Deep learning driven computational methods have profoundly impacted medical imaging by advancing feature extraction, and by establishing a critical foundation for downstream image analysis and interpretation. Co-clinical trials are an emerging area of investigation in which a clinical trial is coupled with a corresponding preclinical study to inform the corresponding clinical trial to enhance translational accuracy and therapeutic development. Within this framework, quantitative imaging and pathology plays a fundamental role in both clinical and preclinical arms of co-clinical trials. Influenza A virus (IAV) poses substantial epidemiological burdens around the world, with an estimated annual global mortality rate in the hundreds of thousands. Its rapid rate of mutation gives it a propensity for causing seasonal epidemics and occasional pandemics. As an airborne virus, IAV is most commonly transmitted through droplets and aerosols. Antibodies play a crucial role in the immune defense against IAV infection. By understanding the mechanisms of antibody-mediated defense and the transmission dynamics of IAV, we can better inform strategies for prevention, vaccination, and treatment, ultimately mitigating its impact on public health. Low back pain (LBP) is the number one musculoskeletal cause of disability with a lifetime prevalence of up to 85%. The causes of LBP are multifactorial with potential contributions from factors such as tissue injury and degeneration as well as social economic and psychological factors. Intervertebral disc (IVD) degeneration is associated with 26-40% of LBP cases and in a 2015 meta-analysis, it was estimated that a total 403 million people suffer from symptomatic IVD degeneration. Chondrocytes, the only native cell population of cartilage, utilize mechanosensitive transmembrane ion channels to sense mechanical forces. Transient Receptor Potential Vanilloid 4 (TRPV4) is activated by physiologic levels of load (resulting in ~10% tissue deformation) while PIEZO1 is activated by supraphysiologic levels of load (resulting in >50% tissue deformation). TRPV4 activation promotes the expression of a limited number of chondrogenic genes and the production of vital cartilaginous matrix proteins, while PIEZO1 activation drives cell death and senescence, cytoskeletal weakening, and production of catabolic mediators, which contribute to the development of osteoarthritis (OA). Advancements in non-invasive imaging have enabled extensive research into higher-order cognitive functions of the brain. Functional Magnetic Resonance Imaging (fMRI) has provided valuable insights into brain networks underlying various cognitive processes. Lignin is a complex, random, and heterogeneous biopolymer in lignocellulosic biomass. It has significant potential for conversion into valuable chemicals and fuels. However, its natural recalcitrant structure has made selective depolymerization challenging. This dissertation presents a detailed investigation into the mechanisms of lignin disassembly and structural evolution under various catalytic and solvent conditions. The research spans three key studies. In Chapter 2, operando magic-angle spinning (MAS) solid-state nuclear magnetic resonance (NMR) spectroscopy was employed to study reaction pathways and kinetics of lignin model polymers during catalytic hydrogenolysis. The landscape of spatial biology research has transformed drastically in recent years allowing a comprehensive understanding of cellular organization and molecular mechanisms in various biological context. This thesis presents three complementary studies that advance our understanding of spatial organization in normal tissue biology and tumor evolution, utilizing cutting-edge spatial multi-omics approaches. The first study presents a comprehensive investigation of kidney sexual dimorphism across the lifespan, integrating six distinct technological platforms: single-nucleus transcriptomics, epigenomics, Visium spatial transcriptomics, Xenium single-cell resolution spatial transcriptomics, conventional immunofluorescence imaging, and co-detection by indexing (CODEX) multiplexed protein imaging. As single-cell omics data sampling and acquisition methods have accumulated at an unprecedented rate, various data analysis pipelines have been developed for the inference of cell types, cell states and their distribution, state transitions, state trajectories, and state interactions. This presents a new opportunity in which single-cell omics data can be utilized to generate high-resolution, high-fidelity computational models. Introduction: Primary Care Providers (PCPs) are seeing growing number of patients presenting for adult attention-deficit/hyperactivity disorder (ADHD) evaluations as a result of social media influences highlighting ADHD. Yet, PCPs report lacking confidence and education with adult ADHD. The purpose of this quality improvement study was to develop educational materials to promote confidence, competence, and adherence to ADHD evidence-based recommendations among integrated care PCPs. The self-disorder model provides a phenomenological framework for understanding how the core symptoms of Schizophrenia Spectrum Disorders (SSDs) are rooted in an instability of minimal selfhood. This instability involves a range of ‘anomalous experiences’: transformations in an individual’s perceptual field and sense of being an agent of action. The explanatory value of this theoretical model can be summarized in two claims about the role of anomalous experiences in self-disorders: (1) anomalous experiences express a common trait-like disturbance that is characteristic of SSDs, and (2) anomalous experiences contribute to the progression of self-disorders into SSDs. The Houge type of X-linked syndromic intellectual developmental disorder (MRXSHG) encompasses a spectrum of neurodevelopmental disorders characterized by intellectual disability (ID), language/speech delay, attention issues, and epilepsy. These conditions arise from hemizygous or heterozygous deletions, along with point mutations, affecting CNKSR2, a gene located at Xp22.12. CNKSR2, also known as CNK2 or MAGUIN, functions as a synaptic scaffolding molecule within the neuronal postsynaptic density (PSD) of the central nervous system. Obesity and type 2 diabetes mellitus (T2DM) are chronic diseases with increasing prevalence, underscoring the urgent need for effective treatment and management strategies. Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) have emerged as an essential class of drugs for managing both obesity and T2DM, offering additional benefits for cardiovascular and kidney health. Twenty-first-century education as a powerful means of knowledge will possibly be shaped by the emphasis it places on having classrooms that are race-conscious and multicultural. The present article is based on a doctoral study that explores an under-researched area: experiences and understanding of multiculturalism in mainstream primary schools situated in the predominantly White places of Southwest England. The study was underpinned by a sociocultural theoretical framework; data were collected through semi-structured interviews with the adult participants, students’ classroom activities, and documentary analysis of school displays. Deviation from protocolized assessment times is commonplace in pragmatic randomized clinical trials. Working with a stakeholder advisory board for a Patient-Centered Outcomes Research Institute®-funded project on statistical methods for handling potential biases introduced by irregular assessment times, we identified reasons for off-schedule or missed assessments. We used the Consolidated Framework for Implementation Research 2.0 to organize our findings. We conjectured that timely completion of outcome assessments is a function of multiple determinants, only some related to participants' health status. Inflammatory bowel disease (IBD) is a chronic condition that affects about 7 million people worldwide, and new therapies are needed. Understanding the complex roles that bile acids (BAs) play in IBD may lead to the development of novel IBD treatments independent of direct immunosuppression. This review discusses the latest discoveries in the roles BAs play in IBD pathogenesis and explores how these discoveries offer promising new therapeutic targets to treat IBD and improve patient outcomes. The maintenance of homeostasis and rapid regeneration of the urothelium following stress are critical for bladder function. Here, we identify a key role for IFRD1 in maintaining urothelial homeostasis in a mouse model. We demonstrate that the murine bladder expresses IFRD1 at homeostasis, particularly in the urothelium, and its loss alters the global transcriptome with significant accumulation of endolysosomes and dysregulated uroplakin expression pattern. We show that IFRD1 interacts with mRNA-translation-regulating factors in human urothelial cells. Loss of Ifrd1 leads to disrupted proteostasis, enhanced endoplasmic reticulum (ER stress) with activation of the PERK arm of the unfolded protein response pathway, …

Many viral proteins form biomolecular condensates via liquid-liquid phase separation (LLPS) to support viral replication and evade host antiviral responses, and thus, they are potential targets for designing antivirals. In the case of nonenveloped positive-sense RNA viruses, forming such condensates for viral replication is unclear and less understood. Human noroviruses (HuNoVs) are positive-sense RNA viruses that cause epidemic and sporadic gastroenteritis worldwide. Here, we show that the RNA-dependent RNA polymerase (RdRp) of pandemic GII.4 HuNoV forms distinct condensates that exhibit all the signature properties of LLPS with sustained polymerase activity and the capability of recruiting components essential for viral replication. …

New awareness of re-thinking childhood in the emergent field of childhood studies inspires production of new discourse on social construction of childhood in literature. This paper looks at contemporary children’s literature in the Philippines from this perspective and within the context of the globalization and transnationalization of care giving. This article examines ten interviews with Polish feminist activists conducted by the Women’s Center “eFka” in Kraków and gathered by the Global Feminisms Project at the Institute for Research on Women and Gender at the University of Michigan in Ann Arbor. Recent AI-based quiz generation efforts have predominantly concentrated on static resources, like textbooks and slides, often overlooking the dynamic and interactive elements of live lectures—contextual cues, discussions, and interactions—that contribute to the learning experience. In this work, we propose a Retrieval-Augmented Generation (RAG) model that processes multimodal inputs by combining text, audio, and video to produce quizzes that capture a fuller context. This paper presents a phenomenological study that explores the embodied experience of vision loss within the context of eye disease, while also providing a detailed discussion of the methodological approaches utilized in the research. Phenomenology is integrated to address clinical epistemic concerns, particularly the limitations of conventional vision tests in fully capturing perception and the lived experiences of vision loss. This interdisciplinary approach builds on the clinical understanding of eye disease by incorporating the broader dimensions of the lifeworld that intersect with visual impairment in degenerative eye diseases.
