Mitochondrial Replacement Therapy And The Regulation Of Reproductive Genetic Technologies In The United States, Bob Zhao
Duke Law & Technology Review
The ability to alter the genes of future generations no longer belongs in the realm of science fiction. The genetic modification capabilities of modern science are advancing rapidly. Mitochondrial replacement therapy (MRT) represents the first crossing of the germline barrier in humans, and as of February 2015, it is the first procedure of its kind to be legalized in the Western world. How Congress decides to regulate MRT will influence future regulation of all genetic manipulation technologies. This brief argues that the current patchwork regulatory framework established in the United States is insufficient to deal with the complex issues MRT …

Development Of A Novel Method To Express And Purify Vaccinia Virus Early Transcription Factors A7 And D6 Using Bacteria, Omkar M. Gandbhir
Seton Hall University Dissertations and Theses (ETDs)
The eradication of Smallpox is one of the greatest human achievements. However other poxviruses still exist infecting humans (Molluscum Contagiosum, monkeypox, and Vaccinia) or livestock (cowpox, sheeppox). Though vaccines and anti-virals exist, the side effects are serious, and viral resistance has been detected. The genome amongst poxviruses is highly conserved, early gene promoters maintain a specific critical consensus region that can be targeted for development of novel broad spectrum therapeutics. Using Vaccinia early transcription factors (VETFs) a novel method of expressing and purifying VETF A7 and D6 was developed. A7 and D6 are required for transcription of early genes recruiting …
The Matriculation Of The Micro-Unit On The College Campus, Barnett L. Horowitz
The Matriculation Of The Micro-Unit On The College Campus, Barnett L. Horowitz
Journal of Collective Bargaining in the Academy
To the extent the world of higher education was following the National Labor Relations Board there was probably limited focus on the Board’s Specialty Healthcare case allowing for the so-called “micro-unit”. Board decisions as to the employee status of tenure track faculty or football players or graduate teaching or research assistants were likely to be of greater interest. Yet the recent holding in Columbia University that students providing instructional services were also employees subject to the Act’s coverage put the micro-unit in play on the college campus. Almost immediately after this decision petitions were filed at Yale seeking representation of …

Domain Coloring And The Argument Principle, Frank A. Farris
Domain Coloring And The Argument Principle, Frank A. Farris
Mathematics and Computer Science
The domain-coloring algorithm allows us to visualize complex-valued functions on the plane in a single image—an alternative to before-and-after mapping diagrams. It helps us see when a function is analytic and aids in understanding contour integrals. The culmination of this article is a visual discovery and subsequent proof of the Argument Principle, which relates the count of poles and zeros of a meromorphic function inside a contour to the accumulated change in argument of the function around the contour. Throughout, I offer connections to standard learning goals of courses in complex variables.

Differential Feedback Regulation Of Δ4-3- Oxosteroid 5Β-Reductase Expression By Bile Acids, Leila Valanejad, Christina Nadolny, Stephanie Shiffka, Yuan Chen, Sangmin You, Ruitang Deng
Biomedical and Pharmaceutical Sciences Faculty Publications
Δ4-3-oxosteroid 5β-reductase is member D1 of the aldo-keto reductase family 1 (AKR1D1), which catalyzes 5β-reduction of molecules with a 3-oxo-4-ene structure. Bile acid intermediates and most of the steroid hormones carry the 3-oxo-4-ene structure. Therefore, AKR1D1 plays critical roles in both bile acid synthesis and steroid hormone metabolism. Currently our understanding on transcriptional regulation of AKR1D1 under physiological and pathological conditions is very limited. In this study, we investigated the regulatory effects of primary bile acids, chenodeoxycholic acid (CDCA) and cholic acid (CA), on AKR1D1 expression. The expression levels of AKR1D1 mRNA and protein in vitro and in …

Understanding observed space environment effects on biological organisms such seeds are important in design of long duration space missions, such as to those planned for Mars, where appropriate storage and growth of food resources is essential. Evaluation of the effects of the real and simulated space environment on seed germination and growth in space is the purpose of this research. Preliminary tests of germination rate and growth of radish seeds in a controlled environment have identified statistically significant differences between control samples and seeds flown for 30 days in low earth orbit on the Bion M-1 launch capsule Russian research …

In this issue of Blood, complimentary studies by J. Li et al1 and H. Li et al2 identify the transcription factor ZEB2 as a critical regulator of multilineage differentiation in both normal and malignant hematopoiesis. In particular, these studies show that ZEB2 is an inhibitor of normal granulocyte production, and in acute myeloid leukemia (AML), antagonizing ZEB2 function releases the granulocytic differentiation block, creating an antileukemic therapeutic effect.

Monofluorophosphate Blocks Internal Polysaccharide Synthesis In Streptococcus Mutans, Miguel Ballicora, Ana M. Demonte, Conrad Naleway, Matias D. Asencion Diez, Alberto A. Iglesias
Chemistry: Faculty Publications and Other Works
Streptococcus mutans is the leading cause of dental caries worldwide by accumulating a glycogen-like internal polysaccharide (IPS) that contributes to cariogenicity when sugars are in excess. Sodium monofluorophosphate (MFP) is an active anticariogenic compound in toothpastes. Herein, we show that MFP inhibits (with an I0.5 of 1.5 mM) the S. mutans ADP-glucose pyrophosphorylase (EC 2.7.7.27), which catalyzes the key step in IPS biosynthesis. Enzyme inhibition by MFP is similar to orthophosphate (Pi), except that the effect caused by MFP is not reverted by fructose-1,6-bisP, as occurs with Pi. Inhibition was correlated with a decrease in acidogenesis and IPS …
Molecular Cloning And Characterization Of A Stilbenoid Prenyltransferase Gene Family In Peanut, Tianhong Yang
Molecular Cloning And Characterization Of A Stilbenoid Prenyltransferase Gene Family In Peanut, Tianhong Yang
Student Theses and Dissertations
Peanut (Arachis hypogaea) produces prenylated stilbenoids as phytoalexins to fight against various pathogens. In addition, the prenylated stilbenoids exhibit several bioactivities with potential applications in human health. Despite their importance, the availability of prenylated stilbenoids is limited and their biosynthetic pathways remain to be elucidated. To date, stilbenoid prenyltransferase genes have not been identified from any plant species. Based on the prenyl unit position on the prenylated stilbenoids, it was hypothesized that different prenyltransferases are involved in the biosynthesis of distinct prenylated stilbenoids in peanut. To this end, an elicitor controlled stilbenoid bioproduction system was developed using hairy roots of …
Habitat And Community Associations And The Population Dynamics Of A Temporary Coral-Reef Fish Ectoparasite, John Michael Artim
Habitat And Community Associations And The Population Dynamics Of A Temporary Coral-Reef Fish Ectoparasite, John Michael Artim
Student Theses and Dissertations
Coral reef is habitat constructed by the scleractinian coral after which the habitat is named. While scleractinian coral colonies are often meters in extent, they are composed of small individual organisms—typically on the scale of millimeters across. Coral reefs that include an abundance of many coral species support a diverse abundance of fish and conspicuous invertebrate life. These macro constituents of coral reefs—coral colonies, reef fish and conspicuous invertebrates—have been the primary focus for much of the existing body of coral reef ecology research. One of the inconspicuous and understudied coral reef invertebrate constituents are gnathiid isopod fish parasites. These …

Changing The Pathobiological Paradigm In Myelodysplastic Syndromes: The Nlrp3 Inflammasome Drives The Mds Phenotype, Ashley Basiorka
USF Tampa Graduate Theses and Dissertations
Note: Portions of this abstract have been previously published in the journal Blood, Basiorka et al. Blood. 2016 Oct 13, and has been reproduced in this manuscript with permission from the publisher.
Myelodysplastic syndromes (MDS) are genetically diverse hematopoietic stem cell malignancies that share a common phenotype of cytological dysplasia, ineffective hematopoiesis and aberrant myeloid lineage maturation. Apoptotic cell death potentiated by inflammatory cytokines has been considered a fundamental feature of MDS for over two decades. However, this non-inflammatory form of cell death cannot account for the inflammatory nature of these disorders. We report that a hallmark of lower-risk …

The production of hydrogen and methane by geochemical reactions associated with the serpentinization of ultramafic rocks can potentially support subsurface microbial ecosystems independent of the photosynthetic biosphere. Methanogenic and methanotrophic microorganisms are abundant in marine hydrothermal systems heavily influenced by serpentinization, but evidence for methane-cycling archaea and bacteria in continental serpentinite springs has been limited. This report provides metagenomic and experimental evidence for active methanogenesis and methanotrophy by microbial communities in serpentinite springs of the Voltri Massif, Italy. Methanogens belonging to family Methanobacteriaceae and methanotrophic bacteria belonging to family Methylococcaceae were heavily enriched in three ultrabasic springs (pH 12). Metagenomic …

Habitat Selection By The Northern Long-Eared Myotis (Myotis Septentrionalis) In The Midwestern United States: Life In A Shredded Farmscape, Jeremy A. White, Patricia Freeman, Cliff A. Lemen
Transactions of the Nebraska Academy of Sciences and Affiliated Societies
Populations of the Northern Long-Eared Myotis (Myotis septentrionalis) have declined dramatically in recent years in eastern North America due to white-nose syndrome. Although M. septentrionalis was once common in parts of eastern North America, few studies have examined habitat selection of this species in an agricultural landscape. We used acoustical methods to quantify bat activity and construct a habitat model of M. septentrionalis in an intensively farmed area in the Midwestern United States, where mortality from white-nose syndrome has not yet been observed. Our study confirms that M. septentrionalis prefers forest and avoids open habitats in this agricultural …
