Neuronal Signal-Regulatory Protein Alpha Drives Microglial Phagocytosis By Limiting Microglial Interaction With Cd47 In The Retina, Danye Jiang, Courtney A Burger, Viktor Akhanov, Justine H Liang, Robert D Mackin, Nicholas E Albrecht, Pilar Andrade, Dorothy P Schafer, Melanie A Samuel

Center on Aging Staff Publications

Microglia utilize their phagocytic activity to prune redundant synapses and refine neural circuits during precise developmental periods. However, the neuronal signals that control this phagocytic clockwork remain largely undefined. Here, we show that neuronal signal-regulatory protein alpha (SIRPα) is a permissive cue for microglial phagocytosis in the developing murine retina. Removal of neuronal, but not microglial, SIRPα reduced microglial phagocytosis, increased synpase numbers, and impaired circuit function. Conversely, prolonging neuronal SIRPα expression extended developmental microglial phagocytosis. These outcomes depended on the interaction of presynaptic SIRPα with postsynaptic CD47. Global CD47 deficiency modestly increased microglial phagocytosis, while CD47 overexpression reduced it. …

GATA-binding factor 1 (GATA1) is a transcription factor that governs the development and function of multiple hematopoietic cell lineages. GATA1 is expressed in hematopoietic stem and progenitor cells (HSPCs) and is essential for erythroid lineage commitment; however, whether it plays a role in hematopoietic stem cell (HSC) biology and the development of myeloid cells, and what that role might be, remains unclear. We initially set out to test the role of eosinophils in experimental autoimmune encephalomyelitis (EAE), a model of central nervous system autoimmunity, using mice lacking a double GATA-site (ΔdblGATA), which lacks eosinophils due to the deletion of the …

Copyright protection provides legal protection to creative industries. One of the creative industries that need to be protected is the video game industry. The development of video games in Indonesia is very fast, therefore legal protection is needed to maintain the development of the video game industry. Basically a video game is an object of creation contained in Law Number 28 of 2014 concerning copyright. In the development of the video game industry, there are problems that occur such as the number of illegal copies of video games. Thus, copyright protection plays a very important role in maintaining the video …

Pastor As Poet Of The Soul, Zachary J. Luben Dec 2022

Pastor As Poet Of The Soul, Zachary J. Luben

Discernment: Theology and the Practice of Ministry

The church finds itself in a culture that challenges the idea of divine action in the world—one of the results of living in a secular age. We are more likely to talk about “good luck” or “coincidence” than we are about God acting in history. Charles Taylor calls this the immanent frame—that we have constructed a social view that frames our lives without the supernatural. In addition, Hartmut Rosa describes modernity as social acceleration, which puts us out-of-sync in our relationships to others and the world. For the pastor, these cultural and social forces at work often lead …

Reclaiming Storage Space By Removal Of Unnecessary Frames From Multi-Frame Photos, Benjamin Azose, Molly Clancy

Defensive Publications Series

Smartphones and other digital cameras enable users to capture photos that include several frames, including some prior to the instant of the user selecting the capture button and some after. This allows motion within the scene to be captured as well as for post-capture selection of a suitable still image, e.g., an image where the subjects are in the correct pose and/or are looking at the camera. However, this is wasteful of storage space when the captured image is of a still object. This disclosure describes techniques to automatically detect such multi-frame photos and enable users to remove excess frames …

Influence Of Environmental Factors On Surface Activity Of Paruroctonus Marksi (Scorpiones: Vaejovidae) In The Mojave Desert, Zia Nisani, Destiny Frederick, Alejandro Garcia-Plascencia, Daian Lopez, Robert Miller, Linhsan Trinh-Nguyen

Euscorpius

Many environmental factors may influence the activity pattern of scorpions in arid ecosystems. We investigated the seasonal variations of Paruroctonus marksi scorpion activity in the Mojave Desert (California, USA). Black light surveys were carried out within two 100 ×100 m plots from August 2017 to September 2018. The majority (85%) of scorpions were found in open areas compared with being in vegetation. The presence of P. marksi was positively correlated with both water vapor pressure and soil temperature, while moon illumination had no significant effect on P. marksi foraging behavior. Scorpions might be using humidity as an indicator of prey …

Towards A Machine Learning-Based Digital Twin For Non-Invasive Human Bio-Signal Fusion, Izaldein Al-Zyoud, Fedwa Laamarti, Xiaocong Ma, Diana Tobón, Abdulmotaleb Elsaddik

Computer Vision Faculty Publications

Human bio-signal fusion is considered a critical technological solution that needs to be advanced to enable modern and secure digital health and well-being applications in the metaverse. To support such efforts, we propose a new data-driven digital twin (DT) system to fuse three human physiological bio-signals: heart rate (HR), breathing rate (BR), and blood oxygen saturation level (SpO2). To accomplish this goal, we design a computer vision technology based on the non-invasive photoplethysmography (PPG) technique to extract raw time-series bio-signal data from facial video frames. Then, we implement machine learning (ML) technology to model and measure the bio-signals. We accurately …

Developing And Utilizing Novel Biofluid N-Glycan Profiling Methods For The Classification Of Suspicious Mammogram Findings, Calvin Blaschke Dec 2022

Developing And Utilizing Novel Biofluid N-Glycan Profiling Methods For The Classification Of Suspicious Mammogram Findings, Calvin Blaschke

MUSC Theses and Dissertations

Biofluids are a great source of biomarkers because they reflect the immune and metabolic status of cells throughout the body and can be collected non-invasively. Changes in the levels and compositions of N-glycans released from serum and plasma glycoproteins have been assessed in many diseases across many large clinical sample cohorts. Assays used for N-glycan profiling in these fluids currently require multiple lengthy processing steps, thus diminishing their potential for use as standard clinical diagnostic assays. In response to this need for rapid, simple, and high-throughput biofluid analysis, a novel slide-based biofluid profiling platform was developed for the detection of …

An Event-Without-Witness: A Nietzschean Theory Of The Digital Will To Power As The Will To Temporalize, Talha Issevenler Dec 2022

An Event-Without-Witness: A Nietzschean Theory Of The Digital Will To Power As The Will To Temporalize, Talha Issevenler

Publications and Research

This article offers a Nietzschean theory of digital will to power to conceptualize the temporality of social media feeds run by algorithms. Stylistic and methodological temporalities of Nietzsche are discussed as well as their influence in subsequent social theory of political technologies. The paradox of heavy investment in both subjective expression and nonhuman temporalization in social media milieus is addressed with the concept of an event-without-witness drawn from Nietzsche’s account of himself as the solitary thinker of catastrophe of nihilism and psychoanalytical and deconstructive literatures on the catastrophes of the 20th century. Nietzsche’s fundamental resistance to and fascination with the …

Noted With Honor: Intersections Between Theology, Music Therapy, Psychotherapy, And Original Music Compositions For Hospice Patients, Stanton Nelson Dec 2022

Noted With Honor: Intersections Between Theology, Music Therapy, Psychotherapy, And Original Music Compositions For Hospice Patients, Stanton Nelson

Doctor of Pastoral Music Projects and Theses

This thesis investigates the intersections between theology, music therapy, and psychotherapy when creating a music composition that honors a hospice patient’s walk of faith. A pioneer organization inspiring this thesis is Swan Songs in Austin, Texas, where musical moments are created for the patient and family. However, its model is primarily based on collating previously composed pieces for recitals without a personalized honoring of the patient’s witness of faith. Noted with Honor is an emerging non-profit organization that creates an original work that reflects upon the testimony of the patient and forms a new narrative towards the end of one’s …

The Presynaptic Role Of Phosphagen Systems, Carlos David Oliva Dec 2022

The Presynaptic Role Of Phosphagen Systems, Carlos David Oliva

Harriet L Wilkes Honors College Theses 2020 - Present

De novo generation of ATP through glycolysis and oxidative phosphorylation is too slow to sustain intense neuronal activity and stabilize ATP levels, suggesting there might be another energy pool at play. The phosphagen system, extensively researched in muscles, is an ATP regeneration pathway; quickly producing ATP through ADP re-phosphorylation. Its presence has been observed in neurons, yet its involvement in neuronal power demands remains poorly understood. To investigate the phosphagen system’s impact on neuronal energy metabolism, we examined Drosophila melanogaster motor neurons, which had arginine kinase knocked down while using genetically-encoded fluorescent reporters of pH, calcium, and ATP. Data obtained …

An Investigation Of Neuronal Cytosolic Ph And Mitochondrial Energy Metabolism In Mdh2 Drosophila Mutants, Frank Andrew Sigui Dec 2022

An Investigation Of Neuronal Cytosolic Ph And Mitochondrial Energy Metabolism In Mdh2 Drosophila Mutants, Frank Andrew Sigui

Harriet L Wilkes Honors College Theses 2020 - Present

Mitochondria are vital organelles essential for proper function of the human body. Mutations in genes encoding mitochondrial proteins lead to mitochondrial dysfunction and severe neurological symptoms. Children reported to have mutations within the gene encoding the enzyme, mitochondrial malate dehydrogenase (MDH2) exhibit symptoms of hypotonia, lactic acidosis and epilepsy. No treatment exists for this disorder and the neurological symptoms stemming from MDH2 mutations require further investigation. Decrease in pH, associated with an increase in lactate production, may contribute to symptoms experienced by children with MDH2 mutations. To investigate pH in individuals with MDH2 mutations, we used CRISPR/Cas9 techniques to introduce …

Place Cells Dynamically Refine Grid Cell Activities To Reduce Error Accumulation During Path Integration In A Continuous Attractor Model, Jose A Fernandez-Leon, Ahmet Kerim Uysal, Daoyun Ji

Faculty, Staff and Students Publications

Navigation is one of the most fundamental skills of animals. During spatial navigation, grid cells in the medial entorhinal cortex process speed and direction of the animal to map the environment. Hippocampal place cells, in turn, encode place using sensory signals and reduce the accumulated error of grid cells for path integration. Although both cell types are part of the path integration system, the dynamic relationship between place and grid cells and the error reduction mechanism is yet to be understood. We implemented a realistic model of grid cells based on a continuous attractor model. The grid cell model was …

Pre-existing diabetes, hypertension and kidney disorders are prominent risk factors of pre-eclampsia (PE). It is a multifactorial pregnancy disorder associated with high blood pressure, proteinuria, and multiorgan failure, which develops after the 20th week of pregnancy. It is one of the most feared pregnancy disorders, as it consumes thousands of fetomaternal lives per annum. According to clinical and pathological studies, the placenta appears to be a key player in the pathogenesis of PE; however, the exact origin of this disorder is still under debate. Defective placentation and angiogenesis are the hallmarks of PE progression. This angiogenic imbalance, together with maternal …

Development Of Functional Markers For Resistance To Smut And Identification Of Genes Differentially Expressed In Response To Brown Rust In Sugarcane, Jose David Cortes

LSU Doctoral Dissertations

Smut, caused by Sporisorium scitamineum and brown rust, caused by Puccinia melanocephala, are two of the most important diseases that affect sugarcane production in Louisiana and worldwide. Smut continues to cause losses in the breeding program in Louisiana. Therefore, a QTL analysis was conducted to identify molecular markers associated with resistance in F1 progeny (162) of a biparental cross between susceptible cultivar L 99-233 and resistant HoCP 96-540. Using 1,574 single-dose SNP markers, a total of 253 linkage groups (LG) were obtained with a genome coverage of 24,580.15 cM. Six QTLs were localized on five LGs. The highest …

Understanding the at-sea movements of wide-ranging seabird species throughout their annual cycle is essential for their conservation and management. Habitat use and resource partitioning of Laysan (Phoebastria immutabilis) and black-footed (Phoebastria nigripes) albatross are well-described during the breeding period but are less understood during the post-breeding period, which represents ~40% of their annual cycle. Resource partitioning may be reduced during post-breeding, when birds are not constrained to return to the nest site regularly and can disperse to reduce competitive pressure. We assessed the degree of spatial segregation in the post-breeding distributions of Laysan (n = 82) and black-footed albatrosses (n …

Does Predation Environment Affect Repeated Responses To Predation Cuesâ In The Fishâ Brachyrhaphis Rhabdophora?, Madeleine S. Nate

Theses and Dissertations

Individual organisms face trade-offs when dealing with predation—more time spent avoiding predators often results in less time allocated to energy acquisition and reproductive-related activities. Individuals that optimize this trade-off and respond appropriately to current risk levels in their environment should be at an advantage. What is less clear is whether this tradeoff changes when individuals are repeatedly exposed to a predation threat. There may be advantages to responding consistently to every signal of threat, but it might also be advantageous to modulate individual behavior. Moreover, it is unclear how evolutionary history of a population might affect such individual responses. Our …

Teaching Social-Emotional Learning To Children With Autism Using Animated Avatar Video Modeling, Emelie Davis Dec 2022

Teaching Social-Emotional Learning To Children With Autism Using Animated Avatar Video Modeling, Emelie Davis

Theses and Dissertations

People with a diagnosis of autism spectrum disorder (ASD) often have difficulties understanding or applying skills related to Social-Emotional Learning (SEL). An individual having a better understanding of SEL concepts is generally associated with more fulfilling connections with others and increased satisfaction in life. Since people with ASD tend to have greater success with learning in structured environments, we created a module to teach these skills using Nearpod. These modules were created with videos of a person embodying a cartoon dog face using Animoji for two reasons; because the animation was meant to appeal to children, and the creation was …
