In Silico Analysis Of Tetrodotoxin Binding In Voltage-Gated Sodium Ion Channels From Toxin-Resistant Animal Lineages, Shana L. Geffeney, Jennie Ann Cordingley, Kenyon Mitchell, Charles T. Hanifin

Multiple animal species have evolved resistance to the neurotoxin tetrodotoxin (TTX) through changes in voltage-gated sodium ion channels (VGSCs). Amino acid substitutions in TTX-resistant lineages appear to be positionally convergent with changes in homologous residues associated with reductions in TTX block. We used homology modeling coupled with docking simulations to test whether positionally convergent substitutions generate functional convergence at the level of TTX–channel interactions. We found little evidence that amino acids at convergent positions generated similar patterns among TTX-resistant animal lineages across several metrics, including number of polar contacts, polar contact position, and estimates of binding energy. The pioneer transcriptional factors (PTFs) of the Oct3/4 network including Oct3/4, Nanog, Sox2, Sall4 (ONSS), have been associated with breast cancer. Regulation of ONSS and other factors in this network were assessed for their role in malignancy. M ethods Triple negative breast cancer cell line (MDA-MB-231) transfected with human Oct3/4-GFP promoter was sorted using FACS. Differentially expressed genes (DEGs) were identified using qPCR and microarray. 3D mammospheres (CSC) from Oct3/4(+) cells were assessed for stable Oct3/4 expression. Studies demonstrated genetic and epigenetic associations with eczema. However, not many studies investigated the interaction of epigenetic and genetic associations with eczema. Additionally, few studies examined gene expression (GE) levels along with epigenetic and genetic associations with eczema. Using the multigenerational epigenetic, genetic, and GE data of Isle of Wight (IOW) birth cohort, I investigated the interplay between DNA methylation (DNAm) and a genetic variant (SNP) with eczema and GE. Invasive Lonicera maackii (L. maackii) is one of the highly successful and problematic bush honeysuckles found in the central and eastern of United States of America, which has been reported to pose a threat to native ecosystems by decreasing biodiversity. The mechanism by which L. maackii negatively impact environments is typically through either the direct effect of increased dominance or the indirect effect of territory modification. Numerous studies have documented the negative effects of L. maackii on native biota and the key traits such as seed dispersal, phenology, resistance to herbivory, rapid growth and environmental plasticity that contribute to invasion …

While many churches today have moved away from the practice of church-based women’s ministries, other churches still see their value and continue their dedication to providing specifically tailored programs to address the spiritual formation in women. The Bible reminds us in Hebrews 10:25 that we are not to forsake “the assembling of ourselves together, as the manner of some…” (King James Bible, 1769/2017, Heb. 10:25). The purpose of this qualitative hermeneutic phenomenology study was to describe the lived experiences of male early childhood teachers in the northeast region of the United States (US). The theory guiding this study was Victor Vroom’s expectancy theory of motivation. Vroom’s theory explains the behavioral process of why individuals choose one behavior over another or one career over another. The central research question in this study addressed the male early childhood teachers’ experiences working in the early childhood education (ECE) career field and motivation to remain in the ECE career field. Deep neural networks have been part of many breakthroughs in computer graphics and vision research. In the context of visual content synthesis, deep learning models have achieved impressive performance in the image domain. However, adapting the successes of image synthesis models to the video domain has been difficult, arguably due to the lack of sufficiently strong inductive biases that encourage the models to capture the temporal-dynamic nature of video data. The incidence of many stress-elicited disorders is markedly influenced by sex. Women are twice as likely as men to develop posttraumatic stress disorder (PTSD), depression, anxiety disorders, and social impairments following exposure to traumatic stress. However, most of the studies in animal models examining putative therapeutics for stress-triggered impairments, including single prolonged stress (SPS), were performed predominantly with males. Previous studies in males demonstrated that intranasal neuropeptide Y (NPY) can provide therapeutic relief of many SPS-triggered symptoms. Nevertheless, the overwhelming majority of studies found that NPY levels in females in many brain regions are lower than in male rodents. NPY …

International Journal of Exercise Science 15(3): 1528-1537, 2022. This study aimed to investigate the effect of biomarkers of oxidative stress (OS) in 8-isoprostane (8-iso) and 8-hydroxy-2'–deoxyguanosine (8-OH-dG) on mild cognitive impairment (MCI) during a 12-week Ruesi Dadton (RD) exercise. A total of 274 enrolled participants were classified into blocks based on age and formal educational years, and randomly assigned into two groups: RD and control (CON). The participants’ cognitive functions were tested using Mini-Mental State Examination and Montreal Cognitive Assessment (MoCA) scores to screen for MCI. In TransUnion, LLC v. Ramirez, the United States Supreme Court held that “in a suit for damages, mere risk of future harm, standing alone, cannot qualify as a concrete harm—at least unless the exposure to the risk of future harm itself causes a separate concrete harm.” Notably, the Court failed to take a position on what common law analog would suffice as a “separate concrete harm,” and instead, affirmed that an emotional distress injury caused by the risk of future harm may be a sufficient concrete injury. Atrial arrhythmias are common in transthyretin cardiac amyloidosis (ATTR-CA), with a prevalence of ≤80%. They are often not well tolerated. We describe 3 patients with decompensated heart failure and cardiogenic shock precipitated by atrial arrhythmias who ultimately received diagnoses of ATTR-CA. (Level of Difficulty: Intermediate.)

This case study was completed in a lake community in the upper midwestern United States. It seeks to understand the social factors that allowed a lake community to establish an effort to protect against a zebra mussel invasion, as well as the perceptions of the different stakeholders and the steps they took to get the effort going. Rural community action is important for the sustainability of many of the world’s natural resources as these communities are often the first line of defense. The use of cephalopod beaks in ecological and population dynamics studies has allowed major advances of our knowledge on the role of cephalopods in marine ecosystems in the last 60 years. Since the 1960’s, with the pioneering research by Malcolm Clarke and colleagues, cephalopod beaks (also named jaws or mandibles) have been described to species level and their measurements have been shown to be related to cephalopod body size and mass, which permitted important information to be obtained on numerous biological and ecological aspects of cephalopods in marine ecosystems. People living with HIV (PLWH) represent a vulnerable population to adverse musculoskeletal outcomes due to HIV infection, antiretroviral therapy (ART), and at-risk alcohol use. Developing measures to prevent skeletal degeneration in this group requires a grasp of the relationship between alcohol use and low bone mass in both the PLWH population and its constituents as defined by sex, age, and race. Coronavirus disease 2019 (COVID-19) has been spreading rapidly and is still threatening human health currently. A series of measures for restraining epidemic spreading has been adopted throughout the world, which seriously impacted the gross domestic product (GDP) globally. However, details of the changes in the GDP and its spatial heterogeneity characteristics on a fine scale worldwide during the pandemic are still uncertain. We designed a novel scheme to simulate a 0.1° × 0.1° resolution grid global GDP map during the COVID-19 pandemic. Not all patients with cancer and severe neutropenia develop fever, and the fecal microbiome may play a role. In a single-center study of patients undergoing hematopoietic cell transplant (n = 119), the fecal microbiome was characterized at onset of severe neutropenia. A total of 63 patients (53%) developed a subsequent fever, and their fecal microbiome displayed increased relative abundances of Akkermansia muciniphila, a species of mucin-degrading bacteria (P = 0.006, corrected for multiple comparisons). Two therapies that induce neutropenia, irradiation and melphalan, similarly expanded A. muciniphila and additionally thinned the colonic mucus layer in mice. Caloric restriction …

Group 2 innate lymphoid cells (ILC2) are innate counterparts of T helper 2 (Th2) cells that maintain tissue homeostasis and respond to injuries through rapid interleukin (IL)-5 and IL-13 secretion. ILC2s depend on availability of arginine and branched-chain amino acids for sustaining cellular fitness, proliferation, and cytokine secretion in both steady state and upon activation. The Ediacara Biota-the oldest communities of complex, macroscopic fossils-consists of three temporally distinct assemblages: the Avalon (ca. 575-560 Ma), White Sea (ca. 560-550 Ma), and Nama (ca. 550-539 Ma). Generic diversity varies among assemblages, with a notable decline at the transition from White Sea to Nama. Preservation and sampling biases, biotic replacement, and environmental perturbation have been proposed as potential mechanisms for this drop in diversity. Here, we compile a global database of the Ediacara Biota, specifically targeting taphonomic and paleoecological characters, to test these hypotheses.
