Schistosomiasis is a neglected tropical disease that is responsible for almost 300,000 deaths annually. Mass drug administration (MDA) is used worldwide for the control of schistosomiasis, but chemotherapy fails to prevent reinfection with schistosomes, so MDA alone is not sufficient to eliminate the disease, and a prophylactic vaccine is required. Herein, we take advantage of recent advances in systems biology and longitudinal studies in schistosomiasis endemic areas in Brazil to pilot an immunomics approach to the discovery of schistosomiasis vaccine antigens. We selected mostly surface-derived proteins, produced them using an in vitro rapid translation system and then printed them to …

The incidence of Clostridium difficile infection (CDI) and associated mortality have increased rapidly worldwide in recent years. Therefore, it is critical to develop new therapies for CDI. In this study, we generated a novel, potently neutralizing, tetravalent, and bispecific antibody composed of 2 heavy-chain-only VH (VHH) binding domains against both TcdA and TcdB (designated “ABA”) that reverses fulminant CDI in mice infected with an epidemic 027 strain after a single injection of the antibody. We demonstrated that ABA bound to both toxins simultaneously and displayed a significantly enhanced neutralizing activity both in vitro and in vivo. Additionally, …

There is a pressing need for modeling of the symbiotic and at times dysbiotic relationship established between bacterial microbiomes and human mucosal surfaces. In particular clinical studies have indicated that the complex vaginal microbiome (VMB) contributes to the protection against sexually-transmitted pathogens including the life-threatening human immunodeficiency virus (HIV-1). The human microbiome project has substantially increased our understanding of the complex bacterial communities in the vagina however, as is the case for most microbiomes, very few of the community member species have been successfully cultivated in the laboratory limiting the types of studies that can be completed. A genetically controlled …

Governing The Wild: Databases, Algorithms, And Population Models As Biopolitics, Irus Braverman Mar 2014

Governing The Wild: Databases, Algorithms, And Population Models As Biopolitics, Irus Braverman

Journal Articles

This essay draws on interviews with conservation biologists to reflect on two interrelated aspects of the in situ – ex situ divide and its increasing integration: database systems and population management models. Specifically, I highlight those databases and software programs used by zoos in ex situ conservation settings, and the parallel, traditionally distinct, in situ databases and risk assessment models. I then explore the evolving technologies that integrate wild-captive databases and population models and, in particular, emerging metapopulation and meta-model approaches to small population management. My central argument is that, while still viewed by many as separate, the in situ …

Molecular Evidence For Vector Implication Of Onchocerca Lupi In Los Angeles County, Ca, Shanna June Bolcen

USF Tampa Graduate Theses and Dissertations

Onchocerca is a genus of roundworm most commonly associated with the human infection onchocerciasis, or river blindness. While typically a zoonotic infection of ungulate populations, canine cases (Onchocerca lupi) have been identified in the United States and Greece. In 2012, Los Angeles County, Veterinary Public Health Program identified 3 cases of Onchocerca spp. infections in domestic canines. Samples from the ensuing blackfly collections were sent to the Global Health Infectious Disease Research Unnasch Laboratory for parasite isolation and vector species identification. Species-specific primers were designed and optimized for O. lupi using a non- specific cytochrome oxidase (COI) gene target (689bp) …

Background: In environmental health interventions addressing water and indoor air quality, multiple determinants contribute to adoption. These may include technology selection, technology distribution and education methods, community engagement with behavior change, and duration and magnitude of implementer engagement. In Rwanda, while the country has the fastest annual reduction in child mortality in the world, the population is still exposed to a disease burden associated with environmental health challenges. Rwanda relies both on direct donor funding and coordination of programs managed by international non-profits and health sector businesses working on these challenges.

Methods and Findings: This paper describes the design, …

Between Amyloids And Aggregation Lies A Connection With Strength And Adhesion, Peter N. Lipke, Caleen Ramsook, Melissa C. Garcia-Sherman, Desmond N. Jackson, Cho X. J. Chan, Michael Bois, Stephen A. Klotz

Publications and Research

We tell of a journey that led to discovery of amyloids formed by yeast cell adhesins and their importance in biofilms and host immunity. We begin with the identification of the adhesin functional amyloid-forming sequences that mediate fiber formation in vitro. Atomic force microscopy and confocal microscopy show 2-dimensional amyloid “nanodomains” on the surface of cells that are activated for adhesion. These nanodomains are arrays of adhesin molecules that bind multivalent ligands with high avidity. Nanodomains form when adhesin molecules are stretched in the AFM or under laminar flow. Treatment with antiamyloid perturbants or mutation of the amyloid sequence prevents …

Statistical And Prognostic Modeling Of Clinical Outcomes With Complex Physiologic Data, Monica A. Puertas Mar 2014

Statistical And Prognostic Modeling Of Clinical Outcomes With Complex Physiologic Data, Monica A. Puertas

USF Tampa Graduate Theses and Dissertations

Laboratory tests are a primary resource for diagnosing patient diseases. However, physicians often make decisions based on a single laboratory result and have a limited perspective of the role of commonly-measured parameters in enhancing the diagnostic process. By providing a dynamic patient profile, the diagnosis could be more accurate and timely, allowing physicians to anticipate changes in the recovery trajectory and intervene more effectively.

The assessment and monitoring of the circulatory system is essential for patients in intensive care units (ICU). One component of this system is the platelet count, which is used in assessing blood clotting. However, platelet counts …

Lead (Pb) Contamination Of Water Drawn From Pitcher Pumps In Eastern Madagascar, David Bradlee Akers Mar 2014

Lead (Pb) Contamination Of Water Drawn From Pitcher Pumps In Eastern Madagascar, David Bradlee Akers

USF Tampa Graduate Theses and Dissertations

Access to safe water supply--a major determinant of public health--is less than 50% in Madagascar, and access to piped, treated water remains out of reach financially for many in the urban and peri-urban areas where available. The Self-supply option of the Pitcher Pump has been meeting the need for household water in coastal areas of Madagascar since the early 1960s and has proven a sustainable option for many. These pumps make use of leaded components in the construction, however, which may pose a health risk for heavy metal intoxication and therefore cause the water to be unsafe for drinking and …

Antiviral Resistance And Dynamic Treatment And Chemoprophylaxis Of Pandemic Influenza, Sandro Paz Mar 2014

Antiviral Resistance And Dynamic Treatment And Chemoprophylaxis Of Pandemic Influenza, Sandro Paz

USF Tampa Graduate Theses and Dissertations

Public health data show the tremendous economic and societal impact of pandemic influenza in the past. Currently, the welfare of society is threatened by the lack of planning to ensure an adequate response to a pandemic. This preparation is difficult because the characteristics of the virus that would cause the pandemic are unknown, but primarily because the response requires tools to support decision-making based on scientific methods. The response to the next pandemic influenza will likely include extensive use of antiviral drugs, which will create an unprecedented selective pressure for the emergence of antiviral resistant strains. Nevertheless, the literature has …

Immune recognition of foreign proteins by T cells hinges on the formation of a ternary complex sandwiching a constituent peptide of the protein between a major histocompatibility complex (MHC) molecule and a T cell receptor (TCR). Viruses have evolved means of "camouflaging" themselves, avoiding immune recognition by reducing the MHC and/or TCR binding of their constituent peptides. Computer-driven T cell epitope mapping tools have been used to evaluate the degree to which articular viruses have used this means of avoiding immune response, but most such analyses focus on MHC-facing ‘agretopes'. Here we set out a new means of evaluating the …

Cd46-Mediated Transduction Of A Species D Adenovirus Vaccine Improves Mucosal Vaccine Efficacy, Zenaido T. Camacho, Michael A. Barry, Eric A. Weaver

Nebraska Center for Virology: Faculty Publications

The high levels of preexisting immunity against Adenovirus type 5 (Ad5) have deemed Ad5 unusable for translation as a human vaccine vector. Low seroprevalent alternative viral vectors may be less impacted by preexisting immunity, but they may also have significantly different phenotypes from that of Ad5. In this study we compare species D Ads (26, 28, and 48) to the species C Ad5. In vitro transduction studies show striking differences between the species C and D viruses. Most notably, Ad26 transduced human dendritic cells much more effectively than Ad5. In vivo imaging studies showed strikingly different transgene expression profiles. The …

Achievement Of Transplantation Tolerance: Novel Approaches And Mechanistic Insights, Joseph Pidala Mar 2014

Achievement Of Transplantation Tolerance: Novel Approaches And Mechanistic Insights, Joseph Pidala

USF Tampa Graduate Theses and Dissertations

Current immune suppressive strategies fail to induce donor-recipient immune tolerance after allogeneic hematopoietic cell transplantation. Accordingly, patients suffer morbidity and mortality from graft vs. host disease (GVHD) and prolonged immune suppressive therapy. Biologic insight into transplantation tolerance is needed, and translation of such insight to novel clinical strategies may improve clinical outcomes. We report original investigation at seminal phases of this process including initial prophylactic immune suppression, onset of acute graft vs. host disease, and ultimate immune suppression discontinuation: In a controlled randomized clinical trial, we demonstrate that sirolimus-based immune suppression reduces risk for acute GVHD, ameliorates the severity of …

Combined Metal-Enhanced Fluorescence-Surface Acoustic Wave (Mef-Saw) Biosensor, Samuel Morrill Mar 2014

Combined Metal-Enhanced Fluorescence-Surface Acoustic Wave (Mef-Saw) Biosensor, Samuel Morrill

USF Tampa Graduate Theses and Dissertations

Immunofluorescence assays are capable of both detecting the amount of a protein and the location of the protein within a cell or tissue section. Unfortunately, the traditional technique is not capable of detecting concentrations on the nanoscale. Also, the technique suffers from non-specific attachment, which can cause false-positives, as well as photobleaching when detecting lower concentrations is attempted. There is also a time constraint problem since the technique can take from many hours to a few days in some cases.

In this work, metal-enhanced fluorescence (MEF) is used to lower the detection limit and reduce photobleaching. Unfortunately, MEF also increases …

A Population Of Langerin-Positive Dendritic Cells In Murine Peyer’S Patches Involved In Sampling B -Glucan Microparticles, Magdia De Jesus Mar 2014

A Population Of Langerin-Positive Dendritic Cells In Murine Peyer’S Patches Involved In Sampling B -Glucan Microparticles, Magdia De Jesus

Biomedical Sciences Faculty Scholarship

Glucan particles (GPs) are 2–4 mm hollow, porous shells composed of 1,3-b-D-glucan that have been effectively used for oral targeted–delivery of a wide range of payloads, including small molecules, siRNA, DNA, and protein antigens. While it has been demonstrated that the transepithelial transport of GPs is mediated by Peyer’s patch M cells, the fate of the GPs once within gut-associated lymphoid tissue (GALT) is not known. Here we report that fluorescently labeled GPs administered to mice by gavage accumulate in CD11c+ DCs situated in Peyer’s patch sub-epithelial dome (SED) regions. GPs appeared in DCs within minutes after gavage and remained …

Observational studies have suggested that with time, some diseases result in a characteristic odor emanating from different sources on the body of a sick individual. Evolutionarily, however, it would be more advantageous if the innate immune response were detectable by healthy individuals as a first line of defense against infection by various pathogens, to optimize avoidance of contagion. We activated the innate immune system in healthy individuals by injecting them with endotoxin (lipopolysaccharide). Within just a few hours, endotoxin-exposed individuals had a more aversive body odor relative to when they were exposed to a placebo. Moreover, this effect was statistically …

Chagas Disease In The 21st Century: A Public Health Success Or An Emerging Threat?, Kevin M. Bonney

Publications and Research

Chagas disease, caused by the protozoan parasite Trypanosoma cruzi, is a major public health burden in Latin America and a potentially serious emerging threat to a number of countries throughout the world. Although public health programs have significantly reduced the prevalence of Chagas disease in Latin America in recent decades, the number of infections in the United States and non-endemic countries in Europe and the Western Pacific Region continues to rise. Moreover, there is still no vaccine or highly effective cure available for the approximately 10 million people currently infected with T. cruzi, a third of which will develop potentially …
