Novel Insights Into The Function And Regulation Of Group X Secretory Phospholipase A2, Joseph D. Layne Jr

Group X secretory phospholipase A2 (GX sPLA2) hydrolyzes membrane phospholipids producing free fatty acids and lysophospholipids. Previous studies from our lab suggest that mice with targeted deletion of GX sPLA2 (GX KO) have increased age-related weight gain due to an increase in overall adiposity. Paradoxically, this increased adiposity is associated with improved age-related glucose intolerance. GX KO mice also demonstrate a reduced inflammatory response to lipopolysaccharide injection. In vitro studies indicate this phenotype may be attributable to blunted macrophage mediated inflammatory responses. © The Author 2013. Deliberately feeding wildlife is an increasingly popular tourism-related activity despite a limited understanding of long-term impacts on the species being fed. As a result, tourist behaviours that may have adverse impacts on imperiled species have often been encouraged without the necessary evaluation or oversight. Here, we report the responses of Northern Bahamian Rock Iguanas (Cyclura cychlura) to human-visitation pressure and associated food provisioning. We compared a variety of blood chemistry parameters of iguanas subjected to supplemental feeding at popular tourist destinations with iguanas occurring on islands where supplemental feeding does not take place. Interpreting variants, especially noncoding ones, in the increasing number of personal genomes is challenging. We used patterns of polymorphisms in functionally annotated regions in 1092 humans to identify deleterious variants; then we experimentally validated candidates. We analyzed both coding and noncoding regions, with the former corroborating the latter. We found regions particularly sensitive to mutations ("ultrasensitive") and variants that are disruptive because of mechanistic effects on transcription-factor binding (that is, " motif-breakers"). We also found variants in regions with higher network centrality tend to be deleterious. Insertions and deletions followed a similar pattern to single-nucleotide variants, with some notable exceptions …

Background:Leishmaniasis remains a global health problem because of the substantial holes that remain in our understanding of sand fly ecology and the failure of traditional vector control methods. The specific larval food source is unknown for all but a few sand fly species, and this is particularly true for the vectors of Leishmania parasites. We provide methods and materials that could be used to understand, and ultimately break, the transmission cycle of zoonotic cutaneous leishmaniasis.Methods and Findings:We demonstrated in laboratory studies that analysis of the stable carbon and nitrogen isotopes found naturally in plant and animal tissues was highly effective …

Prenatal Choline Supplementation And Mk-801 Toxicity: Protecting Memory And Preventing Neurodegeneration, Chelsea Nickerson

Choline is essential to the development and function of the central nervous system. Supplemental choline is neuroprotective against a variety of insults, including neurotoxins like dizocilpine (MK-801). MK-801 is an NMDA receptor antagonist that is frequently used in rodent models of psychological disorders. At low doses, it causes cognitive impairments, and at higher doses it induces motor deficits, anhedonia, and neuronal degeneration. The primary goals of the present study were to investigate whether prenatal choline supplementation protects against the cognitive impairments, motor deficits, and neuropathologies that are precipitated by MK-801 administration in adulthood. Amphibian populations have been declining for several decades, in part due to the emerging fungal skin disease, chytridiomycosis, caused by the chytrid fungus Batrachochytrium dendrobatidis (Bd). Chytridiomycosis has caused extinctions and extirpations in many parts of the world, but its influence varies across species, populations, and individuals. The Green Frog (Lithobates clamitans), which inhabits the eastern half of the United States, seems capable of sustaining Bd infections without experiencing die-offs. It is possible that the Green Frog's epibiotic bacteria are contributing to its defense against Bd as the epibiotic bacteria of several amphibian species are known to play …

Deficiency of micronutrients, particularly of zinc, boron, and iron, in calcareous soils is of great concern. Results indicate a reduction in yield of seed-cotton due to an imbalance of nutrients, particularly micronutrients, on calcareous soils in the last decade. About 90% of Pakistani soils are deficient in micronutrients, particularly zinc, boron, and iron. This paper highlights efforts to overcome these deficiencies in cotton through foliar feeding of micronutrients. Three cultivars of Alternanthera bettzickiana (Regel) G.Nicholson have been evaluated for their tolerance and adaptability potential to salt stress. During the experiment, 5 salt regimes were maintained: 18 (control), 50, 100, 150, and 200 mM NaCl. Salinity adversely affected all growth shoot length and leaves per plant. Root fresh and dry weight decreased with an increase in salinity levels in all 3 cultivars. A. bettzickiana 'Green' was the most tolerant among the cultivars under study, with relatively lower ion leakage through roots, larger vascular region area, and wide metaxylem vessel in roots and stems recorded. A pulsed electric field (PEF) process was applied to unclarified apple juices at 30 and 40 kV cm^{-1} for 50, 100, 150, and 200 pulses after preheating to 40 °C. Afterwards, polyphenol oxidase (PPO) enzyme levels, loss of total phenolic (TP) compounds, and alterations of the physiochemical parameters of the apple juice were determined. Total yeast/mold and mesophilic bacteria loads were periodically evaluated during storage at 25 °C for a 3-month period and the cloud quality of the apple juice was analyzed after storage. The main objective of this study was to develop a spatial model for land suitability assessment for rice (Oryza sativa L.) cultivation using geographic information system (GIS) techniques. The land mapping units, resulting from the overlay of the selected theme layers, have unique land quality information, on which land suitability is based. Results showed that 55.5% of the study area is highly or moderately suitable for rice cultivation, whereas 34% of that area is unsuitable for rice cultivation due to soil and topographic conditions. Dacryodes edulis, with its potential dietary and medicinal uses, is an aging member of the agroforestry system and is classified among the priority species for domestication in Central Africa. Reliable techniques for producing quality planting materials other than seeds are urgently needed. The aims of this study were to produce D. edulis layers and integrate them into the cropping system. To promote rooting and the survival rates of layers, 4 treatments were used. Compared to the other parts of the human body, the gastrointestinal tract harbors a complex microbiota. The impact of the indigenous microbiota on host physiology is most pronounced in the colon, where the primary bile acids chenodeoxycholic acid and cholic acid are converted, via 7α-dehydroxylation, to the toxic secondary bile acids lithocholic acid and deoxycholic acid, respectively, by Clostridium scindens, an obligate anaerobe. Interestingly, other than bile acid dehydroxylation, little is known about the basic physiology of C. scindens. Media has portrayed chocolate milk negatively and has been removed in some schools due to added sugar. However, it contributes only 3% of total added sugars in children's diets (Murphy, Douglas, Johnson, & Spence, 2008). It also provides all nine of the essential nutrients as white milk, three of which are vital shortfall nutrients (Mahan & Escott-Stump, 2008). This research addressed the issues of caregiver stress and feelings towards caregiving. The following research questions were addressed: 1) Do caregivers who participate in yoga exercises feel less stress about their caregiving duties? 2) Do caregivers who participate in yoga view their caregiving duties as a more positive experience than caregivers who do not participate in yoga? 3) Is the relationship between stress level and positive feelings different between the yoga and non- yoga participants? This article concerns the effect of playing rules on efforts to prevent concussions in youth sports. We can significantly reduce rates of sports-related concussions by periodically reviewing safety-based playing rules in light of medical learning, and then by assuring their enforcement by coaches, officials and parents who remain committed to sportsmanship and mutual respect in local games.
