Medicinal Chemistry Publications

In our effort to develop selective sphingosine kinase-2 (SphK2) inhibitors as pharmacological tools, a thiazolidine-2,4-dione analogue, 3-(2-amino-ethyl)-5-[3-(4-butoxyl-phenyl)-propylidene]-thiazolidine-2,4-dione(K145), was synthesized and biologically characterized. Biochemical assay results indicate that K145 is a selective SphK2 inhibitor. Molecular modeling studies also support this notion. In vitro studies using human leukemia U937 cells demonstrated that K145 accumulates in U937 cells, suppresses the S1P level, and inhibits SphK2. K145 also exhibited inhibitory effects on the growth of U937 cells as well as apoptotic effects in U937 cells, and that these effects may be through the inhibition of down-stream ERK and Akt signaling pathways. K145 also significantly …

A Science Of Context: The Qualitative Approach As Fundamental To Strategic Thought, Matthew J. Schmidt

Political Science Faculty Publications

(from book introduction, pp. 6-7) "Schmidt describes the differences between quantitative and qualitative approaches, research questions, and thought processes. Schmidt argues that quantitative methods are appropriate for tactical engagements and can inform strategic thinking, but a completely different thought process (i.e., qualitative) is necessary for success when asking questions with strategic implications. Schmidt also discusses how time delays can affect the perceived impact of qualitative modes of thinking."

Predicting Dyspnea Inducers By Molecular Topology, María Gálvez-Llompart,, Jorge Gálvez, Ramón García-Domenech, Lemont B. Kier

Study of Biological Complexity Publications

QSAR based on molecular topology (MT) is an excellent methodology used in predicting physicochemical and biological properties of compounds. This approach is applied here for the development of a mathematical model capable to recognize drugs showing dyspnea as a side effect. Using linear discriminant analysis, it was found a four-variable regression equations enabling a predictive rate of about 81% and 73% in the training and test sets of compounds, respectively. These results demonstrate that QSAR-MT is an efficient tool to predict the appearance of dyspnea associated with drug consumption.

Bioluminescence imaging (BLI) is a relatively new noninvasive technology used for quantitative assessment of tumor growth and therapeutic effect in living animal models. BLI involves the generation of light by luciferase-expressing cells following administration of the substrate luciferin in the presence of oxygen and ATP. In the present study, the effects of hypoxia, hypoperfusion, and pH on BLI signal (BLS) intensity were evaluated in vitro using cultured cells and in vivo using a xenograft model in nude mice. The intensity of the BLS was significantly reduced in the presence of acute and chronic hypoxia. Changes in cell density, viability, and …

Spectroscopic Study Of The Use Of Lanthanide Metalloporphyrins As Sensors For Benzene And Acetonitrile Detection In Aqueous Studies, Jr Carlos Crawford

Theses

This work entails the research on lanthanide metalloporphyrins for their potential use as chemical sensors for benzene and acetonitrile. This research is of importance due to the health implications that benzene and acetonitrile cause; benzene is a known carcinogen and acetonitrile is a known lung irritant. The use of UV-Vis spectroscopy, Fluorescence spectroscopy, Gaussian DFT, and X-ray diffraction crystallography were used in the characterization and analysis of the lanthanide porphyrin complexes. Europium, terbium, dysprosium, cerium, and gadolinium were the lanthanides used in conjunction with 5,10,15,20-tetraphenylporphyrin, TPP and 5,10,15,20 tetrakissulfonato porphyrin, TBSP. Based on the luminescence spectroscopy and UV-Vis spectroscopy data, …

From Single-Snp To Wide-Locus: Genome-Wide Association Studies Identifying Functionally Related Genes And Intragenic Regions In Small Sample Studies, Knut M. Wittkowski, Vikas Sonakya, Tingting Song, Martin P. Seybold, Mehdi Keddache, Martina Durner

Krueger Laboratory

Background: Genome Wide Association Studies (GWAS) have had limited success when applied to complex diseases. Analyzing SNPs individually requires several large studies to integrate the often divergent results. In the presence of epistasis, multivariate approaches based on the linear model (including stepwise logistic regression) often have low sensitivity and generate an abundance of artifacts. Methods: Recent advances in distributed and parallel processing spurred methodological advances in non-parametric statistics. U-statistics for multivariate data (μStat) are not confounded by unrealistic assumptions (linearity, independence). Results: By incorporating knowledge about relationships between SNPs, μGWAS (GWAS based on μStat) can identify clusters of genes around …

Articles

Background: The protein encoded by GmRLK18-1 (Glyma_18_02680 on chromosome 18) was a receptor like kinase (RLK) encoded within the soybean (Glycine max L. Merr.) Rhg1/Rfs2 locus. The locus underlies resistance to the soybean cyst nematode (SCN) Heterodera glycines (I.) and causal agent of sudden death syndrome (SDS) Fusarium virguliforme (Aoki). Previously the leucine rich repeat (LRR) domain was expressed in Escherichia coli.

Results: The aims here were to evaluate the LRRs ability to; homo-dimerize; bind larger proteins; and bind to small peptides. Western analysis suggested homo-dimers could form after protein extraction from roots. The purified LRR domain, from …

Articles

This study reports a high density genetic linkage map based on the ‘Maryland 96-5722’ by ‘Spencer’ recombinant inbred line (RIL) population of soybean [Glycine max (L.) Merr.] and constructed exclusively with single nucleotide polymorphism (SNP) markers. The Illumina Infinium SoySNP6K BeadChip genotyping array produced 5,376 SNPs in the mapping population, with a 96.75% success rate. Significant level of goodness-of-fit for each locus was tested based on the observed vs. expected ratio (1:1). Out of 5,376 markers, 1,465 SNPs fit the 1:1 segregation rate having ≤20% missing data plus heterozygosity among the RILs. Among this 1,456 just 657 were polymorphic …

Identifying Pathogenic Salmonella Serotypes Isolated From South Central Va Waterways Via Sequential Pcr Analysis, Timothy M. Smith, Jr. Jan 2013

Identifying Pathogenic Salmonella Serotypes Isolated From South Central Va Waterways Via Sequential Pcr Analysis, Timothy M. Smith, Jr.

Theses & Honors Papers

Researchers often debate whether all serovars of Salmonella enterica are pathogenic to humans. Broiler houses containing chickens are a major source of Salmonella and serve as potential reservoirs. Therefore, farmers must take substantial measures in order to avoid contamination with their products. The primary objective of the study was to determine whether the four most clinically relevant Salmonella spp. serovars from poultry houses could be isolated from the environmental waterways in Prince Edward and Cumberland Counties of Virginia. The study adapted a serotyping method designed for typing Salmonella to gain a perspective of possible serovars that may be found in …

On The Sleeve Of The Visual: Race As Face Value, Alessandra Raengo Jan 2013

On The Sleeve Of The Visual: Race As Face Value, Alessandra Raengo

Dartmouth College Press

In this work of critical theory, Black studies, and visual culture studies, the author reads race as a theory of the image. By placing emphasis on the surface of the visual as the repository of its meaning, race presents the most enduring ontological approach to what images are, how they feel, and what they mean. Having established her theoretical concerns, the author's eclectic readings of various artifacts of visual culture, fine arts, cinema, and rhetorical tropes provoke and destabilize readers' visual comfort zone, forcing them to recognize the unstated racial aspects of viewing and the foundational role of race in …

Adoption Awareness In The Elementary Classroom, Claire Laberge Jan 2013

Adoption Awareness In The Elementary Classroom, Claire Laberge

Capstone Projects and Master's Theses

Adoption is a complex and delicate issue, and too often, outdated and biased attitudes toward adoption influence people’s perceptions of adoption and adoptees. Adopted children are particularly vulnerable to negativity toward the adoptive family structure, and adoptees have special emotional needs that must be addressed in the elementary school classroom. Research demonstrates a variety of ways teachers can bring awareness to adoption issues, de-stigmatize adoption in the classroom, and in turn, create a safe and nurturing classroom environment. The researcher designed an anonymous survey to distribute to a sampling of elementary school teachers; the survey results generally supported the researcher’s …

The Perfect Priest: An Examination Of Leviticus 21:17-23, Jared Wilson Jan 2013

The Perfect Priest: An Examination Of Leviticus 21:17-23, Jared Wilson

Seminary Masters Theses

Leviticus 21:17-23 is a text that discriminates against priests who have one of the twelve blemishes listed, approaching the text with the intent to redeem the text for modern application. An exploration in the role of priests in the ancient Near East will show how the priests were human representations of the deity and many cultures and religions had physical restrictions for the priests similar to Lev 21:17-23. Examining the language of Lev 21:17-23 will show that the restrictions were not merely symbolic but intended to stop blemished priests from approaching the altar. However, they were not statements about the …

Parallel Changes In Genital Morphology Delineate Cryptic Diversification Of Planktonic Nudibranchs, Alvin Alejandrino, Celia K. C. Churchill, Ángel Valdés, Diarmaid Ó. Foighil Jan 2013

Parallel Changes In Genital Morphology Delineate Cryptic Diversification Of Planktonic Nudibranchs, Alvin Alejandrino, Celia K. C. Churchill, Ángel Valdés, Diarmaid Ó. Foighil

Biology

The relative roles of geographical and non-geographical barriers in the genesis of genetic isolation are highly debated in evolutionary biology, yet knowing how speciation occurs is essential to our understanding of biodiversity. In the open ocean, differentiating between the two is particularly difficult, because of the high levels of gene flow found in pelagic communities. Here, we use molecular phylogenetics to test the hypothesis that geography is the primary isolating mechanism in a clade of pelagic nudibranchs, Glaucinae. Our results contradict allopatric expectations: the cosmopolitan Glaucus atlanticus is panmictic, whereas the Indo-Pacific Glaucus marginatus contains two pairs of cryptic species …

Cell-Autonomous Sex Determination Outside Of The Gonad, Jenny C. Link, Arthur P. Arnold, Xuqi Chen, Yuichiro Itoh, Karen Reue Jan 2013

Cell-Autonomous Sex Determination Outside Of The Gonad, Jenny C. Link, Arthur P. Arnold, Xuqi Chen, Yuichiro Itoh, Karen Reue

Biology

The classic model of sex determination in mammals states that the sex of the individual is determined by the type of gonad that develops, which in turn determines the gonadal hormonal milieu that creates sex differences outside of the gonads. However, XX and XY cells are intrinsically different because of the cell-autonomous sex-biasing action of X and Y genes. Results: Recent studies of mice, in which sex chromosome complement is independent of gonadal sex, reveal that sex chromosome complement has strong effects contributing to sex differences in phenotypes such as metabolism. Adult mice with two X chromosomes (relative to mice …

Highly Efficient Quenching Of Nanoparticles For The Detection Of Electron-Deficient Nitroaromatics, Patrick Marks, Sage Cohen, Mindy Levine

Chemistry Faculty Publications

Reported herein is the highly efficient quenching of fluorescent organic nanoparticles by 2,4-dinitrotoluene (DNT) and 2,4,6-trinitrotoluene (TNT). These fluorescent nanoparticles are formed from the hydrophobic collapse of fluorescent polymer chains, and display quenching efficiencies that are in line with the highest reported literature values. Moreover, the fluorescent quenching occurs only for the fluorescent nanoparticles, and not for the precursor polymer solutions, which display marked insensitivity to the presence of nitroaromatics. This aggregation-dependent fluorescent quenching has numerous applications for the detection of small-molecule electron-deficient analytes.
