Ormeloxifene is a clinically approved selective estrogen receptor modulator, which has also shown excellent anticancer activity, thus it can be an ideal repurposing pharmacophore. Herein, we report therapeutic effects of ormeloxifene on prostate cancer and elucidate a novel molecular mechanism of its anticancer activity. Ormeloxifene treatment inhibited epithelial-to-mesenchymal transition (EMT) process as evident by repression of N-cadherin, Slug, Snail, vimentin, MMPs (MMP2 and MMP3), β-catenin/TCF-4 transcriptional activity, and induced the expression of pGSK3β. In molecular docking analysis, ormeloxifene showed proficient docking with β-catenin and GSK3β. In addition, ormeloxifene induced apoptosis, inhibited growth and metastatic potential of prostate cancer cells and …

Study On Thermophysical Properties Of Tobacco And Smoke Agent Under Different Temperature, Ma Yaping, Liu Chaoxian, Wang Le, Zhang Mingjian, Zhang Ke, Deng Nan, Lu Duanfeng, Tang Jianguo, Liu Xiangzhen

Food and Machinery

The thermophysical properties of tobacco and smoke agent (the mass ratio of glycerol and propylene glycol is 51) was measured by transient plane heat source method, and the effects of moisture and smoke agent content on reconstituted cut tobacco and flue-cured were measured. The results showed that with the increase of temperature (22~75 ℃), thermal conductivity \[0.06~0.12 W/(m·K) and 0.06~0.16 W/(m·K\] and volume heat capacity \[0.17~2.70 MJ/(m3K) and 0.15~1.45 MJ/(m3K)\] increased; however, the thermal diffusivity decreased (0.33~0.05 mm2 / s and 0.40~0.09 mm2 / s, respectively). With the increase of moisture content (0%~13%) , …

Toxicant Formation In Dabbing: The Terpene Story, Jiries Meehan-Attrash, Wentai Luo, Robert M. Strongin

Chemistry Faculty Publications and Presentations

Inhalable, noncombustible cannabis products are playing a central role in the expansion of the medical and recreational use of cannabis. In particular, the practice of “dabbing” with butane hash oil has emerged with great popularity in states that have legalized cannabis. Despite their growing popularity, the degradation product profiles of these new products have not been extensively investigated. The study herein focuses on the chemistry of myrcene and other common terpenes found in cannabis extracts. Methacrolein, benzene, and several other products of concern to human health were formed under the conditions that simulated realworld dabbing. The terpene degradation products observed …

Insights Into Onchocerca Volvulus Population Biology Through Multilocus Immunophenotyping, Carmelle T Norice-Tra, José Ribeiro, Sasisekhar Bennuru, Michael P Fay, Rahul Tyagi, Makedonka Mitreva, Thomas B Nutman

2010-2019 OA Pubs

We have developed a serologically based immunophenotyping approach to study Onchocerca volvulus (Ov) population diversity. Using genomic sequence data and polymerase chain reaction-based genotyping, we identified nonsynonymous single-nucleotide polymorphisms (SNPs) in the genes of 16 major immunogenic Ov proteins: Ov-CHI-1/Ov-CHI-2, Ov16, Ov-FAR-1, Ov-CPI-1, Ov-B20, Ov-ASP-1, Ov-TMY-1, OvSOD1, OvGST1, Ov-CAL-1, M3/M4, Ov-RAL-1, Ov-RAL-2, Ov-ALT-1, Ov-FBA-1, and Ov-B8. We assessed the immunoreactivity of onchocerciasis patient sera (n = 152) from the Americas, West Africa, Central Africa, and East Africa against peptides derived from 10 of these proteins containing SNPs. Statistically significant variation in immunoreactivity among the regions was seen in SNP-containing peptides …

Heterogeneity Aware Random Forest For Drug Sensitivity Prediction, Raziur Rahman, Kevin Matlock, Souparno Ghosh, Ranadip Pal

Department of Statistics: Faculty Publications

Samples collected in pharmacogenomics databases typically belong to various cancer types. For designing a drug sensitivity predictive model from such a database, a natural question arises whether a model trained on diverse inter-tumor heterogeneous samples will perform similar to a predictive model that takes into consideration the heterogeneity of the samples in model training and prediction. We explore this hypothesis and observe that ensemble model predictions obtained when cancer type is known out-perform predictions when that information is withheld even when the samples sizes for the former is considerably lower than the combined sample size. To incorporate the heterogeneity idea …

Synthesis Of Small Molecule Heterocycles As Antimicrobial And Anticancer Agents, Zakeyah Ali Alsharif

Student Theses and Dissertations

This thesis contains three chapters. In chapter one, the synthesis of dihydropyrido-pyrimidones derivatives by using hexafluoroisopropanol (HFIP) as a solvent was reported and the synthesized compounds were tested against different bacteria but no activity was observed. In the second chapter, the syntheses of thiazoline and thiazole derivatives were reported. In this methodology, HFIP was used to promote for the reaction and sixty new molecules were synthesized. Two of these compounds showed promising activity as a potent anti-cancer agent against several cell lines. In the third chapter, pyrazole derivatives were synthesized and tested against different types of bacteria. Among these compounds, …

The Importance Of Saperda Inornata And Oberea Schaumii (Coleoptera: Cerambycidae) Galleries As Infection Courts Of Hypoxylon Pruinatum In Trembling Aspen, Populus Tremuloides, John C. Nord, Fred B. Knight

The Great Lakes Entomologist

(excerpt) Trembling aspen, Populus tremuloides Michaux, and bigtooth aspen, P. grandidentata Michaux, are hosts of numerous species of injurious insects and microorganisms (Harrison 1959). Only a few of those organisms, however, are directly responsible for mortality of healthy trees. The fungus Hypoxylon pruinatum (Klotzsche) Cke. is most important in that respect, killing 1-2%o f the standing volume annually in the Lake States (Anderson 1964). It invades and spreads in cambial tissue, killing it and eventually the branch or stem by girdling. Initially, a canker appears as a sunken, yellowish-orange area in the bark (Anderson 1956). In a later stage the …

Cross-Reactive Immunity To Porcine Reproductive And Respiratory Syndrome Virus And Its Contribution To Protection, Ignacio Correas

School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research

Due to the vast geographical distribution and significant economic losses generated, porcine reproductive and respiratory syndrome virus (PRRSV) can be considered the most important swine pathogen of contemporary times. Current control and eradication strategies against PRRSV have difficulty succeeding because of their complex nature and the absence of an effective vaccine. A major obstacle for PRRSV vaccine development is the broad heterogeneity of the virus, both at the genetic and antigenic level, its rapid evolution, and an incomplete knowledge of the immune responses responsible for clearing the virus from the host. Specifically, how known correlates of protection against PRRSV—neutralizing antibodies …

Cotesia vestalis was introduced into Kenya from South Africa in March 2003 for biological control of Plutella xylostella with initial release in 2004. To confirm establishment, the rate of spread beyond the release sites and parasitism rates by C. vestalis were assessed between October 2011 and April 2012 in eastern and southeastern Kenya (Kitui, Mwingi/Yatta and Loitokitok areas). Monthly observations on kale infestation by diamondback moth, damage scores, parasitism and parasitoid guild were undertaken on 20 randomly selected plants in farmer-managed kale farms. Sampled larval and pupal stages were placed in containers and taken to the laboratory for parasitoid emergence. …

Synthesis Of Alpha-Galcer Analogues As Inkt-Cell Agonists And Reactions Of 2-Methyleneoxetanes, Donald Raymond Caldwell Aug 2017

Synthesis Of Alpha-Galcer Analogues As Inkt-Cell Agonists And Reactions Of 2-Methyleneoxetanes, Donald Raymond Caldwell

Doctoral Dissertations

Glycolipids, particularly a-galactosylceramides, have gained considerable attention due to their ability to modulate immune responses. The discovery of a synthetic glycolipid antigen, KRN7000, has enhanced our understanding of iNKT cell function. Since KRN7000 stimulates iNKT cells to release both Th1 and Th2 cytokines in relative amounts at which the biological activity of either one is antagonized by the other, tremendous effort has been devoted toward synthesizing analogues of KRN7000 that bias cytokine release, as such targets may offer new approaches to treating viral infections, bacterial infections, cancer, and autoimmune conditions. The first part of this proposal describes the synthesis of …

Injury-Induced Actin Cytoskeleton Reorganization In Podocytes Revealed By Super-Resolution Microscopy, Hani Y. Suleiman, Robyn Roth, Sanjay Jain, John E. Heuser, Andrey S. Shaw, Jeffrey H. Miner

Open Access Publications

The architectural integrity of tissues requires complex interactions, both between cells and between cells and the extracellular matrix. Fundamental to cell and tissue homeostasis are the specific mechanical forces conveyed by the actomyosin cytoskeleton. Here we used super-resolution imaging methods to visualize the actin cytoskeleton in the kidney glomerulus, an organized collection of capillaries that filters the blood to make the primary urine. Our analysis of both mouse and human glomeruli reveals a network of myosin IIA-containing contractile actin cables within podocyte cell bodies and major processes at the outer aspects of the glomerular tuft. These likely exert force on …

Genetic And Biological Studies On Solanum Plants: Insights Into The Leaf Wax Alkane Paleoclimate Proxy, Amanda Lorraine Dorothy Bender Aug 2017

Genetic And Biological Studies On Solanum Plants: Insights Into The Leaf Wax Alkane Paleoclimate Proxy, Amanda Lorraine Dorothy Bender

Arts & Sciences Graduate Student Theses and Dissertations

The hydrogen isotopic composition (δ2H) of precipitation records fluxes of water in the hydrological cycle, and can reflect climatic variables such as temperature and humidity. Leaf wax n-alkanes can record δ2H of precipitation; consequently, n-alkanes extracted from sedimentary archives are commonly used as proxies for paleoenvironmental reconstruction. However, the utility of the leaf wax paleoclimate proxy is modulated by uncertainty about how physiological processes affect hydrogen isotopic compositions during alkane biosynthesis. This work aims to improve our understanding of the leaf wax paleoclimate proxy by examining biological and environmental sources of variability on cuticular leaf waxes. We used Solanum (tomato) …

Specificity Determination By Paralogous Winged Helix-Turn-Helix Transcription Factors, Adam Joyce Aug 2017

Specificity Determination By Paralogous Winged Helix-Turn-Helix Transcription Factors, Adam Joyce

Arts & Sciences Graduate Student Theses and Dissertations

Transcription factors (TFs) localize to regulatory regions throughout the genome, where they exert physical or enzymatic control over the transcriptional machinery and regulate expression of target genes. Despite the substantial diversity of TFs found across all kingdoms of life, most belong to a relatively small number of structural families characterized by homologous DNA-binding domains (DBDs). In homologous DBDs, highly-conserved DNA-contacting residues define a characteristic ‘recognition potential’, or the limited sequence space containing high-affinity binding sites. Specificity-determining residues (SDRs) alter DNA binding preferences to further delineate this sequence space between homologous TFs, enabling functional divergence through the recognition of distinct genomic …

The New Mechanical Philosophy, Stuart Glennan Aug 2017

The New Mechanical Philosophy, Stuart Glennan

Philosophy, Religion, and Classics

The New Mechanical Philosophy argues for a new image of nature and of science--one that understands both natural and social phenomena to be the product of mechanisms, and that casts the work of science as an effort to discover and understand those mechanisms. Drawing on an expanding literature on mechanisms in physical, life, and social sciences, Stuart Glennan offers an account of the nature of mechanisms and of the models used to represent them. A key quality of mechanisms is that they are particulars - located at different places and times, with no one just like another. The crux of …

Colonizing Opportunistic Pathogens (Cops): The Beasts In All Of Us., Lance B Price, Bruce A Hungate, Benjamin J Koch, Gregg S Davis, Cindy M Liu

Environmental and Occupational Health Faculty Publications

Colonizing opportunistic pathogens (COPs) are microbes that asymptomatically colonize the human body and, when the conditions are right, can cause infections. Their ability to persist indefinitely and to be transmitted without detection [1] gives COPs a unique epidemiology that warrants special consideration. There are examples of COPs among bacteria, fungi (e.g., Candida albicans [2]), protozoa (e.g., Blastocystis [3, 4]), and viruses (e.g., Rhinovirus [5]), but bacterial COPs are of particular relevance because of their major contribution to today’s antibiotic resistance crisis. The COPs include a long list of notorious bacteria that …

Fast Tracking Of Single Conjugated Polymer Nanoparticles With High Spatial Resolution, Teeranan Nongnual Aug 2017

Fast Tracking Of Single Conjugated Polymer Nanoparticles With High Spatial Resolution, Teeranan Nongnual

All Dissertations

Highly fluorescent and photostable conjugated polymer nanoparticles that freely diffuse in glycerol/water mixtures were individually tracked at an acquisition rate up to 1 kHz. The average bright fluorescence emission of about 15000 photons per particle per millisecond exposure time (~500 photons detected by sCMOS camera) yields a theoretical localization uncertainty of 10 nm per frame along lateral plane. Axial positional trajectories for 3D particle tracking were determined by defocused imaging, which evaluates the width of fluorescence spot at different displaced focal planes, yielding an axial resolution of 20 nm. The diffusion coefficient of nanoparticles in solution was measured by using …

How Do Former Undergraduate Mentors Evaluate Their Mentoring Experience 3-Years Post-Mentoring: A Phenomenological Study, Kari L. Nelson, Christine E. Cutucache Jul 2017

How Do Former Undergraduate Mentors Evaluate Their Mentoring Experience 3-Years Post-Mentoring: A Phenomenological Study, Kari L. Nelson, Christine E. Cutucache

The Qualitative Report

This phenomenological study involves a unique, longitudinal assessment of the lived experiences of former undergraduate mentors (n=7) in light of their current experiences (i.e., career or advanced schooling). The objective of a phenomenological study is to engage in in-depth probing of a representative number of participants. Specifically, we followed up with graduates of the Nebraska STEM 4U (NE STEM 4U) intervention 3 years post-program, with the overall goal of describing the mentors’ experiences using the lens of their current experiences. This type of longitudinal perspective of mentoring is greatly lacking in the current literature. At the time of the interviews, …

Age Determination Of Pheasants (Phasianus Colchicus) Using Discriminant Analysis, Maureen I.A. Woodburn, John P. Carroll, Peter A. Robertson, Andrew N. Hoodless

National Quail Symposium Proceedings

Most gallinaceous birds can be identified as juveniles or adults using the outermost primaries (P9 and P10) which are retained until after the first breeding season and are often identifiable by colour and wear. The pheasant Phasianus colchicus, however, moults all ten primary feathers during its post-juvenile moult so alternative techniques are required. To date the method most widely used on live birds is measurement of the shaft diameter of the proximal primary feather, P1, which is replaced first before the bird is fully-grown. Using a known-age sample of 752 free-living pheasants, this study presents a discriminant function analysis using …

The Great Lakes Entomologist

Excerpt: This pocket-sized book is indeed a high point in recent entomological literature. Concisely written, and including a remarkable amount of new or recently published information, The Wasps is essentially a comparative natural history of these fascinating animals. Only the "higher" or aculeate wasps are covered, a fact not clear from the title. After a brief survey of the Hymenoptera, five chapters follow: two on solitary species, two on social wasps, and a fascinating final chapter treating the diverse and often intricate biotic relationships of wasps. The book is generously illustrated, with 122 figures about equally divided between ph~tographs and …
