Babesia microti is a tick-transmitted protozoan parasite of wildlife that can also cause serious disease in humans. It is now well established that B. microti represents an assemblage of different strains or species, only some of which are important zoonotic pathogens. Therefore, in order to assess the potential public health risk associated with B. microti in any given location, it is important to determine the strains that are present. This is the first study on the presence and identity of B. microti in Ireland. Overall, 314 wood mice (Apodemus sylvaticus), 243 bank voles (Myodes glareolus) and 634 questing Ixodes ricinus …

During emergency hematopoiesis, hematopoietic stem cells (HSCs) rapidly proliferate to produce myeloid and lymphoid effector cells, a response that is critical against infection or tissue injury. If unresolved, this process leads to sustained inflammation, which can cause life-threatening diseases and cancer. Here, we identify a role of double PHD fingers 2 (DPF2) in modulating inflammation. DPF2 is a defining subunit of the hematopoiesis-specific BAF (SWI/SNF) chromatin-remodeling complex, and it is mutated in multiple cancers and neurological disorders. We uncovered that hematopoiesis-specific Dpf2-KO mice developed leukopenia, severe anemia, and lethal systemic inflammation characterized by histiocytic and fibrotic tissue infiltration resembling a …

Investigating The Antibiotic Potentiating Effects Of A Novel Reactive Oxidant Species-Generating Antimicrobial., Gracious Yoofi Boafo Donkor

Theses and Dissertations

Due to the rise in antimicrobic resistance and the decline in antibiotic discovery, drug development has expanded into alternative antimicrobial compounds, such as silver. Silver-containing compounds have emerged as a promising alternative due to their multi-specific effects on inhibiting bacterial growth. Our work focuses on the antimicrobial effects of AGXX, a novel antimicrobial which has been shown to exert its broad-spectrum effects through the production of highly cytotoxic reactive oxygen species (ROS). Although preliminary studies have demonstrated the potency of AGXX in gram-positive pathogens, its effects on gram-negative pathogens remain largely unknown. The goal of my thesis was to examine …

BACKGROUND: Sepsis-associated acute kidney injury (SA-AKI) is associated with high morbidity, with no current therapies available beyond continuous renal replacement therapy (CRRT). Systemic inflammation and endothelial dysfunction are key drivers of SA-AKI. We sought to measure differences between endothelial dysfunction markers among children with and without SA-AKI, test whether this association varied across inflammatory biomarker-based risk strata, and develop prediction models to identify those at highest risk of SA-AKI.

METHODS: Secondary analyses of prospective observational cohort of pediatric septic shock. Primary outcome of interest was the presence of ≥ Stage II KDIGO SA-AKI on day 3 based on serum creatinine …

Batten disease, one of the most devastating types of neurodegenerative lysosomal storage disorders, is caused by mutations in CLN3. Here, we show that CLN3 is a vesicular trafficking hub connecting the Golgi and lysosome compartments. Proteomic analysis reveals that CLN3 interacts with several endo-lysosomal trafficking proteins, including the cation-independent mannose 6 phosphate receptor (CI-M6PR), which coordinates the targeting of lysosomal enzymes to lysosomes. CLN3 depletion results in mis-trafficking of CI-M6PR, mis-sorting of lysosomal enzymes, and defective autophagic lysosomal reformation. Conversely, CLN3 overexpression promotes the formation of multiple lysosomal tubules, which are autophagy and CI-M6PR-dependent, generating newly formed proto-lysosomes. Together, our …

High-grade serous ovarian cancer (HGSC) is the most lethal histotype of ovarian cancer and the majority of cases present with metastasis and late-stage disease. Over the last few decades, the overall survival for patients has not significantly improved, and there are limited targeted treatment options. We aimed to better characterize the distinctions between primary and metastatic tumors based on short- or long-term survival. We characterized 39 matched primary and metastatic tumors by whole exome and RNA sequencing. Of these, 23 were short-term (ST) survivors (overall survival (OS) < 3.5 years) and 16 were long-term (LT) survivors (OS > 5 years). We compared somatic mutations, copy number alterations, mutational burden, differential gene …

Since the T-box transcription factors (TFs) T-BET and EOMES are necessary for initiation of NK cell development, their ongoing requirement for mature NK cell homeostasis, function, and molecular programming remains unclear. To address this, T-BET and EOMES were deleted in unexpanded primary human NK cells using CRISPR/Cas9. Deleting these TFs compromised in vivo antitumor response of human NK cells. Mechanistically, T-BET and EOMES were required for normal NK cell proliferation and persistence in vivo. NK cells lacking T-BET and EOMES also exhibited defective responses to cytokine stimulation. Single-cell RNA-Seq revealed a specific T-box transcriptional program in human NK cells, which …

Importance: In the 1930s, the government-sponsored Home Owners' Loan Corporation (HOLC) established maps of US neighborhoods that identified mortgage risk (grade A [green] characterizing lowest-risk neighborhoods in the US through mechanisms that transcend traditional risk factors to grade D [red] characterizing highest risk). This practice led to disinvestments and segregation in neighborhoods considered redlined. Very few studies have targeted whether there is an association between redlining and cardiovascular disease.

Objective: To evaluate whether redlining is associated with adverse cardiovascular outcomes in US veterans.

Design, setting, and participants: In this longitudinal cohort study, US veterans were followed up (January 1, 2016, …

Pathogen Emergence As Complex Biological Invasion: Lessons From Dynamical Systems Modeling, Sudam Surasinghe, Marisabel Rodriguez, Victor Meszaros, Jane Molofsky, Salvador Almagro-Moreno, Brandon Ogbunugafor

Northeast Journal of Complex Systems (NEJCS)

Infectious disease emergence has become the target of cross-disciplinary efforts
that aim to understand and predict the shape of outbreaks. The many challenges
involved with the prediction of disease emergence events is a characteristic that in-
fectious diseases share with biological invasions in many subfields of ecology (e.g.,
how certain plants are able to successfully invade a new niche). Like infectious
diseases, biological invasions by plants and animals involve interactions between
agents (pathogens and plants in their respective cases) and a recipient niche. In
this study, we examine the problem of pathogen emergence through the lens of a
framework first …

Radiation Oncology Articles

Here, we investigate radiomics-based characterization of tumor vascular and microenvironmental properties in an orthotopic rat brain tumor model measured using dynamic-contrast-enhanced (DCE) MRI. Thirty-two immune compromised-RNU rats implanted with human U-251N cancer cells were imaged using DCE-MRI (7Tesla, Dual-Gradient-Echo). The aim was to perform pharmacokinetic analysis using a nested model (NM) selection technique to classify brain regions according to vasculature properties considered as the source of truth. A two-dimensional convolutional-based radiomics analysis was performed on the raw-DCE-MRI of the rat brains to generate dynamic radiomics maps. The raw-DCE-MRI and respective radiomics maps were used to build 28 unsupervised Kohonen self-organizing-maps …

Skeletal Anatomy Of The Pectoral Fin In Mudskipper Species From Terrestrial And Aquatic Habitats, Haodong Zhou, Cassandra M. Donatelli, Odette Laneuville, Emily M. Standen

Engineering Faculty Articles and Research

Mudskippers are a group of amphibious fishes in the family Oxudercidae, whose species inhabit a range of habitats from mostly aquatic to mostly terrestrial. Most of our understanding about habitat preference comes from natural history observations, particularly where they are collected (i.e., low intertidal vs. high intertidal regions). Mudskippers have undergone several morphological changes to accommodate a terrestrial life, including major changes to the pectoral and pelvic girdles. These changes result in a novel crutching gait, which mudskippers use to move over land. Though the appendicular morphology and crutching gait of mudskippers have been described in some species, few …

Psychology Faculty Publications and Presentations

The contributions of Black scholars to psychology have been erased or marginalized within mainstream, U.S.-centered psychology. As such, psychologists and trainees have little exposure to strengths-based theories and schools of thought that center and humanize the experiences of people of African descent. This special issue intervenes on anti-Black racism at the epistemic level by curating a review of foundational contributions by diverse Black scholars in psychology and related fields. The special issue is organized around five integrative and overlapping themes: (a) Black scholars who have written on topics related to race, racism, and racial identity; (b) schools of thought that …

Increased vascular permeability is a frequent outcome of viperid snakebite envenomation, leading to local and systemic complications. We reported that snake venom cysteine-rich secretory proteins (svCRiSPs) from North American pit vipers increase vascular permeability both in vitro and in vivo. They also induce acute activation of several adhesion and signaling molecules that may play a critical role in the pathophysiology of snakebites. Extracellular vesicles (EVs) have gained interest for their diverse functions in intercellular communication, regulating cellular processes, blood-endothelium interactions, vascular permeability, and immune modulation. They also hold potential as valuable biomarkers for diagnosing, predicting, and monitoring therapeutic responses in …

Study And Evaluation Of Parameters Influencing Parkison’S Disease Using 3d Cnn, Caroline El Fiorenza J, V. Sellam

Applied Mathematics & Information Sciences

A diagnostic method of determining protein structure in persons for Parkinson’s disease (PD) would be a 3D imaging scan. Convolutional Neural Networks (CNNs), which are efficient when used with spatial data, are suitable candidates for automating this diagnostic procedure to assist medical workers. The protein structure of PD patients was evaluated or diagnosed using a Improved Faster Recurrent Convolutional Neural Network (IFRCNN) ordinal model in this article. A data preprocessing technique was modified to operate PD because IFRCNNs require big datasets to operate acceptably. We take into account the Improved Shortest Paths Ordinal Graph-based Oversampling (ISP-OGO) technique that employs a …

Deep neural networks have been found to be vulnerable to adversarial noise. Recent works show that exploring the impact of adversarial noise on intrinsic components of data can help improve adversarial robustness. However, the pattern closely related to human perception has not been deeply studied. In this paper, inspired by the cognitive science, we investigate the interference of adversarial noise from the perspective of image phase, and find ordinarily-trained models lack enough robustness against phase-level perturbations. Motivated by this, we propose a joint adversarial defense method: a phase-level adversarial training mechanism to enhance the adversarial robustness on the phase pattern; …

Herein we review our work involving dispersed adrenocortical cells from several lizard species: the Eastern Fence Lizard (Sceloporus undulatus), Yarrow's Spiny Lizard (Sceloporus jarrovii), Striped Plateau Lizard (Sceloporus virgatus) and the Yucatán Banded Gecko (Coleonyx elegans). Early work demonstrated changes in steroidogenic function of adrenocortical cells derived from adult S. undulatus associated with seasonal interactions with sex. However, new information suggests that both sexes operate within the same steroidogenic budget over season. The observed sex effect was further explored in orchiectomized and ovariectomized lizards, some supported with exogenous testosterone. Overall, a suppressive effect of testosterone was evident, especially in cells …

The temporal effects of ethanol preservation on the δ13C and δ15N values of tissues excised from European eel Anguilla anguilla were assessed. Preservation significantly enriched 13C values of fin and mucus but not dorsal muscle. The 13C enrichment occurred in the initial 15 days of preservation and was independent of initial eel mass. Tissue preservation effects on δ15N values were negligible. These tissue-specific isotopic shifts should be considered when ethanol-preserved eel samples are used.

Modelling And Optimal Control Strategies Of Corruption Dynamics, Temesgen Duressa Keno, Fekadu Mosisa Legesse

Applied Mathematics & Information Sciences

In this study, we presented a non linear deterministic corruption transmission dynamics using optimal control analysis and cost effective strategies. To begin, we demonstrated that in a given set of initial conditions, the model solution is non-negative and bounded. A basic reproductive number is calculated using the corruption-free equilibrium point via the next generation matrix. The linearization and the Lyapunov function are then used to demonstrate how corruption-free equilibrium is both locally and globally stable. The corruption-free equilibrium point is asymptotically stable both locally and globally if the basic reproduction number is less than one; otherwise, an endemic corruption equilibrium …

Applied Mathematics & Information Sciences

Inthisstudy, anonlinearmathematicalmodelforthetransmissiondynamicsofmastitisdiseasesisformulatedandanalyzed. The local and global stability analysis of mastitis-free equilibrium and endemic equilibrium is obtained using the stability theory of differential equation. It was established that the mastitis-free equilibrium is locally stable if the basic reproduction number is less than unity. The endemic equilibrium, which exists only when the basic reproduction number is greater than unity, is globally asymptotically stable. Sensitivity analysis of the reproduction number suggested that the concentration of bacteria in the environment has a high impact on the dynamics of mastitis. Furthermore, an optimal control problem is formulated by applying Pontryagin’s minimum principle with three control strategies, …

Diversity-Enhancing Generative Network For Few-Shot Hypothesis Adaptation, Ruijiang Dong, Feng Liu, Haoang Chi, Tongliang Liu, Mingming Gong, Gang Niu, Masashi Sugiyama, Bo Han

Generating unlabeled data has been recently shown to help address the few-shot hypothesis adaptation (FHA) problem, where we aim to train a classifier for the target domain with a few labeled target-domain data and a well-trained source-domain classifier (i.e., a source hypothesis), for the additional information of the highly-compatible unlabeled data. However, the generated data of the existing methods are extremely similar or even the same. The strong dependency among the generated data will lead the learning to fail. In this paper, we propose a diversity-enhancing generative network (DEG-Net) for the FHA problem, which can generate diverse unlabeled data with …

Coco: A Coupled Contrastive Framework For Unsupervised Domain Adaptive Graph Classification, Nan Yin, Li Shen, Mengzhu Wang, Long Lan, Zeyu Ma, Chong Chen, Xian Sheng Hua, Xiao Luo

Machine Learning Faculty Publications

Although graph neural networks (GNNs) have achieved impressive achievements in graph classification, they often need abundant task-specific labels, which could be extensively costly to acquire. A credible solution is to explore additional labeled graphs to enhance unsupervised learning on the target domain. However, how to apply GNNs to domain adaptation remains unsolved owing to the insufficient exploration of graph topology and the significant domain discrepancy. In this paper, we propose Coupled Contrastive Graph Representation Learning (CoCo), which extracts the topological information from coupled learning branches and reduces the domain discrepancy with coupled contrastive learning. CoCo contains a graph convolutional network …
