Characterization Of The Roles Of Ppp1r15a (Gadd34) And Ppp1r15b (Crep) In Er Stress-Induced Apoptosis In Vivo, Danielle Hicks Jan 2019

Characterization Of The Roles Of Ppp1r15a (Gadd34) And Ppp1r15b (Crep) In Er Stress-Induced Apoptosis In Vivo, Danielle Hicks

University of the Pacific Theses and Dissertations

Failure to balance synthesis, folding and degradation of secreted proteins results in the accumulation of unfolded proteins in the endoplasmic reticulum, termed ER stress. Cells respond to ER stress by activating a signaling pathway known as the unfolded protein response (UPR). The UPR induces phosphorylation of eIF2α to attenuate global protein translation, allowing the ER to clear misfolded proteins. Opposing this function, eIF2α phosphatases contain a catalytic subunit, Protein Phosphatase 1, and either of two homologous scaffolding subunits, GADD34 and CReP. Inhibition of eIF2α phosphatases has been shown to prolong UPR signaling and promote survival in many cells types and …

Body Cameras Effectiveness In A Large Police Department In The Southeast, Jason D. Armstrong Jan 2019

Body Cameras Effectiveness In A Large Police Department In The Southeast, Jason D. Armstrong

Walden Dissertations and Doctoral Studies

Several recent high profile incidents involving law enforcement officers have resulted in the death of a citizen. In some of these cases, the use of deadly force by police was ruled as justified only to learn later that one or more officers were not truthful about what occurred. These incidents have called into question law enforcement's legitimacy and created a demand for greater transparency by equipping officers with body-worn cameras. Body cameras can act as independent, reliable witnesses with no bias or agenda. Federal, state, and local governments have pledged millions of taxpayer dollars to implement body-worn camera programs in …

Measuring Regulatory And Noncompliance Prevalence Among Maryland Commercial Blue Crab Fishers, Jacquelyn Lee Rachor Hornsby

Walden Dissertations and Doctoral Studies

Few empirical studies exist that compare regulation (R) and fishing crime (VL). The lack of information about R and VL effects stakeholder decision-making. Crime weakens conservation efforts and creates false baseline data. This furthers R and the cycle repeats. The purpose of this correlational study was to determine the statistical association between the number and type of annual commercial blue crab R and VL of the same. The Pearson's R correlation was used to analyze the data because it demonstrated the strength of each relationship. This quantitative study was grounded in enforcement theory. The data was public record and consisted …

Type 2 diabetes is a metabolic disease affecting millions of people around the world. Cognitive and mood impairments are among its many debilitating complications, but disease mechanism(s) remain elusive. Here, we present a series of behavioral tasks that demonstrate impairment of cognitive flexibility in db/db mice, a commonly used type 2 diabetes model. Using immunohistochemistry, we demonstrate disruption of axon initial segments (AIS) and nodes of Ranvier, excitable axonal domains regulating neuronal output, in brain regions associated with cognitive and mood impairments. Finally, we present results of exercise treatment that ameliorates AIS disruption in these animals. Establishing cognitive flexibility deficits …

Classism, Ableism, And The Rise Of Epistemic Injustice Against White, Working-Class Men, Sarah E. Bostic Jan 2019

Classism, Ableism, And The Rise Of Epistemic Injustice Against White, Working-Class Men, Sarah E. Bostic

Browse all Theses and Dissertations

In this thesis, I illustrate how epistemic injustice functions in the divide between white working-class men and the educated elite by discussing the discursive ways in which working-class knowledge and experience are devalued as legitimate sources of knowledge. I demonstrate this by using critical discourse analysis to interpret the underlying attitudes and ideologies in comments made by Clinton and Trump during their 2016 presidential campaigns. I also discuss how these ideologies are positively or negatively perceived by Trump’s working-class base. Using feminist standpoint theory and phenomenology as a lens of interpretation, I argue that white working-class men are increasingly alienated …

Effect Of Hybrid/Complex N-Glycosylation On Cardiac Voltage-Gated Ion Channel Expression, Austin R. Parrish Jan 2019

Effect Of Hybrid/Complex N-Glycosylation On Cardiac Voltage-Gated Ion Channel Expression, Austin R. Parrish

Browse all Theses and Dissertations

Dilated cardiomyopathy (DCM) is one the most common forms of heart failure, with the majority of cases being idiopathic. Dr. Bennett’s laboratory previously showed that deletion of the Mgat1 gene in cardiomyocytes only (Mgat1KO) is sufficient to cause DCM leading to heart failure and early death. The Mgat1 gene product, GlcNAcT1, is responsible for initiating the formation of hybrid/complex N-glycosylation. These data suggest a link between abnormal glycosylation and heart disease; however, there is a mystery of how the specific changes in glycosylation contribute to heart disease etiology and progression. The proper function and gating of voltage-gated Na+ channels (Nav) …

Recognition Of Incomplete Objects Based On Synthesis Of Views Using A Geometric Based Local-Global Graphs, Michael Christopher Robbeloth

Browse all Theses and Dissertations

The recognition of single objects is an old research field with many techniques and robust results. The probabilistic recognition of incomplete objects, however, remains an active field with challenging issues associated to shadows, illumination and other visual characteristics. With object incompleteness, we mean missing parts of a known object and not low-resolution images of that object. The employment of various single machine-learning methodologies for accurate classification of the incomplete objects did not provide a robust answer to the challenging problem. In this dissertation, we present a suite of high-level, model-based computer vision techniques encompassing both geometric and machine learning approaches …

Learned Helplessness In Industrial-Organizational Psychology: Relationships With Locus Of Control, Self-Efficacy, And Feedback-Seeking Behavior, Nicholas Kovacs

Browse all Theses and Dissertations

Researchers have suggested that self-efficacy and feedback-seeking behavior (FSB) are effective in enhancing performance. To improve performance in the workplace, research should focus on how psychologists can enhance these constructs in employees. Though locus of control (LOC) relates to self-efficacy and increased FSB, research has revealed issues in LOC (e.g., failure to predict performance, range restriction, failure to predict behaviors). The current study examined the effects of perceived “lack of control”, learned helplessness, over LOC on both self-efficacy and FSB in two different samples: a student sample (N = 321) and a work sample (N = 794). Learned helplessness accounted …

Rebel Motivations During The Social War And Reasons For Their Actions After Its End, Mark Louis Howard Jan 2019

Rebel Motivations During The Social War And Reasons For Their Actions After Its End, Mark Louis Howard

Browse all Theses and Dissertations

Modern scholarship has fiercely contested the motivations of Italian rebels during the Social War. Generally speaking, three camps have formed concerning this issue: those who strictly follow the sources in arguing that the allies fought against Rome to obtain full citizenship under her rule, those who believe the rebels sought independence rather than citizenship, and those who believe that rebel actions were inspired by differing motivations. By building upon the scholarship of Dart and Salmon, I believe we will see that many of the allies were willing to fight against the empire for a place of privilege within it. Many …

Theses and Dissertations

Streptococcus sanguinis, an abundant and benign inhabitant of the oral cavity, is an important etiologic agent of infective endocarditis, particularly in people with pre-disposing cardiac valvular damage. Although commonly isolated from patients with IE, little is known about the factors that make any particular S. sanguinis isolate more virulent than another or, indeed, whether significant differences in virulence exist among isolates. To investigate the virulence of multiple isolates, a variation of the Bar-seq (barcode sequencing) method was employed. A conserved chromosomal site was identified for subsequent insertion of a barcode identifier, unique for each strain. Barcode insertion did not …
