There is a need for affordable, scalable, and specific blood-based biomarkers for Alzheimer's disease that can be applied to a population level. We have developed and validated disease-specific cell-free transcriptomic blood-based biomarkers composed by a scalable number of transcripts that capture AD pathobiology even in the presymptomatic stages of the disease. Accuracies are in the range of the current CSF and plasma biomarkers, and specificities are high against other neurodegenerative diseases.
The Role Of Pre- And Perinatal Oxycodone Exposure On Postnatal Brain Development And Behavior: Assessing The Impact On Later Life Brain Injuries In Juvenile Rats, Austin M. Gowen
Theses & Dissertations
There has been a long history of research investigating the role of the gestational environment on later life health and disease. The progressive widespread illicit and prescribed use of opioids in the 21st century has seen opioids become one of the most significant gestational insults in the last century. The ability of opioids to pass both the placental barrier and blood-brain barrier makes them a particularly dangerous teratogen capable of altering the peripheral physiology, neurobiology, and behavior of affected children. The works herein investigate the impact of a widely prescribed opioid, oxycodone, on developmental neurobiology in the context of …
Inequality In Opportunities And Outcomes: Indicators To Inform Data-Driven Community Systems For Youth Physical Activity, Ann Essay
Theses & Dissertations
Indicators illuminating whole-of-community and setting system physical activity (PA) outcomes and drivers are needed to inform population health improvement efforts. This multi-study dissertation examines inequalities in PA resources, opportunities, and outcomes and the factors influencing inequalities across multiple temporal and geospatial scales (e.g., community, setting). First, a scoping review examined community-level variability in PA resource and opportunity availability by community sociodemographic characteristics (e.g., race). Across 46 included studies, 41 unique indicators characterizing community PA systems were identified, and 29.8% of relationships assessed indicated greater resource/opportunity availability in communities of higher socioeconomic status or lower minority population prevalence. The indicators synthesized …
Investigating And Targeting Iron Metabolism In Pseudomonas Aeruginosa And Klebsiella Pneumoniae, Zachary Scott
Investigating And Targeting Iron Metabolism In Pseudomonas Aeruginosa And Klebsiella Pneumoniae, Zachary Scott
Theses & Dissertations
Antimicrobial resistance is an incredibly pressing issue in modern medicine. Because of that many researchers are pursuing just as many avenues for treatment of these pathogens. However, due to the nature of bacteria many of these methods are stop gaps and the bacteria can develop typically within years of their deployment. Therefore, more robust methods of treating infections of antimicrobial resistant bacteria that are difficult or impossible to develop resistance to are required. To produce this effect the treatment would need to target and alter or inhibit functions or structures essential to the bacteria that could not be altered, additionally …
Examining Caf Heterogeneity In Pancreatic Cancer, Nidhi V. Dwivedi
Examining Caf Heterogeneity In Pancreatic Cancer, Nidhi V. Dwivedi
Theses & Dissertations
This thesis delves into the complexities of pancreatic ductal adenocarcinoma (PDAC) and its microenvironment. This study centers on unraveling the heterogeneity of pancreatic cancer-associated fibroblasts (CAFs) using a panel of patient-derived pancreatic fibroblasts. Additionally, it delves into the realm of G-protein-coupled receptors (GPCRs), with a specific emphasis on the Endothelin receptors (ETAR and ETBR), all within the context of PDAC.
A key achievement is the development of immortalized patient-derived pancreatic fibroblasts (iPDPFs), replicating CAF heterogeneity in PDAC. The iPDPFs were able to recapitulate the PDAC CAF heterogeneity, plasticity, and the overlapping nature of the tumor stroma. Functional characterization of iPDPFs …
Citrullinated And Malondialdehyde-Acetaldehyde Modified Proteins Promote Cellular Inflammatory And Fibrotic Responses Associated With Rheumatoid Arthritis, Nozima Aripova
Theses & Dissertations
Rheumatoid arthritis (RA) is a systemic, autoimmune disease primarily characterized by perpetual synovial inflammation that results in synovial tissue proliferation and fibrosis (pannus). Although joint involvement is the predominant manifestation of RA, extra-articular features, such as interstitial lung disease (ILD), contribute significantly to the elevated mortality associated with the condition. Post-translational modifications play a significant role in RA and RA-ILD pathogenesis. Circulating anti-citrullinated protein antibodies (ACPAs) are a highly diagnostic tool implicating a potential role for citrullinated antigens in RA and RA-ILD.
Recently, antibodies to malondialdehyde acetaldehyde adducts (MAA) have been markedly elevated in patients with RA and RA-ILD. MAA …

Synergetic Signaling Between The Chlamydia Secreted Effectors Tarp And Tmea Regulates Actin Kinetics And Efficient Entry, Matthew Romero
Synergetic Signaling Between The Chlamydia Secreted Effectors Tarp And Tmea Regulates Actin Kinetics And Efficient Entry, Matthew Romero
Theses & Dissertations
Chlamydia trachomatis is both the leading cause of non-congenital preventable blindness in developing nations and the leading source of bacterial sexually transmitted infections. As an obligate intracellular pathogen, Chlamydia has an absolute requirement on gaining entry into its host, which is accomplished by manipulating the host actin cytoskeleton to assemble engulfment structures that drive pathogen entry. In support of this, Chlamydia secretes the effectors TarP and TmeA via a type-III secretion system, whose respective signaling outputs operate synergistically to generate rapid actin kinetics and efficient pathogen engulfment. Rapid actin recruitment characteristic of invading Chlamydia is derived from a collaborative interaction …

Meningiomas are the most common intracranial brain tumours. These tumours are heterogeneous and encompass a wide spectrum of clinical aggressivity. Treatment options are limited to surgery and radiotherapy and have a risk of post-operative morbidities and radiation neurotoxicity, reflecting the need for new therapies. Three-dimensional (3D) patient-derived cell culture models have been shown to closely recapitulate in vivo tumour biology, including microenvironmental interactions and have emerged as a robust tool for drug development. Here, we established a novel easy-to-use 3D patient-derived meningioma spheroid model using a scaffold-free approach. Patient-derived meningioma spheroids were characterised and compared to patient tissues and traditional …

Research outputs 2022 to 2026
Oxidative dehydrogenation of ethane with CO2 (ODEC) is an attractive reaction for reduction of carbon footprints and ethene production. In this work, we present photothermal catalysis on confined bimetal catalysts for ODEC. Carbon nanotubes confined non-noble bimetal alloy (i.e., CoNi@CNTs and FeNi@CNTs) catalysts were prepared and FeNi@CNTs showed effective performance in photothermal catalytic ODEC to ethene. Experiments and simulations reveal that UV and visible lights (420 – 490 nm) are responsible for ODEC and non-oxidative dehydrogenation of ethane, respectively, to ethene. Additionally, ODEC to ethene is preferred to C-C cracking to methane on FeNi@CNTs in light ( > 490 nm)-induced thermocatalysis. …
