The naturally occurring amino acid, L-Ergothioneine (EGT), has immense potential as a therapeutic, having shown promise in the treatment of other disease models, including neurological disorders. EGT is naturally uptaken into cells via its specific receptor, OCTN1, suggesting that it is highly conserved, to be utilized by cells as an antioxidant and anti-inflammatory, particularly under stressed conditions. In our current study, EGT was administered over a period of 6 months to 25-26 month old CBA/CaJ mice as a possible treatment for age-related hearing loss (ARHL), since presbycusis has been linked to higher levels of cochlear oxidative stress, apoptosis and chronic …

Environmental contamination is accumulating rapidly worldwide due to increasing urbanization rates. Heavy metals are known to be particularly toxic to organisms, to accumulate in biological systems, and to be found in higher concentrations in animals located in areas of high urban levels. However, the physiological impacts of these toxic heavy metals in wildlife remain largely unknown. Lizards are excellent bioindicator species for environmental contaminants due to their trophic level as mesopredators and their susceptibility of exposure via air, water, diet, and soil through egg deposition. Using film in the classroom to teach history has long been endorsed as an effective pedagogical method when the lessons’ purposes and goals are clearly supported with facts. This article, which includes a National Council for the Social Studies C3 inquiry-based lesson plan, is targeted for educators who aspire to help students understand basic European Medieval history and engage in critical thinking. The present research study examines the effectiveness of two types of in two Spanish classrooms for beginner students: planned focus on form and incidental focus on form. Microphototrophs (algae and cyanobacteria) in karst environments have been intensively studied in aquatic epigean habitats. In recent decades knowledge about the communities inhabiting cave entrances and lampenflora has grown substantially, but the data about the communities in aphotic cave zone are scarce. This study aimed to investigate spatio-temporal presence of microphototrophs in the aphotic zone of Veternica Cave (Mt. As COVID-19 has let many students into remote learning environments and exacerbated inequality among marginalized individuals, there is a growing concern about Bias-Based Bullying (BBB) in online spaces among school-aged youths. Learning modality and perceived school fairness may affect youth's likelihood of reporting BBB.

Chronic intraperitoneal injection of thioacetamide (TAA) in rats has been used as an animal model of human cirrhosis to study the effects of the disease on drug metabolism. However, TAA inhibits P450 enzymes directly and independently of cirrhosis. We investigated the effects of chronic cirrhosis in rats, induced by 10 weeks of intraperitoneal TAA, on the P450 enzymes after a 10-day washout period to eliminate TAA. Liver histology and serum biomarkers of hepatic function confirmed cirrhosis in all animals. Microsomal total P450 content, P450 reductase activity and ethoxycoumarin O-deethylase activity, a general marker of P450 activity, were significantly reduced by …

The present study evaluated student academic performance on grammar focused tasks in a Spanish L2 flipped classroom compared to a non-flipped classroom as well as examined learners’ attitudes towards the implementation of the flipped model in the Spanish L2 instruction. This research included a between-group design, which involved a control group being instructed via a flipped approach and a comparison group learning the same material under a non-flipped model; and a within-group design with a group of learners combining these two different methodologies during one semester. It is argued in this article that the legal system, from initial investigation by detectives to final resolution in court by lawyers, judges, and juries, offers a basis for investigating phenomena in the social sciences using mixed methods. We think that this new paradigm combines the components of both the qualitative and quantitative paradigms and provides a practical model for conceptualizing and conducting mixed methods research. Mechanosensitive channels of small conductance, found in many living organisms, open under elevated membrane tension and thus play crucial roles in biological response to mechanical stress. Amongst these channels, MscK is unique in that its activation also requires external potassium ions. To better understand this dual gating mechanism by force and ligand, we elucidate distinct structures of MscK along the gating cycle using cryo-electron microscopy. The heptameric channel comprises three layers: a cytoplasmic domain, a periplasmic gating ring, and a markedly curved transmembrane domain that flattens and expands upon channel opening, which is accompanied by dilation of the periplasmic ring. …

Enterotoxigenic E. coli (ETEC) produce heat-labile (LT) and/or heat-stable (ST) enterotoxins, and commonly cause diarrhea in resource-poor regions. ETEC have been linked repeatedly to sequelae in children including enteropathy, malnutrition, and growth impairment. Although cellular actions of ETEC enterotoxins leading to diarrhea are well-established, their contributions to sequelae remain unclear. LT increases cellular cAMP to activate protein kinase A (PKA) that phosphorylates ion channels driving intestinal export of salt and water resulting in diarrhea. As PKA also modulates transcription of many genes, we interrogated transcriptional profiles of LT-treated intestinal epithelia. In the northern semi-arid region of China, winter survival is always a limiting factor for lucerne production, because low temperatures and a dry climate in winter (Zhou et al., 1993; Ma, 2000; Sun & Gui, 2001; Sun et al., 2003). Diseases partly account for reductions in Astragalus adsurgens, stand longevity. The effect of some cultural practices on the control of pests and diseases have been reported (Hou, 1986; Nan, 1996), but few reports have detailed the relationship among soil fertiliser status, diseases and premature stand decline. To increase the productivity of dairy cattle in the Czech Republic requires an improvement in herbage quality and an increase in the net energy of herbage. The decrease in cattle numbers by 50 % and the expansion of the grassland area both result in a surplus of feedstuffs of low quality. Introduction of strip-seeding of legumes and grasses into grasslands in interaction with fertilisation provides a possible solution to this problem.

Turkmenistan retains a centralized system of livestock production in which many critical assets are owned by the state. Though technically in the temperate zone, the country's climate is harsh and unstable. Groundwater resources are unevenly distributed, leaving many potential grazing areas seasonally inaccessible due to lack of drinking water for livestock. This paper summarizes the results of a three-year study of rangelands, livestock production, flock economics and land tenure at two study sites, one in central and the other in eastern Turkmenistan. This research was aimed at identifying a bacterium that can produce alkaline proteases. As a result, bacteria that produce proteases were isolated from Batu tannery effluents, tested for protease synthesis on skim milk agar plates, and validated with a protease assay. Microscopic and molecular phylogenetic analyses identified Paenibacillus dendritiformis (BT7) as the bacterial isolate with the highest alkaline protease production. The isolate's maximum enzyme production was obtained by 2% inoculum size, 40°C temperature, 9.0 pH, and a 48-hour incubation time with production media components such as glucose, casein, MgCl2, and 2% NaCl. Resolutions and self-promises are two much discussed tools of self-constraint in the face of weakness of will. However, the discussions often begin from a negative and alienated direction, emphasizing self-compulsion, fear of irrationality, or binding ourselves through self-obligation. Jorah Dannenberg has suggested a more optimistic agent-centered account of how to bind ourselves through self-promises. Increasing utilization of nuclear power enhances the risks associated with industrial accidents, occupational hazards, and the threat of nuclear terrorism. Exposure to ionizing radiation interferes with genomic stability and gene expression resulting in the disruption of normal metabolic processes in cells and organs by inducing complex biological responses. Exposure to high-dose radiation causes acute radiation syndrome, which leads to hematopoietic, gastrointestinal, cerebrovascular, and many other organ-specific injuries. Altered genomic variations, gene expression, metabolite concentrations, and microbiota profiles in blood plasma or tissue samples reflect the whole-body radiation injuries. Schistosoma haematobium, the agent of urogenital schistosomiasis, and related schistosomes are transmitted by members of the genus Bulinus. Each of the 38 Bulinus species vary in their ability to vector schistosome species and non-schistosome trematodes resulting in a patchwork of snail-parasite compatibilities. Accurately identifying snail intermediate hosts and the disease-causing parasites they transmit is critical for snail control strategies and the management of human schistosomiasis. Towards these ends, this thesis identifies bulinid species and the parasites they transmit and implicates certain species in the transmission of S. haematobium. Caspase-8 Inactivation Drives Autophagy-Dependent Inflammasome Activation In Myeloid Cells., Yung-Hsuan Wu, Shu-Ting Mo, I-Ting Chen, Fu-Yi Hsieh, Shie-Liang Hsieh, Jianke Zhang, Ming-Zong Lai

Caspase-8 activity controls the switch from cell death to pyroptosis when apoptosis and necroptosis are blocked, yet how caspase-8 inactivation induces inflammasome assembly remains unclear. We show that caspase-8 inhibition via IETD treatment in Toll-like receptor (TLR)-primed Fadd-/-Ripk3-/- myeloid cells promoted interleukin-1β (IL-1β) and IL-18 production through inflammasome activation. Caspase-8, caspase-1/11, and functional GSDMD, but not NLRP3 or RIPK1 activity, proved essential for IETD-triggered inflammasome activation. Autophagy became prominent in IETD-treated Fadd-/-Ripk3-/- macrophages, and inhibiting it attenuated IETD-induced cell death and IL-1β/IL-18 production. "The Village That Learns": A Learning Journey Across Intraventions And Domains Over Two Decades In A Rural Thai Community, Deborah A. Fields, Luis Morales-Navarro, Paulo Blikstein

What does it mean to become a village that learns? In this paper we document the transformative learning journey of a small Thai village over 24 years, becoming a community that identified, tackled, and iterated on problems, altering their everyday practices and lives. In that process the village shifted from a subsistence agricultural community staggeringly in debt to one known for its sustainable environmental, agricultural, and financial initiatives.
