Diabetes is the third most customary non-infectious disease occurring worldwide and leads to untimely disability and mortality. This metabolic disorder is characterized by impaired insulin secretions and high blood glucose levels. Among others, type 2 diabetes is the most common. Various chemical and biochemical hypoglycemic drugs are utilized to control and treat hyperglycemia but required further interventions to reduce their toxicity and side effects on the users. Edible mushrooms have been employed from ancient times to heal many diseases as it composed of anti-oxidant, alkaloids, fibers, triterpenoids, and other phytochemicals. The bioactive secondary metabolites of mushrooms can improve insulin resistance …
Eluication Of Lipid Metabolic Pathways In Differentiating Giardia Lamblia Using High Resolution Mass Spectrometry, Cameron Ellis
Eluication Of Lipid Metabolic Pathways In Differentiating Giardia Lamblia Using High Resolution Mass Spectrometry, Cameron Ellis
Open Access Theses & Dissertations
Giardia lamblia is an intestinal protozoan found worldwide, including the U.S. This parasite exists in two morphologic stages - a replicative trophozoite and a relatively dormant yet viable cyst. While exposures of cysts to gastric acid during passage through the human stomach induces excystation, factors in the small intestine, where trophozoites colonize trigger encystation or cyst formation. Transformation into cyst stage is essential for Giardia to survive in the environment for months before infecting new hosts. Because of its small genome size (11.7 Mb), metabolic pathways in Giardia are highly reduced. As far as lipid metabolism is concerned, only limited …

Tuberculosis (TB) is a communicable disease caused by Mycobacterium tuberculosis (Mtb). It was the leading cause of human death generated by a single infectious agent before the Coronavirus (COVID-19) pandemic. Mtb has a special mechanism to evade the hostâ??s immune system and utilize macrophages as reservoirs. Latent TB infections have been a threat to public health. Liposome has been widely used for drug delivery. Immunoliposomes are special liposome carriers coated with ligands, such as antigens, targeting certain cells or organs to regulate the uptake of liposome. By constructing the liposome components, or surface modification, cargo release of liposome can be …

Characterization Of Esxa/Esxb Heterodimer Membrane Interaction With A Novel Popc/Popg Liposome Model, Salvador Vazquez Vazquez Reyes
Open Access Theses & Dissertations
EsxA and EsxB are co-secreted as a heterodimer and have been shown to play critical roles in phagosome rupture and translocation of Mycobacterium tuberculosis into the cytosol. Current in vitro studies support a model in which the EsxA:EsxB heterodimer is dissociated upon acidification, which might allow EsxA insertion into liposome membranes. While the membrane permeabilizing activity (MPA) of EsxA has been well characterized in liposomes composed of di-oleoyl-phosphatidylcholine (DOPC), the MPA of EsxA:EsxB heterodimer has not been detected with the DOPC liposome. In this study, we established a new in vitro membrane assay to test the MPA of EsxA and …

Reflections On The Adaptation Of A Postgraduate Degree In Water Management From In-Person To Remote Delivery, Murray Clamen, Emma Anderson, Johanna Dipple, Jan Adamowski
Journal of Contemporary Water Research and Education
In early 2020, the COVID-19 pandemic spurred the rapid adaptation of university course delivery to an online format. Though in-person delivery partially resumed in the Fall of 2021, future conditions may favor a return to, or addition of, remote delivery. It is therefore important for instructors, program directors, and institutions to capitalize on this learning opportunity and reflect on adaptation measures’ successes (and failures) to inform future online course design. The reworking of McGill University's Master of Science Program in Integrated Water Resources Management (IWRM) provides a case study to evaluate the adaptation of remote teaching of water resource management. …

What is mathematical modeling and how can inservice and pre-service teachers develop the skills and competencies necessary to increase confidence and interest in teaching relevant mathematical modeling lessons? Mathematical modeling is “the process of choosing and using appropriate mathematics and statistics to analyze empirical situations, to understand them better, and to improve decisions” (CSSM, 2010, p. 72). By providing students with an opportunity to engage in relevant mathematical modeling prompts, we provide them with transferable skills and knowledge. The aim of this paper will be to provide insight into the relevance of teaching mathematical modeling, provide resources for integrating modeling …
Modeling A Pandemic: Investigating Student Learning About Disease Spread In The Context Of Agent-Based Modeling, Siyu Wu
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
The COVID-19 pandemic has highlighted a need for students to learn about public health issues, including the transmission of disease and methods for the prevention of epidemics. This study presents data from a project focused on developing computational microworlds to help middle school students learn about these topics. The microworld is designed to help students model and test their ideas about how a disease spreads through a population and how an epidemic can be prevented. I employed a lab-based case study approach to conduct one-on-one 1.5-hour interviews through Zoom with four middle-school students (ages 12-14). During the interview, the student …
Pesticide-Leaf Interactions And Their Implications For Pesticide Fate Modeling, Ashlie Kinross
Pesticide-Leaf Interactions And Their Implications For Pesticide Fate Modeling, Ashlie Kinross
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
The work described here provides measured data to improve the understanding of the interaction between a pesticide and leaf surface after application. Two methods were developed, one using a newly introduced instrument, for the extraction of pesticides from leaves. This is required to measure the concentration of pesticide in the leaves. Using one of the developed extraction methods, measurements were made to determine how a pesticide equilibrates between a leaf and the air above it. These measurements were incorporated into a pesticide fate model that predicts how a pesticide moves through the environment after application to an agricultural field. The …

Investigating Bacterial Community Composition And Antimicrobial Resistance Genes In Beef Cattle, Arena E. See
Department of Animal Science: Dissertations, Theses, and Student Research
While antibiotics have been critical over the last decade for curbing infectious diseases, they have contributed to the rise of antimicrobial resistance (AMR). Infections resulting from antimicrobial-resistant pathogens are correlated with severe morbidity, mortality, and healthcare costs. With the increasing concerns regarding AMR and its emergence, it is paramount to understand not only the effects of antibiotics on microbial community and diversity, but also to develop a novel tool that gives us a better understanding of the spread and functional traits of AMR genes.
To this end, rumen, fecal, and pen surface samples were collected from 72 animals at United …

Population Health, East Africa
Factors associated with COVID-19 vaccine hesitancy (which we define as refusal to be vaccinated when asked, resulting in delayed or non- vaccination) are poorly studied in sub-Saharan Africa and among refugees, particularly in Kenya. Using survey data from wave five (March to June 2021) of the Kenya Rapid Response Phone Survey (RRPS), a household survey representative of the population of Kenya, we estimated the self-reported rates and factors associated with vaccine hesitancy among non-refugees and refugees in Kenya. Non-refugee households were recruited through sampling of the 2015/16 Kenya Household Budget Survey and random digit dialing. Refugee households were recruited through …

Human lung organoids (hLOs) are useful for disease modelling and drug screening. However, a lack of immune cells in hLOs limits the recapitulation of in vivo cellular physiology. Here, we generated hLOs containing alveolar macrophage (AMφ)-like cells derived from pluripotent stem cells (PSC). To bridge hLOs with advanced human lung high-resolution X-ray computed tomography (CT), we acquired quantitative micro-CT images. Three hLO types were observed during differentiation. Among them, alveolar hLOs highly expressed not only lung epithelial cell markers but also AMφ-specific markers. Furthermore, CD68
Functional Characterization Of The Toxoplasma Gondii Endolysosomal System, Leah Brock Thornton
Functional Characterization Of The Toxoplasma Gondii Endolysosomal System, Leah Brock Thornton
All Dissertations
Toxoplasma gondii is an obligate intracellular parasite that can infect a wide range of mammalian hosts. T. gondii can cause severe disease in immunocompromised individuals as well as pregnant women, in which congenital toxoplasmosis can lead to spontaneous abortion and fetal blindness. T. gondii infects up to roughly one-third of the global human population and is the causative agent of toxoplasmosis. Given this health burden of toxoplasmosis and that antibiotics currently used for the treatment of infection lead to strong side effects, identification of parasite-specific targets are necessary for novel therapeutic strategies.
T. gondii parasites contain a dynamic and active …

Current immunotherapies are only effective in a fraction of cancer patients. One of the main contributing factors to therapy resistance and overall poor prognosis is the immune status of the tumor microenvironment (TME). Tumor-associated macrophages (TAMs) have been shown to have significant influence on the immune status of the TME. As key players in maintaining tissue homeostasis, macrophages exhibit a broad range of phenotypes and functions, making TAMs a promising target for immunomodulatory therapies. As liaisons between innate and adaptive immunity, TAMs can promote a robust anti-tumor response involving both innate and adaptive immunity. This study investigated the potential of …

Characterization Of Kinases And Hypothetical Proteins In The Entamoeba Species, Heather Andrews Walters
All Dissertations
Entamoeba histolytica is a protozoan parasite that causes amebic dysentery and amoebic liver abscess. This pathogen possesses a two-stage life cycle consisting of an environmentally stable latent cyst and a pathogenic amoeboid trophozoite. Since infection is acquired by ingestion of cysts from contaminated food and water, this parasite is prevalent in underdeveloped countries. A reptilian pathogen, Entamoeba invadens, which can encyst in culture, has long-served as a surrogate to study stage conversion. Much remains unclear about stage conversion and the stress response in these parasites and current treatments for amoebiasis are lacking, as they cause severe side effects. Ultimately …

Antimicrobial resistance-encoding mobile genetic elements (MGEs) may contribute to the disease potential of bacterial pathogens. We previously described the association of Group A Streptococcus (GAS) derived from invasive disease with increasingly frequent antimicrobial resistance (AMR). We hypothesized that a 65-kb AMR-encoding MGE (ICESpyM92), highly conserved among closely related emergent invasive emm92 GAS, contributes to GAS disease potential. Here, we provide evidence that a combination of ICESpyM92- and core genome-dependent differential gene expression (DGE) contributes to invasive disease phenotypes of emergent emm92 GAS. Using isogenic ICESpyM92 mutants generated in distinct emm92 genomic backgrounds, we determined the presence of ICESpyM92 enhances GAS …

BACKGROUND: Mitochondrial toxicity resulting in myopathy and lactic acidosis has been described in antiretroviral (ARV)-exposed patients. We hypothesized that myopathy in HIV-infected, ARV-treated children would be associated with metabolic (acylcarnitines) and genetic (variants in metabolic genes) markers of dysfunctional fatty acid oxidation (FAO).
METHODS: Acylcarnitine profiles (ACP) were analyzed for 74 HIV-infected children on nucleoside reverse transcriptase inhibitor (NRTI)-containing ARV. Thirty-seven participants with ≥2 creatine kinase measurements >500 IU (n = 18) or evidence of echocardiographic cardiomyopathy (n = 19) were matched with 37 participants without myopathy. Single nucleotide polymorphisms (SNPs) in FAO genes were also evaluated.
RESULTS: Abnormal ACP …

SARS-CoV-2 Omicron subvariants BA.2.12.1 and BA.4/5 have surged notably to become dominant in the United States and South Africa, respectively1,2. These new subvariants carrying further mutations in their spike proteins raise concerns that they may further evade neutralizing antibodies, thereby further compromising the efficacy of COVID-19 vaccines and therapeutic monoclonals. We now report findings from a systematic antigenic analysis of these surging Omicron subvariants. BA.2.12.1 is only modestly (1.8-fold) more resistant to sera from vaccinated and boosted individuals than BA.2. However, BA.4/5 is substantially (4.2-fold) more resistant and thus more likely to lead to vaccine breakthrough infections. Mutation at spike …

Structural equations modeling, using data from 1,017 American consumers, successfully confirmed the significant influences of trust in media and government on BCI and the corresponding positive effect of BCI on fearfulness. Additionally, the negative effects of fearfulness on intentions to visit …
Mathematical Modeling Of Seir Model With Generalized Incidence Function And The Extension To Covid-19 Model, Shymaa Mohammad Dadoa
Mathematical Modeling Of Seir Model With Generalized Incidence Function And The Extension To Covid-19 Model, Shymaa Mohammad Dadoa
Theses
The COVID-19 pandemic had shown the importance of the SEIR model in predicting the outcome of the disease spread and to find the best strategies to contain the pandemic. As this type of model has a limited number of compartments, many other models were derived from the SEIR model to cover, to the maximum, the complex dynamics of the disease spread. These extensions of the SEIR model bring natural validity questions: How can we validate these models? and how far/close are these extended models from giving us real insights into the pandemic?
This thesis investigates the SEIR epidemic model and …

Cyanobacteria, also known as blue-green algae, are an ancient group of microorganisms that use simple materials, such as sunlight, carbon dioxide and water, to produce energy while providing oxygen to the atmosphere by performing photosynthesis. Synthetic biology approaches have been employed with cyanobacteria as a platform to produce a range of products, such as biofuels, by inserting a series of genes into the cyanobacterial genome that will allow the conversion of metabolic intermediates to such desired products. Although these methods are promising, it is important to understand any potential bio-risk they pose. This research evaluates the potential bio-risk of genetically …

Cyberchondria And Covid-19 Health Threats Related To Attentional Bias And Safety Behaviors, Jacqueline J. Charles
Electronic Theses and Dissertations
Cyberchondria is described as excessively searching online for health information that ultimately increases worry and anxiety about one’s health (Starcevic et al., 2019). Research has demonstrated an increased attentional bias in anxious individuals for threat-related stimuli. Attentional bias for health information exacerbated by the frequent exposure to health threats during the COVID-19 pandemic may contribute to additional health-seeking behavior. The current study aimed to explore the potential relationship between COVID-19 health threats and cyberchondria level on attentional bias toward illness-related stimuli (symptom words) and the intent to perform safety behaviors while controlling for Trait Anxiety and Health Anxiety. Participants ( …

Background:
Some studies have suggested minor changes in the menstrual cycle after COVID-19 vaccination, but more detailed analyses of the menstrual cycle are needed to evaluate more specific changes in the menstrual cycle that are not affected by survey-based recall bias.
Materials and Methods:
Using a pretest–post-test quasi-experimental evaluation of menstrual cycle parameters before and after COVID-19 vaccination, we conducted an anonymous online survey of two groups of North American women who prospectively monitor their menstrual cycle parameters daily including bleeding patterns, urinary hormone levels using the ClearBlue Fertility Monitor, or cervical mucus observations. The primary outcome measures were cycle …
