Can Simple Rules Control Development Of A Pioneer Vertebrate Neuronal Network Generating Behavior?, A Roberts, D Conte, M Hull, R Merrison-Hort, Et Al

School of Engineering, Computing and Mathematics

How do the pioneer networks in the axial core of the vertebrate nervous system first develop? Fundamental to understanding any full-scale neuronal network is knowledge of the constituent neurons, their properties, synaptic interconnections, and normal activity. Our novel strategy uses basic developmental rules to generate model networks that retain individual neuron and synapse resolution and are capable of reproducing correct, whole animal responses. We apply our developmental strategy to young Xenopus tadpoles, whose brainstem and spinal cord share a core vertebrate plan, but at a tractable complexity. Following detailed anatomical and physiological measurements to complete a descriptive library of each …

We recently discovered that the collagen IV scaffolds of bovine, mouse, and Drosophila melanogaster tissues are stabilized by sulfilimine chemical bonds (S = N) (20⇓–22). The bond is a covalent cross-link between methionine-93 (Met93) and lysine-211 or hydroxylysine-211 (Lys211/Hyl211) residues that stabilizes the noncollagenous (NC1) interface of adjoining triple helical protomers (Fig. 1A). Peroxidasin, an animal heme peroxidase embedded in basement membrane, forms these sulfilimine cross-links by producing hypohalous acids as intermediates for Met93 oxidization (21, 22). Here, we explored whether the sulfilimine cross-link of collagen IV is a fundamental requirement in the genesis and evolution of tissues in Eumetazoa. …

Volume Exclusion Agent To Enhance Formation Of Extracellular Matrix, David E. Birk, John Hassell, Bradley Kane, La Tia Etheredge

USF Patents

A method of enhancing the formation of extracellular matrix in culture. Cells in culture secrete most of the collagen into the media as unprocessed procollagen, i.e., the cells do not convert procollagen to collagen. In contrast, normal extracellular matrix deposition involves procollagen processing to collagen, fibril assembly and deposition into the cell layer to form a collagenous extracellular matrix. The addition of certain growth factors and the addition of a thin layer of a certain volume exclusion agent on top of the cells dramatically enhances the conversion of procollagen to collagen and will increase the amount of collagen and extracellular …

Redox-Dependent Stability, Protonation, And Reactivity Of Cysteine-Bound Heme Proteins, Fangfang Zhong, George P. Lisi, Daniel P. Collins, John H. Dawson, Ekaterina V. Pletneva

Dartmouth Scholarship

Cysteine-bound hemes are key components of many enzymes and biological sensors. Protonation (deprotonation) of the Cys ligand often accompanies redox transformations of these centers. To characterize these phenomena, we have engineered a series of Thr78Cys/Lys79Gly/Met80X mutants of yeast cytochrome c (cyt c) in which Cys78 becomes one of the axial ligands to the heme. At neutral pH, the protonation state of the coordinated Cys differs for the ferric and ferrous heme species, with Cys binding as a thiolate and a thiol, respectively. Analysis of redox-dependent stability and alkaline transitions of these model proteins, as well as comparisons to Cys …

A Life History Perspective On Skin Cancer And The Evolution Of Skin Pigmentation, Daniel L. Osborne, Raymond B. Hames Jan 2014

A Life History Perspective On Skin Cancer And The Evolution Of Skin Pigmentation, Daniel L. Osborne, Raymond B. Hames

Department of Anthropology: Faculty Publications

The ancestral state of human skin pigmentation evolved in response to high ultraviolet radiation (UVR) stress. Some argue that pigmentation evolved to limit folate photolysis, therein limiting neural tube defects. Pigmentation also protects against sunburn which decreases the efficiency of sweating and potentiates skin infection. Pigmentation increases the efficacy of skin as a barrier to infection. Skin cancer has been rejected or minimized as a selective pressure because it is believed to have little or no effect on mortality during reproductive years. This argument ignores evidence of human longevity as a derived life history trait and the adaptive value of …

N-Substituted 5-Amino-6-Methylpyrazine-2,3-Dicarbonitriles: Microwave-Assisted Synthesis And Biological Properties, Aidan Coffey, Jiahui Guo, Et. Al.

Department of Biological Sciences Publications

In this work a series of 15 N-benzylamine substituted 5-amino-6-methyl-pyrazine-2,3-dicarbonitriles was prepared by the aminodehalogenation reactions using microwave assisted synthesis with experimentally set and proven conditions. This approach for the aminodehalogenation reaction was chosen due to its higher yields and shorter reaction times. The products of this reaction were characterized by IR, NMR and other analytical data. The compounds were evaluated for their antibacterial, antifungal and herbicidal activity. Compounds 3 (R = 3,4-Cl), 9 (R = 2-Cl) and 11 (R = 4-CF3) showed good antimycobacterial activity against Mycobacterium tuberculosis (MIC = 6.25 µg/mL). It was found that the lipophilicity is …

Light And Affects From A Comparative Point Of View, Kyle Takaki Jan 2014

Light And Affects From A Comparative Point Of View, Kyle Takaki

Comparative Philosophy

Light metaphors occurring in Chinese philosophy and Stoicism are of special interest for the unique ways they channel potentialities of the self. In this paper I apply ideas from cognitive linguistics to examine selected structural features of these metaphors. I also build on these ideas by presenting a framework regarding affects to assist in disclosing what is at stake for differing Chinese and Stoic technologies of the self. The paper adopts a high-level perspective to see these broad philosophical implications, interleaving discussions of Chinese philosophy (mainly views associated with Daoism), Stoicism (bringing into relief important differences from these views), and …

Measurement Issues With Couple-And Family-Level Data, Dean M. Busby, Franklin O. Poulsen

Faculty Publications

Early in my (D.B.) training as a family therapist, I did some co-therapy with a clinical psychology student who wanted to learn how to work with couples. The couple we were working with was quite volatile, and it was not uncommon for the dialogue between the spouses to get heated. While it was challenging enough to keep the interactions between the partners moving in a productive direction, my co-therapist was quite uncomfortable with conflict and would emotionally "check out" of the session as soon as things became intense. Each time a session was challenging and conflict became intense, my co-therapist …

With the trend of an increasing aged population worldwide, Alzheimer’s disease (AD), an age-related neurodegenerative disorder, as one of the major causes of dementia in elderly people is of growing concern. Despite the many hard efforts attempted during the past several decades in trying to elucidate the pathological mechanisms underlying AD and putting forward potential therapeutic strategies, there is still a lack of effective treatments for AD. The efficacy of many potential therapeutic drugs for AD is of main concern in clinical practice. For example, large bodies of evidence show that the antitumor histone deacetylase (HDAC) inhibitor, suberoylanilidehydroxamic acid (SAHA), …

The Emerging Role Of Oxylipins In Thrombosis And Diabetes., Benjamin E Tourdot, Intekhab Ahmed, Michael Holinstat

Division of Endocrinology, Diabetes and Metabolic Diseases Faculty Papers

The prevalence of cardiovascular disease (CVD), the leading cause of death in the US, is predicted to increase due to the shift in age of the general population and increase in CVD risk factors such as obesity and diabetes. New therapies are required to decrease the prevalence of CVD risk factors (obesity and diabetes) as well as reduce atherothrombosis, the major cause of CVD related mortality. Oxylipins, bioactive metabolites derived from the oxygenation of polyunsaturated fatty acids, play a role in the progression of CVD risk factors and thrombosis. Aspirin, a cyclooxygenase-1 inhibitor, decreases atherothrombotic associated mortality by 25%. These …

High-throughput technologies are enabling scientists to profile genomes, transcriptomes, proteomes, and metabolomes at an unprecedented scale [1]. All this “-omics” research (let's call it genomics for simplicity) is exciting—and game changing—but it's also fraught with dangers for the tenderfoot. Here, we've put together a brief “field guide” for those wishing to visit the genomics frontier, in which we use caricatures to illustrate various pitfalls that can beset those who inhabit this new territory. By documenting the behaviors of these common types, we hope to guide researchers in their quest to apply sound practices when designing genomics experiments and analyzing …

Acute kidney injury (AKI) research in the past decade has mostly focused upon development of a standard AKI definition, validation of early novel biomarkers to predict AKI prior to serum creatinine rise and predict AKI severity, and assessment of aspects of renal replacement therapies and their impact on survival. Given the independent association between AKI and mortality in the acute phase, such focus makes imminent sense. More recently, the recognition that AKI is associated with subsequent development of chronic kidney disease and end-stage renal disease, with the attendant increase in mortality, has led to interest in the clinical epidemiology and …
