Disrupting Sumoylation Enhances Transcriptional Function And Ameliorates Polyglutamine Androgen Receptor-Mediated Disease., Jason P Chua, Satya L Reddy, Zhigang Yu, Elisa Giorgetti, Heather L Montie, Sarmistha Mukherjee, Jake Higgins, Richard C Mceachin, Diane M Robins, Diane E Merry, Jorge A Iñiguez-Lluhí, Andrew P Lieberman

Department of Biochemistry and Molecular Biology Faculty Papers

Expansion of the polyglutamine (polyQ) tract within the androgen receptor (AR) causes neuromuscular degeneration in individuals with spinobulbar muscular atrophy (SBMA). PolyQ AR has diminished transcriptional function and exhibits ligand-dependent proteotoxicity, features that have both been implicated in SBMA; however, the extent to which altered AR transcriptional function contributes to pathogenesis remains controversial. Here, we sought to dissociate effects of diminished AR function from polyQ-mediated proteotoxicity by enhancing the transcriptional activity of polyQ AR. To accomplish this, we bypassed the inhibitory effect of AR SUMOylation (where SUMO indicates small ubiquitin-like modifier) by mutating conserved lysines in the polyQ AR that …

Fungal Mediator Tail Subunits Contain Classical Transcriptional Activation Domains, Zhongle Liu, Lawrence C. Myers Feb 2015

Fungal Mediator Tail Subunits Contain Classical Transcriptional Activation Domains, Zhongle Liu, Lawrence C. Myers

Dartmouth Scholarship

Classical activation domains within DNA-bound eukaryotic transcription factors make weak interactions with coactivator complexes, such as Mediator, to stimulate transcription. How these interactions stimulate transcription, however, is unknown. The activation of reporter genes by artificial fusion of Mediator subunits to DNA binding domains that bind to their promoters has been cited as evidence that the primary role of activators is simply to recruit Mediator. We have identified potent classical transcriptional activation domains in the C termini of several tail module subunits of Saccharomyces cerevisiae, Candida albicans, and Candida dubliniensis Mediator, while their N-terminal domains are necessary and sufficient for their …

Same-Sex Marriage And Loving V. Virginia: Analogy Or Disanalogy?, Ronald Turner Feb 2015

Same-Sex Marriage And Loving V. Virginia: Analogy Or Disanalogy?, Ronald Turner

Washington and Lee Law Review Online

In its 1967 decision in Loving v. Virginia, the United States Supreme Court struck down Virginia antimiscegenation laws prohibiting and criminalizing interracial marriages, holding that the challenged laws violated the Due Process and Equal Protection Clauses of the Fourteenth Amendment to the United States Constitution. In recent federal appeals court decisions, Loving has been invoked as an authoritative analogy supporting plaintiffs’ claims that same-sex marriage bans violate the Constitution. This Essay considers the posited Loving analogy and the contentions (1) that different-race marriage and same-sex marriage prohibitions present similar, albeit not identical, instances of unconstitutional state limitations on an …

The Value Of The Follow-Through Derives From Motor Learning Depending On Future Actions., Ian S. Howard, Daniel M. Wolpert, David W. Franklin Feb 2015

The Value Of The Follow-Through Derives From Motor Learning Depending On Future Actions., Ian S. Howard, Daniel M. Wolpert, David W. Franklin

School of Engineering, Computing and Mathematics

In ball sports, we are taught to follow through, despite the inability of events after contact or release to influence the outcome [1, 2]. Here we show that the specific motor memory active at any given moment critically depends on the movement that will be made in the near future. We demonstrate that associating a different follow-through movement with two motor skills that normally interfere [3-7] allows them to be learned simultaneously, suggesting that distinct future actions activate separate motor memories. This implies that when learning a skill, a variable follow-through would activate multiple motor memories across practice, whereas a …

All In The Family: A Retrospective Study Comparing Sibling Bullying And Peer Bullying, Lori A. Hoetger, Katherine P. Hazen, Eve M. Brank

Center on Children, Families, and the Law: Faculty Publications

Extensive bullying research has primarily focused on activities between peers in school settings, but some evidence suggests bullying may occur in other situations. If so, other contexts could potentially benefit from the wealth of peer bullying research. A sample of 392 young adults answered questions about their experiences with sibling and peer bullying behaviors. Participants also provided responses concerning a sibling or peer vignette that focused on reporting bullying behaviors. Results indicated that participants view bullying behaviors between peers and siblings as somewhat similar, but sibling bullying behaviors compared to peer bullying behaviors are reported to be perpetrated and experienced …

Pain Management Via Local Anesthetics And Responsive Hydrogels, Kyle R. Bagshaw, Curt L. Hanenbaum, Erica J. Carbone, Kevin Wh Lo, Cato T. Laurencin, Joseph Walker, Lakshmi S. Nair

Articles - Patient Care

Acute and chronic pain control is a significant clinical challenge that has been largely unmet. Local anesthetics are widely used for the control of post-operative pain and in the therapy of acute and chronic pain. While a variety of approaches are currently used to prolong the duration of action of local anesthetics, an optimal strategy to achieve neural blockage for several hours to days with minimal toxicity has yet to be identified. Several drug delivery systems such as liposomes, microparticles and nanoparticles have been investigated as local anesthetic delivery vehicles to achieve prolonged anesthesia. Recently, injectable responsive hydrogels raise significant …

Hyperactivation Of Bdnf-Trkb Signaling Cascades In Human Hypothalamic Hamartoma (Hh): A Potential Mechanism Contributing To Epileptogenesis, Suzan Semaan, Jie Wu, Yan Gan, Yu Jin, Guo Hui Li, John F. Kerrigan, Yong Chang, Yao Huang Feb 2015

Hyperactivation Of Bdnf-Trkb Signaling Cascades In Human Hypothalamic Hamartoma (Hh): A Potential Mechanism Contributing To Epileptogenesis, Suzan Semaan, Jie Wu, Yan Gan, Yu Jin, Guo Hui Li, John F. Kerrigan, Yong Chang, Yao Huang

Translational Neuroscience

Aims: Although compelling evidence suggests that human hypothalamic hamartoma (HH) is intrinsically epileptogenic for gelastic seizures, the molecular mechanisms responsible for epileptogenesis within HH remain to be elucidated. The aim of this study was to test the hypothesis that hyperactivation of BDNF-TrkB signaling pathways in surgically resected HH tissue is a possible mechanism for downregulation of KCC2 expression, which in turn underlies GABA-mediated excitation within HH. Methods: Activation of three major BDNF-TrkB signaling pathways including MAPKs, Akt, and PLCγ1 were evaluated in surgically resected HH tissue (n = 14) versus human hypothalamic control tissue (n = 8) using combined methodologies …

Creating A Healthier Campus: A Report On The Effectiveness Of Nsu’S “Slimming With The Sharks” Wellness Program, Corey Peacock, Tobin Silver, Pradeep Vanguri

Quadrivium: A Journal of Multidisciplinary Scholarship

With a recent increase in college obesity rates, it is imperative to assess the effectiveness of campus-wide fitness programs while encouraging other institutes of higher education to do so. It is presently uncertain whether these programs are advantageous or disadvantageous for improving campus-wide obesity rates; therefore, assessing campus-wide fitness programs and reporting results may bring awareness to college campus obesity rates and clarify a solution to the problem. NSU’s RecPlex offered “Slimming with the Sharks,” a structured, multiple-component, weight loss initiative available to all students, faculty, and staff. The following is a brief report of “Slimming with the Sharks” data …
