Purpose: Emerging evidence has demonstrated that gut microbiome plays essential roles in the pathogenesis of human diseases in distal organs. Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by the progressive loss of motor neurons. Treatment with the only drug approved by the US Food and Drug Administration for use in ALS, riluzole, extends a patient׳s life span by only a few months. Thus, there is an urgent need to develop novel interventions that for alleviate disease progression and improve quality of life in patients with ALS. Here we present evidence that intestinal dysfunction and dysbiosis may actively …
Induced Pluripotent Stem Cells Derived From Mouse Germ Cells, Nora Khan
Induced Pluripotent Stem Cells Derived From Mouse Germ Cells, Nora Khan
Theses and Dissertations
While embryonic stem cells are well known to give rise to tissues comprising all three germ layers, only recently was it found that embryonic-like stem cells could be derived from the postnatal mice testis in culture. Embryonic-like stem cells from postnatal testes have shown that they can undertake most if not all, the functions of embryonic stem cells. Most recent reports have demonstrated that somatic cells can be induced to pluripotent stem cells, mostly with the addition of genes. However, the aim of this work is to explore the potential of testicular cells to become pluripotent, including expression of pluripotency-associated …

An Historical Analysis Of Lab Techniques As Applies To Current Glial Cell Research, Nathan Ostermann
An Historical Analysis Of Lab Techniques As Applies To Current Glial Cell Research, Nathan Ostermann
JCCC Honors Journal
The primary focus of neurology, since the beginning, has been the neuron. This is understandable, neuronal networks and nerves being the main communication mechanism between the brain and the rest of the body. However, this focus on neurons has overshadowed and delayed research on the other cells in the brain. Glial cells, or neuroglia, are defined as all cells in the brain that are not electrically excitable neurons or vascular cells. This definition of glial cells introduced by Rudolf Ludwig Karl Virchow in 1858. Until relatively recently, the idea that glial cells are simply passive elements of the brain that …

Employing The Enzyme Cofactor Function Of Ascorbic Acid To Affect Oncogenic Pathways In Human Melanoma: Modulating Hypoxia Inducible Factor-1Α And Dna Demethylation To Reduce Malignant Potential, Adam Patrick Fischer
Employing The Enzyme Cofactor Function Of Ascorbic Acid To Affect Oncogenic Pathways In Human Melanoma: Modulating Hypoxia Inducible Factor-1Α And Dna Demethylation To Reduce Malignant Potential, Adam Patrick Fischer
Theses, Dissertations and Capstones
Dioxygenase enzymes such as the HIF hydroxylases (PHD1-3, FIH) and the Ten-eleven translocation (TET1-3) enzymes regulate the activity of the hypoxia inducible factor-1a (HIF1a) transcription factor and the DNA methylation status of cells, respectively. Aberrant accumulation and activation of HIF-1a can allow malignant cells to acquire attributes that promote progression, chemotherapy resistance, and survival, while aberrant hypermethylation of gene promoters can silence the expression of tumor suppressor genes essential to preventing tumorigenesis. Inadequate levels of intracellular ascorbic acid (AA), a necessary cofactor for optimal dioxygenase enzyme function, could potentiate these tumorigenic conditions. In fact, plasma levels …

Clinical practice calls for reliable diagnosis and optimized treatment. However, human errors in health care remain a severe issue even in industrialized countries. The application of clinical decision support systems (CDSS) casts light on this problem. However, given the great improvement in CDSS over the past several years, challenges to their wide-scale application are still present, including: 1) decision making of CDSS is complicated by the complexity of the data regarding human physiology and pathology, which could render the whole process more time-consuming by loading big data related to patients; and 2) information incompatibility among different health information systems (HIS) …

Lived Experiences Of African American Males With Fibromyalgia, Ethel Denise Perry
Lived Experiences Of African American Males With Fibromyalgia, Ethel Denise Perry
Walden Dissertations and Doctoral Studies
Fibromyalgia (FM) is a musculoskeletal and neurological disorder that causes constant pain, cognitive problems with short term memory, and sleep disorders. There are more females diagnosed with FM than males, and research has therefore primarily been conducted with females. Because little is known about African American males with FM, biopsychosocial theory was used in this phenomenological study to examine how African American males with FM are affected biologically, psychologically, and socially. Eight African American males diagnosed with FM were recruited from Houston, Texas and were interviewed about their lived experiences with FM, including experiences with the healthcare system, and how …

A Comprehensive Molecular Phylogeny Of Holocephalans And Identification Of Sex-Specific Genetic Markers In Chondrichthyan Fishes, Jenny Marie Kemper
A Comprehensive Molecular Phylogeny Of Holocephalans And Identification Of Sex-Specific Genetic Markers In Chondrichthyan Fishes, Jenny Marie Kemper
MUSC Theses and Dissertations
Holocephalan fishes occupy an important phylogenetic position as members of the basal clade of jawed vertebrates, as well as the sister group to bony vertebrates. Thus, they can be used to provide an important reference for comparative studies aimed at understanding early gnathostome genome evolution. However, despite this importance, holocephalans have been understudied, and no comprehensive phylogeny for their inter-relationships has been estimated. A phylogeny will provide information about ancestor-descendant relationships, forming a framework that can be used to test future hypotheses by mapping characters, identifying patterns, and estimating ancestral states. A cross-species hybridization capture method was used to sequence …
Low Cost, Multi-Purpose Genotyping Panels For Dairy And Beef Cattle, Michelle Judge
Low Cost, Multi-Purpose Genotyping Panels For Dairy And Beef Cattle, Michelle Judge
Theses
rhe selection of animals based on DNA has revolutionised animal breeding, but the associated high cost of obtaining genotypes has limited its uptake. The objective of this thesis was to develop a low-cost, low-density, multi-purpose genotyping panel for the procurement of reliable genotype information, and to quantify the long-term consequences of using such low-density genotype panels in breeding programs. The objectives were achieved through a combination of real-life cattle genotype data and simulations to mimic a cattle population. The in-silico development of genotype panels was based on actual genotypes from up to 58,705 beef cattle. Alternative novel strategies were used …

Using Nursing Simulation To Improve Early Recognition Of Emergent Situations, Carlene Blais
Walden Dissertations and Doctoral Studies
Nurses' ability to recognize and respond to postoperative patients who require emergent medical care and need immediate assistance during a code blue in the first 10 minutes is essential to improve patient outcomes. This is particularly important for the project site, a 44-bed inpatient surgical specialty hospital located in the Northeast, providing care for patients with head and neck cancer, as the hospital does not have an internal code blue response team. An adjacent facility responds to all code blue emergencies and takes approximately 10 minutes for the team to respond. The purpose of this DNP project was to develop …

Evaluating Student Use Patterns Of Streaming Video Lecture Capture In A Large Undergraduate Classroom, Nathan E. Whitley-Grassi
Walden Dissertations and Doctoral Studies
Large classes that allow smaller amounts of instructor-student interaction have become more common in today's colleges. The best way to provide needed opportunities for students to overcome this lack of interaction with instructors remains unidentified. This research evaluated the use of video lecture capture (VLC) as a supplemental method for teacher-student interaction and what, if any, impact it and attendance have on student performance in large lecture courses. This ex post facto study conducted at a Northeastern research university utilized cognitive and andragogical frameworks to examine the relationships between the independent variables frequency of video viewing, quantity of videos viewed, …

Nitrosative Stress Sensing In Porphyromonas Gingivalis: Structure And Function Of The Heme Binding Transcriptional Regulator Hcpr, Benjamin R. Belvin
Theses and Dissertations
Porphyromonas gingivalis, a Gram negative anaerobe implicated in the progression of periodontal disease, is capable of surviving and causing infection despite high levels of reactive nitrogen species found in the oral cavity due to its efficient nitrosative stress response. HcpR is an important sensor-regulator that plays a vital step in the initiation of the nitrosative stress response in many Gram negative anaerobic bacteria. We employ a combination of X-ray crystallography, SAXS, resonance Raman spectroscopy, UV-Vis spectroscopy, and molecular biology techniques to better understand this key regulator. Knockout of the hcpR gene in W83 P. gingivalis results in the inability of …
The Role Of N-Myc Downstream Regulated Gene 1 In Breast Cancer Lipid Metabolism, Christopher James Sevinsky
The Role Of N-Myc Downstream Regulated Gene 1 In Breast Cancer Lipid Metabolism, Christopher James Sevinsky
Legacy Theses & Dissertations (2009 - 2024)
Altered carbohydrate and lipid metabolism are increasingly well characterized hallmarks of aggressive breast cancers. While aerobic glycolysis, or “the Warburg effect”, is a well-established metabolic adaptation exploited by tumor cells, the understanding of unique aspects of cancer lipid metabolism lags behind. This is especially true regarding the coordination of complex lipid synthesis and trafficking pathways, which remains poorly understood. N-Myc Downstream Regulated Gene1 (NDRG1) is overexpressed in many solid tumors, but its function is unclear. The importance of NDRG1 is best exemplified by the effect of null mutations on human physiology: inactivating mutations give rise to the severe autosomal recessive …
Fundamental Principles Of Tremor Propagation In The Upper Limb, Andrew D. Davidson, Steven Knight Charles
Fundamental Principles Of Tremor Propagation In The Upper Limb, Andrew D. Davidson, Steven Knight Charles
Faculty Publications
Although tremor is the most common movement disorder, there exist few effective tremor-suppressing devices, in part because the characteristics of tremor throughout the upper limb are unknown. To clarify, optimally suppressing tremor requires a knowledge of the mechanical origin, propagation, and distribution of tremor throughout the upper limb. Here we present the first systematic investigation of how tremor propagates between the shoulder, elbow, forearm, and wrist. We simulated tremor propagation using a linear, time-invariant, lumped-parameter model relating joint torques and the resulting joint displacements. The model focused on the seven main degrees of freedom from the shoulder to the wrist …
