Tailoring Cell-Material Interactions Via Poly(Acrylic Acid) Brushes To Enhance Nonviral Substrate-Mediated Gene Delivery, Amy Mantz Jul 2019

Tailoring Cell-Material Interactions Via Poly(Acrylic Acid) Brushes To Enhance Nonviral Substrate-Mediated Gene Delivery, Amy Mantz

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Nonviral gene delivery modifies gene expression by transferring exogenous genetic material into cells and tissues, typically through a bolus of complexes formed by electrostatic interactions between cationic lipid or polymer vectors with negatively charged nucleic acids (e.g. DNA). Although nonviral gene delivery is safer, more cost-effective, and more flexible compared to viral systems, nonviral transfection suffers from low efficiency due to extracellular and intracellular barriers. Much research has focused on tuning physiochemical properties of the complexing vectors to improve transfection, yet the cell-material interface may prove a better platform to immobilize DNA complexes for substrate-mediated delivery (SMD) and modulate the …

Relative Density And Resource Selection Of Urban Red Foxes In Lincoln, Nebraska, Kyle Dougherty Jul 2019

Relative Density And Resource Selection Of Urban Red Foxes In Lincoln, Nebraska, Kyle Dougherty

School of Natural Resources: Dissertations, Theses, and Student Research

Since early reports of urban red fox (Vulpes vulpes) populations in Great Britain, red fox populations have been studied in many large cities throughout Europe, North America, and Australia. However, there has been relatively little research conducted in moderately sized North American cities. To further the ecological understanding of red foxes in moderately sized cities, we investigated relative density and resource selection of the urban fox population in Lincoln, Nebraska. We used presence-only data collected from a citizen science project and inhomogeneous point process models to investigate relative density of urban foxes and deployed GPS collars on ten red …

Sesquicentennial State Park is one of the most popular state parks in South Carolina and is well-known in the Columbia metropolitan area as a refuge from the hustle and bustle of the urban scene. Driving through its entrance gates from busy Two Notch Road, visitors find themselves immediately in the midst of a pine forest. Past the ranger’s kiosk a winding road follows the contours of the gently rolling terrain, offering occasional glimpses of a mysterious fire tower, an evocative two-story log house, and eventually the open vista of a large lake with white concrete buildings and lawns along the …

Biological Sciences: Faculty Publications

Background: With the expansion of soil transmitted helminth (STH) intervention efforts and the corresponding decline in infection prevalence, there is an increased need for sensitive and specific STH diagnostic assays. Previously, through next generation sequencing (NGS)-based identification and targeting of non-coding, high copy-number repetitive DNA sequences, we described the development of a panel of improved quantitative real-time PCR (qPCR)-based assays for the detection of Necator americanus, Ancylostoma duodenale, Ancylostoma ceylanicum, Trichuris trichiura, and Strongyloides stercoralis. However, due to the phenomenon of chromosome diminution, a similar assay based on high copy-number repetitive DNA was not developed for the detection of Ascaris …

Design And Pilot Testing Of Subgoal Labeled Worked Examples For Five Core Concepts In Cs1, Lauren E. Margulieux, Briana B. Morrison, Adrienne Decker

Computer Science Faculty Publications

Subgoal learning has improved student problem-solving performance in programming, but it has been tested for only oneto-two hours of instruction at a time. Our work pioneers implementing subgoal learning throughout an entire introductory programming course. In this paper we discuss the protocol that we used to identify subgoals for core programming procedures, present the subgoal labels created for the course, and outline the subgoal-labeled instructional materials that were designed for a Java-based course. To examine the effect of subgoal labeled materials on student performance in the course, we compared quiz and exam grades between students who learned using subgoal labels …

Aristotle On The Good Of Reproduction, Myrna Gabbe Jul 2019

Aristotle On The Good Of Reproduction, Myrna Gabbe

Philosophy Faculty Publications

This paper discusses Aristotle’s theory of reproduction: specifically, the good that he thinks organisms attain by reproducing. The aim of this paper is to refute the widespread theory that Aristotle believes plants and animals reproduce for the sake of attenuated immortality. This interpretive claim plays an important role in supporting one leading interpretation of Aristotle’s teleology: the theory that Aristotelian nature is teleologically oriented with a view solely to what benefits individual organisms, and what benefits the organism is its survival and well–being. This paper challenges the theories that Aristotle takes plants and animals to reproduce for the sake of …

A Culturally-Sensitive Exploration Of Adversity And Resilience Among Trinidadians, Christabel Léonce Jul 2019

A Culturally-Sensitive Exploration Of Adversity And Resilience Among Trinidadians, Christabel Léonce

Doctor of Psychology (PsyD)

Research done by the Center for Disease Control (CDC) found that adverse childhood experiences (ACEs) such as abuse, household challenges, and neglect were linked to later health risks in life and overall well-being (CDC, 2019). Adverse experiences occur worldwide in a variety of culturally-specific ways. Research involving youth in Trinidad and Tobago outlines emerging concerns with violence in homes, communities, and schools (Baker-Henningham et al., 2009). Currently, however, there is limited data on adverse experiences and their long-term impact in Trinidad and Tobago. Trinidadians (n = 79), born and lived in Trinidad till 18 years was surveyed, ranging from ages …

Preliminary Investigation Of A Diverse Megafossil Floral Assemblage From The Middle Miocene Of Southern Mississippi, Usa, Daniel M. Mcnair, Debra Z. Stults, Brian Axsmith, Mac H. Alford, James E. Starnes

Faculty Publications

Our understanding of Miocene floras in eastern North America is hampered by the rarity of megafossil sites. An early report from the middle Miocene Hattiesburg Formation in Mississippi included palms and Ulmus. A later report listed Taxodium, Salix, either Morus or Celtis, and monocot fragments. The floral assemblage described here was recently recovered from along the Bouie River in southern Mississippi. Ferns are represented by complete Salvinia specimens including attached sporocarps, Woodwardia, and Osmunda. Conifers are represented by branchlets of Taxodium. Angiosperms include leaves attributable to the Lauraceae. Platanus is known from leaves, stipules, and fruits. …

Scale Dependence Of Sex-Specific Movement In A Small-Bodied Stream Fish, Scott R. Clark, Brian R. Kreiser, Jacob F. Schraefer, Laura K. Stewart Jul 2019

Scale Dependence Of Sex-Specific Movement In A Small-Bodied Stream Fish, Scott R. Clark, Brian R. Kreiser, Jacob F. Schraefer, Laura K. Stewart

Faculty Publications

  1. Animal movement at localised scales is often modulated by competing pressures such as avoiding predators while acquiring resources and mates. The relative magnitude of these trade‐offs may affect males and females differently, often resulting in sex‐specific differences in movement.
  2. Sex‐biases in movement have been linked to mating systems (e.g. monogamy or polygamy) in birds and mammals; however, this relationship has received less attention among fishes. Using passive integrated transponder tags and a series of stationary antennas, we evaluated the movement dynamics of a small‐bodied, sexually dimorphic stream fish Fundulus olivaceus over a 30‐day period in a fourth‐order tributary to the …

Of Fire, Mammals, And Rain: Mechanisms Of Plant Invasions, Tara Boyce Bishop Jul 2019

Of Fire, Mammals, And Rain: Mechanisms Of Plant Invasions, Tara Boyce Bishop

Theses and Dissertations

Biological invasions are driving environmental state changes on a global scale. Exotic plant species must be successful at passing several abiotic and biotic filters to establish and disrupt the native plant community assembly. Understanding where exotic plants are on a regional scale and being able to characterize how exotic plants are generally interacting with their environment is crucial information for exotic species management (chapter 1). In the western United States human-related activities are augmenting the spread of exotic plant species by increasing the ignitions of wildfire. Wildfire can lead to nutrient pulses through the removal of intact native communities and …

Characterizing Novel Pathways For Regulation And Function Of Ataxin-2, Elise Spencer Melhado Jul 2019

Characterizing Novel Pathways For Regulation And Function Of Ataxin-2, Elise Spencer Melhado

Theses and Dissertations

Ataxin-2 is an RNA-binding protein that is involved in many crucial cellular processes such as R-loop regulation, mRNA stability, TOR signaling regulation, and stress granule formation. Ataxin-2 is highly conserved, found in organisms ranging from Saccharomyces cerevisiae to Caenorhabditis elegans and Homo sapiens. Recently, ataxin-2 has been linked to the neurodegenerative disease Amyotrophic Lateral Sclerosis (ALS). ALS is a fatal disease that causes loss of motor neurons. In addition to ataxin-2 interacting with known ALS risk factor proteins, research into the relationship between ataxin-2 and ALS shows that polyglutamine expansions in ataxin-2 are gain-of-function mutations that lead to overactivity of …

3d Printed Microfluidic Devices For Bioanalysis, Michael J. Beauchamp

Theses and Dissertations

This work presents the development of 3D printed microfluidic devices and their application to microchip analysis. Initial work was focused on the development of the printer resin as well as the development of the general rules for resolution that can be achieved with stereolithographic 3D printing. The next stage of this work involved the characterization of the printer with a variety of interior and exterior resolution features. I found that the minimum positive and negative feature sizes were about 20 μm in either case. Additionally, micropillar arrays were printed with pillar diameters as small as 16 μm. To demonstrate one …

Personality Inventory Dsm-5: A Spanish Translation For Hispanics In The United States, Jessica Abigail Carmona Jul 2019

Personality Inventory Dsm-5: A Spanish Translation For Hispanics In The United States, Jessica Abigail Carmona

Theses and Dissertations

The Personality Inventory DSM-5 (PID-5) was created to measure personality pathology and help in the development of a dimensional conceptualization of personality disorders (Krueger, Derringer, Markon, Watson, & Skodol, 2012). It measures five maladaptive personality traits: Negative Affect, Detachment, Antagonism, Disinhibition and Psychoticism. The PID-5 has also garnered significant support for its hierarchical structure, five-factor structure across samples and translations, and its ability to predict variance in internalizing and externalizing disorders (Krueger & Markon, 2014). The current study builds on this literature by translating the PID-5 into Spanish spoken in Latin America and testing the replicability of the five-factor structure, …

Use Of Plant Growth Regulators To Expand The Period Of Sagebrush Seed Germination And Reduce The Risk Of Restoration Failure: Laboratory Trials, Chelsea Elizabeth Keefer Jul 2019

Use Of Plant Growth Regulators To Expand The Period Of Sagebrush Seed Germination And Reduce The Risk Of Restoration Failure: Laboratory Trials, Chelsea Elizabeth Keefer

Theses and Dissertations

Seed germination during unhospitable environmental conditions can be a major barrier to direct seeding efforts in dryland systems. In the sagebrush steppe, Artemisia tridentata Nutt. ssp. wyomingensis and Artemisia arbuscula are important shrub species that are being used in restoration, but seeding success is highly sporadic due to inter-annual and intra-seasonal weather variability. Altering and expanding the period of germination, as a form of bet-hedging, may improve plant establishment. Our objective was to determine if we could expand the period of germination using plant growth regulators (PGRs) applied in a conglomerated seed coating treatment. In a laboratory study, the seed …

Landsat Collections Reveal Long-Term Algal Bloom Hot Spots Of Utah Lake, Rachel Shanae Tate Jul 2019

Landsat Collections Reveal Long-Term Algal Bloom Hot Spots Of Utah Lake, Rachel Shanae Tate

Theses and Dissertations

Harmful algal blooms (HABs) and nuisance algal blooms (NABs) are a worldwide phenomenon with implications for human health and safety. HABs occur when algae or bacteria grow in high enough densities to harm animals and humans. A primary component of harmful algal blooms is cyanobacteria, which are aquatic, photosynthesizing microorganisms that produce toxins at high concentrations. Cyanobacterial biomass has increased worldwide in recent decades, raising concern about the future of fresh- and marine-water systems in a changing climate. Understanding the patterns and conditions of past algal blooms can provide useful insights for managing future blooms. Remote sensing can enhance our …

Isolation, Genetic Characterization And Clinical Application Of Bacteriophages Of Pathogenic Bacterial Species, Trever Leon Thurgood Jul 2019

Isolation, Genetic Characterization And Clinical Application Of Bacteriophages Of Pathogenic Bacterial Species, Trever Leon Thurgood

Theses and Dissertations

Bacteriophages (phages) are the smallest biological entity on the planet. They provide vast amounts of valuable knowledge to biologists. Phage genomes are relatively simple compared to the organisms they infect (prokaryotes) and yet continually point to the complexity surrounding molecular- and microbiological mechanisms of life. By studying phages we can learn of the systems of gene expression, protein interaction and DNA organization. Phages are useful not only from an academic perspective, but may also have useful clinical applications. In the face of the rise of antibiotic-resistant bacterial “super pathogens”, scientists and researchers turn to phages as alternative treatments to these …

Dna Capture Via Magnetic Beads In A Microfluidic Platformfor Rapid Detection Of Antibiotic Resistance Genes, David Hyrum Harris Jul 2019

Dna Capture Via Magnetic Beads In A Microfluidic Platformfor Rapid Detection Of Antibiotic Resistance Genes, David Hyrum Harris

Theses and Dissertations

Antibiotic resistant infections are a growing health care concern, with many cases reported annually. Infections can cause irreversible bodily damage or death if they are not diagnosed in a timely matter. To rapidly diagnose antibiotic resistance in infections, it is important to be able to capture and isolate the DNA coding for the resistance genes. This is challenging because bacteria are present in blood in minute concentrations. To enrich the DNA to detectable levels, I modified magnetic microbeads with ssDNA sequences complementary to the target DNA to capture the DNA via hybridization. I compared DNA capture efficiency in three different …

Chlorophyllide A: Fact Or Artifact - Resolution Of The Chlorophyllide A Problem In The Routine Measurement Of Planktonic Chlorophyll A, Sierra Claire Helmann Jul 2019

Chlorophyllide A: Fact Or Artifact - Resolution Of The Chlorophyllide A Problem In The Routine Measurement Of Planktonic Chlorophyll A, Sierra Claire Helmann

Capstone Projects and Master's Theses

Chlorophyll a serves as the routine proxy for most estimates of phytoplankton biomass in limnology and oceanography. It is well known that enzymatic chlorophyllase activity breaks chlorophyll a into the degradation product chlorophyllide a (chlide a). This degradation could result in potentially large underestimates of the true chlorophyll a concentration when the analysis is made by modern chromatographic methods. The goal of this project was to determine proper protocol in the methodological elimination of artifactual chlorophyllide a formed during extraction of chlorophyll pigments through the application of a microwave-assisted solvent extraction technique. This study aimed to answer the question: Is …

The Ambiguity Of Being: Medieval And Modern Cooperation On The Problem Of The Supernatural, Jonathan Robert Heaps Jul 2019

The Ambiguity Of Being: Medieval And Modern Cooperation On The Problem Of The Supernatural, Jonathan Robert Heaps

Dissertations (1934 -)

The recent debate over the supernatural has proved intractable in part because of a failure to distinguish two irreducible-but-linked problems of the supernatural, one medieval and one modern. The first is a metaphysical problem concerning the cooperation of humans with God. Bernard Lonergan’s retrieval of St. Thomas Aquinas’s solution to this problem indicates that a grasp of divine concursus is integral to a theory of nature and grace. A metaphysics of universal cooperation with God implies a pair of ambiguities about creaturely being. The general ambiguity is that, because the fundamental explanatory term for creaturely causation is both universal and …

Revolutionary Bodies: William Blake And The Struggle For Transcendence, Anthony Madia Jul 2019

Revolutionary Bodies: William Blake And The Struggle For Transcendence, Anthony Madia

College of Liberal Arts & Social Sciences Theses and Dissertations

Throughout the course of his work, William Blake used bodies in order to express a desire for a transcendent revolution. This paper examines the different ways that Blake employs bodies to enact revolutionary change. In doing so, the paper discusses how bodies are written, write, and labor. Moreover, the paper refers to the societal constraints placed on the body, be they limits regarding gender, sex, race, or class. By engaging in a discussion of the body's limitations and capabilities coupled with Blake's relationship with the body, it becomes clear that Blake chooses to depict bodies struggle against the normative structure …
