BACKGROUND: The global prevalence of obesity, an established risk and progression factor for colon cancer, is high and rising. Unfortunately, the mechanisms underlying the obesity-colon cancer association are incompletely understood, and new molecular targets enabling more effective intervention strategies to break the obesity-colon cancer link are urgently needed.

OBJECTIVES: This study integrated RNA sequencing data from mouse and human colon tumor samples, as well as human adipose samples, to rigorously establish obesity-associated transcriptomic signatures conserved between the 2 species.

Trauma impacts the life experiences of a sizable portion of the population. Given that so many clients bring trauma into counseling, even when not the presenting issue, knowledge of trauma’s impact and proficiency with trauma treatment are essential topics in counselor education. This study gave voice to counselors in the field to share what they have found effective from working with clients with trauma. The researcher used a phenomenological approach and interviewed six self-identified trauma-specialized counselors who have practiced for five years or longer. Dissolved organic matter (DOM) is a major reservoir of organic carbon in aquatic ecosystems and plays a central role in global carbon cycling through its transformation and remineralization to carbon dioxide (CO₂). As DOM moves from terrestrial environments into streams and rivers, its chemical composition is altered by a combination of microbial and photochemical oxidation processes that regulate both its reactivity and its persistence during transport. Calibration, or students’ ability to predict and postdict their performance on an academic task, has important implications for academic performance. Oceanographic lidar systems remotely characterize the vertical structure of the upper ocean by recording the light backscattered from a laser pulse as it propagates through the water column. Historically these systems have been constrained by limited detection ranges, often capturing only a portion of the illuminated water column, typically fading to noise around the 10% isolume. A primary constraint has been the limited dynamic range of digitizers, which inhibits the ability to resolve both intense near-field and faint far-field backscatter signals within a single acquisition. Family systems theories contextualize violence as occurring across multiple levels of the family, yet much of the existing literature focuses on a single family member’s outcome (i.e., the child) and neglects to include data collected from fathers. This study examined the associations between indicators of functioning at the dyadic-level in the family system—observed parenting, Intimate Partner Violence (IPV) perpetration, and Parent-Child Aggression (PCA) risk—and later overall family dysfunction. In the past decade, a surge in the amount of electronic health record (EHR) data in the United States occurred, driven by a favorable policy environment created by the Health Information Technology for Economic and Clinical Health (HITECH) Act of 2009 and the 21st Century Cures Act of 2016. Clinical notes for patients’ assessments, diagnoses, and treatments are captured in these EHRs in free-form text by physicians, who spend a considerable amount of time entering them. Military sexual trauma (MST) amongst veterans is a growing concern for the Veterans Health Administration (VHA). Veterans who screen positive for MST receive VHA care for any injuries caused as a result of MST, psychological or physical, free of charge. MST is difficult to track, can result in long-term disability, and the burden of illness varies across genders. BACKGROUND: Intellectual developmental disorder with autism and macrocephaly (IDDAM, OMIM #615032) is an autosomal dominant neurodevelopmental disorder characterized primarily by intellectual disability, autism spectrum disorder, macrocephaly, tall stature, gastrointestinal symptoms, and variable neurological manifestations. Most cases result from de novo pathogenic variants in CHD8.

METHODS: We conducted genome sequencing through the Undiagnosed Diseases Network (UDN) in a female proband harboring a CHD8 variant of uncertain significance (VUS), whose clinical presentation was consistent with IDDAM but included atypical features such as ptosis and hearing loss. Variant pathogenicity was further evaluated using EpiSign DNA methylation analysis and structural biology modeling.

BACKGROUND: Age is the strongest factor determining disease expression in multiple sclerosis (MS). We previously demonstrated biological age acceleration in pediatric-onset MS (POMS) compared to controls with both epigenetic clocks (DNAm) and telomere length (TL). It is unknown whether these markers report overlapping or distinct aging-related processes.

BACKGROUND: Cancer-related fatigue (fatigue) is a common and persistent symptom after cancer treatment, yet the role of genetic susceptibility remains unclear.

METHODS: We used data from a prospective cohort study called the ColoCare Study, conducted over 5 US sites and Germany. OBJECTIVE: We sought to evaluate the relationship between neighborhood-level deprivation and perinatal outcomes, particularly preterm birth, as well as COVID-19 outcomes among pregnant patients during the early pandemic.

METHODS: We performed a secondary analysis of a prospective longitudinal cohort study that investigated the impact of prenatal COVID-19 exposure on perinatal outcomes. Pregnant patients were recruited 12/23/20-7/18/22 and serially assessed for COVID-19 exposure during pregnancy, with serum antibody testing, electronic medical record review, and patient report. The expansion of solar photovoltaic energy infrastructure is transforming landscapes worldwide. Increasingly, facilities are planted with native vegetation, yet how vegetation characteristics and landscape variables shape wildlife communities in solar facilities remains poorly understood. Because avian populations have been in decline and are sensitive to habitat changes, understanding how solar facilities influence avian communities is important. The Mississippi Alluvial Plain (MAP) of Arkansas is a landscape where many wetlands have been altered for use as aquaculture ponds or agricultural ditches. Commercial harvest of freshwater turtles within the MAP is not restricted or limited, with reported harvest numbers for 2019 alone exceeding 4000 for spiny softshell turtles (Apalone spinifera) and 39,000 for red-eared sliders (Trachemys scripta elegans). Herein, we attempt to provide baseline estimates of freshwater turtle densities and community composition in aquaculture ponds and agricultural ditches of eastern Arkansas, the habitat types most frequently trapped by commercial harvesters. Cerebral small vessel disease (cSVD) is a leading cause of stroke and dementia with no specific treatment, of which molecular mechanisms remain poorly understood. To identify potential biomarkers and therapeutic targets, we applied Mendelian randomization to examine over 2,500 proteins measured in plasma and, uniquely, cerebrospinal fluid, in relation to magnetic resonance imaging (MRI) markers of cSVD in more than 40,000 individuals. Here we show that 49 proteins are associated with MRI markers of cSVD, most prominently in cerebrospinal fluid. BACKGROUND: The answer to whether females or males have better health, and which sex is the more disadvantaged, has depended in part on the metric and how the inequality is measured. This study introduces a new method for analyzing and interpreting sex inequalities in health outcomes-defined as the avoidable sex differences in health outcomes-that is systematic and potentially more objective. Background: We evaluated whether a large language model could assist in selecting psychopharmacological treatments for adults with treatment-resistant depression. Methods: We generated 20 clinical vignettes reflecting treatment-resistant depression among adults based on distributions drawn from electronic health records. Each vignette was evaluated by 2 expert psychopharmacologists to determine and rank the 5 best next-step pharmacologic interventions, as well as contraindicated or poor next-step treatments. Vignettes were then presented in random order, permuting gender and race, to a large language model (Qwen 2.5:7B), augmented with a synopsis of published treatment guidelines. Purpose of Review: To discuss the biological mechanisms of cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitors, the rationale for using CDK4/6 inhibitors in estrogen receptor-positive (ER+) and human epidermal growth factor receptor-2-negative (HER2-) breast cancer (BC), data from landmark phase III clinical trials that led to U.S. Food and Drug Administration (FDA) approval, and updates on recently published and ongoing clinical trials. Intimate partner violence (IPV) is a global public health issue impacting millions of people each year. Black women constitute 14% of the US population but alarmingly comprise 31% of homicides resulting from IPV. African American women survivors of IPV experience intersecting impacts of racism, sexism, and historical mistrust. This research aimed to raise awareness about their experiences of violence and perceptions of institutional support.

Reforestation is a prominent climate change mitigation strategy, but available global maps of reforestation potential are widely criticized and highly variable, which limits their ability to provide robust estimates of both the locations and total area of opportunity. Here we develop global maps that address common critiques, build on a review of 89 reforestation maps created at multiple scales, and present eight reforestation scenarios with varying objectives, including providing ecosystem services, minimizing social conflicts, and delivering government policies. Zoonotic malaria risk at human-wildlife-environment interfaces requires surveillance that integrates signals from reservoirs, vectors and the environment. We coupled a drone-based environmental DNA (eDNA) canopy swabbing approach with portable quantitative PCR (qPCR) to detect Plasmodium DNA in situ during a 24-h field exercise in the Amazon rainforest. Drone-lowered sterile swabs into the canopy, which were then extracted and subjected to a multiplex pan-Plasmodium assay targeting five human-infecting Plasmodium species (limit of detection 0.2 parasites μL−1). Batten disease is characterized by early-onset blindness, juvenile dementia and death within the second decade of life. The most common genetic cause are mutations in CLN3, encoding a lysosomal protein. Currently, no therapies targeting disease progression are available, largely because its molecular mechanisms remain poorly understood. To understand how CLN3 loss affects cellular signaling, we generated human CLN3 knock-out cells (CLN3-KO) and performed RNA-seq analysis. Our multi-dimensional analysis reveals the transcriptional regulator YAP1 as a key factor in remodeling the transcriptome in CLN3-KO cells. Purpose: Implanted microchips are becoming increasingly common in research for animal identification and have been adopted by commercial vendors for some mouse strains. However, they often contain metal components, which generate magnetic susceptibility artifacts on MR images. Despite this, some microchips are marketed as MR-compatible, even though they are likely to affect image quality.

Methods: We assessed the impact of a radiofrequency identification microchip on MR images of the mouse brain and present a method for precise surgical removal. A handheld magnet was used to locate and stabilize the microchips during removal. Mice were imaged before and after microchip removal. …

Acetaminophen (APAP) overdose is associated with increased transforming growth factor beta 1 (TGFβ1) signaling and elevated oxidative stress, which exacerbate DNA damage. TGFβ1 has been shown to regulate ataxia-telangiectasia mutated (ATM) signaling and DNA repair in other cell types. This study investigates the DNA damage response (DDR) during APAP-induced liver injury, focusing on ATM-mediated regulation of TGFβ1 signaling. APAP administration in vitro and in vivo resulted in DNA damage, increased ATM signaling, accumulation of γH2AX, and activation of phosphorylated ataxia telangiectasia mutated (pATM) and phosphorylated checkpoint kinase 2 (pChk2). Objective: This preliminary study aimed to identify microRNA (miRNA) signatures associated with bipolar disorder (BD) by profiling blood-derived extracellular vesicles (EVs) of both putative neuronal origin and from all sources.

Method: In two parallel studies of individuals with BD and controls (CON), we characterized miRNA expression profiles of blood EVs selected for L1CAM, a putative marker of neuronal origin (n = 20 BD/20 CON), as well as bulk EVs (n = 21 BD/20 CON). For each study, analyses identified miRNAs differentially expressed between groups, followed by functional interrogation and testing for associations with clinical features.
