Type 2 Bovine Viral Diarrhea Virus NPro Suppresses Type I Interferon Pathway Signaling In Bovine Cells And Augments Bovine Respiratory Syncytial Virus Replication, Abdulrahman Abdulaziz A. Alkheraif

School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research

Bovine viral diarrhea virus (BVDV) and bovine respiratory syncytial virus (BRSV) infections contribute to the bovine respiratory disease complex (BRDC), which is a multi-factorial disorder involving co-infections of viruses and bacteria including mycoplasma. BRDC causes great economic losses to the United States feedlot industry. BVDV infection induces immunosuppression in infected animals. BVDV Npro binds and degrades the transcription factor interferon regulatory factor-3 (IRF-3) and effectively blocks type I interferon (type I IFN) expression in host cells. BRSV nonstructural proteins, NS1 and NS2, block activation of IRF-3. In calves, concurrent infection with BVDV and BRSV resulted in more severe clinical …

Schistosomiasis As A Cause Of Acute Cholecystitis, Shaker A. Majrashi

Saudi Medical Journal

Schistosomal cholecystitis is remarkably uncommon disease which is associated with concomitant gallstone disease. The disease prevails with low socio-economic conditions of population. It is still not clear that the widespread fibrocalcific reaction in the wall of gallbladder and the cystic duct, observed for all the reports, predisposes to cholelithiasis or gallstones coexist. In the present study, we report a case of the Saudi male patient affected with schistosomal granulomatous lithiasic cholecystitis. The patient’s gallbladder was badly inflamed therefore performing a safe cholecystectomy in the patient was extremely difficult. Also, infection with schistosoma is related with chronic granulomatous inflammation making the …

Characterization of Metronidazole- and Vancomycin-Resistant Clinical Isolates of Clostridium difficile

Chioma Odo, MS.

Supervisory Professor: Charles Darkoh, Ph.D.

ABSTRACT

The incidence of C. difficile infections (CDI) has been increasing at an alarming rate. This was precipitated by the emergence of strains with increased virulence, disease severity, and high recurrence rates. These strains also exhibit high propensity for resistance to antibiotics such as fluoroquinolones and beta lactams, which has made the treatment of CDI very challenging. Currently, metronidazole and vancomycin are the most commonly used drugs for the treatment of primary CDI. Metronidazole is used for the treatment of mild to …

Genetic Basis For Elevated Rheumatic Heart Disease Susceptibility In Samoa, John Bowen Allen Jul 2018

Genetic Basis For Elevated Rheumatic Heart Disease Susceptibility In Samoa, John Bowen Allen

Theses and Dissertations

Rheumatic heart disease is an inflammatory heart disease that affects millions of people around the world. Especially high rates of the disease can be found in Oceania, including the island nation of Samoa. Genetic studies of immune response genes have provided insight into a possible genetic link to increased susceptibility to rheumatic heart disease, including the genes that code for the toll-like receptor (TLR) protein family. One of the functions of TLR proteins is to recognize the presence of bacteria via identification of bacterial flagella. My evaluation of a Samoan family identified a variant in the TLR-5 gene that would …

The Ability Of Novel Phage To Infect Virulent Bacillus Anthracis Isolates, Hyrum Smith Shumway Jul 2018

The Ability Of Novel Phage To Infect Virulent Bacillus Anthracis Isolates, Hyrum Smith Shumway

Theses and Dissertations

Bacillus anthracis is a soil dwelling microbe with pronounced pathogenic potential. Historically, anthrax has infected livestock and man. In the modern-age, anthrax is a bioterrorism concern with major incidents every decade. While the threat of large scale attacks is currently viewed as unlikely, the threat is consistent and constant. Current methods to defend against such an attack focus on antibiotics and containment of public panic. Antibiotic resistance, while not currently an issue for anthrax, could easily become so with genetically engineered weaponized strains created by rogue states or independent actors. This project evolved from collaborations between the Grose lab and …

Characterizing The Activity Of Antimicrobial Peptides Against The Pathogenic Bacterium Clostridium Difficile In An Anaerobic Environment, Adenrele Mojeed Oludiran

Chemistry & Biochemistry Theses & Dissertations

Clostridium difficile is an anaerobic Gram-positive pathogen with high treatment costs and mortality and very high antibiotic tolerance. Antimicrobial host-defense peptides (HDPs) produced naturally by animal immune systems are promising candidates to develop novel therapies for bacterial infection because they cause oxidative stress that damages multiple targets in bacterial cells, so it is difficult for bacteria to evolve resistance to these attacks.

Piscidins, fish-derived HDPs that can also form complexes with copper (Cu) to enhance their activities, are very active against multiple bacterial species in an aerobic environment. We examined their activity against C. difficile and other species in an …

Endocannabinoid Metabolome Of Human Breast Milk, Adriana V. Gaitan Espinoza Jul 2018

Endocannabinoid Metabolome Of Human Breast Milk, Adriana V. Gaitan Espinoza

LSU Doctoral Dissertations

Human breast milk (HBM) is an extremely complex yet fascinating biofluid tailored to meet an infant’s nutritional requirements for development. Amongst the nutrients present in HBM, the long-chain polyunsaturated fatty acids (LCPUFAs) are of high importance due to the pivotal role they play in infant cognitive and visual development, and growth. In addition, the LCPUFAs are precursors to endocannabinoids (EC) which are endogenous lipid mediators. EC exert metabolic responses including appetite and food intake regulation, and they have been identified to play a role in establishing the suckling response of the newborn that is needed to nurse. Thus, we aimed …

Replacing L-Amino Acid With D-Amino Acid Residues On An Ultrashort Cationic Lipopeptide To Improve Anti-Microbial Activity, Trixie Sabido

The International Student Science Fair 2018

With the overuse, and misuse, of antibiotics, bacteria have become resistant to most commercial drugs. Hence, there is a need to develop new classes of antimicrobial agents. This project aims to evaluate if incorporating D-amino acid residues will improve the antimicrobial activity of lipopeptides. A known antimicrobial agent, Myr-L-(orn)3-NH2 and its mirror-image, myr-D-(orn)3-NH2, were synthesized on a Rink-amide resin using the Fmoc solid phase synthesis strategy. The UV spectra of the Fmoc deprotection solutions confirmed that the Fmoc protecting group was released on each stage of deprotection. Major peaks around 21.6 - 21.8 minutes were observed in the HPLC profiles …

Regulations Of Pathogenic Cd4+ T Helper Lymphocytes In Inflammatory Diseases, Handong Zheng

Biomedical Sciences ETDs

In this comprehensive project, three interrelated studies with distinct foci were employed to understand the regulations of specific CD4+ T helper cell population in inflammatory diseases.

Pathogenic TH17 cells play an essential role in the initiation and development of both human multiple sclerosis (MS) and animal experimental autoimmune encephalomyelitis (EAE). Nevertheless, the underlying mechanism by which the pathogenicity of TH17 cells is controlled in the autoimmune neuro-inflammation remains unclear. In aim 1, we revealed that lumican (Lum), an extracellular matrix (ECM) protein, negatively regulates encephalitic TH17 cell responses. Our findings highlighted a TH17 cell-intrinsic effect of Lum in suppressing …
