Probing Proteomes With Alternative Proteases: Lys-N & Arg-C, Thomas Earle Platé Iv Jan 2014

Probing Proteomes With Alternative Proteases: Lys-N & Arg-C, Thomas Earle Platé Iv

Theses & ETDs

Proteomics, the study of the protein functional complement of the genome, deals with similar issues of amino-acid sequence coverage and depth. Trypsin has been the most common protease used to cleave peptides into manageable units due the many optimum traits of tryptic digestions for mass spectrometry (MS) analyses for the entire history of “shotgun” proteomics. Instrumental and computational methods for proteomic analyses have been tailored for tryptic peptides as a result. This project explored the feasibility of an alternative approach that utilizes a sequential digestion by lesser-used proteases, a C-terminal arginine peptidase (Arg-C or Clostripain) and a specific N-terminal lysine …

Exploring Associations Between Assertiveness, Health, And Gender In Children Ages 6-11, Kelsey Jae Smith Jan 2014

Exploring Associations Between Assertiveness, Health, And Gender In Children Ages 6-11, Kelsey Jae Smith

Theses & ETDs

There is considerable evidence of a gender difference in health among adolescents and adults, however there is a lack of data surrounding the development of these disparities in children. In addition to gender, research in assertiveness shows clear associations with health, but the literature is unclear how gender and assertiveness interact with one another (Sarkova et.al., 2013). The current study sought to fill the gap through exploratory research into children’s assertiveness skills and self-reported health outcomes and by examining sex differences in assertiveness skills when gender socialization, measured by child’s perceptions of parent and peer pressure to conform to a …

Pursuit, Punishment, And Perversion: Variation In Ovid’S Metamorphoses, Ashley Frost Jan 2014

Pursuit, Punishment, And Perversion: Variation In Ovid’S Metamorphoses, Ashley Frost

Theses & ETDs

Ovid’s Metamorphoses compiles more than 250 Greek and Roman myths. This thesis seeks to explain how the poet differentiates tales that share the same theme through close analysis of both the Latin text and an English translation. I show that Ovid individuates myths by controlling how his reader perceives and responds to each tale. In chapter one, which discusses myths that share the theme of unrequited love, including Apollo and Daphne, Pan and Syrinx, Acis, Galatea and Polypehmus, and Glaucis and Scylla, Ovid manipulates every such tale to encompass some degree of humor, either undercutting or compounding the threat of …

“So Long And Thanks For All The Fish”: Human Interactions With A Begging Bottlenose Dolphin (Tursiops Truncatus) And Effects Of Educational Efforts, Madelaine Verbeek Jan 2014

“So Long And Thanks For All The Fish”: Human Interactions With A Begging Bottlenose Dolphin (Tursiops Truncatus) And Effects Of Educational Efforts, Madelaine Verbeek

Theses & ETDs

Human-animal interaction (HI) is defined as any interaction humans and animals have with each other, ranging from cooperation to direct conflict. Although HI is a frequent occurrence, the categories these interactions are divided into range within the scientific community, making comparisons between studies difficult due to inconsistent definitions and approaches among researchers. The “interactor” category, the individual who engages in the interaction, is readily missed during the evaluation process, however knowing the perspective from which the interaction took place is critical to understanding the interaction as a whole. For potentially harmful HI situations, understanding the interactor’s perspective along with the …

The Effects Of Fish Waste Nutrient Enrichment On Thalassia Testudinum Banks Ex König In Sarasota Bay, Florida, Kyle Mccormick Jan 2014

The Effects Of Fish Waste Nutrient Enrichment On Thalassia Testudinum Banks Ex König In Sarasota Bay, Florida, Kyle Mccormick

Theses & ETDs

Seagrass has numerous vital functions in marine ecosystems worldwide that range from binding substrate to providing food and shelter for fauna. However, seagrass loss is at a dangerously high rate across the globe. Anthropogenic factors such as dredging and pollution have caused seagrass beds to decline in recent years. The lack of methodical restoration efforts as well as the general variability of open ocean restorations has made restoration efforts difficult, but new emerging techniques and advances in technology give hope for improvement. In this experiment the use of nutrient enrichment to affect Thalassia testudinum colonization was studied in the New …

Small Steps By Mankind, Giant Leaps For Animal Rights: State-Level Farm Animal Welfare Legislation, Malorie Sneed Jan 2014

Small Steps By Mankind, Giant Leaps For Animal Rights: State-Level Farm Animal Welfare Legislation, Malorie Sneed

Theses & ETDs

Farm animal welfare legislation regulates the well-being of animals during their lives on factory farms. The recent passage of citizen-initiated ballot measures and legislation in several states suggests that certain factors play a role in the passage of this legislation. The goal of this thesis was to understand which factors have contributed to the success of this legislation. This study posits that education, agribusiness activity, political ideology, and party identification influence the passage of farm animal welfare legislation. I conducted quantitative correlational tests between these four factors and the presence of farm animal welfare legislation. I found that ideology appears …

The Economics And Sustainability Of Compost At New College Of Florida: A Cost Benefit Analysis, David Smith Jan 2014

The Economics And Sustainability Of Compost At New College Of Florida: A Cost Benefit Analysis, David Smith

Theses & ETDs

This thesis is a cost-benefit analysis of the compost system at New College of Florida. During the spring semester of 2013, data were collected regarding the amount of food that was diverted from the waste stream and composted, as well as the amount of finished compost produced. Benefits to the system were calculated using the known cost of trash removal for the school, the value of the finished compost, and the estimated value of methane sequestration that occurs during the compost process. The cost of the system and operating expenses were then deducted from the benefits. The conclusion found that …

Creating A Spotted Wing Drosophila, Drosophila Suzukii (Matsumura), Fruit Puree Diet To Facilitate Integrated Pest Management Techniques, Lynn Marie Gusman Jan 2014

Creating A Spotted Wing Drosophila, Drosophila Suzukii (Matsumura), Fruit Puree Diet To Facilitate Integrated Pest Management Techniques, Lynn Marie Gusman

Theses & ETDs

Spotted Wing Drosophila, Drosophila suzukii (Matsumura), is an invasive fruit fly of Asian origins. It uses a serrated ovipositor to lay eggs in soft-skin fruit such as strawberries, blueberries, grapes, and caneberries, causing fruit to rot early. Because of this damage, it has been deemed an economically important pest that must be controlled by Integrated Pest Management practices. The goal of this study was to determine if different fruits impact the fecundity and overall success of SWD. To rear large numbers of SWD to facilitate testing, we designed puree diets from strawberries and grapes for ease of rearing and fungus …

Unjust Deserts: Food Insecurity, Structural Inequality, And Community Agency In Newtown, Fl, Juliana Dearr Jan 2014

Unjust Deserts: Food Insecurity, Structural Inequality, And Community Agency In Newtown, Fl, Juliana Dearr

Theses & ETDs

This thesis focuses on the issues surrounding food access in Newtown, the historic black neighborhood of Sarasota, FL. Poor nutrition, particularly in economically distressed areas, reinforces and maintains the violent cycle of poverty, as nutrition problems increase the risk of chronic health conditions. Ethnography and quantitative data collection on food availability and transportation access are the key methods. This research will show Newtown's consumption of healthy food is hindered by the sparse availability of fresh food, lacking transportation, education gaps, and time and money constraints. These barriers must be addressed to create systemic improvements to Newtown's welfare. Several community-based organizations, …

Gardens As Educational Tools In Urban Public Schools, Michelle Diane Wheat Jan 2014

Gardens As Educational Tools In Urban Public Schools, Michelle Diane Wheat

Theses & ETDs

Urban public school education has become increasingly decontextualized from the natural world over time. This causes a division within students that impacts how they learn, what they learn, and their mental health. Children's knowledge is increasingly devoid of environmental awareness. In addition ADHD, depression, and obesity have been linked to this separation. These problems are exacerbated by a culture of standardized tests motivated by financial concerns, and the gearing of education toward career preparation. Children learn to fear nature and reap none of the benefits that immersion in nature can foster. School gardens, especially in urban areas, can provide a …

Benzyl Isothiocyanate As An Adjuvant Chemotherapy Option For Head And Neck Squamous Cell Carcinoma, Mary Allison Wolf Jan 2014

Benzyl Isothiocyanate As An Adjuvant Chemotherapy Option For Head And Neck Squamous Cell Carcinoma, Mary Allison Wolf

Theses, Dissertations and Capstones

Isothiocyanates (ITCs) are natural phytochemicals produced by cruciferous vegetables. Recent evidence supports that, in addition to cancer prevention, ITCs can use various mechanisms to target malignant cells. Current therapies for cancer often provoke detrimental side effects, however clinical evidence supports that ITCs have little to no side effects in patients. Consequently, ITCs may be a promising treatment option for cancer patients, especially patients suffering from head and neck squamous cell carcinoma (HNSCC).

Despite recent improvements in cancer treatment, overall survival of advanced HNSCC has not improved in the past three decades. Metastasis and chemoresistance represent two detrimental events that greatly …

Severe Morbidity And Mortality Risk From Malaria In The United States, 1985–2011, Jimee Hwang, Karen A. Cullen, S. Patrick Kachur, Paul M. Arguin, J. Kevin Baird

United States Public Health Resources

Background. Recent reports of Plasmodium vivax associated with severe syndromes and mortality from malaria endemic areas questions the “benign” course of non-falciparum malarias.

Methods. We retrospectively analyzed data from patients reported to the US Centers for Disease Control and Prevention with a diagnosis of malaria parasite single-species infection between 1985 and 2011. Patients classified as having severe illness were further classified according to outcome (survival versus death) and clinical syndrome.

Results. Among all cases, .9% of Plasmodium falciparum cases resulted in death and 9.3% were classified as severe, whereas .09% of P. vivax cases resulted in death and 1.3% were …

State-Specific Estimates Of Complete Smoke-Free Home Rules Among Postpartum Women, 2010, Van T. Tong, Yalonda Hutchings, Sherry L. Farr, Denise D'Angelo, Stephen Babb

United States Public Health Resources

Background: Secondhand smoke exposure increases an infant's risk of morbidity and mortality. We provide state-specific estimates for and characterize postpartum women with complete smoke-free home rules.

Methods: Data were analyzed from 26 states and New York City (n= 37,698) from the 2010 Pregnancy Risk Assessment Monitoring System, a population-based survey of women who recently delivered live-born infants. We calculated state-specific estimates of complete rules and assessed associations between complete rules and selected characteristics.

Results: Overall, 93.6% (95% CI: 93.1–94.1) of women with recent live births had complete smoke-free home rules (86.8% [West Virginia] to 98.6% [Utah]). Demographic groups with the …

Bottle-Feeding Practices During Early Infancy And Eating Behaviors At 6 Years Of Age, Ruowei Li, Kelley S. Scanlon, Ashleigh May, Chelsea Rose, Leann L. Birch

United States Public Health Resources

BACKGROUND: Evidence suggests an association of breastfeeding with a maternal feeding style (MFS) that is less controlling than formula feeding, which, in turn, may improve a child’s self-regulation of eating. This study examines associations of bottle-feeding practices during infancy with MFS and children’s eating behavior (CEB) at 6 years old.

METHODS: We linked data from the Infant Feeding Practices Study II to the Year 6 Follow-Up, which include 8 MFS and CEB measures adapted from previous validated instruments. Bottle-feeding practices during the first 6 months estimated by using the Infant Feeding Practices Study II were bottle-feeding intensity (BFI), mother’s …

Rapid Detection Of Shigella And Enteroinvasive Escherichia Coli In Produce Enrichments By A Conventional Multiplex Pcr Assay, Rachel Binet, Deanne M. Deer, Samantha J. Uhlfelder

United States Public Health Resources

Faster detection of contaminated foods can prevent adulterated foods from being consumed and minimize the risk of an outbreak of foodborne illness. A sensitive molecular detection method is especially important for Shigella because ingestion of as few as 10 of these bacterial pathogens can cause disease. The objectives of this study were to compare the ability of four DNA extraction methods to detect Shigella in six types of produce, post-enrichment, and to evaluate a new and rapid conventional multiplex assay that targets the Shigella ipaH, virB and mxiC virulence genes. This assay can detect less than two Shigella cells …

Detection Of Five Shiga Toxin-Producing Escherichia Coli Genes With Multiplex Pcr, Insook Son, Rachel Binet, Anna Maounounen-Laasri, Andrew Lin, Thomas S. Hammack, Julie A. Kase

United States Public Health Resources

Escherichia coli serogroup O157 is the pathogen most commonly associated with foodborne disease outbreaks, but epidemiological studies suggest that non-O157 Shiga toxin-producing E. coli (STEC) is a major player as well. The ten most clinically relevant STECs belong to serogroups O26, O103, O111, O145, O157, O91, O113, O128, O45, and O121; but emerging strains, such as O104:H4 that was identified with the 2011 German outbreak, could become more prevalent in the future. A 75-min conventional multiplex PCR assay, IS-5P, targeting the four virulence factors stx1, stx2, eae, and ehxA plus the O157:H7- specific +93 uidA single nucleotide …
