External stressors modulate the oligomerization state of photosystem I (PSI) in cyanobacteria. The number of red chlorophylls (Chls), pigments lower in energy than the P₇₀₀ reaction center, depends on the oligomerization state of PSI. Here, we use ultrafast transient absorption spectroscopy to interrogate the effective connectivity of the red Chls in excitonic energy pathways in trimeric PSI in native thylakoid membranes of the model cyanobacterium Synechocystis sp. PCC 6803, including emergent dynamics, as red Chls increase in number and proximity. Fluence-dependent dynamics indicate singlet–singlet annihilation within energetically connected red Chl sites in the PSI antenna but not within bulk Chl …

Salmonella enterica serovar Typhimurium is a facultative intracellular pathogen that utilizes its type III secretion systems (T3SSs) to inject virulence factors into host cells and colonize the host. In turn, a subset of cytosolic immune receptors respond to T3SS ligands by forming multimeric signaling complexes called inflammasomes, which activate caspases that induce interleukin-1 (IL-1) family cytokine release and an inflammatory form of cell death called pyroptosis. Human macrophages mount a multifaceted inflammasome response to Salmonella infection that ultimately restricts intracellular bacterial replication. However, how inflammasomes restrict Salmonella replication remains unknown. We find that caspase-1 is essential for mediating inflammasome responses …

A Study To Identify The Causal Rare Genetic Variants In Primary Open And Closed Angle Glaucoma, Pseudoexfoliation Syndrome, And Associated Glaucoma, Suganya K

Theses and Dissertations

BACKGROUND: Glaucoma is the second most common cause of blindness globally typically diagnosed with a triad of clinical symptoms of increased intraocular pressure (IOP) with associated optic disc, optic nerve head (ONH) changes, and visual field defects. Genetic and environmental factors are some of the strong aetiology factors for glaucoma and identification of these factors has a potential implication in the management of the disease and its outcome. There is a paradigm shift towards understanding the genetics of glaucoma, wherein the variants in the nuclear, mitochondrial genome and other regulatory regions are being identified as contributing risk factors.

Induced Mutagenesis In West Nile Virus Non-Structural Protein 3 With A Discussion Of The Mutation's Effects On Viral Properties, Caden Connealy

Honors Program: Senior Projects (Public)

West Nile Virus (WNV), a member of the Flaviviridae family and introduced to the Western hemisphere in 1999, presents a unique and emerging threat to human health. Infection in humans is often mild but can occasionally result in severe neurological impairment and even death. West Nile Virus non-structural protein 3 (NS3) plays an important role in the process of viral genome amplification. More specifically, the C-terminal domain of the protein folds into a helicase enzyme, which is responsible for the “unwinding” of double-stranded RNA intermediates and RNA secondary structure on the WNV genome. This is a vitally important step in …

Department of Biochemistry and Molecular Biology Faculty Papers

Maackia amurensis lectins, including MASL, MAA, and MAL2, are widely utilized in biochemical and medicinal research. However, the structural and functional differences between these lectins have not been defined. Here, we present a high-resolution cryo-EM structure of MASL revealing that its tetrameric assembly is directed by two intersubunit disulfide bridges. These bridges, formed by C272 residues, are central to the dimer-of-dimers assembly of a MASL tetramer. This cryo-EM structure also identifies residues involved in stabilizing the dimer interface, multiple glycosylation sites, and calcium and manganese atoms in the sugar-binding pockets of MASL. Notably, our analysis reveals that Y250 in the …

Soft Robotic Brittle Star Shows The Influence Of Mass Distribution On Underwater Walking, Zach J. Patterson Mar 2025

Soft Robotic Brittle Star Shows The Influence Of Mass Distribution On Underwater Walking, Zach J. Patterson

Faculty Scholarship

Most walking organisms tend to have relatively light limbs and heavy bodies in order to facilitate rapid limb motion. However, the limbs of brittle stars (Class Ophiuroidea) are primarily comprised of dense skeletal elements, with potentially much higher mass and density compared to the body disk. To date, little is understood about how the relatively unique distribution of mass in these animals influences their locomotion. In this work, we use a brittle star inspired soft robot and computational modeling to examine how the distribution of mass and density in brittle stars affects their movement. The soft robot is fully untethered, …

Identifying Herbicides To Manage Weeds In A Buckwheat Cover Crop And For The Control Of Volunteers, Taylor Randell-Singleton, Hannah E. Wright-Smith, Lavesta C. Hand, Jenna C. Vance, A. Stanley Culpepper

Horticulture Faculty Publications and Presentations

Buckwheat (Fagopyrum esculentum) is a fast-growing plant that quickly produces a dense groundcover. The utilization of buckwheat as a cover crop in vegetable production could be beneficial to Georgia producers, however for successful adoption, weed control in the cover crop coupled with control of buckwheat volunteers must be achievable. A preemergence (PRE) and a postemergence (POST) experiment were each conducted three times in Ty Ty, GA (2020-2021) addressing these objectives. In the PRE study, acetochlor at 0.56 lb ai acre(-1) injured buckwheat up to 16% without negatively influencing growth, suggesting potential for use in buckwheat for weed control. Barriers And Breakthroughs In The Evolution Of Aggregative Multicellularity, Israt Jahan Mar 2025

Barriers And Breakthroughs In The Evolution Of Aggregative Multicellularity, Israt Jahan

Arts & Sciences Graduate Student Theses and Dissertations

The evolution of multicellular organization represents a key innovation in the history of life, yet fundamental questions remain about the evolutionary mechanisms that allowed some lineages to achieve remarkable complexity, while constrained others in their elaboration. The widely accepted single-cell bottleneck model provides a compelling explanation for the success of clonal multicellularity by reducing internal conflict and facilitating developmental integration. However, this perspective does not fully account for the stability and repeated evolution of aggregative multicellularity, where cooperation emerges among previously independent cells that may not be genetic relatives. Aggregative multicellularity demonstrates that the evolution of multicellularity is not a …

Management Of Parkinson's Disease Psychosis: First-Line Antipsychotic Selection And Rationale For Continuing, Combining, Or Switching, Stuart H. Isaacson, Henry Nasrallah, Rajesh Pahwa, Gustavo Alva, Daniel Kremens, Stephen M. Stahl

Department of Neurology Faculty Papers

INTRODUCTION: The past decade has seen a paradigm shift in the evaluation and management of Parkinson's disease psychosis (PDP), with the first approval of an antipsychotic in the US in 2016. An evidence-based review by the Movement Disorder Society found pimavanserin and clozapine to be clinically useful, (low-dose) quetiapine to be possibly useful, and all other antipsychotics to be avoided due to motor worsening. Clozapine and quetiapine use can be limited by provoking Parkinson's disease (PD) nonmotor symptoms of somnolence and hypotension. Quetiapine may also be limited by its risk in cognitive impairment. Pimavanserin is not associated with these symptoms. …

Small mustelids are difficult to survey due to their low density and cryptic nature. Population status of North American weasels (Mustela erminea, Mustela nivalis, and Neogale frenata) are believed to be in decline, but there are no standardized monitoring protocols to evaluate their status. To support weasel monitoring, we compared the attractiveness of various combinations of baits and lures to weasels in sites located throughout the eastern and central USA. We baited a total of 122 clusters of 4 camera traps, across 14 states, with random combinations of 4 baits and 3 scent lures in the winters of 2022 and …

The Prideful Ass: Using Augustine's Neoplatonic Theory Of Sin To Investigate Curiosity In Apuleius' Golden Ass, Danielle E. Booth

USF Tampa Graduate Theses and Dissertations

Ancient discourse about the beneficial yet predominantly dangerous role of curiosity inattaining knowledge was prevalent in the Roman Empire around the 2nd century C.E. when the North African author, Apuleius, composed his Latin novel, the Golden Ass. Although there is no shortage of modern scholarship that focuses on how Apuleius dramatizes obsessive intrigue by means of his protagonist, Lucius, contemporary publications seldom reorient the discussion to consider how unclear he is about the unstated source of curiosity, as I do in the following thesis. Approaching this topic through the context of ancient Neoplatonism, I argue Augustine of Hippo – a …

Awful, Yet Awe-Full: The Sublime Dystopia Of Blade Runner 2049, Charles E. Butt

USF Tampa Graduate Theses and Dissertations

Blockbuster dystopian sci-fi features stories of environmental collapse, authoritarian governments, out of control corporations, unbounded consumerism, and other awful possibilities. Scholars of science fiction broadly agree that this genre explores collective fears, anxieties, and speculations about the present and future. At the same time, film scholars have established that dystopian sci-fi cinema is compelling, and audiences respond to their effects-driven visuals, photorealistic mise-en-scene, and dramatic narratives. This creates an apparent paradox: Blockbuster cinematic aesthetics portray undesirable worlds desirably. I resolve this paradox by arguing that the worldbuilding of spectacle-driven, dystopian sci-fi cinema is Romanticized. That is, the provocative style of …

Computational Methods And Optimization For Naturalistic Functional Brain Mapping Using Diffuse Optical Tomography, Morgan Fogarty Mar 2025

Computational Methods And Optimization For Naturalistic Functional Brain Mapping Using Diffuse Optical Tomography, Morgan Fogarty

McKelvey School of Engineering Graduate Student Theses & Dissertations

Naturalistic neuroimaging has significantly advanced our understanding of the brain in everyday situations. Stimulus designs that previously targeted individual brain regions are now being replaced by activities such as watching movies, engaging in conversations, and interacting with virtual reality environments. However, traditional functional neuroimaging techniques like functional magnetic resonance imaging (fMRI) are inherently unnaturalistic, as participants must lay down and remain still within the magnet's bore. Consequently, these naturalistic paradigms require an equally naturalistic neuroimaging modality. High-density diffuse optical tomography (HD-DOT) offers a wearable alternative to fMRI by utilizing overlapping optical measurements to densely sample cortical brain activity. HD-DOT has …
