Circulating tumor cells (CTCs) from peripheral blood account genetic information for cancer diagnosis and overall disease monitoring. Analysis of “liquid biopsy” holds immense promise as it may lead to new approaches for cancer treatment. The study reports effective and continuous flow microchannel system for isolating CTCs using transferrin conjugated 3D matrix synthesized by crosslinking polyethylene glycol-Fe3O4 nanostructures for rapid and efficient capturing of CTCs. The platform provides option of using multiple microchannel units in series that can influence higher cell-capture efficiency due to increasing cell-substrate contact frequency. CTCs are captured with high efficiency even at low concentration of target cells …

An Investigation Into Spike-Based Neuromorphic Approaches For Artificial Olfactory Systems, Anup Vanarse, Adam Osseiran, Alexander Rassau
Research outputs 2014 to 2021
The implementation of neuromorphic methods has delivered promising results for vision and auditory sensors. These methods focus on mimicking the neuro-biological architecture to generate and process spike-based information with minimal power consumption. With increasing interest in developing low-power and robust chemical sensors, the application of neuromorphic engineering concepts for electronic noses has provided an impetus for research focusing on improving these instruments. While conventional e-noses apply computationally expensive and power-consuming data-processing strategies, neuromorphic olfactory sensors implement the biological olfaction principles found in humans and insects to simplify the handling of multivariate sensory data by generating and processing spike-based information. Studies On Development In Euphilomedes Ostracods: Embryology, Nervous System Development, And The Genetics Of Sexually Dimorphic Eye Development, Kristina Koyama
University of the Pacific Theses and Dissertations
Model organism studies have been fundamental in understanding evolutionary and developmental biology. However, non-model organisms present opportunities to study unique characteristics and as comparisons to model organisms, leading us toward broader and more relevant perspectives on diversity. The Euphilomedes genus of ostracods is an example of a non-model group with potential for evolutionary and developmental studies.
Ostracoda is an ancient, basally branching lineage of Crustaceans with a complete and prodigious fossil record. Despite the group’s promise for evolutionary studies, much remains unknown about the basic biology of this clade. There are a limited number of embryogenesis studies in Ostracoda; here, …
A Review Of Random Matrix Theory With An Application To Biological Data, Jesse Aaron Marks
A Review Of Random Matrix Theory With An Application To Biological Data, Jesse Aaron Marks
Masters Theses
"Random matrix theory (RMT) is an area of study that has applications in a wide variety of scientific disciplines. The foundation of RMT is based on the analysis of the eigenvalue behavior of matrices. The eigenvalues of a random matrix (a matrix with stochastic entries) will behave differently than the eigenvalues from a matrix with non-random properties. Studying this bifurcation of the eigenvalue behavior provides the means to which system-specific signals can be distinguished from randomness. In particular, RMT provides an algorithmic approach to objectively remove noise from matrices with embedded signals.

A case study in unconsolidated carbonates on San Salvador Island, The Bahamas, utilized high-frequency (800 MHz) georadar imaging to augment existing methodologies (burrow counts and measurements, casting) in brachyuran bioturbation research (Ocypode quadrata and Gecarcinus lateralis), and as part of a new dataset characterizing blue land crab (Cardisoma guanhumi) burrows. Non-invasive techniques such as ground-penetrating radar (GPR) can complement traditional field surveys aimed at quantifying mesoscale bioturbation in modern settings. These methods can establish diagnostic features for tracemaker identification and refine existing ichnofacies models. Drone-mounted aerial coverage provided the first high-resolution images of the micro-topography and …

Human Serine Racemase Structure/Activity Relationship Studies Provide Mechanistic Insight And Point To Position 84 As A Hot Spot For Β-Elimination Function, David L. Nelson, Greg A. Applegate, Matthew L. Beio, Danielle L. Graham, David B. Berkowitz
Department of Chemistry: Faculty Publications
There is currently great interest in human serine racemase, the enzyme responsible for producing the NMDA co-agonist D-serine. Reported correlation of D-serine levels with disorders including Alzheimer’s disease, ALS, and ischemic brain damage (elevated D-serine) and schizophrenia (reduced D-serine) has further piqued this interest. Reported here is a structure/activity relationship study of position Ser84, the putative re-face base. In the most extreme case of functional reprogramming, the S84D mutant displays a dramatic reversal of β-elimination substrate specificity in favor of L-serine over the normally preferred L-serine-O-sulfate (~1200-fold change in kcat/Km …

Metal-free magnetic resonance imaging (MRI) agents could overcome the established toxicity associated with metal-based agents in some patient populations and enable new modes of functional MRI in vivo. Herein, we report nitroxidefunctionalized brush-arm star polymer organic radical contrast agents (BASP-ORCAs) that overcome the low contrast and poor in vivo stability associated with nitroxide-based MRI contrast agents. As a consequence of their unique nanoarchitectures, BASP-ORCAs possess per-nitroxide transverse relaxivities up to ∼44-fold greater than common nitroxides, exceptional stability in highly reducing environments, and low toxicity. These features combine to provide for accumulation of a sufficient concentration of BASP-ORCA in murine subcutaneous …

Fluorescent Probe Development For Fructose Specific Transporters In Cancer, Joseph Fedie
Fluorescent Probe Development For Fructose Specific Transporters In Cancer, Joseph Fedie
Dissertations, Master's Theses and Master's Reports
Carbohydrate transporters or GLUTs of the major facilitator superfamily (MFS) are responsible for transporting sugars into the cell and have been of research interest for decades. Disruptions, mutations, and over-activations of GLUTs have been linked to a number of major diseases including cancer, obesity, and diabetes. Differentiating between transporters is incredibly difficult due to highly conserved structures, and so specific targeting between transporters has proven a complex challenge. GLUTs are highly flexible in their conformations however exactly what will and will not pass through the transporter is ambiguous at best, and many attempt to target these transporters have failed.

The Human Mitochondrial Chaperonin: It Takes Two Single-Rings To Tango, Adrian Sergio Enriquez
The Human Mitochondrial Chaperonin: It Takes Two Single-Rings To Tango, Adrian Sergio Enriquez
Open Access Theses & Dissertations
The human mitochondrial chaperonin is a macromolecular machine that catalyzes the proper folding and assembly of newly imported mitochondrial proteins into their biologically active state. It is composed of two proteins from the highly conserved heat shock protein family, hsp10 and hsp60, that assemble into large oligomeric complexes responsible for mediating the folding of non-native polypeptides in an ATP dependent manner. In addition to its innate role in protein folding, human mitochondrial hsp60 has been implicated in numerous moonlighting cellular activities that have been linked to diseases conditions such as cancer and neurodegeneracy. In light of its cellular importance, the …

Nest-To-Surf Mortality Of Loggerhead (Caretta Caretta) Sea Turtle Hatchlings On Florida’S East Coast In 2016, Victoria Erb
Electronic Theses and Dissertations 2010 - 2019
Worldwide, sea turtles are especially vulnerable immediately after emerging from nests. Many monitoring programs measure hatchling production from nest inventories. These inventories rarely account for mortality occurring post-emergence, leaving an incomplete estimate of hatchling production. This study addresses the nest-to-surf data gap for Florida’s east coast nesting assemblages of loggerhead sea turtles (Caretta caretta). Five locations were surveyed during the 2016 nesting season by using infrared time-lapse imagery, night vision optics, and track maps. Over all beaches, 7.6% of the observed hatchlings did not survive to reach the water. Mortality sources varied by location. Observed predators included: foxes, bobcats, yellow-crowned …
