Transcriptional Pausing Factor M1bp Regulates Cellular Homeostasis By Suppressing Autophagy And Apoptosis In Drosophila Eye, Anuradha Venkatakrishnan Chimata, Hannah Darnell, Akanksha Raj, Madhuri Kango-Singh

Biology Faculty Publications

During organogenesis cellular homeostasis plays a crucial role in patterning and growth. The role of promoter proximal pausing of RNA polymerase II, which regulates transcription of several developmental genes by GAGA factor or Motif 1 Binding Protein (M1BP), has not been fully understood in cellular homeostasis. Earlier, we reported that M1BP, a functional homolog of ZKSCAN3, regulates wingless (wg) and caspase-dependent cell death (apoptosis) in the Drosophila eye. Further, blocking apoptosis does not fully rescue the M1BPRNAi phenotype of reduced eye. Therefore, we looked for other possible mechanism(s). In a forward genetic screen, members of the Jun-amino-terminal-(NH2)-Kinase (JNK) pathway …

Alaskan Educator Perceptions Of Preparedness To Identify Diverse Gifted Learners: A Phenomenological Study, Heather J. Stewart

Student Theses and Dissertations

The problem addressed in this qualitative study is that classroom teachers are not adequately prepared to identify gifted characteristics, which leads to the underreferral of students from diverse socioeconomic, ethnic, and language backgrounds. The purpose of this study was to explore the perceptions of public K-6 general education classroom teachers regarding their understanding of giftedness as it relates to their abilities to identify potentially gifted students and their awareness of programs and services offered by their district. The study’s guiding theoretical framework was based on key theories of intelligence and giftedness. In a phenomenological approach, 15 elementary educators from the …

Why We Work: Exploring The Relationships Between Work Rewards, Burnout, And Intention To Leave For Professional Nurses, Jacqueline Christianson Sep 2023

Why We Work: Exploring The Relationships Between Work Rewards, Burnout, And Intention To Leave For Professional Nurses, Jacqueline Christianson

Dissertations

Nurse burnout leads to attrition from hospital nursing positions and the nursing profession prior to typical retirement age. Yet some nurses choose to stay despite burnout. Previous research indicates that nurses stay due to the rewards they receive from work but the relationships between different types of work rewards, work-related burnout, and intention to leave are poorly described. The ability to implement or execute altruistic behaviors may also represent an under-recognized work reward. Thus, the purpose of this study is to investigate the relationships between work rewards, work-related burnout, and intention to leave nursing positions and the nursing profession and …

Is Climate Change Causing The Range Contraction Of Cape Rock-Jumpers (Chaetops Frenatus)?, Gregory D. Duckworth, Raquel A. Garcia, Rheinhardt Scholtz, Res Altwegg

Department of Agronomy and Horticulture: Faculty Publications

Species distribution models often suggest strong links between climate and species' distribution boundaries and project large distribution shifts in response to climate change. However, attributing distribution shifts to climate change requires more than correlative models. One idea is to examine correlates of the processes that cause distribution shifts, namely colonization and local extinction, by using dynamic occupancy models. The Cape Rock-jumper (Chaetops frenatus) has disappeared over most of its distribution where temperatures are the highest. We used dynamic occupancy models to analyse Cape Rock-jumper distribution with respect to climate (mean temperature and precipitation over the warmest annual quarter), …

The epidermal growth factor receptor (EGFR) plays crucial roles in several important biological functions such as embryogenesis, epithelial tissue development, and cellular regeneration. However, in multiple solid tumor types overexpression and/or activating mutations of the EGFR gene frequently occur, thus hijacking the EGFR signaling pathway to promote tumorigenesis. Non-small cell lung cancer (NSCLC) tumors in particular often contain prevalent and shared EGFR mutations that provide an ideal source for public neoantigens (NeoAg). Studies in both humans and animal models have confirmed the immunogenicity of some of these NeoAg peptides, suggesting that they may constitute viable targets for cancer immunotherapies. Peptide …

Prognostic Evaluation Of Re-Resection For Recurrent Glioblastoma Using The Novel Rano Classification For Extent Of Resection: A Report Of The Rano Resect Group, Philipp Karschnia, Antonio Dono, Jacob S Young, Stephanie T Juenger, Nico Teske, Levin Häni, Tommaso Sciortino, Christine Y Mau, Francesco Bruno, Luis Nunez, Ramin A Morshed, Alexander F Haddad, Michael Weller, Martin Van Den Bent, Juergen Beck, Shawn Hervey-Jumper, Annette M Molinaro, Nitin Tandon, Roberta Rudà, Michael A Vogelbaum, Lorenzo Bello, Oliver Schnell, Stefan J Grau, Susan M Chang, Mitchel S Berger, Yoshua Esquenazi, Joerg-Christian Tonn Sep 2023

BACKGROUND: The value of re-resection in recurrent glioblastoma remains controversial as a randomized trial that specifies intentional incomplete resection cannot be justified ethically. Here, we aimed to (1) explore the prognostic role of extent of re-resection using the previously proposed Response Assessment in Neuro-Oncology (RANO) classification (based upon residual contrast-enhancing (CE) and non-CE tumor), and to (2) define factors consolidating the surgical effects on outcome.

METHODS: The RANO resect group retrospectively compiled an 8-center cohort of patients with first recurrence from previously resected glioblastomas. The associations of re-resection and other clinical factors with outcome were analyzed. Propensity score-matched analyses were …

Faculty, Staff and Student Publications

Fetal alcohol spectrum disorders (FASD) are leading causes of neurodevelopmental disability. The mechanisms by which alcohol (EtOH) disrupts fetal brain development are incompletely understood, as are the genetic factors that modify individual vulnerability. Because the phenotype abnormalities of FASD are so varied and widespread, we investigated whether fetal exposure to EtOH disrupts ribosome biogenesis and the processing of pre-ribosomal RNAs and ribosome assembly, by determining the effect of exposure to EtOH on the developmental expression of 18S rRNA and its cleaved forms, members of a novel class of short non-coding RNAs (srRNAs). In vitro neuronal cultures and fetal brains (11–22 …

Recent studies have determined that the nervous system can sense and respond to signals from skeletal tissue, a process known as skeletal interoception, which is crucial for maintaining bone homeostasis. The hypothalamus, located in the central nervous system (CNS), plays a key role in processing interoceptive signals and regulating bone homeostasis through the autonomic nervous system, neuropeptide release, and neuroendocrine mechanisms. These mechanisms control the differentiation of mesenchymal stem cells into osteoblasts (OBs), the activation of osteoclasts (OCs), and the functional activities of bone cells. Sensory nerves extensively innervate skeletal tissues, facilitating the transmission of interoceptive signals to the CNS. …

Evolutionary Action-Machine Learning Model Identifies Candidate Genes Associated With Early-Onset Coronary Artery Disease, Dillon Shapiro, Kwanghyuk Lee, Jennifer Asmussen, Thomas Bourquard, Olivier Lichtarge

Faculty, Staff and Students Publications

Background Coronary artery disease is a primary cause of death around the world, with both genetic and environmental risk factors. Although genome-wide association studies have linked >100 unique loci to its genetic basis, these only explain a fraction of disease heritability. Methods and Results To find additional gene drivers of coronary artery disease, we applied machine learning to quantitative evolutionary information on the impact of coding variants in whole exomes from the Myocardial Infarction Genetics Consortium. Using ensemble-based supervised learning, the Evolutionary Action-Machine Learning framework ranked each gene's ability to classify case and control samples and identified 79 significant associations. …

Across multiancestry groups, we analyzed Human Leukocyte Antigen (HLA) associations in over 176,000 individuals with Parkinson’s disease (PD) and Alzheimer’s disease (AD) versus controls. We demonstrate that the two diseases share the same protective association at the HLA locus. HLA-specific fine-mapping showed that hierarchical protective effects of HLA-DRB1*04 subtypes best accounted for the association, strongest with HLA-DRB1*04:04 and HLA-DRB1*04:07, and intermediary with HLA-DRB1*04:01 and HLA-DRB1*04:03. The same signal was associated with decreased neurofibrillary tangles in postmortem brains and was associated with reduced tau levels in cerebrospinal fluid and to a lower extent with increased Aβ42. …

Erythrophagocytosis In Autoimmune Immunoglobulin A-Mediated Hemolysis, Jahnavi Gollamudi, Swapan Kumar Dasgupta, Perumal Thiagarajan

Faculty, Staff and Students Publications

INTRODUCTION: Warm antibody-mediated autoimmune hemolysis (WAIHA) is most often due to immunoglobulin G (IgG) antibodies and is detected by direct antiglobulin test (DAT). However, about 10% cases of hemolytic anemia are DAT negative. Herein, we describe a patient with DAT-negative hemolytic anemia due to an anti-IgA antibody. We investigate if isolated IgA can promote erythrophagocytosis.

METHODS: We isolated patient and control IgA on Jacalin agarose. Isolated IgA was used to sensitize healthy ABO/Rh-matched donor red blood cells (RBC). Antibody binding was examined by flowcytometry. The effect of IgA on erythrophagocytosis was evaluated using Macrophage colony stimulating factor 1 (M-CSF)-differentiated autologous …

Elevated levels of circulating tumor necrosis factor receptors 1 and 2 (cTNFR1/2) predict chronic kidney disease (CKD) progression; however, the mechanisms of their release remain unknown. Whether acute kidney injury (AKI) drives cTNFR1/2 elevations and whether they predict disease outcomes after AKI remain unknown. In this study, we used AKI patient serum and urine samples, mouse models of kidney injury (ischemic, obstructive, and toxic), and progression to fibrosis, nephrectomy, and related single-cell RNA-sequencing datasets to experimentally test the role of kidney injury on cTNFR1/2 levels. We show that TNFR1/2 serum and urine levels are highly elevated in all of the …

G-protein-coupled receptors (GPCRs) are critical regulators of cardiac physiology and a key therapeutic target for the treatment of heart disease. Ectopic olfactory receptors (ORs) are GPCRs expressed in extra-nasal tissues which have recently emerged as new mediators in the metabolic control of cardiac function. The goals of this study were to profile OR gene expression in the human heart, to identify ORs dysregulated by heart failure caused by ischemic cardiomyopathy, and to provide evidence suggestive of a role for those altered ORs in the pathogenesis of heart failure. Left ventricular tissue from heart failure patients (

Lem2 Is Essential For Cardiac Development By Maintaining Nuclear Integrity, Jacob A Ross, Didier Hodzic, Et Al.

2020-Current year OA Pubs

AIMS: Nuclear envelope integrity is essential for the compartmentalization of the nucleus and cytoplasm. Importantly, mutations in genes encoding nuclear envelope (NE) and associated proteins are the second highest cause of familial dilated cardiomyopathy. One such NE protein that causes cardiomyopathy in humans and affects mouse heart development is Lem2. However, its role in the heart remains poorly understood.

METHODS AND RESULTS: We generated mice in which Lem2 was specifically ablated either in embryonic cardiomyocytes (Lem2 cKO) or in adult cardiomyocytes (Lem2 iCKO) and carried out detailed physiological, tissue, and cellular analyses. High-resolution episcopic microscopy was used for three-dimensional reconstructions …

Periodical Cicadas (Magicicada Spp.): Predator Satiation, Or Too Much Of A Good Thing?, John R. Cooley, Greg Holmes

The Great Lakes Entomologist

We report a novel mode of ecdysis failure in periodical cicadas (Magicicada spp. Davis) tied to the cicadas’ reliance on high densities for survival in the face of predation. Under extremely high densities, cicadas can disrupt others undergoing ecdysis by knocking them loose from their perches, trapping them, or causing physical damage. Our observations raise the possibility that while periodical cicadas thrive at high densities, it is possible that some densities are so high that they inhibit success.

An Implementation Of The Method Of Moments On Chemical Systems With Constant And Time-Dependent Rates, Emmanuel O. Adara, Roger B. Sidje Sep 2023

An Implementation Of The Method Of Moments On Chemical Systems With Constant And Time-Dependent Rates, Emmanuel O. Adara, Roger B. Sidje

Northeast Journal of Complex Systems (NEJCS)

Among numerical techniques used to facilitate the analysis of biochemical reactions, we can use the method of moments to directly approximate statistics such as the mean numbers of molecules. The method is computationally viable in time and memory, compared to solving the chemical master equation (CME) which is notoriously expensive. In this study, we apply the method of moments to a chemical system with a constant rate representing a vascular endothelial growth factor (VEGF) model, as well as another system with time-dependent propensities representing the susceptible, infected, and recovered (SIR) model with periodic contact rate. We assess the accuracy of …

Abundance And Diversity Of Bees Visiting Flowering Pennycress, A New Oilseed Crop In The Midwestern Usa, Frank Forcella, Zachary M. Portman, Samantha S. Wells, William Perry, Russ W. Gesch, Yesuf Mohammed, Cody Hoerning, Alex Hard, Tad L. Wesley, Winthrop B. Phippen

The Great Lakes Entomologist

Oilseed pennycress (Thlaspi arvense) is a new, autumn-sown, “cash cover crop” for the Midwestern USA and elsewhere. Anthesis occurs in early spring when few other plants bloom, and its flowers attract early-emerging bees. However, the taxonomic composition of these bees was unknown. Consequently, we systematically captured and identified the genera and species of bees visiting pennycress flowers throughout anthesis at five site-years: two in Illinois and three in Minnesota. A cumulative total of 28 bee species were found across site-years. The most common genera were Andrena (10 species), Lasioglossum (12 species), and Halictus (2 species). Rarer genera were …

Limoniic Acid Is A Sex Attractant Pheromone Component Of Limonius Agonus (Coleoptera: Elateridae), Wim Van Herk, Regine Gries, Jocelyn Smith, Ian Scott, Gerhard Gries

The Great Lakes Entomologist

Recently, (E)-4-ethyloct-4-enoic acid (limoniic acid) has been reported as the major sex attractant pheromone component of L. canus and L. californicus (Coleoptera: Elateridae) in western North America. Our objective was to determine whether limoniic acid is also a sex attractant pheromone component of the eastern field wireworm, Limonius agonus (Say). In gas chromatographic-electroantennographic detection (GC-EAD) analyses of headspace volatiles from L. agonus females, antennae from male L. agonus responded to limoniic acid as a trace component. In field experiments, traps baited with synthetic limoniic acid, or its analog (E)-5-ethyloct-4-enoic acid, afforded captures of male L. …

Observations On Two Stem-Boring Coleoptera Of Prickly-Ash (Zanthoxylum Americanum) In Michigan: Micracis Suturalis (Curculionidae: Scolytinae) And Sternidius Alpha (Cerambycidae), Robert A. Haack Sep 2023

Observations On Two Stem-Boring Coleoptera Of Prickly-Ash (Zanthoxylum Americanum) In Michigan: Micracis Suturalis (Curculionidae: Scolytinae) And Sternidius Alpha (Cerambycidae), Robert A. Haack

The Great Lakes Entomologist

The xylophagous scolytine Micracis suturalis LeConte and the cerambycid beetle Sternidius alpha (Say) were reared or collected from trunk sections of prickly-ash, Zanthoxylum americanum Mill., in Ingham County, Michigan during 1986 to 1988. Most M. suturalis gallery systems were initiated by August in host material cut in late May 1987. Micracis suturalis gallery-system density data are presented. No M. suturalis were reared from caged host material one-year after cutting. Sternidius alpha was univoltine, with adults emerging from mid-May to mid-July from host material cut the previous spring. From 1 to 8 S. alpha larvae originated from individual oviposition sites, averaging …

Alderfly (Megaloptera: Sialidae) Larval Emergence And Pupation Site Selection At Intermediate Lake, Antrim County, Michigan, Robert A. Haack Sep 2023

Alderfly (Megaloptera: Sialidae) Larval Emergence And Pupation Site Selection At Intermediate Lake, Antrim County, Michigan, Robert A. Haack

The Great Lakes Entomologist

Larvae of the alderfly Sialis mohri Ross were collected in barrier pitfall traps along the shoreline of Intermediate Lake during a 3-year period (2015–2017). Two barriers (each 1-m long) were used per trap in 2015 and 2016, being placed on opposite sides of a central collection cup that was about 1 m from the shoreline at time of deployment. In 2015, with two traps monitored at 4–to 18-day intervals from 19 April to 29 June, larvae were collected first during the 19 April – 7 May period and last during 16–31 May. In 2016, using the same two trap locations, …

Differences In The Critical Thermal Maximum Between Two Size Classes Of Stenonema Femoratum (Ephemeroptera: Heptageniidae), Jaiden Frantz, David Houghton

The Great Lakes Entomologist

Temperature is an important variable affecting the behavior and survival of aquatic organisms; however, little is known about the effects of size and corresponding developmental differences on aquatic insect temperature tolerance. We tested the critical thermal maximum (CTM) of large (head capsule width mean = 3.5 mm) and small (1.9 mm) specimens of Stenonema femoratum (Say) by raising the experimental temperature by 0.1, 0.3, and 0.5ºC per minute during laboratory CTM trials. Size class and temperature increase rate combinations were randomized, and each combination was tested over four trials, for a total of 24 trials of four specimens each. Two-way …

The Caddisflies (Trichoptera) Of Finch Creek, Antrim County, Michigan (Usa), David Houghton, Robert Haack

The Great Lakes Entomologist

The caddisfly assemblages of Finch Creek, a small woodland stream in northern Lower Michigan, was sampled along its entire continuum during May, June, July, and September 2021–2022 using ultraviolet blacklight traps. A total of 98 species representing 15 families and 49 genera were collected, including two species [Lepidostoma prominens (Banks) and L. sommermanae (Ross)] not previously reported from Michigan, and several others either not previously found in the Lower Peninsula or not reported from the state for >70 years. A non-metric multidimensional scaling ordination determined distinct species assemblages corresponding to the spring, summer, and fall months. Shredders dominated the …
