Identification Of Lethal Inhibitors And Inhibitor Combinations For Mono-Driver Versus Multi-Driver Triple-Negative Breast Cancer Cells, Geng Chia Ku, Abygail G. Chapdelaine, Marina K. Ayrapetov, Gongqin Sun

There are no signaling-based targeted therapies for triple-negative breast cancer. The development of targeted cancer therapy relies on identifying oncogenic signaling drivers, understanding their contributions to oncogenesis and developing inhibitors to block such drivers. In this study, we determine that DU-4475 is a mono-driver cancer cell line relying on BRAF and the mitogen-activated protein kinase pathway for viability and proliferation. It is fully and lethally inhibited by BRAF or Mek inhibitors at low nM concentrations, but it is resistant to inhibitors targeting other signaling pathways. Free-roaming domestic cats (i.e., cats that are owned or unowned and are considered ‘at large’) are globally distributed non-native species that have marked impacts on biodiversity and human health. Despite clear scientific evidence of these impacts, free-roaming cats are either unmanaged or managed using scientifically unsupported and ineffective approaches (e.g., trap-neuter-release [TNR]) in many jurisdictions around the world. A critical first initiative for effective, science-driven management of cats must be broader political and legislative recognition of free-roaming cats as a non-native, invasive species. Center for Coastal and Ocean Mapping

The sediment-water interface in the coastal ocean is a highly dynamic zone controlling biogeochemical fluxes of greenhouse gases, nutrients, and metals. Processes in the sediment mixed layer (SML) control the transfer and reactivity of both particulate and dissolved matter in coastal interfaces. Here we map the global distribution of the coastal SML based on excess 210Pb (210Pbex) profiles and then use a neural network model to upscale these observations. We show that highly dynamic regions such as large estuaries have thicker SMLs than most oceanic sediments. Six-Transmembrane Epithelial Antigen of the Prostate 1-4 (STEAP1-4) compose a family of metalloproteinases involved in iron and copper homeostasis and other cellular processes. Thus far, five homologs are known: STEAP1, STEAP1B, STEAP2, STEAP3, and STEAP4. In prostate cancer, STEAP1, STEAP2, and STEAP4 are overexpressed, while STEAP3 expression is downregulated. Although the metalloreductase activities of STEAP1-4 are well documented, their other biological functions are not. Furthermore, the properties and expression levels of STEAP heterotrimers, homotrimers, heterodimers, and homodimers are not well understood. Background: Anecdotal experience and studies have shown that most pediatric patients fail to reach target therapeutic vancomycin trough levels (VTLs) and required higher total daily doses (TDD). This retrospective study aims to evaluate the frequency of hospitalized children who achieved target VTLs with a vancomycin (VNCO) dosing regimen of 40-60 mg/kg/d q6h and to assess the VNCO-TDD required to attain the target and their effects on clinical outcomes in pediatric patients.

Introduction

Negative affective symptoms (e.g., anxiety, depression, and anger) are correlated and have parallel associations with outcomes, as do related personality traits (i.e., facets of neuroticism), often prompting statistical control (i.e., partialing) to determine independent effects. However, such adjustments among predictor variables can alter their construct validity. In three studies, the interpersonal circumplex (IPC) and a related analytic approach (i.e., Structural Summary Method) were used to evaluate changes in interpersonal correlates of negative affective characteristics resulting from partialing.

BRICK is a semi-empirical model for global and local mean sea-level change (Wong et al., 2017). The core model includes component sub-models for the major contributors to global mean sea-level change - glaciers and ice caps, thermal expansion, land water storage, and the Greenland and Antarctic ice sheets. The resulting global mean sea levels can be downscaled via a data set that represents the “fingerprint” of each sea-level component on local mean sea level (Slangen et al., 2014). Under the context of China's carbon peak and carbon neutrality, the treatment of municipal solid waste (MSW) is facing new challenge to reduce carbon emission. This study suggests that significant carbon emission reduction and dual cycle of mass and energy can be realized, under the context of MSW classification, by the collaborative treatment of multi-source solid waste in Eco-Industrial Park. Results showed that in 2020, the incineration and landfilling of MSW in China reduced 5.18 million tons of CO2 emission, at the same year, 15 million tons of CO2 emission could have been reduced if the wet waste was treated …

The number of Middle Eastern immigrant women (MEIW) living in Canada has significantly increased. However, this group of women is underrepresented in health research and there is a gap in knowledge of their cancer survivorship experiences in Canada. This phenomenological qualitative approach was employed to uncover the meaning of the lived experiences of MEIW during their cancer survivorship journey (CSJ). Data were collected through unstructured interviews and one written description from three MEIW. Early-stage pancreatic cancer remains challenging to detect, leading to a poor five-year patient survival rate. This obstacle necessitates the development of early detection approaches based on novel technologies and materials. In this work, the presence of a specific pancreatic cancer-derived miRNA (pre-miR-132) is detected using the fluorescence properties of biocompatible nitrogen-doped graphene quantum dots (NGQDs) synthesized using a bottom-up approach from a single glucosamine precursor. Six Ways To Misunderstand Precarity: Reflections On Social Angst And Its Political Offspring, Albena Azmanova

Emancipations: A Journal of Critical Social Analysis

These reflections develop a comprehensive conceptualization of precarity as a condition of politically and systemically generated economic and social vulnerability caused by insecurity of livelihoods – a form of disempowerment, experienced as incapacity to cope, itself rooted in a misalignment between responsibility and power. Precarity, thus understood, is a transversal social injustice cutting across differences in social class, education, employment, and income. It harms people’s material and psychological welfare and hampers society’s capacity to manage adversity and govern itself.

The article elaborates on the above conceptualization by addressing six fallacies in debates on precarity:

Pilitropic and flagellotropic phages adsorb to bacterial pili and flagella. These phages have long been used to investigate multiple aspects of bacterial physiology, such as the cell cycle control in the Caulobacterales. Targeting cellular appendages for adsorption effectively constrains the population of infectable hosts, suggesting that phages may have developed strategies to maximize their infective yield. Brevundimonas phage vB_BsubS-Delta is a recently characterized pilitropic phage infecting the Alphaproteobacterium Brevundimonas subvibrioides. Like other Caulobacterales, B. subvibrioides divides asymmetrically and its cell cycle is governed by multiple transcriptional regulators, including the master regulator CtrA. Self-enhancement (SE) is often overlooked as a fundamental cognitive ability mediated via the Prefrontal Cortex (PFC). Here, we present research that establishes the relationship between the PFC, SE, and the potential evolved beneficial mechanisms. Specifically, we believe there is now enough evidence to speculate that SE exists to provide significant benefits and should be considered a normal aspect of the self. Whatever the metabolic or social cost, the upside of SE is great enough that it is a core and fundamental psychological construct. Divergent selection of populations in contrasting environments leads to functional genomic divergence. However, the genomic architecture underlying heterogeneous genomic differentiation remains poorly understood. Here, we de novo assembled two high-quality wild barley (Hordeum spontaneum K. Koch) genomes and examined genomic differentiation and gene expression patterns under abiotic stress in two populations. These two populations had a shared ancestry and originated in close geographic proximity but experienced different selective pressures due to their contrasting micro-environments.
