Expression of the tumor suppressor protein BRCA1 is frequently lost in breast cancer patients, and the loss of its expression is associated with disruption of various critical functions in cells and cancer development. In the present study, we demonstrate through microarray analysis that cells with tumor suppressor candidate 4 (NPRL2/TUSC4) knockdown show critical changes to cell cycle, cell death pathways and a global impact on cancer development. More importantly, we observed a clear cluster pattern of NPRL2/TUSC4-knockdown gene profiles with established homologous recombination (HR) repair defect signature. Visual working memory (VWM) is the temporary retention of visual information and a key component of cognitive processing. The classical paradigm for studying VWM and its encoding limitations has been change detection. Early work focused on how many items could be stored in VWM, leading to the popular theory that humans could remember no more than 4±1 items. More recently, proposals have suggested that VWM is a noisy, continuous resource distributed across virtually all items in the visual field, resulting in diminished memory quality rather than limited quantity. The Ca2+-permeable TRPV4 channel is predominantly expressed in the distal nephron (DN) and its activity is essential for [Ca2+]i elevations in response to increased tubular flow. Here, I probed the physiological mechanisms controlling TRPV4 function and expression in the DN. I found that renal TRPV4 expression and mRNA levels were significantly increased by high K+ diet (5%) and decreased by dietary K+ restriction (0.003%). In contrast, variations in Na+ regimen had no apparent effect on TRPV4 expression and mRNA levels. Regulation of TRPV4 protein expression by K+ diet was independent of …

HER2-positive breast cancer, which is characterized by the over-expression of the HER2 onco-protein, accounts for approximately 20% of all breast cancer cases. Trastuzumab (Herceptin), the first targeted therapy approved for HER2-positive disease, potently prevents the activation of signaling pathways downstream of HER2 and significantly improves patients’ outcomes. However, resistance to trastuzumab is inevitable; such resistance can occur through reduced expression of PTEN protein.
Jab1 is over-expressed in 50% of primary cancers and 90% of metastatic tumors. Our lab previously showed that depletion of Jab1 in combination with trastuzumab treatment up-regulated PTEN in mouse xenografts refractory to trastuzumab. PTEN was not …

Src family kinases (SFKs) are commonly over-expressed and/or activated in glioblastoma multiforme (GBM), where they serve as key mediators of GBM cell proliferation, survival, invasion and angiogenesis. Mechanisms of allosteric SFK activation are well described; however, the SFK Fyn is commonly up-regulated at the mRNA level in multiple human cancers, including GBM, where the mode of increased expression is poorly understood. Since activating mutations in the epidermal growth factor receptor (EGFR) are commonly occurring in GBM, we examined whether EGFR could induce Fyn expression. Ovarian cancer (OC) is a highly metastatic disease, but no effective strategies to this process currently are known. Here, an integrated computational analysis of The Cancer Genome Atlas ovarian cancer dataset coupled with experimental validation identified a novel zinc finger transcriptional factor 304 (ZNF304) as one of the key factors for ovarian cancer metastasis. High tumoral ZNF304 expression was associated with poor overall survival in ovarian cancer patients. Through reverse phase protein array analysis, we demonstrated that ZNF304 promotes multiple proto-oncogenic pathways important for cell survival, migration, and invasion. ZNF304 transcriptionally regulates β1 integrin, which subsequently regulates Src/focal adhesion kinase …

This thesis considers the problem of detecting the eating activities (e.g. meals, snacks) of people by tracking their wrist motion. Our goal is to automatically de- tect the start and end times of eating activities during free-living. It builds upon previous work done by our research group [6] [7] [8] [21]. The detection is done by segmenting data into segments then classifying the segments as EA (eating activity) and NonEA (non-eating activity) using a Bayesian classier. Prospective memory failures (or failures to remember a future intention) can result in a wide range of negative consequences. The use of reminders has been shown to improve the rate of PM successes. The aim of the current study was to examine the effectiveness of reminders based on their type (text or picture) and their timing. We hypothesized that successful PM performance would be successfully maintained over longer anticipatory intervals when paired with picture reminders rather than with simple text reminders because of the inherent distinctiveness of pictures. Human noroviruses (NoV) are a leading cause of acute gastroenteritis, causing nearly 58% of foodborne disease in the USA. The aim of our study was to determine the prevalence of NoV on select bathroom surfaces in food establishments in South Carolina (SC). All SC counties (N=46) were sorted by population density into high-, medium-, and low-density categories. Three counties were randomly selected from each population category. Lists of all foodservice establishments in each selected county (N=9) were obtained from the Department of Health and Environmental Control and a total of 120 foodservice establishments were randomly selected. This thesis examines the possibility of grouping charitable donors by income level to develop a set of models that can more accurately predict charitable donations. Previous work is inconsistent in predicting charitable donations. This work helps to determine if these inconsistencies are a result of methodological differences between researchers, or if group membership is an important factor in predicting charitable donations as suggested by some researchers. This research only found four variables that were common to all three income groups, frequency of church attendance, family income, age, and years of education. This study seeks to quantify runoff volume generation and peak flow rates from the urban Sand River Headwaters to determine the most effective placement of additional green infrastructure in Aiken, SC. ArcMap 10.1, HEC-GeoHMS, and HEC-HMS were used to delineate a total outlet watershed along with subwatershed(s) for urban stormwater infrastructure system by 'burning' the stormwater system at an artificial elevation below the existing topologically-based Digital Elevation Model (DEM). Structural Health Monitoring (SHM) is a field of research that seeks to identify the onset of damage in infrastructure systems such that catastrophic failures can be averted. Acoustic Wavenumber Spectroscopy (AWS) is an emerging SHM technique that can identify and locate change of thickness resulting from damage in thin plates through the estimation of the characteristic wavenumber. AWS measures the ultrasonically induced vibrations of thin plates by laser scanners and monitors the propagation of Rayleigh-Lamb waves through the structure. Designs must ensure high biocompatibility, drug compatibility, accuracy and small power consumption. The focus of this thesis was to fabricate a prototype magnetic nanoparticle membrane for eventual incorporation into a biomedical pump and then determine the relationship between this membrane deflection and applied pneumatic or magnetic force. Vibrio cholerae, a Gram-negative bacterium found in natural aquatic environments, is the causative agent of cholera, a severe diarrheal disease most commonly spread through drinking water. An essential component to this pathogen's success and persistence in the environment is its ability to attach to both biotic and abiotic surfaces via biofilm formation. Biofilms not only aid in surface attachment, but also provide a barrier that protects and enhances survival. Water-soluble extracts from the North American cranberry (Vaccinium macrocarpon) were found to dramatically inhibit V. cholerae biofilm formation at a low concentration of 2mg/ml. Chlorinated ethenes are known or suspected carcinogens. They are also among the most frequently detected organic groundwater contaminants because of wide use and poor disposal practices. Within anaerobic aquifer systems, Fe(III) reduction is a prevalent terminal electron accepting process and has often been reported as a competitive electron acceptor with respect to trichloroethene (TCE), cis-dichloroethene (cis-DCE), and vinyl chloride (VC) reduction. Often, practitioners add excessive amounts of electron donor to compensate for this competition in an effort to avoid incomplete dechlorination or "cis-DCE and VC stall." With the increasing attention and support behind plug in hybrid electric vehicles, research must be conducted to examine the impacts of vehicles on electric distribution and transmission systems. This research aims first to model the behavior of vehicle battery chargers during system disturbances and mitigate any impacts. Myocardial infarction (MI) affects nearly 600,000 individuals each year, and the resulting damage initiates a pathophysiological progression towards congestive heart failure (CHF). A shortage of donor organs precludes heart transplantation as a practical solution, and neither surgical intervention nor stem cell therapy have yielded consistent and sufficiently positive results in clinical investigation. To prevent CHF, prospective therapies must target the cause of the maladaptive cardiac remodeling which precedes it--the nonfunctional, dyskinetic infarct scar--and aim to replace it with functional cardiac muscle. The high demand for more efficient purification processes with increased automation and throughput pushes the development of more advanced preparative, pilot, and process scale HPLC instrumentation that is capable of achieving higher purities in a shorter amount of time than are currently achieved using one dimensional separations. A preparative scale 2D HPLC system was designed and reduced to practice in order to demonstrate the capacity for scalability of on-line comprehensive 2D HPLC separations of basic compounds from a challenging natural product extract of Oplopanax horridus. Each of the three chapters of this dissertation makes a unique contribution to the fields of public finance or corporate finance. In chapter one I show that tax-price is increasing in workplace risk due to a positive wage-risk response that is observed in the labor supply price for hazardous industries. This result implies that, holding human capital constant, workers in more dangerous industries will demand a relatively smaller public sector. Currently the backbone of the world's energy supply is composed of fossil fuels. However, the combustion of fossil fuels results in the production of enormous quantities of particulate pollutants. The smog resulting from these particulate pollutants causes significant health problem for city dwellers. Wet scrubbers, which use a water spray to scavenge airborne particles, is one of the most widely used devices to control particulate pollutants. Typical wet scrubbers can scavenge particles with diameters bigger than 10 um, but it is inefficient in scavenging particles with diameters on the order of 1 um. Research has linked neighborhoods of concentrated poverty with high crime, low employment, poor health, and low educational achievement. Because of these linkages, federal housing policy over the past few decades has often tried to 'deconcentrate' or disperse the poor from these neighborhoods into more affluent neighborhoods with the hope that better institutions and better neighbors will motivate these families to improve their lives.
