Predicting The Unexpected: Applying Advanced Underwriting To Accurately Predict Early Duration Claims In Life Insurance, Jessica Howe May 2020

Predicting The Unexpected: Applying Advanced Underwriting To Accurately Predict Early Duration Claims In Life Insurance, Jessica Howe

Honors Scholar Theses

Early duration claims, which are defined as claims that are filed within the first five years of a life insurance policy's issuance, are a large potential risk with high costs that life insurers remain exposed to, yet there has not been any significant advancement on how to properly budget for these claims. The early duration claims model seeks to address this problem by creating a more thorough underwriting procedure that specifically assesses an applicant’s likelihood of early death. The underwriting questions aim to capture risks that are not assessed by the current life insurance underwriting process and may be utilized …

Characterization Of A Mycoplasma Pneumoniae Cards Toxin Mutant, Nikaash Pasnoori May 2020

Characterization Of A Mycoplasma Pneumoniae Cards Toxin Mutant, Nikaash Pasnoori

Honors Scholar Theses

Mycoplasma pneumoniae is a high-burden pathogen which causes mild to significant infections of the respiratory system. According to the CDC, an estimated two million cases occur yearly in the United States alone, demonstrating the widespread effect of the pathogen. In addition to being the cause of respiratory infections, M. pneumoniae has also been implicated in exacerbating pre-existing asthma conditions. These morbidities make finding a vaccine candidate a vital part of easing the healthcare burden caused by the pathogen. The current mechanism of infection is unknown, but recent evidence points to the Community Acquired Respiratory Distress Syndrome (CARDS) toxin as being …

Detection And Enumeration Of Bacterial Pathogens In The American Oyster (Crassostrea Virginica), Mohammad Maruf Billah May 2020

Detection And Enumeration Of Bacterial Pathogens In The American Oyster (Crassostrea Virginica), Mohammad Maruf Billah

Theses and Dissertations

American oyster (Crassostrea virginica) is a popular seafood for its delicacy and high nutritional value. Based on increasing concern about contamination of bacterial pathogens in raw oyster, my research objectives have been focused on detection and enumeration of two important bacterial pathogens, Escherichia coli and Salmonella spp. in the American oyster in south Texas waters, local markets and controlled laboratory studies. Immunohistochemical and RT-PCR analyses showed substantial bacterial pathogen’s presence in gills and digestive glands of oysters collected from San Martin Lake and South Padre Island as well as local markets. Laboratory studies showed increasing trend of both …

The Molluscum Contagiosum Virus Targets Host Innate Immune Signaling Networks At Multiple Points Using Viral Proteins Mc160 And Mc163, Catherine D. Suarez May 2020

The Molluscum Contagiosum Virus Targets Host Innate Immune Signaling Networks At Multiple Points Using Viral Proteins Mc160 And Mc163, Catherine D. Suarez

Seton Hall University Dissertations and Theses (ETDs)

The Molluscum Contagiosum Virus (MCV) causes a common skin infection. A member of the Poxviridae family, MCV spreads through skin to skin contact and produces benign skin lesions that persist for months in otherwise healthy individuals. Despite its prevalence, MCV remains understudied, and its pathogenicity is poorly understood. Therefore, a more detailed understanding of MCV-host interactions is of great importance. The MCV genome encodes a variety of immune evasion molecules to antagonize host cell innate immune responses (IIRs). Two such MCV immune evasion molecule are the MC160 and MC163 proteins. MC160 contains two tandem death effector domains (DEDs). DEDs are …

Challenging Social Systems Under The Threat Of Pollution: Replication And Extension Of Eadeh And Chang (2019), Dylan J. Pieper May 2020

Challenging Social Systems Under The Threat Of Pollution: Replication And Extension Of Eadeh And Chang (2019), Dylan J. Pieper

Dissertations and Theses @ UNI

I replicated and extended Eadeh and Chang (2019; Study 2) who found that reading a story about pollution harming a child increased self-reported liberalism and increased support for environmental regulation policies through greater negative affect (i.e., anger and disgust) (Hypothesis 1). Eadeh and Chang’s (2019) threat-affordance model posits that perceived threat can increase greater support for liberalism or greater support for conservatism, depending on the type of threat. In contrast, Jost and colleagues’ (2003) system justification theory posits that perceived threat asymmetrically predicts greater support for conservatism and greater systemsupporting collective action. This article advances a synthesis of these two …

Comparative Host Response And Gene Regulation In Deer Mice And Syrian Golden Hamsters During Acute Infection With Modoc Virus, Tyler Sherman May 2020

Comparative Host Response And Gene Regulation In Deer Mice And Syrian Golden Hamsters During Acute Infection With Modoc Virus, Tyler Sherman

Dissertations

Flaviviruses (family Flaviviridae, genus Flavivirus) comprise a diverse group of globally-distributed viruses that may specifically infect vertebrates, invertebrates, or both. Of those that can replicate in both invertebrates (such as ticks and mosquitoes) as well as vertebrates (such as mammals), many represent important human pathogens. Examples of these include dengue virus (DENV), which may cause severe hemorrhagic fevers, West Nile virus (WNV), Japanese encephalitis virus (JEV), and Zika virus (ZIKV), all of which may cause febrile illness with neurologic involvement, and yellow fever virus (YFV), which may induce hepatic dysfunction. Given that these viruses (among others) are carried and transmitted …

The Impact Of State-Level Laws On Syringe Service Program Access And Risk Environment Of People Who Inject Drugs (Pwid), Samuel Pettyjohn May 2020

The Impact Of State-Level Laws On Syringe Service Program Access And Risk Environment Of People Who Inject Drugs (Pwid), Samuel Pettyjohn

Electronic Theses and Dissertations

Background: Understanding concentrated areas with high rates of opioid use disorder (OUD) allows for improved placement of Narcan access points through syringe services programs (SSPs). People Who Inject Drugs (PWID) have lower risk of contracting infectious diseases the closer they are to SSPs. Tennessee law prohibits SSPs within 2000ft of a school or park, impacting the placement of SSPs in non-urban areas. Testing factors related to SSP siting placement within a system dynamic model can better determine the relationship between PWID risk environment and SSP access and utility.

Methods: We identified areas of greatest need for harm reduction interventions within …

Assessing The Effect Of Bile And Oxygen Availability On The Redox Status Of Listeria Monocytogenes Strain F2365, Sukriti Bhattarai May 2020

Assessing The Effect Of Bile And Oxygen Availability On The Redox Status Of Listeria Monocytogenes Strain F2365, Sukriti Bhattarai

Honors Theses

Although bile is a bactericidal agent able to disrupt membrane structure and cellular homeostasis, including the induction of oxidative stress, Listeria can tolerate bile and also utilize it as a signal to enhance infection and virulence. Preliminary findings showed that under anaerobic conditions, exposure to bile significantly lowered the amount of oxidative damage present in bile-resistant strain F2365 cells. Similarly, Listeria further elicits an adaptive immune response, wherein pre-exposure of the bacterium to stress during food processing or in the host prior to entry into the intestine increases bile tolerance. Based on these previous studies, we hypothesized that bile induces …

Parallel Evolution Of Tobramycin Resistance Across Species And Environments, Michelle R. Scribner, Alfonso Santos-Lopez, Christopher Marshall, Christopher Deitrick, Vaughn S. Cooper

Biological Sciences Faculty Research and Publications

Different species exposed to a common stress may adapt by mutations in shared pathways or in unique systems, depending on how past environments have molded their genomes. Understanding how diverse bacterial pathogens evolve in response to an antimicrobial treatment is a pressing example of this problem, where discovery of molecular parallelism could lead to clinically useful predictions. Evolution experiments with pathogens in environments containing antibiotics, combined with periodic whole-population genome sequencing, can be used to identify many contending routes to antimicrobial resistance. We separately propagated two clinically relevant Gram-negative pathogens, Pseudomonas aeruginosa and Acinetobacter baumannii, in increasing concentrations of …

Vestigial-Like 1 Is A Shared Targetable Cancer-Placenta Antigen Expressed By Pancreatic And Basal-Like Breast Cancers, Sherille Denae Bradley

Dissertations and Theses (Open Access)

Cytotoxic T lymphocyte (CTL)-based cancer immunotherapies have shown great promise for inducing clinical regression by targeting tumor-associated antigens (TAA). To expand the TAA landscape of pancreatic ductal adenocarcinoma (PDAC), we performed tandem mass spectrometry analysis of HLA class I-bound peptides from tumors of PDAC patients. This led to the identification of a shared HLA-A*0101 restricted peptide derived from co-transcriptional activator Vestigial-like 1 (VGLL1), a novel putative TAA demonstrating overexpression in multiple tumor types and low or absent transcript expression in normal tissues with the exception of placenta. VGLL1-specific CTL isolated and expanded from the blood of a male PDAC patient …

Heterogeneous Nuclear Ribonucleoprotein K (Hnrnp K) Overexpression And Its Interaction With Runx1 Rna In Acute Myeloid Leukemia, Marisa Aitken

Dissertations and Theses (Open Access)

Acute myeloid leukemia (AML) is an often devastating hematologic malignancy with 5-year overall survival lingering near 20%. Acquiring a deeper understanding of molecular underpinnings of leukemogenesis will provide a basis for developing more effective therapeutic strategies for patients with AML.

Here, we identified overexpression of hnRNP K as a recurrent abnormality in a subset (~20%) of AML patients. High levels of this RNA-binding protein associated with inferior clinical outcomes in de novo AML. Thus, to evaluate its putative oncogenic capacity in myeloid disease, we overexpressed hnRNP K in murine hematopoietic stem and progenitor cells isolated from fetal liver cells (FLCs). …

Impact Of Epa And Dha Supplementation And 15-Lox-1 Expression On Colitis And Colitis-Associated Colorectal Cancer, Jonathan Jaoude

Dissertations and Theses (Open Access)

Inflammatory bowel disease (IBD) patients not only suffer from colitis but also from increased morbidity and mortality of colitis-associated colorectal cancer (CAC). The enzyme 15-lipoxygenase-1 (15-LOX-1) is crucial to converting omega-3 fatty acid derivatives eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) to resolvins, potent anti-inflammatory products. 15-LOX-1 effects on the conversion of EPA and DHA to resolvins that subsequently exert anti-inflammatory and anti-tumorigenic effects have received little attention. To address this knowledge gap, we hypothesize that 15-LOX-1 expression in colonic epithelial cells is essential for resolvin biosynthesis from EPA and DHA to modulate immunophenotype, limit inflammation, promote resolution, and help …

Exosomal Communication By Metastatic Osteosarcoma Cells Modulates Alveolar Macrophages To An M2 Tumor-Promoting Phenotype And Inhibits Tumoricidal Functions, Kerri Wolf May 2020

Exosomal Communication By Metastatic Osteosarcoma Cells Modulates Alveolar Macrophages To An M2 Tumor-Promoting Phenotype And Inhibits Tumoricidal Functions, Kerri Wolf

Dissertations and Theses (Open Access)

Osteosarcoma metastasizes to the lung, and there is a link between the predominance of tumor promoting immunosuppressive M2 macrophages in the metastases and poor patient survival. By contrast, M1macrophage predominance correlates with longer survival. M2 macrophages can be induced by various stimuli in the tumor microenvironment, including exosomes, which are 40- to 150-nm vesicles that are involved in intercellular communication and contribute to tumor progression and immune evasion. Recognizing that tumor cells can influence the tumor microenvironment to make it more permissive and because of the link between M2 dominance and curtailed patient survival, we evaluated the effect of …

Loss Of Caspase-8 Function In Combination With Smac Mimetic Treatment Sensitizes Head And Neck Squamous Carcinoma To Radiation Through Induction Of Necroptosis., Burak Uzunparmak

Dissertations and Theses (Open Access)

Caspase-8 (CASP8) is one of the most frequently mutated genes in Head and Neck Squamous Carcinomas (HNSCC), and mutations of CASP8 are associated with poor overall survival. The distribution of these mutations in HNSCC suggests that they are likely to be inactivating. Inhibition of CASP8 has been reported to sensitize cancer cells to necroptosis, a unique cell death mechanism. Here, we evaluated how CASP8 regulates necroptosis in HSNCC using cell line models and syngeneic mouse xenografts. In vitro, knockdown of CASP8 rendered HNSCCs susceptible to necroptosis induced by a second mitochondria-derived activator of caspase (SMAC) mimetic, Birinapant, when combined …

Public Healthcare Crisis In Venezuela, Valerie Bittleston May 2020

Public Healthcare Crisis In Venezuela, Valerie Bittleston

Ballard Brief

The public healthcare crisis in Venezuela has been compared to the emergency status primarily declared on countries at war. Lack of investment in their free public healthcare system has depleted the country of medical personnel, infrastructure, and supplies. Limited to no access to essential medicines and healthcare services have increased the spread of infectious diseases. Venezuelans often migrate in order to seek aid elsewhere, carrying previously eradicated diseases with them. Mother and infant health is of utmost concern as mortality rates are increasing at exorbitant rates with no end in site. The government implemented a public healthcare program in 2003 …

Indemnifying Precaution: Economic Insights For Regulation Of A Highly Infectious Disease, Christopher Robertson, K Aleks Schaefer, Daniel Scheitrum, Sergio Puig, Keith Joiner May 2020

Indemnifying Precaution: Economic Insights For Regulation Of A Highly Infectious Disease, Christopher Robertson, K Aleks Schaefer, Daniel Scheitrum, Sergio Puig, Keith Joiner

Faculty Scholarship

Economic insights are powerful for understanding the challenge of managing a highly infectious disease, such as COVID-19, through behavioral precautions including social distancing. One problem is a form of moral hazard, which arises when some individuals face less personal risk of harm or bear greater personal costs of taking precautions. Without legal intervention, some individuals will see socially risky behaviors as personally less costly than socially beneficial behaviors, a balance that makes those beneficial behaviors unsustainable. For insights, we review health insurance moral hazard, agricultural infectious disease policy, and deterrence theory, but find that classic enforcement strategies of punishing noncompliant …

Characterization Of Microbial Communities Across Disease States And Environmental Conditions In Kemp’S Ridley (Lepidochelys Kempii) And Green Sea Turtles (Chelonia Mydas), Kerry L. Mcnally May 2020

Characterization Of Microbial Communities Across Disease States And Environmental Conditions In Kemp’S Ridley (Lepidochelys Kempii) And Green Sea Turtles (Chelonia Mydas), Kerry L. Mcnally

Graduate Doctoral Dissertations

All species of sea turtles are threatened or endangered, with various diseases and conditions affecting populations around the world. Understanding healthy populations as well as populations beset by disease conditions, such as fibropapillomatosis and cold-stunning, could lead to helpful tools in the conservation management and medical treatment needed to protect these species. Microbial communities, or the microbiome, at different body sites of sea turtles likely play important roles in the health of these animals, from aiding in digestion to immune system regulation. Disruption of these communities, either through disease and/or environmental factors, may play a role in disease processes and …

Examination Of The Polymicrobial Interaction: Inhibitory Effects Of Alcaligenes Species On Members Of The Candida Species, Madelyn Whitlock May 2020

Examination Of The Polymicrobial Interaction: Inhibitory Effects Of Alcaligenes Species On Members Of The Candida Species, Madelyn Whitlock

Undergraduate Honors Theses

Candida species are commonly found in the human normal flora, however they are a major cause of nosocomial infections that can be life threatening. This fungal species is an opportunistic pathogen and causes infection in individuals who are immunosuppressed. A key characteristic of Candida’svirulence is the ability to change its morphology from ovoid yeast to filamentous hyphae. Alcaligenes species are common bacteria found in the environment that rarely, if at all, cause infections in humans. It has been observed that when allowed to interact, Alcaligenes faecalis changes the morphology of Candida albicans from yeast cells to hyphal cells. When …

Synergistic Inhibition Of Resistant Enterobacteriaceae Using A Possible Klebsiella Secreted Bacteriocin With Broad-Spectrum Antibiotic, Andrew Robbins May 2020

Synergistic Inhibition Of Resistant Enterobacteriaceae Using A Possible Klebsiella Secreted Bacteriocin With Broad-Spectrum Antibiotic, Andrew Robbins

Undergraduate Honors Theses

Due to the increasing prevalence of multi-drug resistant (MDR) bacteria, it is now important to begin the search for novel means of defending against such resistant infections. Enterobacteriaceae is a clinically relevant family of bacteria that has shown extensive resistance to many antibiotics, especially after biofilm formation. Inhibitory poly-microbial interactions within this family have been observed. It is known that Citrobacter freundii (CF) growth is significantly inhibited by Klebsiella pneumoniae (KP) through a secreted protein. In this study, the potential KP bacteriocin was screened for its inhibitory effects on CF at various phases of biofilm development. The suspected KP bacteriocin …

Chloroquine (CQ) and Hydroxychloroquine (HCQ) have been proposed to be effective at treating COVID-19 patients. We, and others, have previously reported on the capacity of CQ to reduce HIV-1 replication in vitro. We tested CQ administration in post-partum mothers on influencing HIV-1 viral loads in human milk as a means of lowering mother to child transmission. A Phase I/II, randomized, placebo-controlled study to evaluate chloroquine administration to reduce HIV-1 RNA levels in human milk: the CHARGE study. Thirty HIV-1 positive pregnant Rwandese women (CQ n = 20; placebo n = 10) were enrolled in a 16-week study, with the treatment …

Chronic Pain And History Of Trauma, Lauren K. Grizzard May 2020

Chronic Pain And History Of Trauma, Lauren K. Grizzard

Electronic Theses and Dissertations

As literature has shown, chronic pain patients present with a unique challenge to the medical field. The existing question is if the patient’s pain is unmanageable because of the severity of the pain or if the patient lacks coping mechanisms. If there is a lack of coping mechanisms, then that raises a plethora of new questions for the medical teams caring for them. After IRB approval, the current study was conducted on a sample of three patients that were referred to the Hendrick Medical Center Pathways team for pain management. It sought to determine if there was any association between …

Efficient Conditional Inactivation Of Rdh10 Reveals Important Role Of Retinoic Acid During Lung Branching Morphogenesis., Nhut Quang Huy Tran

Electronic Theses and Dissertations

Birth defects are complicated pathological processes. Many birth defects can be traced back to disrupted regulation of molecular signaling pathways during embryogenesis. One of the most important gene regulatory factors during embryo development is retinoic acid (RA). However, studying the specific roles of RA in each organ during embryogenesis in vivo is challenging. The Sandell laboratory has a conditional mutant mouse model that can potentially be used for studying the role of RA during embryogenesis by inducing stage-specific RA deficiency. This model allows retinol dehydrogenase 10 (Rdh10), a gene required for RA production, to be inactivated at a …

A larger number of people reside outside their country of birth today than in any other time in history, and these levels are expected to continue to rise in the future (International Organization for Migration, 2011). The United States is currently the most popular immigrant destination in the world, with more immigrants than any other country. Immigrants statistics indicate that the immigrant population in the United States reached a record of 44.4 million in 2017 (Pew Research Centre, 2019). Correspondingly, the percentage of immigrants who were admitted into the U.S. as spouses of citizens has increased over of the last …

Development Of In Vitro Models To Study The Rapid Extraintestinal Dissemination Of Salmonella., Adarsh Gopinath May 2020

Development Of In Vitro Models To Study The Rapid Extraintestinal Dissemination Of Salmonella., Adarsh Gopinath

Electronic Theses and Dissertations

Salmonella appears in the bloodstream of mice in as little as 15 minutes after oral inoculation and establishes persistent colonies in the spleen and liver. While its pathway to blood is undetermined, this phenomenon is dependent on the activity of Salmonella pathogenicity island 2 (SPI-2) coded type III secretion system (T3SS) and CD18+ phagocytes. We hypothesize that dendritic cells associated with the basal face of the gut epithelium, that are naturally migratory and known to sample for luminal antigens directly transport Salmonella to the bloodstream. This process comprises of at least two phases, dissociation and reverse transmigration. We define dissociation …

Development Of Rationally Designed Live Attenuated Vaccines For Lassa Fever And Venezuelan Equine Encephalitis., Dylan Macgregor Johnson

Electronic Theses and Dissertations

Lassa Virus (LASV) and Venezuelan Equine Encephalitis Virus (VEEV) are two single stranded RNA viruses belonging to the Arenavirus and Alphavirus families, respectively. Both are emerging pathogens without approved vaccines or treatments. VEEV is an important biothreat pathogen because it has been weaponized and is extremely infectious as an aerosol. VEEV causes biphasic febrile illness that can progress to a viral encephalitis with low mortality, but high morbidity. LASV causes a viral hemorrhagic fever, Lassa Fever (LF), which is endemic in West Africa, and responsible for between 100-500k annual infections with a 1-2% overall mortality rate. ML29 and VEEV TC-83 …

Context: Unhealthy lifestyle habits are a significant risk factor for chronic disease and death in the United States of America. The healthcare system is largely focused on symptom-management and disease treatment with seemingly little regard for prevention, but physical therapists are particularly well-placed to improve patients’ health through lifestyle counseling and modification. Objectives: To assess the current state of holistic health promotion among physical therapists and to identify barriers to improving these practices. Design: An electronic survey was completed by physical therapists in and around the state of Kentucky. Descriptive statistics were used to assess participant demographics and the state …

Reversible Inhibition Of Mycobacterial Dnab Protein Splicing By Zinc, Gabrielle Hardison May 2020

Reversible Inhibition Of Mycobacterial Dnab Protein Splicing By Zinc, Gabrielle Hardison

Honors College Theses

Inteins are emerging as post-translational regulatory elements, undergoing conditional protein splicing in response to a variety of environmental cues. Inteins are seamlessly removed by self-splicing from exteins, or flanking portions of the host protein, which they interrupt. DnaB of Mycobacterium smegmatis, a helicase essential for replication, harbors two inteins known as DnaBi1 and DnaBi2, each with discrete structural characteristics and insertion positions. DnaBi1 was used here to design a reporter system which links splicing with resistance to the antibiotic kanamycin. We built a construct that strictly requires catalytically active DnaBi1 for survival in the presence of kanamycin and used …
