The HSV-1 (VC2) live-attenuated vaccine strain was engineered with specific deletions in the amino termini of glycoprotein K (gK) and membrane protein UL20, rendering the virus unable to enter neurons and establish latency. VC2 replicates efficiently in epithelial cell culture but produces lower viral titers and smaller viral plaques than its parental HSV-1 (F) wild-type virus. VC2 is an effective live-attenuated vaccine against HSV-1 and HSV-2 infections in mice and guinea pigs and an anti-tumor immunotherapeutic and oncolytic virus against melanoma and breast cancer in mouse models. Previously, we reported that the gK/UL20 complex interacts with the UL37 tegument protein, …

Comparison Of High-Resolution Naip And Unmanned Aerial Vehicle (Uav) Imagery For Natural Vegetation Communities Classification Using Machine Learning Approaches, Parth Bhatt, Ann Maclean
Michigan Tech Publications, Part 1
To map and manage forest vegetation including wetland communities, remote sensing technology has been shown to be a valid and widely employed technology. In this paper, two ecologically different study areas were evaluated using free and widely available high-resolution multispectral National Agriculture Imagery Program (NAIP) and ultra-high-resolution multispectral unmanned aerial vehicle (UAV) imagery located in the Upper Great Lakes Laurentian Mixed Forest. Three different machine learning algorithms, random forest (RF), support vector machine (SVM), and averaged neural network (avNNet), were evaluated to classify complex natural habitat communities as defined by the Michigan Natural Features Inventory. Accurate training sets were developed …

Soil Organic Matter Diagenetic State Informs Boreal Forest Ecosystem Feedbacks To Climate Change, Allison N. Myers-Pigg, Karl Kaiser, Ronald Benner, Susan E. Ziegler
Faculty Publications
The fate of soil organic carbon (SOC) in boreal forests is dependent on the integrative ecosystem response to climate change. For example, boreal forest productivity is often nitrogen (N) limited, and climate warming can enhance N cycling and primary productivity. However, the net effect of this feedback on the SOC reservoir and its longevity with climate change remain unclear due to difficulty in detecting small differences between large and variable carbon (C) fluxes needed to determine net changes in soil reservoirs. The diagenetic state of SOC – resulting from the physicochemical and biological transformations that alter the original biomolecular composition …

Adoption Ouroboros: Repeating The Cycle Of Adoption As Rescue, Malinda L. Seymore
Faculty Scholarship
Ouroboros—the circular symbol of the snake eating its tail; an endless cycle. As the U.S. recently withdrew from Afghanistan in chaos and Russia invaded Ukraine, the attention of Americans turned, as it frequently has in times of international conflict, to the plight of children in need of rescue. For many Americans, rescue is synonymous with adoption. The history of international adoption began with rescues following America’s wars in Europe and Asia and continues today through other violent upheavals. International adoption is an ouroboros, repeating the pattern of adoption as a response to humanitarian crises. But as human and charitable as …

Originalism: Erasing Women From The Body Politic, Malinda L. Seymore
Faculty Scholarship
In Dobbs v. Jackson Women's Health, the Court relied on originalism to excise women from the Constitution. Originalism is purposefully backward-looking. With cherry-picked history, the Court created a future that looks to the past: a past where unwed pregnancy is shameful and can be redeemed only by secret adoption. Yet the case has revealed originalism as a flawed method, harmed the legitimacy of the Court, and energized those supporting abortion rights.

‘Chinese Virus’: A Critical Discourse Analysis Of Anti-Asian Racist Discourse During The Covid-19 Pandemic, Peiwen Wang, Theresa Catalano
Department of Teaching, Learning, and Teacher Education: Faculty Publications
Since the emergence of COVID-19, researchers have documented an increase in cases of anti-Asian racism and hate crimes. Research shows a possible connection between the ‘Chinese virus’ discourse of the Trump administration and violence in society (Arora and Kim 2020:387). Drawing from critical discourse studies we explore 2,071 comments from one YouTube video which documents anti-China rhetoric by the Trump administration in order to understand the underlying strategies commenters relied on in their reproduction and defence of this discourse. Findings show the trickle-down influence of Trump’s discourse on YouTube commenters, but also ways in which social media created a platform …

Nitric oxide (NO), a small free radical molecule, turned out to be pervasive in biology and was shown to have a substantial influence on a range of biological activities, including cell growth and apoptosis. This molecule is involved in signaling and affects a number of physiologic functions. In recent decades, several processes related to cancer, such as angiogenesis, programmed cell death, infiltration, cell cycle progression, and metastasis, have been linked with nitric oxide. In addition, other parallel work showed that NO also has the potential to operate as an anti-cancer agent. As a result, it has gained attention in cancer-related …
Widespread Variation In Salt Tolerance Within Freshwater Zooplankton Species Reduces The Predictability Of Community-Level Salt Tolerance, Matthew Schuler
Widespread Variation In Salt Tolerance Within Freshwater Zooplankton Species Reduces The Predictability Of Community-Level Salt Tolerance, Matthew Schuler
Department of Biology Faculty Scholarship and Creative Works
The salinization of freshwaters is a global threat to aquatic biodiversity. We quantified variation in chloride (Cl−) tolerance of 19 freshwater zooplankton species in four countries to answer three questions: (1) How much variation in Cl− tolerance is present among populations? (2) What factors predict intraspecific variation in Cl− tolerance? (3) Must we account for intraspecific variation to accurately predict community Cl− tolerance? We conducted field mesocosm experiments at 16 sites and compiled acute LC50s from published laboratory studies. We found high variation in LC50s for Cl− tolerance in multiple species, which, in the …

Transracial families are part of our society whether people like it or not. There are not enough adoptive families of any kind, and children need support, love, and acceptance. Last August the Census Bureau reported that the number of non-Hispanic Americans who identify as multiracial jumped by 127 percent over the last decade, and for people who identified as Hispanic, the increase was even higher. Also, from 2017 to 2019, half of all U.S. adoptions were of non-white children, and half of these adoptions were transracial. The result? An increase in transracial adoptions in the United States and the subsequent …

Alcohol (ethanol) exposure during pregnancy can adversely affect development, with long-lasting consequences that include neuroimmune, cognitive, and behavioral dysfunction. Alcohol-induced alterations in cytokine levels in the hippocampus may contribute to abnormal cognitive and behavioral outcomes in individuals with fetal alcohol spectrum disorders (FASD). Nutritional intervention with the essential nutrient choline can improve hippocampal-dependent behavioral impairments and may also influence neuroimmune function. Thus, we examined the effects of choline supplementation on hippocampal cytokine levels in adolescent and adult rats exposed to alcohol early in development. From postnatal day (PD) 4-9 (third trimester-equivalent), Sprague-Dawley rat pups received ethanol (5.25 g/kg/day) or sham …

The present investigation deals with some aspects of the diversity of fire ants (Hymenoptera: Formicidae) in their native range. The Red Imported Fire Ant Solenopsis invicta is native to the tropical and subtropical inland territories of South America. In Brazil, it mainly occurs around the Pantanal region and across the Paraguay river, a region composed of grasslands which are seasonally flooded. Recent studies have evidenced this fire ant species is gradually spreading to other regions of Brazil. In the present investigation, we surveyed the molecular diversity of S. invicta populations across fragments of Atlantic Forest in Sao Paulo, Brazil, using …

Galactose is a secondary fermentable sugar that requires specific regulatory and structural genes for its assimilation, which are under catabolite repression by glucose. When glucose is absent, the catabolic repression is attenuated, and the structural GAL genes are fully activated. In Saccharomyces cerevisiae, the GAL pathway is under selection in environments where galactose is present. However, it is unclear the adaptive strategies in response to long-term propagation in galactose as a sole carbon source in laboratory evolution experiments. Here, we performed a 4,000-generation evolution experiment using 48 diploid Saccharomyces cerevisiae populations to study adaptation in galactose. We show that fitness …

Multiple Behaviors For Turning Performance Of Pacific Bluefin Tuna (Thunnus Orientalis), Abigail M. Downs, Allison Kolpas, Barbara A. Block, Frank E. Fish
Biology Faculty Publications
Tuna are known for exceptional swimming speeds, which are possible because of their thunniform lift-based propulsion, large muscle mass and rigid fusiform body. A rigid body should restrict maneuverability with regard to turn radius and turn rate. To test if turning maneuvers by the Pacific bluefin tuna (Thunnus orientalis) are constrained by rigidity, captive animals were videorecorded overhead as the animals routinely swam around a large circular tank or during feeding bouts. Turning performance was classified into three different types: (1) glide turns, where the tuna uses the caudal fin as a rudder; (2) powered turns, where the animal uses …

Insect species are subjected to disparate selection pressure due to various biotic and abiotic stresses. Management practices including the heavy use of chemical insecticides and introduction of insect-resistant plant cultivars have been found to accelerate these processes. Clearly, natural selection coupled with human intervention have led to insect adaptations that alter phenotypes and genetic structure over time, producing distinct individuals with specialized traits, within the populations, commonly defined as biotypes. Biotypes are commonly found to have better fitness in the new environment and, in the case of aphids, the most commonly studied system for biotypes, have the ability to successfully …

PURPOSE: Glioblastoma(GBM) is a lethal disease characterized by inevitable recurrence. Here we investigate the molecular pathways mediating resistance, with the goal of identifying novel therapeutic opportunities.
EXPERIMENTAL DESIGN: We developed a longitudinal in vivo recurrence model utilizing patient-derived explants to produce paired specimens(pre- and post-recurrence) following temozolomide(TMZ) and radiation(IR). These specimens were evaluated for treatment response and to identify gene expression pathways driving treatment resistance. Findings were clinically validated using spatial transcriptomics of human GBMs.
RESULTS: These studies reveal in replicate cohorts, a gene expression profile characterized by upregulation of mesenchymal and stem-like genes at recurrence. Analyses of clinical databases …

White sturgeon (Acipenser transmontanus) in the Lower Fraser River are the focus of a catch-and-release angling fishery in British Columbia, Canada. However, the lower region of the catch area includes areas where tidal waters invade, and the consequence of salinity levels on recovery from an angling challenge are not characterized in sturgeon, despite theoretical implications of its import. We acclimated white sturgeon to various salinities (0, 10 and 20 (parts per thousand)) to investigate the effects of acclimation on recovery from stimulated angling stress that was induced through manual chasing. This challenge elicited the traditional physiological responses such as ion …

Although human oral fluid has become more routine for quantitative drug detection in pain management, detecting a large scope of medications and substances is costly and technically challenging for laboratories. This paper presents a quantitative assay for 64 pain medications, illicit substances, and drug metabolites in human oral fluid. The novelty of this assay is that it was developed on an older model AB SCIEX 4000 instrument and renders obscure the need for more technical and expensive laboratory equipment. This method includes addition of internal standard and a 2-step liquid-liquid extraction and dry-down step to concentrate and clean the samples. …

The prevalence and distribution of African alphaviruses such as chikungunya have increased in recent years. Therefore, a better understanding of the local distribution of alphaviruses in vectors across the African continent is important. Here, entomological surveillance was performed from 2014 to 2018 at selected sites in north-eastern parts of South Africa where alphaviruses have been identified during outbreaks in humans and animals in the past. Mosquitoes were collected using a net, CDC-light, and BG-traps. An alphavirus genus-specific nested RT-PCR was used for screening, and positive pools were confirmed by sequencing and phylogenetic analysis. We collected 64,603 mosquitoes from 11 genera, …
