A subset of basal breast cancer patients has tumors overexpressing EGFR, loss of PTEN expression and mutated p53; these alterations cooperate and result in a poor prognosis for these patients. PTEN is a direct negative regulator of PI3’Kinase enzymatic activity and loss of PTEN expression is correlated with elevated PI3’Kinase pathway activation. In the presented dissertation, we utilized a panel of PTEN-null basal breast cancer cell lines containing constitutive AKT activation and elevated levels of EGFR to model this subset of breast cancer patients. We first investigated the response of AKT phosphorylation to PTEN re-expression in the cell line panel. …

This work describes the synthesis and characterization of novel bioplastic composites by using novel additive manufacturing (AM) approaches. As a background, global plastic consumption has become a problem with the accumulation of plastics in landfills and the ocean. Recently, bioplastics have emerged as a viable option to petrochemicals. Mindfulness has been associated with various benefits, including enhanced cognitive performance and disengagement from problematic emotions. Previous research has indicated that engaging in a brief mindfulness exercise may enhance performance on the Color Word Stroop, neutral Stroop, and emotional Stroop. Participants in the current study were 201 students (139 female), mean age 19.7 years (SD=1.4) from a Midwestern university, who were randomly assigned to listen to a 9- minute mindfulness exercise, progressive muscle relaxation (PMR) exercise, or interview control condition before completing the Color-Word Stroop, Neutral Stroop, and Emotional Stroop. In Birchfield v. North Dakota (2016), the Supreme Court broke new Fourth Amendment ground by establishing that law enforcement’s collection of information can be cause for “anxiety,” meriting constitutional protection, even if subsequent uses of the information are tightly restricted. This change is significant. While the Court has long recognized the reality that police cannot always be trusted to follow constitutional rules, Birchfield changes how that concern is implemented in Fourth Amendment law, and importantly, in a manner that acknowledges the new realities of data-driven policing. The Wisconsin State Laboratory of Hygiene challenged Wisconsin laboratories to examine their biosafety practices and improve their culture of biosafety. One hundred three clinical and public health laboratories completed a questionnaire-based, microbiology-focused biosafety risk assessment. Greater than 96% of the respondents performed activities related to specimen processing, direct microscopic examination, and rapid nonmolecular testing, while approximately 60% performed culture interpretation. Although they are important to the assessment of risk, data specific to patient occupation, symptoms, and travel history were often unavailable to the laboratory and, therefore, less contributory to a microbiology-focused biosafety risk assessment than information on the specimen source …

The interplay between biological and cultural factors has remained understudied among ethnic minority groups, including Latinos. To the best of our knowledge, minimal to no research has examined the relationship between acculturative stressors and diurnal cortisol among Latina women. This initial exploratory study sought to examine the relationship between cortisol and acculturative stress. Among a small sample of adult Latina women, salivary cortisol was collected at 3 time points (waking, 30 min postwaking, and bedtime) to assess the cortisol awakening response (CAR) and diurnal cortisol response. Although sorghum [Sorghum bicolor (L.) Moench ssp. bicolor] is the fifth most important grain crop in terms of global production, no commercial hybrids carry genetically engineered (GE) traits for resistance to insect pests or herbicides due to regulatory concerns about gene flow to weedy relatives. However, non-GE herbicide resistance currently is being developed in grain sorghum and will likely transfer to related weeds. Pearl millet is a non-model grain and fodder crop adapted to extremely hot and dry environments globally. In India, a great deal of public and private sectors’ investment has focused on developing pearl millet single cross hybrids based on the cytoplasmic-genetic male sterility (CMS) system, while in Africa most pearl millet production relies on open pollinated varieties. Pearl millet lines were phenotyped for both the inbred parents and hybrids stage. Many breeding efforts focus on phenotypic selection of inbred parents to generate improved parental lines and hybrids. Plant and soil microbiome studies are becoming increasingly important for understanding the roles microorganisms play in agricultural productivity. The purpose of this manuscript is to provide detail on how to rapidly sample soil, rhizosphere, and endosphere of replicated field trials and analyze changes that may occur in the microbial communities due to sample type, treatment, and plant genotype. The experiment used to demonstrate these methods consists of replicated field plots containing two, pure, warm-season grasses (Panicum virgatum and Andropogon gerardii) and a low-diversity grass mixture (A. gerardii, Sorghastrum nutans, and Bouteloua curtipendula). Story County (estimated population 92,406 in 2013) lies in the heart of central Iowa, a state renowned for its remarkable agricultural productivity. Iowa leads all states for production of corn, soybean, and hogs. Revenues from agricultural products in Iowa total more than $30 billion annually according the 2012 Agricultural Census (USDA-NASS 2014). This productivity stems from a favorable natural and political environment. Afforestation is often viewed as the purposeful planting of trees in historically non-forested grasslands, but an unintended consequence is woody encroachment, which should be considered part of the afforestation process. In North America’s temperate grassland biome, Eastern redcedar (Juniperus virginiana L.) is a native species used in tree plantings that aggressively invades in the absence of controlling processes. Triterpenes are thirty-carbon compounds derived from the universal five-carbon prenyl precursors isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP). Normally, triterpenes are synthesized via the mevalonate (MVA) pathway operating in the cytoplasm of eukaryotes where DMAPP is condensed with two IPPs to yield farnesyl diphosphate (FPP), catalyzed by FPP synthase (FPS). Squalene synthase (SQS) condenses two molecules of FPP to generate the symmetrical product squalene, the first committed precursor to sterols and most other triterpenes. In the green algae Botryococcus braunii, two FPP molecules can also be condensed in an asymmetric manner yielding the more highly branched triterpene, botryococcene. Botryococcene …

Genomic prediction (GP) is now routinely performed in crop plants to predict unobserved phenotypes. The use of predicted phenotypes to make selections is an active area of research. Here, we evaluate GP for predicting grain yield and compare genomic and phenotypic selection by tracking lines advanced. We examined four independent nurseries of F3:6 and F3:7 lines trialed at 6 to 10 locations each year. Yield was analyzed using mixed models that accounted for experimental design and spatial variations. Genotype-by-sequencing provided nearly 27,000 high-quality SNPs. Rice yield is highly sensitive to increased temperature. Given the trend of increasing global temperatures, this sensitivity to higher temperatures poses a challenge for achieving global food security. Early seed development in rice is highly sensitive to unfavorable environmental conditions. Heat stress (HS) during this stage decreases seed size and fertility, thus reducing yield. Here, we explore the transgenerational phenotypic consequences of HS during early seed development on seed viability, germination, and establishment. The purpose of this dissertation is to explore how a mainstream theory of psychological practice might inadvertently conceal and ignore contemporary values and ideologies and their pathological consequences. Through a hermeneutic approach, I interpreted Leslie Greenberg’s Emotion-focused therapy: Coaching clients to work through their feelings (2nd ed), a popular and widely used theory in psychotherapy. As a practitioner with humanistic foundations, this was also an opportunity for the author to understand his own unexamined values as a therapist. Two landmark accidents invigorated the discussion regarding pilot mental health. Other incidents may also have a mental health component that is related to, but not the direct cause of, the event. The purpose of this study is to investigate the relationship between dementia caregivers’ communication behaviors (information seeking and forwarding) and their outcomes (coping outcomes: e.g., dealing better with negative feelings or improved medical outcomes). A survey data set of dementia patients’ caregivers substantiates the effects of communication behaviors about dementia illness on coping outcomes, as well as the mediating role of emotion-focused and problem-focused coping processes. Using structural equation modeling (SEM), this study found positive effects of communication behaviors on outcomes through coping processes. Nitrates are extensively used in agricultural practices as fertilizers, applied as either animal manures or derived slurries or as chemical fertilizers in the form of ammonium nitrate. Most of the toxicologically relevant nitrate enters the human body through consumption of vegetables, fruits, processed food products, and drinking water. Activated partial thromboplastin times (APTT) and factor deficiency assays were performed on the plasma of twenty American bullfrogs (Lithobates catesbeianus) to test for potential changes and regulation of the intrinsic coagulation pathway during and after hibernation. Bullfrogs (pre-hibernating n=4, hibernating n=6, post-hibernating n=10) were housed in 10 gallon aquariums filled with RO water and placed in a temperaturecontrolled room. Hibernation was gradually induced by decreasing the temperature of the room from 24 C to 3 C in increments of 3 C per week. Low molecular-weight gelators (LMWGs) are a fascinating and interesting class of compounds that can self-assemble and form supramolecular structures in appropriate solvents. The gelators form fibrous networks which entrap the solvent and result in a supramolecular gel. Carbohydrate-derived LMWGs are especially useful since they are more likely to be biocompatible and biodegradable. Dendritic gelators are branch-shaped molecules with precise molecular weight that are between small molecules and polymer gelators. Glycoclusters with sugar moieties at the periphery of the molecules are important classes of bioactive compounds due to the multivalency effect. This dissertation examined experiences of alienation among Black African youth in Waterloo region schools. This study is timely and relevant considering the rapid influx of Black Africans into the region in response to government initiatives to redirect immigrants to smaller communities. The research addressed the dearth of scholarship on experiences of Black Africans outside the major metropolitan areas by employing Afrocentric and critical race theories to explore relationships between race and youth experiences of alienation. Attachment theory has made substantial contributions to the understanding of close relationships. The purpose of this study was to inquire whether an attachment-informed psychoeducational program is a feasible and effective intervention for couples expecting their first child. The overarching question was: Is an attachment-informed relationship enhancement program, such as Hold Me Tight® (HMT), helpful to couples in strengthening their relationship and increasing their confidence in becoming first-time parents? The research question was addressed using a mixed-methods approach.
