All-Optical Photoacoustic Detection Of Absorbers In Tissue Phantoms, Jami Johnson, Michelle Sabick, Kasper Vanwijk
All-Optical Photoacoustic Detection Of Absorbers In Tissue Phantoms, Jami Johnson, Michelle Sabick, Kasper Vanwijk
Mechanical and Biomedical Engineering Faculty Publications and Presentations
Visualizing and characterizing vascular structures is important for many areas of health care, from accessing difficult veins and arteries for laboratory testing, to diagnosis and treatment of cardiovascular disease. Photoacoustic (PA) imaging, one of the fastest growing fields of biomedical imaging, is well suited for this task. PA imaging is based on the photoacoustic effect, which starts with a pulsed laser source incident on biological tissue. If the wavelength of the source matches an absorption wavelength of a chromophore within the tissue, a portion of the pulse energy is absorbed by the chromophore and converted into heat. A subsequent increase …

Status Of Nearshore Finfish Stocks In South-Western Western Australia: Part 1: Australian Herring, Kimberley A. Smith, Joshua Brown, Paul Lewis, Chris Dowling, A Howard, Rod J. Lenanton, Brett Molony
Fisheries Research Reports
The status of the popular nearshore finfish resource in the West Coast Bioregion (WCB) of Western Australia (WA) was largely unknown prior to this study. Previously, declining catches of several nearshore species had highlighted the risk to their sustainability and the need for greater certainty about their status. Recently, the risk further increased due to management changes in the WCB aimed at reducing the catch of demersal scalefish, which are likely to result in a shift in targeting towards nearshore species. This increase in fishing pressure on nearshore species will be on top of any increase due to the continuing …

In angiosperms, the endosperm provides nutrients for embryogenesis and seed germination and is the primary tissue where gene imprinting occurs. To identify the imprintome of early developing maize (Zea mays) endosperm, we performed highthroughput transcriptome sequencing of whole kernels at 0, 3, and 5 d after pollination (DAP) and endosperms at 7, 10, and 15 DAP, using B73 byMo17reciprocal crosses.Weobserved gradually increased expression of paternal transcripts in 3- and 5-DAP kernels. In 7-DAP endosperm, the majority of the genes tested reached a 2:1 maternal versus paternal ratio, suggesting that paternal genes are nearly fully activated by 7 DAP. …

Fisheries Research Report No. 249 - An Integrated System To Survey Boat-Based Recreational Fishing In Western Australia 2011/12, Karina L. Ryan, Brent S. Wise, Norman G. Hall, K H. Pollock, Elena H. Sulin, Daniel J. Gaughan
Fisheries Research Reports
The Department of Fisheries developed an integrated system involving several survey methods to provide a more robust approach for obtaining annual estimates of recreational catch by boat based fishers at both state-wide and bioregional levels. These surveys, which used the recently implemented Recreational Fishing from Boat Licence (RFBL) as the basis for sampling, were the most comprehensive ever conducted in Western Australia. They not only provided estimates of catch and effort but provided the information for the validation of these estimates by enabling comparisons across the various methods.

Walden Faculty and Staff Publications
Autism is an increasingly prevalent developmental neurological condition that manifests in pervasive impairments in social interaction, communication, and stereotypic behavior and interests. Early identification leads to positive long-term outcomes. At present, all standardized rating instruments are written in English; therefore, the purpose of the present study was to adapt the Gilliam Autism Rating Scale-2 from English into Spanish using the state-of-the-art procedures described in the literature. The English instrument was translated into Spanish, back-translated into English, and then revised using an iterative process. The adapted instrument's psychometric qualities were substantiated. An alpha coefficient of .96 verified the adapted instrument's internal …

Bacterial Endophytes Enhance Competition By Invasive Plants, Marnie E. Rout, Thomas H. Chrzanowski, Tara K. Westlie, Thomas H. Deluca, Ragan M. Callaway, William E. Holben
Biological Sciences Faculty Publications
- Premise of the study: Invasive plants can alter soil microbial communities and profoundly alter ecosystem processes. In the invasive grass Sorghum halepense, these disruptions are consequences of rhizome-associated bacterial endophytes. We describe the effects of N2-fixing bacterial strains from S. halepense (Rout and Chrzanowski, 2009) on plant growth and show that bacteria interact with the plant to alter soil nutrient cycles, enabling persistence of the invasive.
- Methods: We assessed fluxes in soil nutrients for ~4 yr across a site invaded by S. halepense. We assayed the N2-fixing bacteria in vitro for phosphate solubilization, …

Status Of Nearshore Finfish Stocks In South-Western Western Australia Part 3: Whiting (Sillaginidae): Nrm Project 09003, Joshua Brown, Christopher Dowling, Alexander Hesp, Kimberley A. Smith, Brett W. Molony
Fisheries Research Reports
The whiting family (Sillaginidae) is common in nearshore waters of south-western Western Australia (WA) where it is highly valued by recreational and commercial fishers. The importance of these species to recreational fishers is reflected in this category of fishes being ranked as the most or 2nd most retained finfish species group by boat- and shore-based fishers in several recreational fishing surveys in WA. Whiting are commercially important in a number of fisheries, with total state catches averaging over 200 tonnes per annum since 1980. The composition of the whiting catch, however, is largely unknown, with a number of whiting species …

Department of Cancer Biology Faculty Papers
The CCND1 gene, which is frequently overexpressed in cancers, encodes the regulatory subunit of a holoenzyme that phosphorylates the retinoblastoma protein. Although it is known that cyclin D1 regulates estrogen receptor (ER)α transactivation using heterologous reporter systems, the in vivo biological significance of cyclin D1 to estrogen-dependent signaling, and the molecular mechanisms by which cyclin D1 is involved, are yet to be elucidated. Herein, genome-wide expression profiling conducted of 17β-estradiol-treated castrated virgin mice deleted of the Ccnd1 gene demonstrated that cyclin D1 determines estrogen-dependent gene expression for 88% of estrogen-responsive genes in vivo. In addition, expression profiling of 17β-estradiol-stimulated cyclin …

Draft Genome Sequence Of A Stable Mucoid Strain Of Pseudomonas Aeruginosa Pao581 With A Muca25 Mutation, Yeshi Yin, T. Ryan Withers, John R. W. Govan, Shannon L. Johnson, Hongwei D. Yu
Biochemistry and Microbiology
A mutation in the mucA gene, which encodes a negative regulator of alginate production in Pseudomonas aeruginosa, is the main mechanism underlying the conversion to mucoidy in clinical isolates from patients with cystic fibrosis (CF). Here, we announce the draft genome sequence of the stable alginate-overproducing mucoid strain P. aeruginosa PAO581 with a mucA25 mutation, a derivative from the nonmucoid strains P. aeruginosa PAO381 and PAO1.

Stress-inducible phosphoprotein 1 (STI1) is part of the chaperone machinery, but it also functions as an extracellular ligand for the prion protein. However, the physiological relevance of these STI1 activities in vivo is unknown. Here, we show that in the absence of embryonic STI1, several Hsp90 client proteins are decreased by 50%, although Hsp90 levels are unaffected. Mutant STI1 mice showed increased caspase-3 activation and 50% impairment in cellular proliferation. Moreover, placental disruption and lack of cellular viability were linked to embryonic death by E10.5 in STI1-mutant mice. Rescue of embryonic lethality in these mutants, by transgenic expression of the …

Our understanding of the sources of Mycobacterium avium infection is partially based on genotypic matching of pathogen isolates from cases and environmental sources. These approaches assume that genotypic identity is rare in isolates from unlinked cases or sources. To test this assumption, a high-resolution PCR-based genotyping approach, large-sequence polymorphism (LSP)-mycobacterial interspersed repetitive unit–variable-number tandem repeat (MIRU-VNTR), was selected and used to analyze clinical and environmental isolates of M. avium from geographically diverse sources. Among 127 clinical isolates from seven locations in North America, South America, and Europe, 42 genotypes were observed. Among 12 of these genotypes, matches were seen in …

The first and most important step of molecular techniques is to isolate the high quality and standard quantity of DNA. The DNA extracted using the recommended method could successfully amplify the regions of interest and demonstrated reliable results can be applied in other molecular assays. Moreover, the designed primers of ITS1-UM2 and ITS4-UM2 were perfectly matched with the species of Basidiomycetes, can be used in phylogenetic studies of other mushrooms. We here evaluated the quality and quantity of DNA using a spectrophotometer, showed reliable OD260/280, and concentration. The protocol is efficient, rapid, low-cost, and simple, needs low amount of sample, …

Stress And Alcohol Consumption In College Students: Do Tangible And Belonging Support Matter?, Marion Danh, Amanda Brouwer
Student Research and Creative Projects 2013-2014
College students to cope with stress often use alcohol. Social support has been shown to buffer the negative outcomes of stress, but specific forms of support such as tangible and belonging support have been understudied in the buffering hypothesis. Therefore, I examined how belonging and tangible social support affect alcohol use depending on one’s stress level. Participants (N=212, Mage= 21.51, SD=2.96) were emailed a survey and responded to demographic, stress, social support, and alcohol consumption questions. The moderation effect was tested using multiple regression. There was a significant interaction between tangible support and perceived stress on hours spent drinking and …

Babesia microti is the causative agent for babesiosis, a blood-based parasitic disease that causes malaria-like symptoms in humans. It is passed through Ixodes scapularis (deer ticks) and is rising in incidence in the upper Midwest. Ticks that carry Borrelia, the causative agent for Lyme disease, may also carry Babesia microti. There are two major goals in this project: determine the prevalence of Babesia microti in ticks in the Winona area and determine whether those ticks that carry Borrelia are more likely to carry Babesia microti. The first part of the project requires development of a PCR assay that works well …
