The Human Disease-Associated Ab Amyloid Core Sequence Forms Functional Amyloids In A Fungal Adhesin, Rachele D. Rameau, Desmond N. Jackson, Audrey Beaussary, Yves F. Dufrêne, Peter N. Lipke
Publications and Research
There is increasing evidence that many amyloids in living cells have physiological functions. On the surfaces of fungal cells, amyloid core sequences in adhesins can aggregate into 100- to 1,000-nm-wide patches to form high-avidity adhesion nanodomains on the cell surface. The nanodomains form through interactions that have amyloid-like properties: binding of amyloid dyes, perturbation by antiamyloid agents, and interaction with homologous sequences. To test whether these functional interactions are mediated by typical amyloid interactions, we substituted an amyloid core sequence, LVFFA, from human A protein for the native sequence IVIVA in the 1,419-residue Candida albicans adhesin Als5p. The chimeric protein …

The greenhead horse fly, Tabanus nigrovittatus Macquart, is frequently found in coastal marshes of the Eastern United States. The greenhead horse fly larvae are top predators in the marsh and thus vulnerable to changes in the environment, and the adults potentially are attracted to polarized surfaces like oil. Therefore, horse fly populations could serve as bioindicators of marsh health and toxic effects of oil intrusion. In this study, we describe the impact of the April 2010 Deep Water Horizon oil spill in the Gulf of Mexico on tabanid population abundance and genetics as well as mating structure. Horse fly populations …

Prior research has found high levels of depression and stress among persons who are incarcerated in the United States (U.S.). However, little is known about changes in depression and stress levels among inmates post-incarceration. The aim of this study was to examine changes in levels of depression and stress during and after incarceration in a tobacco-free facility. Questionnaires that included valid and reliable measures of depression and stress were completed by 208 male and female inmates approximately eight weeks before and three weeks after release from a northeastern U.S. prison. Although most inmates improved after prison, 30.8% had a worsening …

Targeted Curing Of All Lysogenic Bacteriophage From Streptococcus Pyogenes Using A Novel Counter-Selection Technique, Chad W. Euler, Barbara Juncosa, Patricia A. Ryan, Douglas R. Deutsch, W. Michael Mcshan, Vincent A. Fischetti
Publications and Research
Streptococcus pyogenes is a human commensal and a bacterial pathogen responsible for a wide variety of human diseases differing in symptoms, severity, and tissue tropism. The completed genome sequences of >37 strains of S. pyogenes, representing diverse disease- causing serotypes, have been published. The greatest genetic variation among these strains is attributed to numerous integrated prophage and prophage-like elements, encoding several virulence factors. A comparison of isogenic strains, differing in prophage content, would reveal the effects of these elements on streptococcal pathogenesis. However, curing strains of prophage is often difficult and sometimes unattainable. We have applied a novel counter-selection approach …

Common Sense Beliefs About The Central Self, Moral Character, And The Brain, D. Fernandez-Duque, Barry Schwartz
Psychology Faculty Works
To assess lay beliefs about self and brain, we probed people's opinions about the central self, in relation to morality, willful control, and brain relevance. In study 1, 172 participants compared the central self to the peripheral self. The central self, construed at this abstract level, was seen as more brain-based than the peripheral self, less changeable through willful control, and yet more indicative of moral character. In study 2, 210 participants described 18 specific personality traits on 6 dimensions: centrality to self, moral relevance, willful control, brain dependence, temporal stability, and desirability. Consistent with Study 1, centrality to the …

Highly Distinct Chromosomal Structures In Cowpea (Vigna Unguiculata), As Revealed By Molecular Cytogenetic Analysis, Aiko Iwata-Otsubo, Jer-Young Lin, Navdeep Gill, Scott Jackson
Biology Faculty Articles
Cowpea (Vigna unguiculata (L.) Walp) is an important legume, particularly in developing countries. However, little is known about its genome or chromosome structure. We used molecular cytogenetics to characterize the structure of pachytene chromosomes to advance our knowledge of chromosome and genome organization of cowpea. Our data showed that cowpea has highly distinct chromosomal structures that are cytologically visible as brightly DAPI-stained heterochromatic regions. Analysis of the repetitive fraction of the cowpea genome present at centromeric and pericentromeric regions confirmed that two retrotransposons are major components of pericentromeric regions and that a 455-bp tandem repeat is found at seven …
The Regulatory Life Of Threatened Species Lists, Irus Braverman
The Regulatory Life Of Threatened Species Lists, Irus Braverman
Contributions to Books
Published as Chapter 1 in Animals, Biopolitics, Law: Lively Legalities, Irus Braverman, ed.
“The Regulatory Life of Threatened Species Lists” explores a prominent technology for the legal regulation of nonhuman life: the threatened species list. I argue that threatened species lists are biopolitical technologies: they produce and reinforce underlying species ontologies by creating, calculating, and governing the boundaries between various nonhuman species. Such a differentiated treatment of the life and death of nonhuman species through their en-listing, down- and up-listing, multi-listing, and un-listing translates into the positive protection and active governance of such species. Listing threatened species thus becomes a …

New Hampshire Agricultural Experiment Station Publications
Background: With its simple library preparation and robust approach to genome reduction, genotyping-by-sequencing (GBS) is a flexible and cost-effective strategy for SNP discovery and genotyping, provided an appropriate reference genome is available. For resource-limited curation, research, and breeding programs of underutilized plant genetic resources, however, even low-depth references may not be within reach, despite declining sequencing costs. Such programs would find value in an open-source bioinformatics pipeline that can maximize GBS data usage and perform high-density SNP genotyping in the absence of a reference.
Results: The GBS SNP-Calling Reference Optional Pipeline (GBS-SNP-CROP) developed and presented here adopts a clustering strategy …

Malignant phyllodes tumor is a rare breast malignancy with sarcomatous overgrowth and with limited effective treatment options for recurrent and metastatic cases. Recent clinical trials indicated a potential for anti-angiogenic, anti-EGFR and immunotherapeutic approaches for patients with sarcomas, which led us to investigate these and other targetable pathways in malignant phyllodes tumor of the breast. Thirty-six malignant phyllodes tumors (including 8 metastatic tumors with two cases having matched primary and metastatic tumors) were profiled using gene sequencing, gene copy number analysis, whole genome expression, and protein expression. Whole genome expression analysis demonstrated consistent over-expression of genes involved in angiogenesis including …

We have developed a novel Neisseria meningitidis serogroup-specific loop-mediated isothermal amplification (LAMP) assay for six of the most common meningococcal serogroups (A, B, C, W, X, and Y). The assay was evaluated using a set of 31 meningococcal LAMP assay positive cerebrospinal fluid (CSF) specimens from 1574 children with suspected meningitis identified in prospective surveillance between 1998 and 2002 in Vietnam, China, and Korea. Primer specificity was validated using 15 N. meningitidis strains (including serogroups A, B, C, E, W, X, Y, and Z) and 19 non-N. meningitidis species. The N. meningitidis serogroup LAMP detected down to ten copies …

Ovarian cancer constitutes one of the most lethal gynaecological malignancies worldwide and currently no satisfactory therapeutic approaches have been established. Therefore, elucidation of molecular mechanisms to develop targeted therapy of ovarian cancer is crucial. PDLIM2 is critical to promote ubiquitination of nuclear p65 and thus its role in inflammation has been highlighted recently. We demonstrate that PDLIM2 is decreased in both ovarian high-grade serous carcinoma and in various human ovarian cancer cell lines compared with normal ovary tissues and human ovarian surface epithelial cells (HOSE). Further functional analysis revealed that PDLIM2 is epigenetically repressed in ovarian cancer development and inhibition …

Feedstock recalcitrance is the most important barrier impeding cost-effective production of cellulosic biofuels. Pioneer commercial cellulosic ethanol facilities employ thermochemical pretreatment and addition of fungal cellulase, reflecting the main research emphasis in the field. However, it has been suggested that it may be possible to process cellulosic biomass without thermochemical pretreatment using thermophilic, cellulolytic bacteria. To further explore this idea, we examine the ability of various biocatalysts to solubilize autoclaved but otherwise unpretreated cellulosic biomass under controlled but not industrial conditions.
Reconsidering State-Society Relations In South Asia: A Himalayan Case Study, Sanjog Rupakheti
Reconsidering State-Society Relations In South Asia: A Himalayan Case Study, Sanjog Rupakheti
HIMALAYA, the Journal of the Association for Nepal and Himalayan Studies
Since the mid-eighteenth century when armies serving the English East India Company (EIC) clashed with the Gorkhali power, British officers depicted Nepal as an example of classical Hindu despotism. Subsequent scholars of the region have not challenged these representations, taking such colonial descriptors as ‘facts.’ The portrayal of the centralized and ‘despotic’ state in South Asian pasts rests upon a certain understanding of the state’s relationship with society. It calls for imagining the Gorkhali regime as alien to the rest of society and supposing that ethnicity and caste are inflexible from one century to the next. The excessive attention given …

On The Distribution Of The Genus Rhopalurus Thorell, 1876 (Scorpiones: Buthidae) In The Southern Caribbean Islands, Rolando Teruel, Michiel A.C. Cozijn
Euscorpius
In the present note, we report on the occurrence of the genus Rhopalurus Thorell, 1898, in the southern Caribbean islands offshore Venezuela. The only published records are from Isla Margarita and Los Roques, but our study of new specimens (including an important collection assembled by the late Pieter Wagenaar Hummelinck) proved this genus to be widely distributed along several archipelagos such as Los Testigos, Los Frailes, and Los Hermanos, as well as the larger, separate islands of Margarita, Cubagua, La Tortuga, and Coche. These specimens are tentatively referred here to Rhopalurus laticauda Thorell, 1876, but their precise identity still warrants …
