Weak Interactions Between Salmonella Enterica Flhb And Other Flagellar Export Apparatus Proteins Govern Type Iii Secretion Dynamics, Jonathan L. Mcmurry, Tohru Minamino, Yukio Furukawa, Joshua W. Francis

Faculty Articles

The bacterial flagellum contains its own type III secretion apparatus that coordinates protein export with assembly at the distal end. While many interactions among export apparatus proteins have been reported, few have been examined with respect to the differential affinities and dynamic relationships that must govern the mechanism of export. FlhB, an integral membrane protein, plays critical roles in both export and the substrate specificity switching that occurs upon hook completion. Reported herein is the quantitative characterization of interactions between the cytoplasmic domain of FlhB (FlhBC) and other export apparatus proteins including FliK, FlhAC and FliI. FliK and FlhAC bound …

BACKGROUND: Reproductive success depends on a functional oviduct for gamete storage, maturation, fertilization, and early embryonic development. The ovarian-derived steroids estrogen and progesterone are key regulators of oviductal function. The objective of this study was to investigate luteal and follicular phase-specific oviductal epithelial cell function by using microarray-based transcriptional profiling, to increase our understanding of mRNAs regulating epithelial cell processes, and to identify novel genes and biochemical pathways that may be found to affect fertility in the future.

METHODS: Six normally cycling Angus heifers were assigned to either luteal phase (LP, n = 3) or follicular phase (FP, …

Reverse transcriptase-quantitative polymerase chain reaction (RT-qPCR) is a reliable technique for measuring and evaluating gene expression during variable biological processes. To facilitate gene expression studies, normalization of genes of interest relative to stable reference genes is crucial. The western flower thrips Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), the main vector of tomato spotted wilt virus (TSWV), is a destructive invasive species. In this study, the expression profiles of 11 candidate reference genes from nonviruliferous and viruliferous F. occidentalis were investigated. Five distinct algorithms, geNorm, NormFinder, BestKeeper, the ΔCt method, and RefFinder, were used to determine the performance …

Earlier efforts have been concerned with the association of Ruthenium octahedral complexes with DNA. Since the positively charged ruthenium species has been found to associate with the electron rich major groove of double-stranded DNA, it was proposed the addition of cationic sites on the ligands attached to ruthenium would facilitate such association. Thus, we have synthesized several series of octahedral ruthenium complexes bearing ligands having within themselves cationic sites. These have been investigated in their interaction with calf thymus DNA using fluorescence titration analysis. The introduction of the cationic ligands has been found to exhibit enhanced association of the ruthenium …

The Role Of Visualization And 3-D Printing In Biological Data Mining, Talia L. Weiss, Amanda Zieselman, Douglas P. Hill, Solomon G. Diamond, Li Shen, Andrew J. Saykin, Jason H. Moore

Dartmouth Scholarship

Background:

Biological data mining is a powerful tool that can provide a wealth of information about patterns of genetic and genomic biomarkers of health and disease. A potential disadvantage of data mining is volume and complexity of the results that can often be overwhelming. It is our working hypothesis that visualization methods can greatly enhance our ability to make sense of data mining results. More specifically, we propose that 3-D printing has an important role to play as a visualization technology in biological data mining. We provide here a brief review of 3-D printing along with a case study to …

Crispr/Cas9-Based Generation Of Knockdown Mice By Intronic Insertion Of Artificial Microrna Using Longer Single-Stranded Dna., Hiromi Miura, Channabasavaiah B. Gurumurthy, Takehito Sato, Masahiro Sato, Masato Ohtsuka

Journal Articles: Genetics, Cell Biology & Anatomy

Knockdown mouse models, where gene dosages can be modulated, provide valuable insights into gene function. Typically, such models are generated by embryonic stem (ES) cell-based targeted insertion, or pronuclear injection, of the knockdown expression cassette. However, these methods are associated with laborious and time-consuming steps, such as the generation of large constructs with elements needed for expression of a functional RNAi-cassette, ES-cell handling, or screening for mice with the desired knockdown effect. Here, we demonstrate that reliable knockdown models can be generated by targeted insertion of artificial microRNA (amiRNA) sequences into a specific locus in the genome [such as intronic …

Investigating The Impact Of Sibling Foster Care On Placement Stability, Jeffrey David Waid Aug 2015

Investigating The Impact Of Sibling Foster Care On Placement Stability, Jeffrey David Waid

Dissertations and Theses

Sibling relationships are an important, yet under investigated aspect of foster care research. Despite the fact that between 65-85% of children in care have brothers and sisters, only recently have child welfare researchers begun to explore the complex and dynamic nature of sibling relationships in substitute care settings. Although cross-sectional and longitudinal studies suggest differences in placement stability and permanency outcomes for siblings placed together versus those placed separately, the conditions under which sibling relationships influence placement stability, permanency, and well-being in foster care settings remain unknown.

This dissertation investigated how family dynamics and home setting characteristics influenced the likelihood …

Catholic Teachers’ Postgraduate Qualifications And Students’ End Of Schooling Outcomes: A Large Scale Queensland Based Comparative Study, Judy Smeed, Beryl Exley

eJournal of Catholic Education in Australasia

Abstract

A key feature of the current era of Australian schooling is the dominance of publically available student, school and teacher performance data. Our paper examines the intersection of data on teachers’ postgraduate qualifications and students’ end of schooling outcomes in 26 Catholic Systemic Secondary Schools and 18 Catholic Independent Secondary Schools throughout the State of Queensland. We introduce and justify taking up a new socially-just measurement model of students’ end of schooling outcomes, called the ‘Tracking and Academic Management Index’, otherwise known as ‘TAMI’. Additional analysis is focused on the outcomes of top-end students vis-à-vis all students who are …

Adaptive Dynamic Walking And Motion Optimization For Humanoid Robots, Andreas Seekircher Aug 2015

Adaptive Dynamic Walking And Motion Optimization For Humanoid Robots, Andreas Seekircher

Open Access Dissertations

An essential ability of a robot is to act in its environment by generating motions for locomotion or manipulation. This can be a challenging problem on a robot with high degrees of freedom. Although biped robots have shown drastic improvements with regard to motion skills over the past few years, many approaches for generating motions still require tedious and time-consuming manual calibration due to variances in the hardware or inaccurate sensors and actuators. This research focuses on generating motions for humanoid robots automatically without manual calibration. The first approach discussed uses parameter optimization (e.g. CMA-ES, PSO) to directly optimize the …

Evaluation Of 5 Μm Superficially Porous Particles For Capillary And Microfluidic Lc Columns, James Grinias, Robert T. Kennedy Aug 2015

Evaluation Of 5 Μm Superficially Porous Particles For Capillary And Microfluidic Lc Columns, James Grinias, Robert T. Kennedy

College of Science & Mathematics Departmental Research

Large-size (4–5 µm) superficially porous particles yield lower plate heights (e.g., the minimal reduced plate height or hmin ≈ 1.5) than fully porous particles of a similar size when packed into large-bore columns. This property allows for better chromatographic performance without the higher pressures required for smaller particles. This study explores the use of such particles in microfluidic LC columns where materials and fitting pressure limits can constrain the size of particle used. The theoretically predicted performance improvements compared to fully porous particles were not demonstrated in capillary columns (with hmin ≈ 2 for both particle types), in …

Comparative Foliar Water Uptake And Leaf Hydrophobicity In Island Versus Mainland Ferns, Victoria M. Lekson, Helen I. Holmlund, Nicole A. Nakamatsu, Amanda M. Burns, Stephen D. Davis

Biology

In 2014, Southern California experienced the worst drought in the last 1,200 years. In this study, the mechanisms of drought tolerance for eight species of ferns in the Santa Monica Mountains were assessed with a focus on foliar water uptake and hydrophobicity of frond (leaf) surfaces. We measured gravimetric foliar water uptake and angle of contact for a drop of water on a frond. Seasonal shifts were tracked as a method of following drought response patterns.

Fog is becoming more important due to limited ground water. Fog is far more common on Santa Cruz Island off the coast of California …

Beclin-1 Expression Is Retained In High-Grade Serous Ovarian Cancer Yet Is Not Essential For Autophagy Induction In Vitro, Rohann J.M. Correa, Yudith R. Valdes, Trevor G. Shepherd, Gabriel E. Dimattia

Anatomy and Cell Biology Publications

Background: Autophagy is a conserved cellular self-digestion mechanism that can either suppress or promote cancer in a context-dependent manner. In ovarian cancer, prevalent mono-allelic deletion of BECN1 (a canonical autophagy-inducer) suggests that autophagy is impaired to promote carcinogenesis and that Beclin-1 is a haploinsufficient tumor suppressor. Nonetheless, autophagy is known to be readily inducible in ovarian cancer cells. We sought to clarify whether Beclin-1 expression is in fact disrupted in ovarian cancer and whether this impacts autophagy regulation.

Methods: BECN1 expression levels were assessed using The Cancer Genome Atlas (TCGA) datasets from 398 ovarian high-grade serous cystadenocarcinomas (HGSC) and protein …

Current strategies for early detection of breast and other cancers are limited in part because some lesions identified as potentially malignant do not develop into aggressive tumors. Acid pH has been suggested as a key characteristic of aggressive tumors that might distinguish aggressive lesions from more indolent pathology. We therefore investigated the novel class of molecules, pH low insertion peptides (pHLIPs), as markers of low pH in tumor allografts and of malignant lesions in a mouse model of spontaneous breast cancer, BALB/neu-T. pHLIP Variant 3 (Var3) conjugated with fluorescent Alexa546 was shown to insert into tumor spheroids in a sequence-specific …

A Case Study Of The Impediments To The Commercialization Of Research At The University Of Kentucky, Nathan L. Vanderford, Elizabeth Marcinkowski

Markey Cancer Center Faculty Publications

The commercialization of university-based research occurs to varying degrees between academic institutions. Previous studies have found that multiple barriers can impede the effectiveness and efficiency by which academic research is commercialized. This case study was designed to better understand the impediments to research commercialization at the University of Kentucky via a survey and interview with three successful academic entrepreneurs. The study also garnered insight from the individuals as to how the commercialization process could be improved. Issues with commercialization infrastructure; a lack of emphasis, at the university level, on the importance of research commercialization; a void in an entrepreneurial culture …

Large Amplitude, Short Wave Peristalsis And Its Implications For Transport, Lindsay D. Waldrop, Laura A. Miller Aug 2015

Large Amplitude, Short Wave Peristalsis And Its Implications For Transport, Lindsay D. Waldrop, Laura A. Miller

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

Valveless, tubular pumps are widespread in the animal kingdom, but the mechanism by which these pumps generate fluid flow is often in dispute. Where the pumping mechanism of many organs was once described as peristalsis, other mechanisms, such as dynamic suction pumping, have been suggested as possible alternative mechanisms. Peristalsis is often evaluated using criteria established in a technical definition for mechanical pumps, but this definition is based on a small-amplitude, long-wave approximation which biological pumps often violate. In this study, we use a direct numerical simulation of large-amplitude, short-wave peristalsis to investigate the relationships between fluid flow, compression frequency, …

Work In Progress -- Does A Technology-Rich Transportation Engineering Experiences Increase Interest In Civil Engineering, Marissa K. Orr, Sanjay Tewari, David E. Hall, Norman D. Pumphrey

Civil, Architectural and Environmental Engineering Faculty Research & Creative Works

This paper examines whether a technology-rich transportation engineering experience generates interest among college freshman students towards transportation related degrees and careers, specifically, whether it increases interest in civil engineering. Louisiana Tech’s first-year engineering experience provides relevant, project-focused education for over 500 students each year and an opportunity to introduce transportation-related projects. The six semester-hour, three-course sequence, called “Living with the Lab,” boosts experiential learning through student ownership of inexpensive laboratory equipment. In one section of the Living with the Lab course, a transportation engineer visited the class and the students were led through a technology-rich transportation engineering activity. They used …

State Funding Of Nontherapeutic Abortions; Medicaid Plans; Equal Protection; Right To Choose An Abortion; Beal V. Doe, Maher V. Roe, Poelker V. Doe, Constance Leistiko

Akron Law Review

In Beal v. Doe the United States Supreme Court held that Title XIX of the Social Security Act permits but does not require states participating in the Medicaid program established by that Act to fund nontherapeutic abortions. In the companion cases of Maher v. Roe and Poelker v. Doe, the same majority held in Maher that the Equal Protection Clause does not require a state that funds childbirth and therapeutic abortions to also fund the costs of nontherapeutic abortions, and in Poelker, that the Constitution does not prohibit a state or city from forbidding the performance of elective …

Forensic Autopsy. The Particularities Of Consent For Research, Sorin Hostiuc Aug 2015

Forensic Autopsy. The Particularities Of Consent For Research, Sorin Hostiuc

Journal of Health Ethics

Forensic autopsy has as its main goal to find out the truth about the causes and circumstances of death, when there is a reasonable suspicion to be caused by a violent act. As the legal guardian cannot refuse a forensic autopsy, the forensic physician is free to perform any procedures considered to be necessary for finding out the truth (in order to answer to the objectives asked by the authority). However the use of biological samples or data in research is limited and needs an additional consent, from the legal guardian, which can lead to difficulties in the interpretation of …
