The basement membrane (BM) is a form of extracellular matrix that underlies cell layers in nearly all animal tissues. Type IV collagen, a major constituent of BMs, is critical for tissue development and architecture. The enzyme peroxidasin (Pxdn), an extracellular matrix-associated protein, catalyzes the formation of structurally reinforcing sulfilimine cross-links within the collagen IV network, an event essential to basement membrane integrity. Although the catalytic function of Pxdn is known, the regulation of its activity remains unclear. In this work we show through N-terminal sequencing, pharmacologic studies, and mutational analysis that proprotein convertases (PCs) proteolytically process human Pxdn at Arg-1336, …

Background: Although canine clinical manifestations of Lyme disease vary widely, cutaneous manifestations are not well documented in dogs. In contrast, a variety of cutaneous manifestations are reported in human Lyme disease caused by the spirochete Borrelia burgdorferi. A recently recognized dermopathy associated with tickborne illness known as Morgellons disease is characterized by brightly-colored filamentous inclusions and projections detected in ulcerative lesions and under unbroken skin. Recent studies have demonstrated that the dermal filaments are collagen and keratin biofibers produced by epithelial cells in response to spirochetal infection. BACKGROUND: We sought to assess physician interest in molecular prognosic testing for patients with early stage colon cancer, and identify factors associated with the likelihood of test adoption.

METHODS: We identified physicians who care for patients with early-stage (pN0) colon cancer patients, mailed them a survey, and analyzed survey responses to assess clinician receptivity to the use of a new molecular test (GUCY2C) that identifies patients at risk for recurrence, and clinician readiness to act on abnormal test results.

RESULTS: Of 104 eligible potential respondents, 41 completed and returned the survey. Among responding physicians, 56 % were receptive to using …

Analysis of individual cells at the subcellular level is important for understanding diseases and accelerating drug discovery. Nanoscale endoscopes allow minimally invasive probing of individual cell interiors. Several such instruments have been presented previously, but they are either too complex to fabricate or require sophisticated external detectors because of low signal collection efficiency. Here we present a nanoendoscope that can locally excite fluorescence in labelled cell organelles and collect the emitted signal for spectral analysis. Finite Difference Time Domain (FDTD) simulations have shown that with an optimized nanoendoscope taper profile, the light emission and collection was localized within ~100 nm. …

Introduction

Rhythmic auditory cueing improves certain gait symptoms of Parkinson's disease (PD). Cues are typically stimuli or beats with a fixed inter-beat interval. We show that isochronous cueing has an unwanted side-effect in that it exacerbates one of the motor symptoms characteristic of advanced PD. Whereas the parameters of the stride cycle of healthy walkers and early patients possess a persistent correlation in time, or long-range correlation (LRC), isochronous cueing renders stride-to-stride variability random. Random stride cycle variability is also associated with reduced gait stability and lack of flexibility.

The yeast homolog of DJ-1, Hsp31, is a multifunctional protein that is involved in several cellular pathways including detoxification of the toxic metabolite methylglyoxal and as a protein deglycase. Prior studies ascribed Hsp31 as a molecular chaperone that can inhibit a-Syn aggregation in vitro and alleviate its toxicity in vivo. It was also shown that Hsp31 inhibits Sup35 aggregate formation in yeast, however, it is unknown if Hsp31 can modulate [PSIC] phenotype and Sup35 prionogenesis. Previous studies have investigated carotid artery intima-media thickness (IMT) and blood pressure and found a direct correlation between the two. It is known that adult females have better cardiovascular health than males until a certain stage of life, yet limited research has examined gender differences in vascular function. Thus, the purpose of this study was to investigate vascular structure and function, blood pressure, and blood glucose/cholesterol levels in relation to gender differences in young healthy adults. Recent advances in wearable robotic devices have demonstrated the ability to reduce the metabolic cost of walking by assisting the ankle joint. To achieve greater gains in the future it will be important to determine optimal actuation parameters and explore the effect of assisting other joints. The persistence of altruism throughout the evolutionary process has been explained by some on the basis of assortation, which requires the ability to detect dispositional altruism in others and voluntary interaction, resulting in altruism homophily. Numerous studies have identified the ability to detect dispositional altruism in strangers, but few have investigated this ability and altruism homophily in social networks. Impact Of Interviews On Heterosexual Students' Expressions Of Cultural Competency, Carol Isaac, Linda Behar-Horenstein

The Qualitative Report

The purpose of this qualitative study was to understand the effects of a cultural competency intervention on dental pre-doctoral students’ attitudes toward individuals of a different sexual orientation. 22 heterosexual students interviewed gay or lesbian individuals and wrote reflective text. Results illustrated that participants found that their interviewees had “surprisingly similar” beliefs and values – especially in the areas of religion and family. Because of their “similar values,” these students expressed respect toward their interviewees who were “so different” than themselves. This conclusion of “sameness” forced them to see homosexuals as people, rather than a stigmatized invisible outgroup, mitigating sexual …

The mediation of religious narratives through sacred texts is intimately bound to the power relations involved in their transmission and maintenance. Those who possess such mediated messages and control their access and interpretation have historically held privileged positions of authority, especially when those positions are not easily contested. The 2010 film The Book of Eli uniquely engages these elements by placing the alleged last copy of the King James Version of the Christian Bible at the forefront of a clash between different individuals in a post-nuclear wasteland. The 1990s were a politically, socially, and economically turbulent decade for Cuba. It is neither surprising that it was during these years that the state amended its approach to religious freedom nor that it was during this time that Pope John Paul II made his historic visit to the island. Following the pontiff’s visit, the state amended the constitution and declared itself secular rather than Marxist, thus removing much of the stigma that believers had previously faced.
