Schistosomiasis is an important parasitic disease, touching roughly 200 million people worldwide. The causative agents are different Schistosoma species. Schistosomes have a complex life cycle, with a freshwater snail as intermediate host. After infection, sporocysts develop inside the snail host and give rise to human dwelling larvae. We present here a detailed step-by-step video instruction in English, French, Spanish and Portuguese that shows how these sporocysts can be manipulated and transferred from one snail to another. This procedure provides a technical basis for different types of ex vivo modifications, such as those used in functional genomics studies.

Characterizing the transmitted/founder (T/F) viruses of multi-variant SIV infection may shed new light on the understanding of mucosal transmission. We intrarectally inoculated six Chinese rhesus macaques with a single high dose of SIVmac251 (3.1 × 104 TCID50) and obtained 985 full-length env sequences from multiple tissues at 6 and 10 days post-infection by single genome amplification (SGA). All 6 monkeys were infected with a range of 2 to 8 T/F viruses and the dominant variants from the inoculum were still dominant in different tissues from each monkey. Interestingly, our data showed that a cluster of rare T/F viruses was unequally …

HTLV-1-Associated Myelopathy (HAM/TSP) is a progressive neuroinflammatory disorder for which no disease-modifying treatment exists. Modest clinical benefit from type I interferons (IFN-α/β) in HAM/TSP contrasts with its recently identified IFN-inducible gene signature. In addition, -α treatment in vivo decreases proviral load and immune activation in HAM/TSP, whereas IFN-β therapy decreases tax mRNA and lymphoproliferation. We hypothesize this "IFN paradox" in HAM/TSP might be explained by both cell type- and gene-specific effects of type I IFN in HTLV-1-associated pathogenesis. Therefore, we analyzed ex vivo transcriptomes of CD4+ T cells, PBMCs and whole blood in healthy controls, HTLV-1-infected individuals, and HAM/TSP patients. …

The purple sea urchin, Strongylocentrotus purpuratus, has a complex and robust immune system that is mediated by a number of multi-gene families including the SpTransformer (SpTrf) gene family (formerly Sp185/333). In response to immune challenge from bacteria and various pathogen-associated molecular patterns, the SpTrf genes are up-regulated in sea urchin phagocytes and express a diverse array of SpTrf proteins. We show here that SpTrf proteins from coelomocytes and isolated by nickel affinity (cNi-SpTrf) bind to Gram-positive and Gram-negative bacteria and to Baker’s yeast, Saccharomyces cerevisiae, with saturable kinetics and specificity. cNi-SpTrf opsonization of the marine bacteria, Vibrio diazotrophicus, augments phagocytosis, …

The diagnosis of strongyloidiasis by coprological methods has a low sensitivity, underestimating the prevalence of Strongyloides stercoralis in endemic areas. Serodiagnostic tests for strongyloidiasis have shown robust diagnostic properties. However, these methods require a blood draw, an invasive and labor-intensive sample collection method, especially in the resource-limited settings where S. stercoralis is endemic. Our study examines a urine-based assay for strongyloidiasis and compares its diagnostic accuracy with coprological and serological methods. Receiver operating characteristic (ROC) curve analyses determined the diagnostic sensitivity (D-Sn) and specificity (D-Sp) of the urine ELISA, as well as estimates its positive predictive value and diagnostic risk. …

An abundance of research has documented health inequalities by race and socioeconomic position (SEP) in the United States. However, conceptual and methodological challenges complicate the interpretation of study findings, thereby limiting progress in understanding health inequalities and in achieving health equity. Fundamental to these challenges is a lack of clarity about what race is and the implications of that ambiguity for scientific inquiry. Additionally, there is wide variability in how SEP is conceptualized and measured, resulting in a lack of comparability across studies and significant misclassification of risk. The objectives of this review are to synthesize the literature regarding common …

Neurofibromatosis type 1 (NF1), a common genetic disorder with a birth incidence of 1:2,000–3,000, is characterized by a highly variable clinical presentation. To date, only two clinically relevant intragenic genotype-phenotype correlations have been reported for NF1 missense mutations affecting p.Arg1809 and a single amino acid deletion p.Met922del. Both variants predispose to a distinct mild NF1 phenotype with neither externally visible cutaneous/plexiform neurofibromas nor other tumors. Here, we report 162 individuals (129 unrelated probands and 33 affected relatives) heterozygous for a constitutional missense mutation affecting one of five neighboring NF1 codons—Leu844, Cys845, Ala846, Leu847, and Gly848—located in the cysteine-serine-rich domain (CSRD). …

The incidence of pre-diabetes (PD) and Type-2 Diabetes Mellitus (T2D) is a worldwide epidemic. African American (AA) individuals are disproportionately more likely to become diabetic than other ethnic groups. Over the long-term, metabolic complications related to diabetes result in significant alterations in growth hormone (GH) and insulin-like growth factor-1 (IGF-1). Considering the limited exercise-related studies in the area of gene expression changes with disease progression, the objective of this study was to examine differences in exercise-induced gene expression related to the GH and IGF-1 pathways in peripheral blood mononuclear cells (PBMCs) of healthy (CON) and PD AA individuals. Design: Ten …

Genetic variation is associated with a number of lifestyle behaviours; it may be associated with adherence and individual responses to exercise training. We tested single nucleotide polymorphisms (SNPs) in the acid ceramidase gene (ASAH1) for association with subject adherence and physiologic benefit with exercise training in two well-characterised randomised, controlled 8-month exercise interventions: STRRIDE I (n = 239) and STRRIDE II (n = 246). Three ASAH1 non-coding SNPs in a linkage disequilibrium block were associated with non-completion: rs2898458(G/T), rs7508(A/G), and rs3810(A/G) were associated with non-completion in both additive (OR = 1.8, 1.8, 2.0; P < 0.05 all) and dominant (OR = 2.5, 2.6, 3.5; P < 0.05 all) models; with less skeletal muscle ASAH …

Through the work done by Shaw & McKay (1942), Bursik (1989), Messner & Sampson (1991), and Sampson & Wilson (1995), social disorganization and focus on inner-city crime became a major point of interest for researchers. The idea of social disorganization looks at the weakening of social institutions, both formal and informal, that eventually lead to crime and the study of crime patterns in urban areas. This framework has been used to study crime and pinpoint areas where decisions from governmental officials and local communities have eroded the collective efficacy of neighborhoods, leading to crime. In 2003, the FCC made changes …

The Oppressive Nexus Of Socioeconomic Status And Gender In Egyptian Society: A Marxist Exposition, Stephanie Attar
Theses - Graduate Programs in Economic Theory and Policy
The main purpose of this paper is to argue that women from lower socioeconomic status (SES) position are, on the whole, subjected to higher rates of exploitation within the household. This is done by exploring the implications and flaws within traditional neoclassical economic models of the household, and offering a Marxian model of domestic production as an alternative. I argue that incidence of lower SES exacerbates the rate of exploitation by lowering women’s bargaining power within the household. Two forms of bargaining power are posited to be particularly important: (1) the bargaining process over the distribution of resources within the …

Introduction: Biomedical applications of nanoparticles (NPs) as enzyme inhibitors have recently come to light. Oxides of metals native to the physiological environment (eg, Fe, Zn, Mg, etc.) are of particular interest—especially the functional consequences of their enzyme interaction. Materials and methods: Here, Fe 2 O 3 , zinc oxide (ZnO), magnesium oxide (MgO) and nickel oxide (NiO) NPs are compared to copper (Cu) and boron carbide (B 4 C) NPs. The functional impact of NP interaction to the model enzyme luciferase is determined by 2-dimensional fluorescence difference spectroscopy (2-D FDS) and 2-dimensional photoluminescence difference spectroscopy (2-D PLDS). By 2-D FDS …

The gills of juvenile freshwater bivalves undergo a complex morphogenesis that may correlate with changes in feeding ecology, but ontogenic studies on juvenile mussels are rare. Scanning electron microscopy was used to examine the ultrastructure and ontogeny of 117 juvenile freshwater pearl mussels (Margaritifera margaritifera) ranging in age from 1-44 months and length from 0.49-8.90 mm. Three stages of gill development are described. In Stage 1 (5-9 inner demibranch filaments), only unreflected inner demibranch filaments were present. In Stage 2 (9-17 inner demibranch filaments), inner demibranch filaments began to reflect when shell length exceeded 1.13 mm, at 13-16 months old. …

Automated Droplet Manipulation Through Electrowetting On Dielectric For Solid-Phase Oligonucleotide Synthesis, Hee Tae An
Journal of the Microelectronic Engineering Conference
Electrowetting on dielectric (EWOD) discrete microfluidic (DMF) devices have shown favorable properties for use in microreactor lab-on-a-chip applications. The following work looks toward the design and fabrication of EWOD devices for the automated droplet manipulation necessary in the ultimate goal of oligonucleotide synthesis. By modifying the previous work on mechanical filtration on EWOD devices by Schertzer et al. to be compatible with processing in the RIT Semiconductor & Microsystems Fabrication Laboratory, DI water droplets were successfully able to be moved from electrode to electrode or created from reservoir electrodes using a gap between devices of approximately 100 - 250 micrometer …

Thin film, high-k capacitors are processed via ALD (atomic layer deposition). At a temperature of 200 C, the deposition recipe realized rates of 0.97 and 0.95 °A/cycle for alumina and hafnia, respectively. 31.8 and 34.7 nm alumina and hafnia films were measured by ellipsometry to have refractive indices of 1.69 and 2.07. Capacitance-voltage tests confirmed dielectric constant values of 8.36 and 24.8 while current-voltage tests lead to the derivation of the following ranges for dielectric strength: 17.0-24.5 and 16.8-27.0 MV/cm for alumina and hafnia. The dielectric strength of alumina was found to be ˜100x that dictated in literature. The dielectric …

Rheumatoid arthritis (RA) is a chronic auto-inflammatory condition that affects multiple joints in the body, causing synovial thickening, joint swelling and significant bone erosion. The inflammatory effects of the disease are known to cause significant pain, disability and depression in many patients. Mouse models of RA, including a transgenic mouse model that over-expresses the inflammatory cytokine, TNF-α (i.e. the TNFtg mouse strain), have been used to explore the pathogenesis of RA. However, the role that TNF-α over-expression plays on behavior is understood poorly. The purpose of the present study was to quantify the progression of disease in TNFtg mice using …

Health And Outdoor Settings: A Summary Of The Coalition For Education In The Outdoors Pre-Symposium Workshop, 2018, Kristina R. Anderson, Alan Ewert
Research in Outdoor Education
At the Coalition for Education in the Outdoors (CEO) 14th Biennial Research Symposium, researchers and practitioners explored the intersection between outdoor education settings and practices and human health during a pre-symposium workshop. Guided by a supposition that outdoor education experiences impact one or more dimensions of health, participants first aligned around a collective foundation of 1) definitions of nature and health and 2) formative scholarship in outdoor exposure and natural elements.
With this foundation, existing paradigms were questioned: Are mainstay methodologies used in outdoor education efficacious, particularly if researchers are to engage with cross-disciplinary research teams or seek new funding …

A World From Dust is a popular science book about the chemical sequence behind the evolution of creation.
It’s about how geology, biology, and chemistry worked together over billions of years, providing a hidden order under the random flow of genes and lava and water.
It’s about the chemical job that each element takes up in life, and how that job is predictable from its place on the periodic table.
It can be told as the story of many elements: how iron and sulfur gave a spark of life; how manganese was a key for oxygen; and how copper and …
