a random sample of agricultural producers about their preferences for the 1995 farm bill. Similar surveys were undertaken in 1984 and 1989, prior to the last two farm bills. Each Nebraska survey was part of a larger, national effort. This year, 15 states, scattered across all regions of the United States, participated.

Most quantitative investigations of spermatogenesis requites identification of stages of the cycle of the seminiferous epithelium. The current "tubular morphology" system of staging for the rabbit permits identification of only eight stages and is not applicable to all experimental conditions. A more advantageous staging system, based on changes in the acrosome and morphology of the developing spermatids, had not been developed for the rabbit. Knowledge of spermatogonial kinetics, including the identity of spermatogonial subtypes and the number and timing of spermatogonial divisions, is also important, but had not been fully elucidated in this species. This Extension Circular reviews what is known about the effects of rotations, tillage practices, and planting and harvest dates on crop insect management, focusing on major crops in Nebraska.

Before the development of synthetic organic insecticides (pre-DDT era), rotations, tillage practices, planting and harvest dates, and other nonchemical cultural controls were commonly recommended for insect management. Research focused on crop rotation and other cultural practices for insect management. With the development of DDT and later insecticides however, research on nonchemical controls decreased markedly.

This publication describes the most common types of ventilation fans and compares their physical and performance characteristics.

Two general types of ventilation fans are commonly used for agricultural applications. Each type has many different blade diameters, materials, blade shapes, speeds, and motor horsepowers available. The desired airflow rate and the resistance to airflow through the ventilation system, or static pressure capabilities, are the two primary considerations when selecting a ventilation fan. Other considerations include maintenance requirements, noise levels, and energy efficiency.

The purpose of this study was to describe extension agricultural agents' perceptions of sustainable agriculture in the Southern Region of the United States. The population included Cooperative Extension county/parish agents with major responsibility in agriculture working in the 13 states of the Southern Region of the United States. These individuals were either employed by the thirteen institutions established by the Morrill Act of 1862 (the 1862 Cooperative Extension Services) or by eleven institutions established by the Second Morrill Act of 1890 (the 1890 Cooperative Extension Services). Flooding is a common natural stress occurring in wetland/bottomland forests. Submergence, total immersion, is a special case of flooding, which has received limited attention in the literature. This study provides information on the impacts of three submergence treatments on three bottomland tree species. The results indicated that submergence would inhibit shoot and root growth for the three tested species during the submergence period. However, the survival of these species under submergence conditions were different. Taxodium distichum (L.) Rich survived even after 30 days of water coverage and survival continued to be high during the recovery period. Quercus nuttallii Palmer was …

Mechanisms Of Resistance To Aflatoxin Accumulation By Aspergillus Flavus In Maize Genotype Gt-Mas:Gk., Baozhu Guo

Maize genotype differences in aflatoxin production were observed when kernels of resistant GT-MAS:gk and thirteen susceptible commercial hybrids were inoculated with Aspergillus flavus using a laboratory kernel screening assay. GT-MAS:gk supported the lowest levels of aflatoxin in both intact and endosperm-wounded kernels. Treating intact kernels with KOH effected substantial increases in aflatoxin accumulation in GT-MAS:gk, but only marginal increases in Pioneer 3154. Removing wax from the surface of GT-MAS:gk kernels greatly increased aflatoxin accumulation. These results indicated that GT-MAS:gk resistance was associated with an intact pericarp (wax and cutin layers), acting as a physical barrier, along with internal biochemical factors. …

Reactions of soil arsenic with arsenic addition and the effects of soil arsenic on canola were studied because of a lack of information on this subject. The reactions of different pools of soil arsenic to arsenic addition were studied. The effects of soil arsenic and arsenic form and concentration in solution on canola growth and nutrient uptake were also investigated and an attempt to model arsenic uptake with a mechanistic computer model was made. In a solution study, rate of inorganic arsenic did not appear to effect arsenic accumulation in roots and shoots of canola. However, shoot and root arsenic …

Automated analyzers have revolutionized diagnostic hematology in mammalian species. These commercial instruments utilize flow cytometric technology to enumerate blood cell concentrations. Because of the nuclei present in most non-mammalian erythrocytes and thrombocytes, these instruments are unable to calculate leukocyte counts in birds, amphibians, reptiles, and fish. These investigations sought to determine if three commonly used methodologies in flow cytometry could sufficiently differentiate avian erythrocytes, leukocytes, and thrombocytes, and ultimately form a basis for performing total leukocyte counts. The effects of extrusion temperature and post extrusion holding time, microwave heating, and gamma irradiation on antioxidants and lipid composition in rice bran were studied. Post extrusion holding time had no apparent effect on rice bran stability. Increased holding times reduced (p $<$ 0.05) total vitamin E. Oryzanol concentration was lower (p $<$ 0.05) only with 6 min holding time. Inactivation of lipolytic enzymes was obtained by extrusion temperatures of 120, 130, and 140$\sp\circ$C. Increased extrusion temperatures reduced the retention of vitamin E and oryzanol during storage. An extrusion temperature of 110$\sp\circ$C provided the highest retention of vitamin E and oryzanol during storage although FFA level was higher (p $<$ 0.05) than that of the other extrusion temperatures from 105 to 375 days. There were no significant (p $>$ 0.05) decreases in individual vitamin E vitamers until 4 min microwave heating. After 8 min heating, individual E vitamers decreased significantly (p $$ 0.05) in FFA level by 24 weeks of storage with microwave heating for 12 min. Microwave heating for 4, 8, and 12 min did not provided adequate inactivation of lipolytic enzymes. Total vitamin …

The ability of uterine isolates of Streptococcus zooepidemicus to resist phagocytosis was explored by the organism's ability to grow in equine fresh blood. Although isolates grew in fresh blood of many horses, any given isolate was killed by blood of certain horses. Killing required leukocytes, heat labile and heat stable components of plasma and appeared to follow phagocytosis of the organism by neutrophils. This led to the working hypothesis that killing was mediated by phagocytosis requiring complement and isolate-specific antibody to variable surface antigens. Red drum juveniles of mean initial weight 9.23 $\pm$ 0.27 g were kept in indoor aquaria. The water was maintained at a salinity of 12 $\pm$ 0.5 ppt. The fish were fed six isonitrogenous (38% crude protein) and isocaloric (1.36 $\times$ 10$\sp4$ kJ/kg), purified diets differing in lipid content, for nine weeks, to investigate the effects of selected dietary essential fatty acids (EFA) on growth performance and tissue fatty acid composition.
