Periodically, WellBeing News (WBN) carries a story on using animals to test the safety of chemicals (see May 2021 and December 2021). Advocacy campaigns to switch from animal tests to non-animal alternatives started over a hundred years ago. Public policy initiatives and technological advances leading to the implementation of alternatives began several decades ago. The WBN December 2021 story included an illustration of five selected countries showing animal use in laboratories increased rapidly from 1900 to the mid-1970s and then declined almost as rapidly to around 20-40% of peak laboratory animal use. The Gulf Coast tick, Amblyomma maculatum, is a vector of several tick-borne pathogens, including Rickettsia parkeri. The ability of R. parkeri to persist within the tick population through transovarial and transstadial transmission, without apparently harming the ticks, contributes to the pathogen’s perpetuation in the tick population. Previous studies have shown that the R. parkeri load in A. maculatum is regulated by the tick tissues’ oxidant/antioxidant balance and the non-pathogenic tick microbiome. Children with Prader-Willi syndrome (PWS) face a multitude of potential health challenges including life-threatening obesity, endocrinopathies, behavioral and emotional dysregulation, developmental delays, and sleep disorders. In the current perspective piece, we provide a focused review of the condition's etiology and clinical findings, as well as a more in-depth discussion of sleep disorders frequently associated with PWS. In particular, we highlight and discuss difficult clinical scenarios frequently encountered by the pediatric sleep physician caring for this patient population, including diagnosis and treatment of complex sleep-related breathing disorders, considerations for sleep apnea surgery, the interplay between growth hormone and sleep apnea, diagnostic …

This study presents a linguistic profile of a heritage speaker of Russian and recounts the efforts to revitalize his Russian after he had nearly stopped speaking it. The study was conducted over the course of almost two years, starting when the subject was twelve years old and ending when he was fourteen. Although this study found that the subject displayed many of the linguistic features typical of heritage speakers, the subject's Russian had suffered less attrition than is common for heritage speakers of Russian in the United States. Sponges are increasingly recognized as an ecologically important taxon on coral reefs, representing significant biomass and biodiversity where sponges have replaced scleractinian corals. Most sponge species can be divided into two symbiotic states based on symbiont community structure and abundance (i.e., the microbiome), and are characterized as high microbial abundance (HMA) or low microbial abundance (LMA) sponges. Across the Caribbean, sponge species of the HMA or LMA symbiotic states differ in metabolic capacity, as well as their trophic ecology. A metagenetic analysis of symbiont 16 S rRNA and metagenomes showed that HMA sponge microbiomes are more functionally diverse than LMA …

Oropharyngeal and esophageal dysmotility can cause serious clinical complications such as aspiration pneumonia, cachexia, and sarcopenia, with a resulting increase in mortality and disability. The current standard of care for the treatment of SSc-associated swallowing dysfunction is mainly supportive, although severe cases are usually refractory to conventional management. Recent studies have shown that the abnormal production of functional autoantibodies such as anti-cholinergic muscarinic receptor III antibodies may participate in the pathogenesis of SSc-associated gastrointestinal dysmotility and may provide a novel target for therapeutic intervention. This study examines the importance of medical waste management activities for developing a sustainable green healthcare environment. This study applied a multiple methodological approach as follows. A thorough review of the literature was performed to delineate the factors that have been explored for reducing medical waste; hospital staff who handle medical waste were surveyed to obtain their opinions on these factors; the analytic hierarchy process (AHP) was applied to determine the priorities among the identified key factors; and experts’ opinions were consulted to assess the actual applicability of the results derived by the AHP. Targeting The Mdm2-P53 Pathway In Dedifferentiated Liposarcoma, Raymond S Traweek, Brandon M Cope, Christina L Roland, Emily Z Keung, Elise F Nassif, Derek J Erstad

Dedifferentiated liposarcoma (DDLPS) is an aggressive adipogenic cancer with poor prognosis. DDLPS tumors are only modestly sensitive to chemotherapy and radiation, and there is a need for more effective therapies. Genetically, DDLPS is characterized by a low tumor mutational burden and frequent chromosomal structural abnormalities including amplification of the 12q13-15 chromosomal region and the MDM2 gene, which are defining features of DDLPS. The MDM2 protein is an E3 ubiquitin ligase that targets the tumor suppressor, p53, for proteasomal degradation. MDM2 amplification or overexpression in human malignancies is associated with cell-cycle progression and worse prognosis. The MDM2-p53 interaction has thus garnered …

Reorganization of the primary somatosensory cortex (S1) that follows deafferentation may play a role in phantom limb pain (PLP). An animal model of forelimb amputation and subsequent S1 reorganization, like the rat, could advance research in the underlying mechanisms of PLP. The rat grimace scale (RGS) is a method of behavioral analysis used to quantify the affective state. In this study, 8 amputee and 4 sham-operated rats were observed for a minimum of one week pre-surgery and up to 11 weeks post-surgery to score the facial behaviors using RGS. The purpose of this study was to design, develop, and test the effects of digital game-based learning on undergraduate students’ learning motivation, mathematics achievement, and learning retention in a college algebra course. Digital games utilize interactive digital media that offer rules-based challenges and react to the individual’s choices. Digital games have been used in preschool, K-12, higher education, and workplace environments in a wide range of subject areas, including language arts, secondary language acquisition, social studies, science, and math. Neuroblastoma (NB) is a pediatric tumor of the peripheral nervous system. Approximately 80% of relapsed NB show RAS-MAPK pathway mutations that activate ERK, resulting in the promotion of cell proliferation and drug resistance. Ulixertinib, a first-in-class ERK-specific inhibitor, has shown promising antitumor activity in phase 1 clinical trials for advanced solid tumors. Here, we show that ulixertinib significantly and dose-dependently inhibits cell proliferation and colony formation in different NB cell lines, including PDX cells. Transcriptomic analysis revealed that ulixertinib extensively inhibits different oncogenic and neuronal developmental pathways, including EGFR, VEGF, WNT, MAPK, NGF, and NTRK1. These remarks were delivered as part of the program entitled The Presidential Succession Act at 75: Praise It or Bury It?, which was held on April 6, 2022, and hosted by the Fordham University School of Law. The Presidential Succession Act sets out the presidential line of succession and other procedures for situations in which the president and vice president have both died, resigned, been removed, or become unable to discharge the presidency’s powers and duties. Riparian zones are among the most biologically diverse ecosystems in the Intermountain West, USA, and provide valuable ecosystem services, including high rates of biotic productivity, nutrient processing, and carbon storage. Thus, their sustainability is a high priority for land managers. Large ungulates affect composition and structure of riparian/stream ecosystems through herbivory and physical effects, via trailing and trampling. Bison (Bison bison) in Yellowstone National Park (YNP) have been characterized as “ecosystem engineers” because of their demonstrated effects on phenology, aboveground productivity of grasses, and woody vegetation structure. Sous vide cooking is a method of food preparation in which food is vacuum sealed and cooked in a water bath that is set to a precise temperature and circulated by a sous vide device. Due to ease of use and affordability, this cooking method has grown increasingly popular in food service kitchens and domestic settings. In this thesis, polycarbonate (PC) and acrylic (PMMA) surfaces were treated with carbon tetrafluoride (CF4), oxygen (O2), and air plasmas with a low-pressure plasma method in order to change their surface properties such as surface free energy and wettability. Moreover, different pattern sized, line, triangular and square shaped surface patterns were used to determine the shape and size-dependent changes. Alzheimer’s disease (AD) is a progressive neurodegenerative disease that is most prevalent in females. While estrogen provides neuroprotection in females, sex mediated differences in the development of AD pathology are not fully elucidated. Therefore, comparing events between sexes in early-stage AD pathology may reveal more effective therapeutic targets of intervention. To address sex differences, we analyzed early-stage 9-month male and female TgF344-AD (Tg-AD) rats, an AD model carrying the APPswe and Presenilin 1 (PS1ΔE9) mutations that develops progressive age-dependent AD pathology similar to humans. The edible long-horned grasshopper Ruspolia differens Serville (Orthoptera:Tettigoniidae) is a highly nutritious food source consumed in over 20 African countries. Its occurrence is highly seasonal, and wild harvesting is carried out using locally designed and inefficient light traps, thus limiting sustainable utilization as an important food source. To ensure year-round production and availability of R. differens, we evaluated the effects of low-cost and affordable diets based on agricultural by-products on their growth performance, survival, fecundity, and longevity.
